RT-PCR amplification of ginseng ?-amyrin synthase gene was successfully performed based on the total RNAs extracted from ginseng hairy roots induced by Agrobacterium rhizogenes A4 using a modified guanidine isothiocyanate-method. Sequence analysis of this gene revealed that its sequence was consistent with the sequence of a previously reported ginseng ?-amyrin synthase gene (GenBank No. AB009030). This gene was inserted into pMD-119T simple vector and transformed into E.coli DH5?. Furthermore antisense plant expression vector of this gene was constructed using the pBI121 vector, laying foundation for studies on antisense regulation of ginseng ?-amyrin synthase gene.

Genetic engineering is a powerful tool in Panax ginseng breeding.Genetic transformation and plant regeneration are the premise and foundation involved in genetic engineering of Panax ginseng.Ginseng can be regenerated through organogenesis or somatic embryogenesis and indirect somatic embryogenesis is mainly used for its regeneration.Summurized the factors influencing plant regeneration such as different explants,different carbohydrates,somatic embryo optimization and hormone-free approach.Ginseng transformation has been achieved by Agrobacterium tumefaciens and Agrobacterium rhizogenes and transgenic ginseng with good characters was obtained by introducing genes associated with biosynthesis of ginsenosides or herbicide gene.Hairy root culture system can supply large scale of ginsenosides,thus effect of rolC genes on ginseng hairy root induction,regeneration and bioreactor culture of hairy root were discussed.Additionally,problems that are present in genetic engineering of Panax ginseng were also discussed in this review.
5.Effect of continuous femoral nerve block in analgesia and the early rehabilitation after total knee replacement.

OBJECTIVETo evaluate the benefit and safety of continuous femoral nerve block in patients undergoing total knee arthroplasty.
METHODSFrom December 2008 to August 2009, 80 patients with total knee arthroplasty were randomly divided into two groups, 40 patients in each group. In group A the pain-control was dominanted by continuous femoral nerve block, there were 5 males adn 35 females with an average age of (65.0 +/- 4.2) years old; In group B the pump of vein odynolysis was used, there were 5 males and 35 females with an average age of (64.7 +/- 8.5) years old. The treatment of relieve pain continued for 3 days. The pain of visual analog scale (VAS), sleep condition, early-stage rehabilitation and adverse effects were recorded.
RESULTSThe VAS scores of group A was lower than that of group B at 2, 6, 24, 36, 48, 56, 72 h after operation (P < 0.05 or P < 0.01); The sleep condition and early-stage rehabilitation of group A were better than that of group B. The complications of group B occurrenced more than group A; The analgetica of group B were used more than that of group A.
CONCLUSIONThe continuous femoral nerve block is an effective pain relieve method and is benefical to rehabilitation from total knee arthroplasty early.

6.Influence of polarized bias and porous silicon morphology on the electrical behavior of Au-porous silicon contacts.

This paper reports the surface morphology and I-V curves of porous silicon (PS) samples and related devices. The observed fabrics on the PS surface were found to affect the electrical property of PS devices. When the devices were operated under different external bias (10 V or 3 V) for 10 min, their observed obvious differences in electrical properties may be due to the different control mechanisms in the Al/PS interface and PS matrix morphology.

7.The clinical evaluation of patients with pituitary adenomas undergone transsphenoidal microsurgery.

OBJECTIVETo evaluate the overall effect of transsphenoidal microsurgery for pituitary adenomas in recent 5 years and to discuss the surgical technique, application of new technology and postoperative follow-up results.
METHODSThe clinical presentation, image characteristics, endocrinal findings, pathological types, tumor removal percentage, postoperative complication and follow-up of 1 462 patients with pituitary adenomas who underwent the transsphenoidal microsurgery from 1997 to 2002 were analysed retrospectively.
RESULTSTotal rate of tumor removal for the patients achieved 97.0% in the patients with Hardy I adenomas, 95.2% with Hardy II, 90.5% with Hardy III, and 47.4% with Hardy IV respectively. A significant postoperative improvement both in clinical symptoms and endocrinal parameters was achieved. The tumor recurrence rate was 0.3%.
CONCLUSIONSWith the improvement of microsurgical technique and application of novel technology, the indication of transsphenoidal microsurgery for pituitary adenomas was increasingly extended. Endoscope and(/or) neuronavigation-assisted microsurgery via transsphenoidal approach should be of the first choice for the treatment of pituitary adenomas. The routine postoperative radiotherapy is not required for patients with total tumor removal.

8.Amplitude Changes of Low Frequency Fluctuation in Brain Spontaneous Nervous Activities Induced by Needling at Hand Taiyin Lung Channel.

