1.Content Determination of Six Types of Heavy Metals in Pian-Zai-Huang Gan-Bao Tablets by ICP-MS

This study was aimed to develop an ICP-MS method to determine contents of six types of heavy metals, which include Cr, Pb, Cd, As, Hg and Cu, in Pian-Zai-Huang Gan-Bao (PZHGB) tablets. The samples were pro-cessed by microwave digestion. And then, they were determined by ICP-MS. The results showed that the working curves of Cr, Cu, As, Cd, Hg and Pb had good linear relationships with the correlation coefficient which was more than 0.9993. The average recovery rate of Cr was 101.2%. The relative standard deviation (RSD) was 7.6%. The av-erage recovery rate of Cu was 96.4%, with the RSD of 3.5%. The average recovery rate of As was 101.3%, with the RSD of 1.8%. The average recovery rate of Cd was 100.2%, with the RSD of 0.3%. The average recovery rate of Hg was 100.5%, with the RSD of 4.2%. The average recovery rate of Pb was 104.2%, with the RSD of 5.6%. It was concluded that the method was stable and reliable, with high sensitivity and repeatability. This method can be used in the content determination of six types of heavy metals, which include Cr, Pb, Cd, As, Hg and Cu in PZHGB tablets.

Objective To investigate the relationship of diet habits and health consciousness with essential hypertension in the rural residents of different economic levels in Hebei Province. 3.Risk Factors in Rural Residents with Essential Hypertension of Different Social Economic Levels

Objective To explore the difference of the risk factors among the rural residents with hypertension with different social economic levels.Methods Risk factors survey were conducted in 469 new cases with hypertension(aged over 35) with different economic-social levels in Jizhou city,Hebei province using self-made questionnaire,2799 people(aged over 35) as the controls.The results were analyzed with Logistic regression analysis.Results OR of the risk factors in higher economic level group were:family history of hypertension(2.863),body mass index(BMI,1.286),preserved foods(1.263),the amount of eggs taken daily(1.200) and the ages of the patients(1.052).The OR of risk factors in lower economic levels' group were:family history of hypertension(3.990),smoking(1.767),the amount of eggs taken daily(1.753),drinking(1.728),education background(1.532),sex(1.448),daily amount of meat intake(1.276),BMI(1.205) and the age of the patients(1.068).The higher annual income of the family and the amount of fresh fruits intake daily were the protective factors of hypertension with the OR were 0.708 and 0.788 respectively.Conclusion The results show that the patients at different economic levels have different risk factors of hypertension.
4.Validity and reliability of the General Alienation Scale in college students

Objective:To test the validity and reliability of the General Alienation Scale (GAS)in college students sample.Methods:Totally 1200 college students were surveyed,using classical test theory (CTT)and i-tem response theory (IRT)to test the psychometric properties of GAS.Under the framework of CTT,item selec-tion was performed with all data.Then,the sample was divided into two groups based on the input order to exam-ine the construct validity.Exploratory factor analysis (EFA)and parallel analysis (PA)were performed with odd group,while confirmatory factor analysis (CFA)was performed with even group.Criterion validity was examined with the short-form of the University of California Los Angeles Loneliness Scale (ULS-8),General Self-Efficacy Scale (GSES),Rosenberg Self-Esteem Scale (RSES).Under the framework of IRT,all data was fitted the rat-ing scale model (RSM),including the principal component analysis of residuals (PCAR),the item fit indexes (Infit and Outfit),measurement reliability (item separation index,item reliability,person separation index and person reliability)and the Wright map.Results:Under the framework of CTT,the GAS was retained 13 items except item 2 and 5.The results of both EFA and PA showed that 13 items of GAS was one-dimensional,and there was a good fit of CFA (χ2 /df=2.689,P <0.001,CFI =0.897,TLI =0.877,RMSEA =0.055,SRMR =0.046).The correlation coefficients were 0.57, -0.60 and -0.40 with the GAS,ULS-8,GSES,RSES
scores respectively (Ps <0.001 ).The Cronbach 's αcoefficient and confidence interval were 0.81 (0.80, 0.83).Under the framework of IRT,the ratio of first-paired eigenvalue was 1.6 in PCAR.The infit and outfit of retaining 13 items in CTT were from 0.6 to 1.4 excluding item 4.The item separation index,item reliability,per-son separation index and person reliability of the 12-item GAS were10.91,0.99,2.14 and 0.82 respectively.The item difficulty can matched with person ability,and they were closed to the normal distribution.Conclusion:It suggests that the General Alienation Scale (GAS)is of good validity and reliability,and it could be used as an ef-fective tool for measuring the alienation of college students.
5.Purification of human nasal epithelial cells by using immunomagnetic beads

