3.Effects of high hydrostatic pressure on virulence and antigenicity of Coxsackie virus B_3

Objective To evaluate the influences of high hydrostatic pressure (HHP) on the virulence and antigenicity of Coxsackie virus B3 (CB3 V) , to explore a new physiological method to develop an active vaccine against Coxsackie virus. Methods The phagocytosis of the macrophages in mice to the CB3 V and the TCID50 of the CB3V before and after treated by HHP were examined. Results TCID50 increased from 10-5 to 10-4 when treated by 680 MPa; viruses were inactivated at 700 MPa. Viruses were also inactivated by the pressure less than 700 MPa, but needed prolonged time. 4.Optimizing super pressure extracting technique for ginsenoside from Panax ginseng by orthogonal test

Objective To study the optimum procedure of extracting ginsenoside from Panax ginseng by super pressure extracting technique (SPET). Methods SPET was used to extract ginsenoside from P. ginseng at the normal temperature. The optimum extracting process was selected by the orthogonal test. The contents of ginsenoside in P. ginseng were determined by UV-spectrophotometry. Results The optimum condition of SPET was as follows: when the solid dissolved in 50% ethanol, ratio of raw P. ginseng (g) and solvent (mL) was 1∶75, the pressure was kept at 500 MPa for 2 min; the high yield of ginsenoside was up to 7.76%. Conclusion SPET has a series of advantages, such as higher efficiency, shorter extracting time, and lower exhausting energy, at the same time, the impurity is little, etc.
5.Ultra-high pressure extraction of Astragalus polysaccharides

AIM: To study the optimum technological parameters for extraction of Astragalus polysaccharides by ultra-high pressure extraction. METHODS: Measured concentration of total astragalus polysacharides served as the marker,the design of uniform distribution was applied to evaluating the ultra-high pressure extration efficiency in comparison with reflux and ultrasonic extraction efficiency. RESULTS: The optimal parameters of ultra-high pressure extraction was as follows: 350 MPa pressure,60 mL/g ratio of solvent to solid material,5 h soaking time,2 min extraction time.Under the conditions,the high yield was up to 24.2 %. CONCLUSION: Ultra-high pressure extraction has a series of advantages,such as higher yield,less impurity,easier separation-purification,shorter extraction time and lower energy input.
6.Inhibitory effects of piroxicam on the transplanted sarcoma S180 of mice and its effect on the expression of COX-2,VEGF, FGF-2 and MVD

AIM To investigate the effects of piroxicam on transplanted Sarcoma S180 and the expression of COX 2,VEGF,FGF 2 and microvessel density (MVD) in tumor tissue METHODS Kunming mice were randomizedly divided into control group, FT207 positive group and 5, 2 5, 1 mg?kg -1 piroxicam groups One day after inoculation of 0 2 ml S180 cell suspension, FT207 and piroxicam were given by gastric intubation for 9 days The inhibitory rate on S180 was calculated routinely The expression of COX 2,VEGF,FGF 2 and MVD was detected by immunohistochemistry RESULTS The growth of S180 was significantly inhibited by piroxicam at the doses of 5, 2 5, 1 mg?kg -1 with the inhibitory rate of 31 4%,40 7% and 34 9% respectively The expression of COX 2 in the tumor tissue was also inhibited by piroxicam. Accordingly the expression of VEGF,FGF 2 and MVD was markedly inhibited in dose dependent manner by piroxicam CONCLUSIONS The results suggest that piroxicam has inhibitory effects on S180,and it also decreases the expression of COX 2 in tumor tissue. There is a relation ship between the expression of COX 2 and angiogenesis related factor Antiangiogenesis may be another mechanism for piroxicam to exert its chemopreventive and treatment effects.
7.Epidemiological analysis of imported malaria cases in 20 counties at border region of Yunnan Province from 2012 to 2014

Objective To analyze the epidemiological characteristics of the imported malaria cases in 20 counties at the bor?der region of Yunnan Province from 2012 to 2014,so as to provide the evidence?based proof for adjusting the strategies in the elimination stage. Methods The malaria epidemic data of the 20 border counties in Yunnan Province from 2012 to 2014 were collected and analyzed by using Microsoft Excel 2010. Results From 2012 to 2014,a total of 1 558 malaria cases were report?ed in the 20 border counties in Yunnan Province,among which,1 336 were imported cases,accounting for 85.75%(1 336/1 558),and 222 were indigenous cases,accounting for 14.25%(222/1 558). The number of the imported cases in the above years took up 80.00%(544/680),89.10%(425/477)and 91.52%(367/401)of the total reported cases in the whole year,re?spectively. Among all the 1 336 imported cases,1 045(78.22%)were infected with Plasmodium vivax,284(21.26%)were in?fected with P. falciparum,3 were infected with P. malariae,3 were mixed infection and 1 was an unclassified case;2 patients died. And 95.58%of the cases were mainly infected in Myanmar(1 277 cases). Young and middle?aged adult of 20-40 years who worked overseas were the predominant(802 cases,60.03%)and most of the cases occurred from April to June of the year (679 cases,50.82%). Those cases mainly distributed in Tengchong(459 cases),Ruili(366 cases),Yingjiang(191 cases)and Mangshi(78 cases). Conclusions The epidemic situation of imported malaria is serious in the border region of Yunnan Prov?ince. Therefore,the surveillance system of malaria control needs to be well planned and managed to ensure timely case detection and prompt response at the elimination and post?elimination stage.
8.Application of Traditional Chinese Medicine and Psychological Theory to Scale Development of Social Adaptation Degrees of Special Service Personnel

