4. Evaluation of wound healing of skin defect in mice with radiation injury

Objective To establish mouse models of total body irradiation (TBI) with different doses of 60 Co radiation combined with skin defect, so as to investigate the influence of TBI on wound healing and the pathophysiological changes in combined radiation injury and skin defect. Methods A total of 180 female Kunming mice were irradiated with a single dose of 4, 6 or 8 Gy 60 Co ray. Within 30 min after irradiation,a full thickness square wounds (1.5cm×1.5cm) was made on the back of mice to establish animal models of TBI combined with skin defect (n=50), another 30 mice with pure skin defect were used as controls (n=30). Mice in each group were sacrificed on the 3rd, 5th, 7th, 10th and 14th day after injury, each time 6 mice, and the full thickness wounds were harvested. Histological method was used to evaluate the changes of inflammatory cells, fibro blasts and new blood capillaries in the wounds. Image analysis system was used to analyze the areas of the residual wounds. The survival rates and body weight changes of mice within 14 days were analyzed in all groups. Results On the 7th and 14th day after injury, the survival rates of mice in the 6 Gy group were 75% and 55%, respectively. While in the 8 Gy group the survival rate of mice was only 33% on the 7th day,and all the animals died by the 10th day. Within 14 days after injury, the body weight loss of mice demonstrated an increasing tendency with the increase of radiation doses (4,6,and8 Gy groups). TBI delayed wound healing in mice with the increase of irradiation dose.The unhealed areas in the 6 and 8 Gy groups were larger than that in the control group on the 2nd day (P<0.01), and that in 4 Gy group was significantly larger than that in the control group on the 8th day (P<0.05). H-E staining showed that the early inflammatory responses were inhibited, the increase of fibroblasts and new capillaries were greatly delayed, and the granulation tissue formation and reepithelialization were slowed down in the TBI group compared with the simple wound group.Conclusion Animal models of TBI with 6 Gy 60 Co ray combined with skin defect can serve as a platform to study the mechanism of difficult healing and early treatment of radiation injury combined with skin defect.

6.An experimental osteoarthritis induced by implanting composite material into subchondral bone of rabbit knee

[Objective]To investigate the pathogenesis of osteoarthritis by increasing stiffness of subchondral bone to simulate subchondral bone sclerosis to induce the osteoarthritis.[Methods]The proportion between polymethylmethacrylate(PMMA),methylmethacrylate(MMA),hydroxyapatite(HA) and distilled water was adjusted to keep the maximum reaction temperature under 40℃.The composite material PMMA/HA was obtained and compressed on a mechanical material machine to determine its ultimate strength and stiffness,which were compared with those of subchondral bone.The subchondral bone of medial tibial plateau was scraped,where the composite material PMMA /HA was implanted into.Rabbits were sacrificed at 3,6,9 and 12 weeks,respectively.The articular cartilages of the medial tibial plateau were harvested for histological observation.Immunohistochemical analysis was adopted to record the expression and distribution of collagen type Ⅱ and MMP-1 in cartilage at protein level,which were compared with the blank and control groups.Transmission electron microscope was used to examine the ultramicroscopic changes of chondrocytes in blank and 6,12-week-postsurgery groups.[Results]With the time going,the experimental groups gradually presented degeneration,and had increased histological Mankin score.TEM demonstrated degenerative changes of chondrocytes.Collagen type Ⅱ expressed mainly in transition zone and upper deep zone,and MMP-1 expressed predominantly in the superficial and upper intermediate layers of cartilage.Both expressions steadily increased with time passing.[Conclusion]The increased stiffness of subchondral bone induced osteoarthritis in rabbits,which suggests that subcondral bone sclerosis may induce cartilage damage,and that subcondral bone sclerosis is one of the initial pathogenesis of osteoarthritis.
7.Study of Projection Angle for Multiple Pelvic Fractures

Objective To explore the multiple pelvic fractures projection methods and value. Methods 50 cases of pelvic trauma patients were divided into two groups for control photography, and underwent X-ray examination with different projection angles. Results The display effect of the image of the image by 35? projection was better than that of the control group. Conclusion 35? projection results in the images satisfying clinical requirements.

OBJECTIVETo explore the molecular mechanisms of main active ingredients of Siwu decoction analogous formulae for treating primary dysmenorrhea of gynecology blood stasis syndrome by network pharmacology study, and to investigate the correlations between multi-compounds, multi targets and multi pathways.
METHODMajor active compounds from Siwu decoction analogous formulae, including ligustilide, butylidene phthalide, senkyunolide, ferulic acid, gallic acid, peoniflorin, jioglutin A, catalpol, transanethole, zingiberone, commiphoric acid, eugenol, isorhamnetin-3-O-neohesperidoside, wulingzhic acid, alpha-cyperone, cyperene, costunolide, costic acid, tetrahydropalmatine, protopine, amygdalin, 24-methylene cycloartanol, oleic acid, linoleic acid, 3-p-coumaroylquinic acid, hydroxysafflor yellow A, coptisine, berberine, jatrorrhizine, baicalein, baicalin, wogonin were collected to build component-protein networks based on PharmMapper database. The targets information access was used to construct and visualize components-targets-pathways network model using the kyoto encyclopedia of genes and genomes (KEGG) pathway database and Cytoscape software.
RESULT AND CONCLUSIONSerine threonine protein kinases play an important role in the process of cells. They were potential targets in the effect of Siwu decoction analogous formulae. The effect of main active ingredients involved 51 the pathway. Besides the same ones, Shaofu Zhuyu decoction had more effect on lipid metabolism, Xiangfu Siwu decoction on amino acid metabolism pathways, Taohong Siwu decoction on carbohydrate metabolism, while, Qinlian Siwu decoction on ErbB, VEGF signal transduction pathway. Siwu decoction and its derived formulae not only had common targets and pathways, but also had their own emphasis. This reflected the formulae effect mode of multi-ingredients, multi-targets and multi-pathways. It may provide clues to deeper study of molecular mechanism of Siwu decoction analogous formulae action.