Objective To establish a high purity primary culture methods of human nasal epithelial cells (HNEC) in vitro and to provide a successful primary culture model for evaluation experiments of the nasal preparation .Methods Primary culture of human nasal epithelial cells were performed with enzymatic dissociation of isolated tissue and cultured in serum-free medium .HNEC were separated through magnetic field by immunomagnetic beads .We determined the purity of the separated cells by light microscopy and flow cytometry .The morphology of HNEC was observed with a scanning electron microscope .Results Under an inverted phase microscope ,the cells morphology was paving stone shaped .Under the scanning electron microscopy ,abundant microvilli and cilia differentiation were observed .Flow cytometry showed the epithelial cells accounted for 99% .Conclusion The highly purified HNEC can be directly isolated by the magnetic cell sorting system .The cell model can be used for the basic research of nasal cavity preparation .
6.Optimization of extraction technology of Psoralea corylifolia and Salvia miltiorrhiza by orthogonal design with multi-targets.

OBJECTIVETo optimize the extraction technology for Psoralea corylifolia and Salvia miltiorrhiza.
METHODAn orthogonal test was adopted in this study. The contents of Tanshinone IIA, Salvianolic acid B, Psoralen and Isopsoralen were used as indices of appraisal. Data processing was carried out with the multiple guidelines grading method for optimizing the extraction condition.
RESULTThe best extraction technology was to extract for 2 times with 8 fold ethanol solution (70%) and 1.5 h each time.
CONCLUSIONThe optimized extraction technology was rational and the extraction rate of the active principles is high.

7.Study on in vitro mucosal permeation properties and factors affecting mucosal permeability of paeonol.

OBJECTIVETo study the in vitro mucosal permeation properties of paeonol and the factors that affect such permeability.
METHODBullfrog skin was selected as the permeation model, and the modified Franz diffusion cell was adopted to investigate the percutaneous adsorption in vitro. Content of paeonol was quantified by HPLC. Kinds of receiving solution, parts of bullfrog skin and impacts of drug concentration on percutaneous adsorption in vitro were systematically studied.
RESULTPercutaneous permeation of paeonol was to some extent affected by osmotic pressure and pH value of the receiving solution. Abdominal skin, lateral abdominal skin and dorsal skin, with the permeability coefficient form high to low, had great influence on the permeation of paeonol. There existed a good percutaneous permeation of paeonol. And the relationship between the cumulative permeability rate and time was coincided with the first order kenetical process.
CONCLUSIONTake the in vitro abdominal skin of bullfrog as the permeation model, some similarities are revealed between in vitro percutaneous permeation of paeonol and mucosal adsorption.

8.A correlative study of ultrashortwave therapy affecting survival of rat tail replantation