Objective To develop a scale of social adaptation degrees of special service personnel (referred to as scale) applying Traditional Chinese Medicine and psychological theory and to assess its reliability and validity. Methods By literature review, job analysis, inventory survey, topics interview, this scale was determined in dimensions, items and items pool based on theoretical model. Results After specialist important analysis and combining the different results of statistical analysis, including Cronbach's α coefficient, resolution factor, correlation analysis and confirmatory factor analysis, 6 dimensions, 14 factors and 40 items were finally kept in the scale. The dimensions were psychological characteristics, physical condition, intelligence level, interpersonal relationships, environmental factors and moral character. The factors were self-control, self-esteem character bias, self-confidence, physiological function, learning intelligence, creating intelligence, leadership intelligence, gregariousness, cooperative, confidence feeling, natural environment, social environment and moral character. Conclusion This scale was made on the basis of general international principle, it had good reliability and validity meeting measurement standard of traditional Chinese medicine and psychological theory, and it is an effective and practical evaluation tool of social adaptation degrees of special service personnel.
9.Microembolic signals and outcome in patients with acute ischemic stroke: a prospective case series study

Objective To investigate the correlation of microembolic signals (MES) and outcome in patients with acute ischemic stroke.Methods The patients with acute ischemic stroke were enrolled in the study.The MES of middle cerebral artery was monitored dynamically using transcranial color Doppler ultrasound.The early lesions of ischemic stroke were evaluated by MRI.The National Institutes of Health Stroke Scale (NIHSS) was used to evaluate neurological deficits.The modified Rankin scale was used to evaluate the outcome,and the stroke recurrence was recorded.Results A total of 135 patients with acute ischemic stroke were enrolled,in which,33 were cardiogenic cerebral embolism,49 were large artery atherosclerotic stroke,24 were small arterial occlusive stroke,and 29 were other clear causes or cryptogenic stroke.Multivariate logistic regression analysis showed that coronary heart disease (odds ratio [OR],5.862,95% confidence interval [CI] 2.008-17.114; P =0.000) was the independent risk factor for positive MES within 48 hours after stroke onset,while the history of antithrombotic treatment (OR 0.376,95% CI 0.141-0.998; P =0.045) was its independent protective factor.In addition,coronary heart disease (OR 4.879,95% CI 1.257-18.939; P =0.033),hypertension (OR 4.958,95% CI 1.029-23.882; P =0.030),and diabetes (OR 3.659,95% CI 1.027-13.034; P =0.050) were the independent risk factors for positive MES within 1 week after stroke onset.The NIHSS scores of the patients of the positive MES at baseline and 1 week and the clinical outcome at 3 months had no significant differences with the patients of negative MES,however,stroke recurrence and deaths increased significantly (P =0.019).Conclusions MES within 48 hours of onset was not associated with the outcome in patients with acute ischemic stroke at 3 months,however,the incidence of endpoint events such as recurrence and death was significantly higher in patients of positive MES within 3 months.
10.The role of integrin involved heparin-binding protein secretion in acute lung injury

Objective To study the relationship between heparin-binding protein(HBP) and β2 integrin on the surface of neutrophil in acute lung injury.Methods A total of 30 mice from animal center of Tongji University were divided into control group,CLP group and antibody-treatment group by random number method.Acute lung injury animal model was established by cecal Ligation and puncture in the mice of CLP group and antibody-treatment group.Sham operations were performed on the mice in control group.Mice in antibody-treatment group received anti-CD18 antibody injected via tail vein 30 min before establishing acute lung injury animal model.Twenty-four hours after operation,mice were sacrificed and lung tissue was taken.After HE staining,lung injury were evaluated by Smith score.Lung tissue wet/dry weight ratio,BALF protein level,plasma HBP level and β2 integrin on neutrophil were measured.T test were used to compare the difference among groups.Pearson correlation was used to study the correlation between HBP,integrin and lung injury index.Results Compared with control group,mice in CLP group had higher Smith score(t=10.607,P<0.01),lung wet/dry ratio(t=3.968,P=0.001),BALF protein level(t=4.331,P<0.01) and as well as higher plasma HBP(t=3.515,P=0.002) and β2 integrin(t=4.816,P<0.01) level.After CD18 antibody treatment,anti-treat group mice had lower Smith score (t=2.307,P=0.033),lung wet/dry ratio(t=3.080,P=0.006),BALF protein level(t=2.484,P=0.023) and as well as higher plasma HBP level(t=2.218,P=0.046) than mice in CLP group.Pearson correlation analysis showed HBP had obvious correlation with wet/dry ratio(r=0.527,P=0.017),BALF protein(r=0.508,P=0.022) and as well as β2 integrin(r=0.674,P=0.001).Conclusions Both HBP and β2 integrin involved in the lung injury pathogenesis.HBP level is interrelated with the severity of lung injury.The β2 integrin is associated with the release of HBP by neutrophil.Inhabiting the function of integrin could decrease HBP level and alleviate the severity of lung injury.
