1.Influence of Chinese medicine of Jiangzhi Zhuanggu on bone morphogenetic protein and estrogen receptor expression in bone tissue of hyperlipidemia-induced osteoporosis rats

BACKGROUND: Active bone morphogenetic protein (BMP-2) reduction can cause severe symptoms like osteoporosis. In addition, there is estrogen receptor (ER) in osteoblast, osteoclast and bone marrow stromal cells, indicating a direct effect of estrogen on bone tissues.OBJECTIVE: To investigate the effect of Chinese medicine of Jiangzhi Zhuanggu on BMP-2 and ER expression in rats with prey.hyperlipidemia induced osteoporosis.METHODS: A total of 27 adult SD rats, of either gender, weighing 180-230 g, were randomly divided into three groups. In the normal control group, rats were intragastrically infused with 5 mL/kg normal saline every morning and afternoon. In the model group, the rats were infused with 5 mL/kg high-fat diet in the morning and 5 mL/kg normal saline in the afternoon. In the Chinese medicine group, 5 mL/kg high-fat diet was infused in the morning and 5 mL/kg Chinese medicine of Jiangzhi Zhuanggu water extract in the afternoon. After 8 weeks, expression levels of BMP-2 and ER in bone tissue was detected with immunohistological methods, and ER mRNA level of bone tissue in rats was detected by in situ hybridization.RESULTS AND CONCLUSION: Compared with the model group, the BMP-2 and ER expression in the bone tissue was significantly increased (P < 0.01), and ER mRNA level increased following Chinese medicine treatment. Results show that Chinese medicine of Jiangzhi Zhuanggu could increase BMP-2 and ER expression in the osteoporosis bone tissue, and improve osteoporosis effectively.
2.Effect of the serum containing Jiang Zhi Zhuang Gu solution on BMP-2 expression of rat bone marrow stromal cells

Objective To study osteoporosis associated with hyperlipidemia;To research high cholesterol se-rum and Jiangzhizhuanggu-containing serum intervention rat bone marrow stromal cells BMP-2 expression changes. Methods Adult rat femur by density gradient centrifugation separation of bone marrow stromal cells, cultured cells to the third generation, and was identified by flow cytometry. The third generation cells, were randomly divided into three groups. Normal serum control group: two percent blank senun (volume ratio); High cholesterol serum injury group: fi-nal concentration of 4 mmol/L, high cholesterol serum; medicine + pretreatment serum high cholesterol serum injury, group :2% senun Chinese medicine(volume ratio) after pretreatment 2 hours + final concentration of 4 mmol/L, high cholesterol serum. Total RNA was isolated from cells recovered. BMP-2 mRNA expression was detected. And analyzed statistically. Results (1) Blank serum compared with the control group,high cholesterol serum cultured bone mar-row stromal cells proliferation was inhibited. BMP-2 mRNA expression significantly decreased(P < 0.01) . (2) Jiang-zhizhuanggu solution containing serum protection two hours later to join a high cholesterol serum group, the cell growth conditions improved significantly, BMP-2 mRNA expression significantly increased than pure high cholesterol serum injured group(P <0.01) . Conclusion Solution's Jiangzhizhuanggu containing serum can enhance high cholesterol serum environment bone manow stromal cells BMP-2 mRNA expression, significantly improved bone marrow stromal cell growth state is conducive to the treatment of osteoporosis.
3.Regulation of Insulin Signaling in the Hypothalamus of Spleen Yin Deficiency Diabetes Rats Treated with Zibu Piyin Recipe

