1.The clinical analysis of neonatal testicular torsion

Objective To investigate the clinical characteristics and choices of management of neonatal testicular torsion (NTF).Methods Between January 2013 and December 2015,the clinical data of 7 cases of neonatal testicular torsion who were confirmed by surgery and pathological examination in the department of pediatric surgery of our hospital was retrospectively reviewed.Of the 7 NTTs (4 in right,2 in left and 1 in both sides),the median age and onset time of NNTs was 3 d (1-21 d) and 1 d (1-21 d),respectively.The presentation of NTTs included swelling of scrotum,scrotal mass and discoloration of scrotum.Features of ultrasonography included enlargement of testicular volume,echo heterogenicity or enhancement of testis,and decreased or disappeared testicular blood flow.Results All NTTs were performed by surgery and confirmed by pathological examination.In the operation,5 patients with extravaginal torsion (1 bilateral) and 2 with intravaginal torsion were detected.The degree of twisted testis was from 270° to 720°(average 438.8°) and resection rate for necrosis testis was 87.5% (7/8).The median time of follow-up was 12 months (3-36 months),and none of testis underwent re-torsion and the findings of testicular ultrasonography was not abnormal.Conclusions The presentation of neonatal testicular torsion is non-specific.Once the scrotum shows discoloration,swelling,or enlargement of testis,testicular torsion is suspected.Urgent surgical exploration is recommended in order to make an effort to save the testis.

3.Apoptosis in gastric cancer cells induced by trichosanthin is related with down-expression of bcl-2

Objective To investigate the apoptosis in gastric cancer induced by trichosanthin, and the relationship between this apoptosis and expression of bcl2. Methods In in vitro experiments, morphologic test and TUNEL staining method were used to quantitatively and qualitively detect the apoptosis status of gastric adenocarcinoma cell line SGC7901 before and after the trichosanthin treatment. Immunohistochemical staining method and Northern Blot hybridization were used for detecting expression status of apoptosisrelated genes bcl2, before and after trichosanthin treatment. Results When SGC7901 cells were treated with trichosanthin (0.1 ?g/ml, 36 h), they presented some typical apoptotic morphologic changes observed by fluorescent staining. These morphologic changes include nuclear condensation, nucleosomal fragments forming a lunate body under nuclear membrane, etc. When SGC7901 cells were treated with trichosanthin at the concentration 0.1 ?g/ml for 36 h,42 h and 48 h, respectively, TUNEL staining showed a significant increase of apoptotic index (AI), from 3.78%?1.11%, 3.98%?1.12%,3.85%?1.08%, respectively, to 11.30% ? 2.33%, 10.22% ?2.00%,11.18%?1.85%(P
4.Construction of recombinant plasmid harboring human peptide antibiotic gene hPAB-? and its expression in E. coli

Objective To construct the recombinant plasmid with a human peptide antibiotic hPAB-? gene and to make it expressed in E. coli. Methods To replace the CNBr cleavage site in plasmid pFAST-hPAB-? (CNBr), a pair of primers containing the hydroxylamine cleavage site were designed, and the amplified PCR fragments were cloned into pFAST-HTa plasmid to produce pFAST-hPAB-? (HA), which was then transformed into E. coli DH10B. The constructed plasmid was identified by Ehe Ⅰ/Hind Ⅲ digestion and direct DNA sequencing. An Ehe Ⅰ/Hind Ⅲ digested fragment from pFAST-hPAB-? (HA) was subcloned into pQE32-CP to construct pQE32-CP-hPAB-?, which was transformed into E. coli JM109. The bacteria containing the expression plasmid were induced to express the fusion protein by IPTG. SDS-PAGE was carried out to analyze the molecular weight, expression quantity and expression form of the target fusion protein. After captured by Ni-NTA affinity column, the fusion protein was subjected to hydroxylamine cleavage analysis. Results An expected 230bp fragment was obtained by digesting pFAST-hPAB-? with Ehe Ⅰ/Hind Ⅲ. After this fragment was cloned into pQE32-CP, the recombinant plasmid was confirmed to contain the correct target sequence by DNA sequencing. The recombinant plasmid pQE32-CP-hPAB-? could express a desired protein with a relative molecular weight about 27kD, and its expression level reached 43 percent of the total bacterial proteins. The inclusion bodies were lysed by 8mol/L urea, and the fusion protein could then be captured by Ni-NTA column and cleaved by 2mol/L hydroxylamine at pH9.0. Conclusion The recombinant plasmid pQE32-CP-hPAB-? has been successfully constructed, and it can express the desired hPAB-? fusion protein in E. coli JM109 at high level. These results provide the foundation for future research.

Persister cells are dormant or slowly grown variations in microbial populations .They are highly tolerant to antibiot-ics but the tolerance are not inherited as the genetic resistance , when persisters are inoculated to fresh medium , most bacteria are still susceptive to antibiotic , while only a small fractiont become persisters again .Persisters are believed to be closely related to clinic bio-film forming and the recurrence and recalcitrance of chronic infections .Different persisters have been found in Escherichia coli , Pseud-omonas aeruginosa , Staphyococcus aureus , Mycobacterium tuberculosis , Candida albicans and so on , and a few mechanisms about per-sisters formation have been studied .This article reviews the current progresses of research methods and achievements on persisters from different levels .

8.GC-MS Analysis of Essential Oil from Mosla Chinensis Seeds

OBJECTIVE:To analyze the chemical constituents of the essential oil extracted from Mosla chinensis seeds. METHODS: The essential oil was extracted from Mosla chinensis seeds by steam distillation method and identified by GC-MS, and its relative content was determined using normalization method. RESULTS: A total of 20 chromatographic peaks were detected and 17 chemical compounds were identified, which made up 99.06% of the total essential oil content. The major chemical constituents were carvacrol(57.01%), thymol(30.72%), thymol acetate(2.56%), carvacrol acetate(2.14%) and ?-caryophyllene(1.62%). CONCLUSION: The results in our study provides scientific basis for further exploitation and utilization of Mosla chinensis and serves as reference for establishing quality standard of Mosla chinensis herbal medicine.

10.Study on relationship between changes of DNA content in human spleen nuclei and the postmortem interval by image analysis

Objective To study changes of DNA content in human spleen nuclei and seek an experimental method for estimation of the postmortem interval(PMI)using computerized image-analysis technique(CIAT).Methods Smear sections from spleen sampled were collected in 36 cadavers with known the accurate PMI respectively every hour within the first 36 hours after death,which were then fixed with cold Carony fixation.The smeared sections were stained by Feulgen-van's staining method.3 indices for spleen nucleic DNA including integral optical density(IOD),average optical density(AOD)and average gray(AG)were measured using the CIAT.Results IOD and AOD in the spleen nuclei declined regularly,whereas AG increased with the extension of PMIs in 36 hours.Conclusion There are definite relationships between the PMI and gray parameters(IOD、AOD and AG)representing the DAN content of nucleic DNA in the spleen in 36 hours after death,which may be used for estimation of PMI.
