Objective To explore the effect of Renzhu Jianwei Granules(Granule of Semen Coicis and Rhizoma Atractylodis Macro- cephalae for strengthening the stomach)on the patients with precancerous lesions of gastric cancer(PLGC)and its mechanism. Methods The 58 patients of chronic atrophic gastritis(CAG)complicated with intestinal metaplasia(IM)and/or dysplasia(Dys) were randomized into treatment group and control group with 29 in each.The treatment group was administered Renzhu Jianwei Gran- ules while the control group was given Weifuchun Tablets.The numbers of positive cells of carcinoembryonic antigen(CEA)and cy- clooxygenase 2(COX-2)of gastric mueosa before and after treatment were observed.Results The scores of positive cells of CEA and COX-2 of gastric mucosa of both groups were greatly reduced after treatment(P0.05).Conclusion Renzhu Jianwei Granules can reduce CEA and COX-2 of PLGC patients,possibly being one of the mechanisms in treating PLGC.

Objective: To screen the risk factors for coronary artery diseases (CAD) before undergoing operation for rheumatic heart valve disease(RHVD) in elder patients and to establish a logistic model for predicting CAD, so as to guide the selection of coronary angiography. Methods: A total of 585 elder patients who underwent routine coronary angiography before heart valve operations for RHVD during 1998-2009 were analyzed in the present study. Logistic regression was employed to screen the risk factors for CAD and to establish a bootstrap logistic regression model, with P<0.05 used for identifying the low-risk patients. The receiver operating characteristic (ROC) curves were used to compare the diagnosis efficacies of the established model and other similar models. Results: It was found that the independent predictors of CAD in elder RHVD patients included age, male sex, angina, hypertension, and hypercholesterolemia. A predicting logistic model was successfully established. According to the model, 155 patients were designated as low risk, with 4 (2.6%) complicated with single-vessel disease CAD. The model showed better discrimination efficacy compared with the existing ones, with the area under the ROC curve being 0.793. Conclusion: We have successfully established a predicting model for CAD in elder patients undergoing operation for RHVD, which may help to screen CAD in elder patients before receiving heart valve operations for RHVD.

Objective: To transplant the autologous mesenchymal stem cells (MSCs) carrying human hyperpolarization activated cyclic nucleoside gated channel 2 (hHCN2) gene into the His-bundle in porcine model of complete heart block (CHB), so as to assess the possibility of establishing autologous biological pacing. Methods: We constructed the recombinant adenovirus containing hHCN2 gene to transfect the porcine MSCs. After autotransplantation into the Hie-bundle in CHB model, the genetically-altered MSCs were tested for their ability to provide a stable and catecholamine-responsive heart rhythm. Histological and immunofluorescence analyses were also performed for the myocardium of the injection site. Results: Recombinant adenovirus pAd. hHCN2 was successfully constructed. Porcine MSCs were transfected by the adenovirus. After autotransplantation, transgenic MSCs significantly enhanced the heart rates in porcine CHB model compared with the control group (P < 0.01), and the cardiac rhythms in the transgenic MSC group were catecholamine responsive. Tissues obtained from the transplanted heart sites showed that the MSCs survived in the myocardium and overexpressed hHCN2 protein. Conclusion: Transplantation of autologous genetically-altered MSCs into the His-bundle in porcine CHB model can serve as a short-term catecholamine-responsive biological pacemaker.

AIM: To study the effect of wild-type p53 gene on the differentiation, apoptosis and expression of scavenger receptor CD36 in U937 cells. METHODS: Recombinant adenovirus vector with wild-type p53 gene was constructed and used to transfect U937 cells. With the expression of wild-type p53 gene following adenoviral infection, transfected U937 cells were largely promoted to differentiate into macrophages. RESUITS: Trypanblue-staining test demonstrated that the percentage of positive cells increased from (14.2±5.5)% to (64.6±9.2)% and nitroblue tetrazolium (NBT) reduction test reached similar results (6.3±1.8)% vs (49.7±12.6)%. Furthermore, CD36 mRNA was up-regulated as confirmed by RT-PCR. The increased expression level of CD 36 was also detected by flow cytometry analysis. CONCLUSION: These results suggest that wild-type p53 gene can affect U937 cells differentiation and apoptosis, up-regulate expression of scavenger receptor CD36. It may have a potential significance on atherogenesis.

The way of "extracting-salting-chromatography" was used to purify the phycoerythrin and phycocyanin from Porphyra yezoensis in process scale-up.First,by comprehensive comparison of efficiency,the Sephadex G-25 was selected from four resins (Sephadex G-25、G-100、S-300 and CL-6B) as the best choice used in crude extract desalting of phycobiliprotein.Then the preparation process of phycobiliprotein was scaled-up with raw material(Porphyra yezoensis) increased from 1g to 20g,and finally to 400g.The results indicated that the yields of purified phycoerythrin and phycocyanin (absorption spectra purity above 3.2) increased during according to process scale-up,with 0.323% phycoerythrin and 0.148% phycocyanin obtained from 400g frozen Porphyra yezoensis blades respectively.It is no doubt that the process involved in the experiment is a potential way for large scale preparation of phycobiliproteins of high purity.