Objective To research the effect mechanism of ultrashortwave in the correlation of ultrashortwave and the tail replantation, provide the experiment basis of clinical practice of prevention and cure for the vascular crisis after micromodule anastomosis. Methods Eighty Sprague-Dawley(SD) rats of clean grade were 3-month-old,female,and were divided into four groups:control group (group 0),model group (group 1 ),contrast group (group 2),ultrashortwave (USW) group (group 3).The preparation of tail replantation model was cut off soft tissue except for caudal veins on both sides of the tail. The coccyx was not broken away from tail.At last,the audal artery under abdominal main centre ditch was anastomosed.In experiment process, the USW group was divided into high dosage group (group 3A) and low dosage group (group 3B). The caudal arterys were ligated and not anastomosed in the group 0. Caudal arterys in other groups were anastomosed.Rats in the group 0 and group 1 received no treatment,normal management after the operation. Rats in the group 2 were given abdominal cavity injection of papaverin liquid immediately,then once a day to 5 days after the tail replantation.Rats in the group 3 were immediately given USW therapy of twenty minutes on the anastomosis section,and then once a day for 5 days after the tail replantation.The USW dosage of group 3A was 3th grade and 50mA. The USW dosage of group 3B was 2th grade and 28mA.The survival rate of the rat tails was observed after the tail replantation for 10 day.Before being grouped,it was measured that the tail skin temperature diference between near and far side of anastomosis section.After the tail relpantation, the temperature diference was inspected daily for 10 postoperative days hence. Before rats were grouped and the eighth hour after the tail replantation, rats were collected blood plasma specimens and measured contents of nitric oxide with destination colorimetric mathods of nitric oxide.Results Carrying out comparison of survival rate of every group,the output weve:between tail cutting off group (group 0,0) and tail replantation group (group 1,2,3,43.94%) to compare P > 0.05; between each group of the tail replantation groups (1,2,3A,3B group) to compare P> 0.05,group 3B < 2 < 1 < 3A; between group 3B and group 1 to compare P > 0.05; between group 2 and group 1 to compare P > 0.05. Each group were compared with the change daily between postoperative and preceding operative the skin temperature diference,single-factor analysis of variance (One-Way ANOVA) analysis:Postoperative 1 day,group 3A < 1,P < 0.05.Postoperative 6 day:3A < 3B < 1 < 2,P > 0.05.Postoperative 7 day:group 3B < 1 < 3A < 2,P < 0.05.Each group were compared with the change of the content of nitric oxide between postoperative 8 hour and preceding operative,with rank-sum test:group 3B > 3A > 2 > 1 > 0,H =33.760,P < 0.05,shows statistically significant.Conclusions USW therapy,especially USW low-dose therapy,can reduce vascular crisis and improve the survival rate of replanted rat tails,after the postoperative 1,6,7 days,reduce skin temperature,improve blood supply,improve nitric oxide at postoperative eighth hour,prevent vascular crisis.Rat tail replantation model in this experiment is feasible.
9.Dissolution of tanshinone IIA from xianling gubao capsule.

OBJECTIVETo establish a method to determine the dissolution of Tanshinone IIA in xianling gubao capsules, and compare the dissolution of Xianling Gubao capsules prepared by different processes.
METHODThe small cup method was used at a rotary speed of 100 r x min(-1), the dissolubility was inspected in the following 6 mediums: (1) water; (2) 0.1 mol x L(-1) hydrochloric acid solution; (3) pH 6.8 phosphate buffer; (4) pH 6.8 phosphate buffer solution + 0.5% cholic acid; (5) pH 6.8 phosphate buffer solution + 1.0% cholic acid; (6) pH 7.6 phosphate buffer solution + 4. 0% cholic acid. A HPLC method was used to determine the dissolution samples of tanshione IIA in different dissolution mediums. The accumulative dissolution percentages were calculated, and the resemblance of release curves were compared by similarity factors (f2).
RESULTThe accumulative dissolution percentage was the highest while pH 7.6 phosphate buffer solution + 4.0% cholic acid was used as medium. The similarity factor values (f2) of tanshinone IIA dissolution curves of different preparation processes of Xianling Gubao capsules were all less than 50.
CONCLUSIONThere is significant difference in tanshinone IIA dissolution between Xianling Gubao capsules of different preparation processes. The accumulative dissolution percentages of the tanshinone IIA in new Xianling Gubao capsules is higher than that in the capsules prepared by other preparation processes.

10.Comparative study on dissolution of Xianlinggubao capsules prepared by different processes.

OBJECTIVETo study the dissolution characteristics of seven active components of Xianlinggubao capsules in vitro, and compare the dissolution of Xianlinggubao capsules prepared by different processes.
METHODThe dissolution of Xianlinggubao capsules was determined by small cup method with the rotating speed of 100 r x min(-1) and phosphate buffer (pH 6.8) as dissolution medium. The different dissolution rates in vitro of seven kinds of active components (icariin, epimedin C, asperosaponin VI, psoralen, isopsoralen, salvianolic acid B and tanshinone IIA) of Xianlinggubao capsules were investigated. The contents of active components were determined by HPLC. The accumulative dissolution percentages were calculated, and the resemblance of release curves were compared by similarity factors (f2).
RESULTThe results of determination showed that the similarity factor values (f2) of the dissolution curves of seven active components after different preparation processes were all less than 50.
CONCLUSIONThere is significant difference in dissolution of active components between Xianlinggubao capsules of different preparation processes. The accumulative dissolution percentages of the active components in new Xianlinggubao capsules are higher than that of Xianlinggubao capsules.
