1,3-propanediol (1,3-PD) is an important material for chemical industry, therefore, there is much interest in the production of 1,3-PD. The gene dhaT encoding 1, 3-propanediol dehydrogenase ( PDOR) of Citrobacter freundii was amplified by PCR. Sequence analysis of the similarity at the nucleotide and amino acid level between the gene encoding C. freundii PDOR and that of C. freundii ( U09771 ) were 78% and 90% , respectively. The recombinant plasmid pSE-dhaT was constructed by inserting dhaT gene into expression vector pSE380 and then transformed E. coli JM109. The recombinant strain was induced by IPTG to express dhaT. Further more the recombinant enzyme was purifed from recombinant E. coli by Ni-nitrilotriacetate affinity chromatography followed by Sephacral S-300 gel filtration. A single obvious protein about 42kDa could be obtained by the sodium dodecyl sulfate-polyacrylamide gel electrophoresis of recombinant enzyme. The purified enzyme was used to determined enzyme property on the substrate of propionaldehyde and 1, 3-PD. The optimal temperature and optimal pH of the purified enzyme were 37℃, 8.0 for reduction and 25℃, 10. 5 for oxidation, respectively; and the kinetic property of PDOR about Km and V max were 10. 05mmol/L, 37. 27?mol/min/mg for propionaldehyde and 1. 28mmol/L, 25. 55?mol/min/mg for 1,3-PD, respectively; The deduced dhaT gene product (388 amino acids) showed a specific reduction activity of 49. 50U/mg and oxidation activity of 79. 92U/ mg. There also have a putative iron-binding motif ( G-XX-H-X-X-A-H-X-X-G-X-X-X-X-X-P-H-G) as a fingerprint pattern in the recombinant enzyme, the motif is fully conserved among these 1, 3-propanediol dehydrogenase. It is beneficial to the researches of high producing 1, 3-propanediol by gene engineering strain.
4.Cheiro-oral syndrome: A reappraisal of the etiology and outcome

Objective: This is a review of our cases and published literature on cheiro-oral syndrome (COS), to
better understand its localization, etiology and outcome. Methods: In addition to our database, we
reviewed the medical database (including PUBMED, BIOSIS, EMBASE, and SCOPUS) and other
sources, searched by the keyword of “cheiro-oral”. The defi nition of COS was a subjective or an
objective sensory disturbance confi ned to the perioral area and the fi nger(s)/hand without a detectable
abnormality in mental, motor or cerebellar function. Only cases of COS where the clinicoanatomic
correlation could be identifi ed by neuroimaging study, autopsy or stereotatic surgery was included.
Results: There were a total of 174 patients; 85 patients from our database, 76 patients from medical
database, and 13 patients from other sources. They were 111 men and 63 women. Their age ranged
from 12 to 85 years; average being 58.2 years. Stroke is the leading etiology and constituted 74% of
the patients. The most common location of lesion was thalamus, followed by pons and cortex. Classical
unilateral COS was seen in 81% of patients, atypical COS in 19%. Whereas the lesions were from
cortex to cervical spinal cord in unilateral COS, atypical COS was associated with lesions in pons or
medulla oblongata. An early deterioration was seen in 16.5% of patients, especially in large cortical
infarction and subdural hemorrhage. Structural lesions were found in 85% of patients.
Conclusion: Classical unilateral COS do not have a high localizing value, the atypical COS is associated
with lesion in pons or medulla.
5.Features of intestinal flora in children with food protein-induced proctocolitis based on high-throughput sequencing.

OBJECTIVES:
To study the features of intestinal flora in children with food protein-induced proctocolitis (FPIP) by high-throughput sequencing.
METHODS:
A total of 31 children, aged <6 months, who experienced FPIP after exclusive breastfeeding and attended the outpatient service of the Third Affiliated Hospital of Zunyi Medical University from October 2018 to February 2021 were enrolled as the FPIP group. Thirty-one healthy infants were enrolled as the control group. Fecal samples were collected to extract DNA for PCR amplification. High-throughput sequencing was used to perform a bioinformatics analysis of 16S rDNA V3-V4 fragments in fecal samples.
RESULTS:
The diversity analysis of intestinal flora showed that compared with the control group, the FPIP group had a lower Shannon index for diversity (P>0.05) and a significantly higher Chao index for abundance (P<0.01). At the phylum level, the intestinal flora in both groups were composed of Firmicutes, Actinobacteria, Proteobacteria, and Bacteroidetes. Compared with the control group, the FPIP group had a significant reduction in the composition ratio of Actinobacteria (P<0.001) and a significant increase in the composition ratio of Proteobacteria (P<0.05). At the genus level, the intestinal flora in the FPIP group were mainly composed of Escherichia, Clostridium, Enterococcus, Klebsiella, and Bifidobacterium, and the intestinal flora in the control group were mainly composed of Bifidobacterium and Streptococcus. Compared with the control group, the FPIP group had a significant reduction in the composition ratio of Bifidobacterium and Ruminococcus (P<0.05) and significant increases in the composition ratios of Clostridium and Shigella (P<0.05).
CONCLUSIONS
Compared with the control group, the FPIP group has a reduction in the diversity of intestinal flora and an increase in their abundance, and there are certain differences in several bacterial genera. These results suggest that changes in the composition of intestinal flora at genus level may play an important role in the development and progression of FPIP.