OBJECTIVETo observe amplitude changes of low frequency fluctuation in brain spontaneous nervous activities induced by needling at Hand Taiyin Lung Channel, and to preliminarily explore the possible brain function network of Hand Taiyin Lung Channel.
METHODSBy using functional magnetic resonance imaging (fMRI), 16 healthy volunteers underwent resting-state scanning (R1) and scanning with retained acupuncture at Hand Taiyin Lung Channel (acupuncture, AP). Data of fMRI collected were statistically calculated using amplitude of low frequency fluctuations (ALFF).
RESULTSUnder R1 significantly enhanced ALFF occurred in right precuneus, left inferior parietal lobule, bilateral superior temporal gyrus, bilateral middle frontal gyrus, left superior frontal gyrus, left inferior frontal gyrus, left medial frontal gyrus. Under AP significantly enhanced ALFF occurred in right precuneus, bilateral superior frontal gyrus, cerebellum, bilateral middle frontal gyrus, right medial frontal gyrus, and so on. Compared with R1, needing at Hand Taiyin Lung Channel could significantly enhance ALFF in right gyrus subcallosum and right inferior frontal gyrus. Significant decreased ALFF appeared in right postcentral gyrus, left precuneus, left superior temporal gyrus, left middle temporal gyrus, and so on.
CONCLUSIONNeeding at Hand Taiyin Lung Channel could significantly change fixed activities of cerebral cortex, especially in right subcallosal gyrus, right inferior frontal gyrus, and so on.

9.Effect of topographical factors on podophyllotoxin content in Sinopodophyllum hexandrum and study on ecological suitability.

In order to find the optimal topographical factor for regionslization, the content of cimetidine in 116 Sinopodophyllum hexandrum sample collected from Sichuan, Qinghai, Gansu, Tibet, Yunnan and Shaanxi provinces, was determined. Using mathematical statistics and geographical spatial analysis of GIS analysis, the relationship between content of podophyllotoxin and influencing factors including altitude gradient and gradient position was analyzed. It is found that the optimal altitude was 2 800 m to 3 600 m, the aspect of slope north or northeast and northwest and the slope 12 degrees to 65 degrees with a high suitability degree. Considering the artificial planting, the suitable planting area for S. hexandrum is comfirmed. The topographical factor is important for S. hexandrum regionalization, but has hardly effect on podophyllotoxin content. The results of the study provide an important scientific basis for S. hexandrum production development. But there are many factors which affect suitability index and podophyllotoxin content of S. hexandrum, it is necessary to consider other factors like climate and soil while exploitation and protection of S. hexandrum.

10.Dihydromyricetin inhibits PC12 cell injury induced by 1-methyl-4-phenylpyridinium by up-regulating autophagy activity

OBJECTIVE To observe the effect of dihydromyricetin (DMY) on cell injury induced by 1-methyl-4-phenylpyridinium (MPP+) in PC12 cells and explore the possible mechanism. METHODS After being pretreated with DMY 5, 10, 20, 40 and 80 μmol · L-1 for 0.5 h, PC12 cells were incubated with DMEM culture medium including autophagy inhibitor 3-methyladenine (3-MA) 10 mmol · L-1 and MPP+500μmol·L-1 for 48 h. MTT Assay was used to test the viability of PC12 cells. The level of lactate dehy-dregenase (LDH) was measured colorimetrically. The ultrastructure of PC12 cells was observed under transmission electron microscope. Expressions of autophagy related protein, Beclin-1, microtubule-associated protein light chain 3 (LC3) and p62 were measured by Western blotting. RESULTS Compared with cell control group, the cell survival rate was significantly decreased, the level of LDH was signifi-cantly increased, the number of autophagosomes was obviously decreased, the percentage of autophagic vacuoles in the total cytoplasm area was significantly decreased (P<0.05), expression of Beclin-1 and LC3-Ⅱ and the ratio of LC3-Ⅱ to LC3-Ⅰ were significantly down-regulated (P<0.05) and the expres-sion of p62 was significantly up-regulated in MPP+group (P<0.05). Compared with MPP+group, the cell survival rate was significantly increased, while the level of LDH was significantly decreased in MPP++DMY 10-80 μmol · L-1 group (P<0.05). Compared with MPP+ group, the number of autophagosomes obviously increased, the percentage of autophagic vacuoles in the total cytoplasm area was significantly increased, the expression of Beclin-1 and LC3-Ⅱ and the ratio of LC3-Ⅱ to LC3-Ⅰwere significantly up-regulated and the expression of p62 was significantly down-regulated in MPP++DMY 20 μmol · L-1 group (P<0.05). Compared with MPP++DMY 20μmol · L-1 group, the cell survival rate was significantly decreased, but the level of LDH was significantly increased in MPP ++DMY+3-MA group (P<0.05). CONCLUSION DMY may inhibit the cell injury induced by MPP+in PC12 cells, and the mechanism may be related to the up-regulation of autophagy activity induced by DMY.