This study was aimed to observe changes of key molecular in insulin signaling pathway in the hypothala-mus of rats to explore the mechanism of spleen yin deficiency diabetes-associated cognitive decline (DACD) and Zibu Piyin Recipe (ZBPYR) in order to provide new ideas and new clues for treatment of DACD. Rats were randomly divided into five groups, which were the control (Cont) group, diabetes (DM) group, spleen yin deficiency (pi) group, spleen yin deficiency diabetes (piDM) group and the ZBPYR group. Insulin receptor substrate 1 (IRS-1) serine phos-phorylation levels and protein kinase B (PKB/Akt) serine phosphorylation levels which were involved in the insulin signaling were observed by western blotting in the hypothalamus to determine whether there were insulin signaling obstacles in the hypothalamus of rats. The results showed that the expression of p-IRS-1ser in the DM group, pi group and piDM group was increased compared with the Cont group (P< 0.05); while the p-Akt expression of the DM group and piDM group was decreased (P< 0.05). The expression of p-IRS-1ser in the ZBPYR group decreased compared with the DM group and piDM group (P< 0.05); while the level of p-Akt increased compared with the DM group and piDM group (P < 0.05). It was concluded that insulin signaling was not transduced normally in the hy-pothalamus of the DM group, pi group and piDM group. Insulin resistance may occur in the hypothalamus. And ZBPYR can correct insulin signaling transduction disorder.
4.Effects of Zibu Piyin Recipe on Endoplasmic Reticulum Stress in Cortex of Spleen Yin Deficiency Diabetes-associated Cognitive Decline Rats

Objective To clarify the pathogenesis of spleen yin deficiency diabetes-associated cognitive decline (DACD) and mechanism of Zinu Piyin Recipe (ZBPYR) by observing the expression of phosphorylated RNA-dependent protein kinase-like endoplasmic reticulum kinase (PERK), eukaryotic initiation factor 2α subunit (eIF2α), p-eIF2α, glucose-regulated protein 78 (GRP78) in cerebral cortex of spleen yin deficiency diabetes mellitus rats. Methods The rats were randomly divided into control group, diabetes mellitus group, spleen yin deficiency group, spleen yin deficiency diabetes mellitus group (disease-syndrome group) and spleen yin deficiency diabetes mellitus+ZBPYR group (treatment group). Type 2 diabetes mellitus models were established by high-fat food feeding for 4 weeks and low dose STZ intraperitoneal injection. Then the classical compound method was used to construct spleen yin deficiency rats model by improper diet, over exertion and yin fluids exhaustion. The reatment group was given ZBPYR and other groups were given equal volume of normal saline for 15 days, then cerebral cortex was obtained. The expression of p-PERK, p-eIF2α, eIF2α and GRP78 were observed by Western Blot and RT-PCR. Results The protein expression of p-PERK, p-eIF2α, and mRNA expression of GRP78 of diabetes mellitus group, spleen yin deficiency group and disease-syndrome group was enhanced compared with control group (P<0.05). GRP78 protein expression of diabetes mellitus group, disease-syndrome group was increased compared with control group (P<0.05). The protein expression of p-PERK, p-eIF2α, GRP78 and mRNA expression of GRP78 of treatment group was decreased compared with diabetes mellitus group and disease-syndrome group (P<0.05). Conclusion Endoplasmic reticulum stress is involved in spleen yin deficiency DACD. ZBPYR improves learning and memory ability by reducing endoplasmic reticulum stress.
5.Lnc-BM Promotes Gastric Cancer Progression by Regulating Mitochondrial Respiratory Function Through FASTK/MT-ND6 Axis

Objective To explore the role and molecular mechanism of Lnc-BM in the occurrence and development of gastric cancer (GC). Methods GC tissues and paired adjacent normal tissues of 36 GC patients were collected, and the expression of Lnc-BM was detected by RT-qPCR. Colony formation and CCK-8 assays were used to investigate the proliferation of GC cells. The migration and invasion properties of GC cells were investigated via Transwell assay. RNA pull-down assay was applied to confirm the interaction between FASTK and Lnc-BM. Western blot assay was used to detect FASTK protein level in Lnc-BM overexpressing or knockdown cells. Mitochondrial respiratory capacity and the related proteins expression levels were detected by Seahorse and Western blot assays, respectively. Lnc-BM stably overexpressing GC cells were constructed and then injected subcutaneously into nude mice. The tumor growth was observed. Results Lnc-BM was highly expressed in GC tissues compared with their paired adjacent normal tissues. 6.Application of MALDI-TOF MS for identification of common bacteria in clinical microbiological test

