Objective To establish a rapid, sensitive, and specific quantitative method to detect hepatitis C virus. Methods A primer set targeting HCV 5'UTR was designed. The isothermal amplification was performed by the Bst DNA polymerase and AMV reverse transcriptase, under the temperature of 60℃ for 60 min. The signal was monitored by SYBR Green Ⅰ. Results One hundred and twenty positive serum samples, confirmed by the real-time PCR. All were detected by the isothermal amplification, while 110 healthy subjects' samples were negative by the both methods. The lower detect limit was determined to 10 IU/ml HCV-RNA, by the assay on serial dilutions of the quality control standards obtained from clinical investigation center of MOH. Conclusion A real time reverse loop-mediated isothermal amplification method was developed to detect HCV, with the characteristic of rapidity, high sensitivity and specificity.

Objective NAG6, NAG 7 and BRD7 genes were novel tumor related genes identified recently. This study was designed to examine the expression of these genes in gastric cancer and matched paracancerous tissues and to investigate their roles in the occurrence and development of gastric carcinoma. Methods The expression levels of NAG6, NAG 7 and BRD7 in 34 cases of human gastric cancer and matched paracancerous tissues were analyzed by RT PCR, Northern blot analysis and Dot hybridization. Results The expression of NAG6 was absent in 61.8% gastric cancer tissues(21/34). The expression of NAG6 was significantly down regulated in gastric cancer tissues compared with that in paracancerous tissues ( P 0.05 ). The expression rates of NAG 7 in gastric cancer and paracancerous tissues were 88.2% (30/34) and 82.3% (28/34) respectively. BRD7 was expressed in all gastric cancers and matched paracancerous tissues. The results of RT PCR, Northern blot analysis and Dot hybridization all showed that the expressions of NAG 7 and BRD7 did not display any difference between gastric cancer and matched paracancerous tissues. Meanwhile, Dot hybridization also confirmed that NAG6 was significantly down expressed in gastric cancer. Conclusions NAG6 gene was significantly down regulated in gastric cancer tissues. The down regulation of this gene may play a key role in occurrence and development of gastric cancer, however it may be not associated with lymph node or distance metastasis. The expression levels of NAG 7 and BRD7 were not altered in gastric cancer. It seems that NAG 7 and BRD7 gene may not contribute to the occurrence and development of gastric carcinoma.

Objective A retrospective study of the results of surgical or non-operative treatment in patients with herniation of lumbar disc 10 years or longer before,in order to evaluate the efficacy of different therapeatic strategies for lumbar disc herniation. Method Definite diagnosis of lumbar disc herniation was made in all cases before treatment, and the follow-up period was 10 to 38 years. The patients were divided into three groups according to different treatment methods: non-operative group, simple nucleus resection group, and nucleus resection combined with space fusion group. The long term therapeutic effects and image change of the three groups were observed. The difference of height of intervertebral space at the herniation site was measured before treatment and at the latest follow-up. Adjacent spaces were also observed. The data were analyzed statistically by t -test and ? 2 -test. Results Among 98 cases in non-operative group,85 cases enjoyed almost a normal life and work;vertebral canal decompression was performed for the rest 13 cases because of intermittent claudication. The protruded space was narrowed for 3.17?1.65mm in height in average after non-operative treatment, and there was degeneration of adjacent spaces in 13 cases. Among the 84 cases in simple nucleus resection group, relative good outcome was seen in 71 patients, and the remaining 13 cases had to receive a second operation because of symptom relapse. The protruded space was decreased 5.50?1.51mm in height in average as measured at the latest follow-up comparing with that of before operation. The degeneration of the adjacent space was observed in 15 cases. In 71 patients who received nucleus resection combined with space fusion experienced a good relief of symptoms. The space, where disc herniation had occurred, fused very well and no re-narrowing occurred. Degeneration of the adjacent space was observed in 39 cases, but the patients were symptomless. Conclusions The effect of either non-operative therapy or operation for patients with lumbar disc herniation was good after a long-term follow-up if the indication was chosen properly. We suggest that non-operative therapy should be the first choice for the lumbar disc herniation. The relatively normal nucleus should be preserved as much as possible when nucleus resection was performed. Intervertebral fusion was a relative better treatment comparing with the other two treatments.

Parkinson's disease (PD) is one of the most prevalent neurodegenerative movement disorder in human being, characterized by progressive degeneration and lost of dopamine (DA)-producing neurons in the substantia nigra pars compacta (SNpc),causing low level of DA in the nigrostriatal pathway, eventually leading to motor dysfunctions including tremor, rigidity, bradykinesia, and postural instability. The pathology of PD is complex and the etiology remains poorly understood. In recent years, inflammation has been suggested to be involved in the occurrence and development of PD, by activation of microglia and overexpression of the inflammatory factors. And anti-inflammatory drugs has been developed to delay or prevent the progressive course of PD, becoming a new hotspot in treatment of PD. In this review, the latest progresses on the inflammatory mechanisms and anti-inflammatory treatments of PD was introduced.

Objective To investigate the status of soil fertility of Good Agricultural Practices ( GAP) base for Spatholobus suberectus Dunn. (SSD) in Pingyuan county of Guangdong province, thus to provide reference for GAP research and the subsequent fertilization for SSD. Methods The deep layer and superficial layer of GAP soil were collected for the physiochemical detection and nutrient assay. Compared with the classification standard of the second national general soil investigation, single base soil fertility index was diagnosed and the comprehensive soil fertility was evaluated with modified Nemoro Index. Results The soil pH value and the contents of exchangeable calcium and magnesium were unbalanced, and the contents of macroelements of nitrogen and phosphonium, microelements, and organic matter were low. Therefore, the measures for improving the base soil fertility should be as follows: ( 1) soil amendments of bentonite, gypsum and slaked lime should be used to adjust the soil pH value; ( 2) each plant should be given 10 kg of slaked organic fertilizer as base fertilizer; ( 3) in the process of nurturing, some special micro-fertilizer solution should be used to treat the cut slips, and 5 kg of urea should be used for every 667 meter square of land; ( 4) besides compound fertilizer, every 667 meter square of land should be fertilized with 15 kg of ammonium dihydrogen phosphate for the supplement of nitrogen and phosphorus, and slaked lime and magnesium carbonate should be used for the supplement of soil moderate-quantity elements after transplantation. Conclusion The comprehensive fertility of Pingyuan GAP base for Spatholobus suberectus Dunn. is at low level, and should be improved in combination with GAP requirements.