7.Metabolomics Study of Exhaled Breath Condensate in Childhood Asthma by Gas Chromatography-Mass Spectrometry

Exhaled breath condensate (EBC) was analyzed by gas chromatography-mass spectrometry (GC-MS / MS) in childhood asthma and healthy control, aiming to find the potential markers of EBC in children with asthma, and provide a scientific reference for its pathogenesis and early screening. EBC samples were collected from 21 asthmatic children (age (8. 2 ±1. 6) years) and 17 healthy children ( age (8. 1 ±1. 3) years). GC-MS / MS was used to obtain the full scan data of chemical components. Cluster analysis was performed on the two groups of metabolites by principal component analysis (PCA), and potential biomarkers were found using Metaboanalyst 3. 0 attributable metabolic pathways. The results showed that the EBC metabolic maps of asthmatic group and normal group were very different, and eight endogenous potential biomarkers were identified, suggesting that starch and sucrose metabolism, lysine degradation, aminoglycan nucleoside metabolism, phenylalanine metabolism may play important roles in the development of asthma in children.
8.Phylogenetic analysis and PCR-restriction fragment length polymophism identification of Salmonella based on grOEL gene sequence.

OBJECTIVETo investigate the value of grOEL gene sequence in phylogenetic analysis and typing of Salmonella.
METHODSThe grOEL gene was amplified by PCR, sequenced and analyzed using Bioedit and DNAstar software. The Salmonella strains were identified using PCR-restriction fragment length polymophism (PCR-RFLP).
RESULTSThe conservative and variable regions of grOEL gene of Salmonella serogroup were separately distributed and most of the small mutant regions distributed intermittently among the conservative regions. The phylogenetic tree of Salmonella based on the nucleotides differed from that generated based on the amino acid sequence. O8, O9 and O10 had the closest consanguinity, and 5 patterns were identified by PCR-RFLP.
CONCLUSIONThe grOEL gene can be used as a genetic marker for phylogenetic analysis of Salmonella and also as a target sequence for Salmonella typing identification.

9.Cardiac electromechanical mapping in analyzing the mechanism of left ventricular remodeling immediately after percutaneous transluminal septal ablation in patients with hypertrophic obstructive cardiomyopathy.

BACKGROUNDEffect of percutaneous transluminal septal ablation (PTSA) with ethanol injection on electromechanical remodeling of left ventricule still remains unknown. This study was conducted to assess the potential significance of cardiac electromechanical mapping (CEMM) in analyzing the left ventricular remodeling before and immediately after percutaneous transseptal myocardial ablation (PTSMA) in patients with hypertrophic obstructive cardiomyopathy (HOCM).
METHODSEight patients with drug-refractory HOCM and 6 patients with hypertrophic cardiopathy (HM) without increased left ventricular outtract gradien (LVOTG) were enrolled into the present study. CEMM was undergone in patients with HOCM before and immediately after PTSA procedure, and in patients with HM.
RESULTSPTSA was successful in all patients with HOCM, LVOTG significantly decreased from (62.87 +/- 21.16) mmHg to (12.73 +/- 3.05) mmHg immediately after ablation procedure. Value of UVP in septal-base segment in HM group was higher than that in HOCM group [(22.79 +/- 2.34) mV vs (18.54 +/- 1.76) mV]. In patients with HOCM, lateral-middle and -base segments had lowest value of UVP [(15.93 +/- 1.11) mV and (15.83 +/- 1.07) mV] before PTSA. Value of UVP at posterior-middle segment decreased from (23.58 +/- 2.21) mV pre-PTSA to (18.89 +/- 1.91) mV post-procedure, PTSA led to significant increase of UVP at lateral-middle segment. Septal region in patients with HM and septal-middle, septal-base, posterior-base segments in HOCM had lower value of local linear shortening (LLS) among all patients in both HOCM and HM groups. PTSA resulted in significant reduction of LLS in anterior region and at septal-apex segment. Anterior-base and septal-middle segments in patients with HM had lowest value of local active time (LAT), and significantly differentiated from that in patients with HOCM [(-8.57 +/- 0.68) ms vs (-18.61 +/- 1.02) ms, (-6.75 +/- 0.37)ms vs (-21.90 +/- 0.96) ms, respectively]. LAT at septal-middle and -base segments in patients with HOCM was decreased significantly [(-21.90 +/- 0.96) ms vs (-13.80 +/- 1.04) ms, P < 0.002; and (-15.20 +/- 1.06) ms vs (-6.33 +/- 0.52) ms, respectively] immediately after PTSA.
CONCLUSIONSPosterior-lateral and anterior region probably played important roles in electromechanical remodeling. Significant electromechanical remodeling disassociation (uncoupling) was detected in most left ventricular regions, which would be important in differentiating of HOCM from HM, and in predicting the prognosis in patients with HOCM after PTSA procedure.

10.Application of Excel programs of Fisher exact probability test for medical data.

OBJECTIVETo accomplish the computation of Fisher exact probability test for fourfold table data in Excel.
METHODThe computing program of exact probability method for medical data in fourfold table design was edited by employing the IF statement and the relevant functions, such as SUM, FACT, DSUM, etc in Excel. The computational results are compared and evaluated according to the case studies.
RESULTSThe output of Fisher Exact Probability was generated and presented correctly following the input of four numerical values into the computation program in the setting of Excel. The parametric outcomes are in agreement with those produced by SAS and SPSS, in the combination tables containing the P value, two-tailed cumulative P value, left-tailed P-value, right-tailed P-value, Chi2 values and P values both for direct Chi-squared test and corrected Chi-squared test.
CONCLUSIONSDirect Chi-squared test, corrected Chi-squared test combined with Fisher Exact Probability test for fourfold table data can be conveniently, rapidly, and accurately accomplished in Excel.
