8. Breeding of Dendrobium officinale var. 'Xianhu 2' and its characteristics
Chinese Pharmaceutical Journal 2013;48(19):1677-1680
OBJECTIVE: To select and breed elite variety of Dendrobium officinale, thus to conserve the wild germplasms and promote the healthy developing of D. officinale industry. METHODS: System breeding method was employed for the breeding of new variety developed from wild germplasm oi Dendrobium officinale. Yield of fresh stems and leaves, quality characters including dried stem rate and raw polysaccharide content etc. were tested. ISSR molecular marker technique was used to distinguish genetic difference between new variety and other D. officinale materials. RESULTS: New elite variety 'Xianhu 2' was bred successfully with high polysaccharide content, fine adaptability, and high yield. 'Xianhu 2' had significantly higher yields of fresh stems-leaves and stems than D. officinale var. 'Yunnan Ruanjiao' and D. officinale var. 'Xianhu 1' and significantly higher dried stem ratio and raw polysaccharide content than 'Yunnan Ruanjiao' at 12, 24, and 36 m, respectively. ISSR analysis indicated that there were distinct differences in DNA level between 'Xianhu 2' and other D. officinale varieties and lines. D. officinale var. 'Xianhu 2' got its certificate for commerce releasing by Zhejiang Province in December 2011. CONCLUSION: 'Xianhu 2' is a new elite variety with good synthetic agronomic traits. Its breeding is beneficial for the cultivation of D. officinale.

Objective To isolate and identify the pathogenic bacteria from peripheral blood of a patient with septicemia of unknown etiology and to analyze their drug resistance genes. Methods Two peripheral blood samples were collected from the patient after having fever.Several assays including culturing bacteria on blood agar plates by using streaking technique,Gram-staining of bacterial colonies and microscopic observation,VITEK 2-compact automatic bacterium detection and analysis system as well as a sequencing analysis of the 16s rRNA gene were performed to identify the bacterial pathogens in blood samples.Microdilution test was performed to detect the drug susceptibilities of isolated bacteria to antibiotics.Confirmatory tests were performed to detect the production of β-lactamase and extended spectrum β-lactamase by the isolated strains and the phenotypes of AmpC enzyme and carbapenemase.PCR was used to identify the β-lactamase-encoding genes in the isolated strains by using the primers of 19 common β-lactamase-,AmpC enzyme-and carbapenemase-encoding genes in Enterobacteriaceae strains and the primers of 21 annotated gene sequences encoding the β-lactamase of a Ralstonia mannitolilytica strain. The PCR products were sequenced and analyzed after T-A cloning. Results Ralstonia mannitolilytica strains were isolated from the two peripheral blood samples. The isolated strains were sensitive to ceftriaxone, cefepime, ciprofloxacin, ofloxacin, tigecycline and compound sulfamethoxazole (SMZ-TMP),but resistant to the other 11 tested antibiotics. Results of PCR amplification by using the primers of common genes encoding β-lactamase of Enterobacteriaceae strains were all negative.Fragments of genes encoding the β-lactamase of the isolated Ralstonia mannitolilytica strain were successfully amplified, which were TK49_09850,TK49_12955,TK49_14470,TK49_14495and TK49_18990.The sequences of the amplified gene fragments were not similar to those of the common β-lactamase-encoding genes in Enterobacteriaceae strains. Conclusion The patient was infected with Ralstonia mannitolilytica. The isolated Ralstonia mannitolilytica strain showed a high-level drug resistance with a noticeable diversity against different β-lactam antibiotics. The genes encoding β-lactamase of the isolated Ralstonia mannitolilytica strain were completely different to those of Enterobacteriaceae strains.

Objective To compare the effect of botulinum toxin A(BTX-A)applied according to experi- ence with its effect when the application is guided by electrical stimulation on spasticity in the ankle plantarflexors of children with cerebral palsy(CP).Methods Forty-five children with CP were randomly assigned into 2 groups to receive injections of BTX-A guided by electrical stimulation,or injections of BTX-A guided by experience. All chil- dren received a local injection in the ankle plantar flexors.Physiotherapy and ankle-foot orthoses were applied by a physical therapist 3 days after the BTX-A injections. After the first 10 days, the therapy was administered by the patient's family.Clinical assessments included the patient's passive range of movement(PROM),scoring on the Ash- worth scale(MAS),the composite spasticity scale(CSS),and the D and E dimensions of the gross motor function measure(GMFM),and walking velocity(WV).Assessments were performed before treatment and at 3 days,2 weeks,1,2,and 3 months following the injection with BTX-A.Results All children showed significant decrease in spasticity(PROM,MAS and CSS)after 3 days. The improvement was maintained at 3 months. When compared with the results before the injection, the improvements in standing and walking(GMFM)and in walking velocity were statistically significant after 2 weeks of treatment for both groups, and were maintained at 3 months. The differences in PROM and CSS scores at 3 days,2 weeks,1,2,and 3 months following the injection were statistically significant between the 2 groups.Significant differences were also found between the 2 groups in MAS scores at 3 days,2 and 3 months after treatment, and in GMFM and WV at 2 and 3 months after treatment. Conclusions A BTX-A injec- tion, whether guided by electrical stimulation or experience,in combination with physiotherapy, can reduce spasticity in the ankle plantarflexors of ambulant children with CP and improve their functional performance.BTX-A injection guided by electrical stimulation was more effective than an injection guided by experience.