Objective To evaluate the application value of MALDI-TOF MS system in clinical routine separation microorganism by comparing two kinds of microorganism identification system MALDI-TOF MS and Vitek 2 Compact.Methods The bacterial species database of MALDI-TOF MS system and the Vitek 2 Compact system were compared,the new strains from MALDI-TOF MS library were screened out,and the number and frequency of new strains detected from May 2015 when MALDI-TOF MS was put in use to December 2016,and the frequency of detection of common pathogenic bacteria with high confidence were counted.Results There were 205 more new strains in the MALDI-TOF MS library,compared with Vitek 2 Compact.From May 2015 to December 2016,206 times were detected in clinical microbiological examination,and 286 cases if 4 kinds of clinical common pathogenic bacteria were identified by MALDI-TOF MS system with high confidence.Conclusion MALDI-TOF MS strain database is more accurate and large,which can meet the needs of clinical microbiological examination,and is suitable for wider application in clinical microbial identification.
7.Advancement of maggot including living body to treat chronic infected wounds.

Human has used maggot to treat diseases for thousands of years. In recent years, with abuse of antibiotic and the rising incidence of antibiotic resistance, maggot therapy, as a surgical alternative, is mainly applied to treat chronic infected wounds on account of its low cost, efficacy and safety. Its mechanisms are disinfection, bio-debridement and enhancement of tissue regeneration. Maggot therapy which serves as a kind of biological therapy is promising. However, living maggot therapy could result in inevitable complications, so that we should apply traditional Chinese medicine theory to investigate and develop new delivery method of maggot. The review summarizes the past and present of maggot therapy.

Objective To observe the value of augmented reality(AR)navigation system for assisting CT-guided puncture of pulmonary nodules in dog models.Methods Five healthy dogs were selected,and 4 target lung rings were implanted in each dog to build pulmonary nodule models.Deferring to crossover design,CT-guided punctures were performed with or without AR navigation 2 and 4 weeks after successful modeling,respectively,while punctures with AR navigation were regarded as AR group and the others as conventional group,respectively.The time duration of puncturing,the times of CT scanning,of needle adjustment,and the deviation distance between needle pinpoint to the center of pulmonary nodule shown on three-dimensional reconstruction were compared between groups.Results The duration time of puncture in AR group and conventional group was(13.62±5.11)min and(20.16±4.76)min,respectively.In AR group,the times of CT scanning,of needle adjustment,and the deviation distance was 2.40±0.50,2.75±0.44 and(2.94±1.92)mm,respectively,while in conventional group was 3.10±0.64,3.70±0.57 and(4.90±3.38)mm,respectively.The introduction of AR navigation was helpful to shortening the duration of puncture,reducing times of CT scanning and needle adjustment,also decreasing positioning error of needle pinpoint(all P<0.05).In contrast,the variance of puncture sequences and dogs had no obvious effect on the results(both P>0.05).Conclusion AR navigation system could improve accuracy and efficiency in CT-guided puncture of pulmonary nodules in dog models.
10.The accuracy of augmented reality-based percutaneous angle localization system in liver puncture

Objective To evaluate the application value of percutaneous angle positioning system based on augmented reality in improving the accuracy of liver puncture.Methods A canine liver with an embedded marking ring was used as the target for puncture.A skilled physician with over 5 years of experience in liver puncture and a novice physician with limited experience in liver puncture separately performed liver puncture using either the augmented reality-based percutaneous angle localization system(navigation)or CT-guided technique alone(non-navigation).The corresponding puncturing data of non-navigation skilled group(Group A),non-navigation non-skilled group(Group B),navigation skilled group(Group C),and navigation non-skilled group(Group D)were obtained.The differences in the evaluation indicators,including the number of CT scans,number of needle adjustment,time spent for operation,and distance of error,between Group A and Group B,between Group C and Group D,between Group A and Group C,and between Group B and Group D,were analyzed.Results Statistically significant differences in the number of CT scans,number of needle adjustment,time spent for operation,and distance of error existed between Group A and Group B,between Group A and Group C,and between Group B and Group D(all P<0.0 5),while the differences in the number of CT scans,number of needle adjustment,time spent for operation,and distance of error between Group C and Group D were not statistically significant(all P>0.05)Conclusion In performing liver puncture,the use of percutaneous angle localization system can reduce the number of CT scans,number of needle adjustment,time spent for operation and distance of error,and improve the puncture accuracy as well,which provides a basis for the clinical utilization of this system and the employment of this system-guided puncture technology in primary hospitals.(J Intervent Radiol,2024,33:507-511)
