BACKGROUND:Chemokines can promote (MSCs) the secretion of vasoactive factors from mesenchymal stem cel s (MSCs) through paracrine mechanism, which have important role in accelerating angiogenesis. OBJECTIVE:Under the high glucose environment, to the effect of the supernatant of MSCs stimulated by chemokine (C-X-C motif) ligand 8 (CXCL-8) on human umbilical vein endothelial cel s (HUVECs), and to analyze the mechanism of Sonic Hedgehog signaling pathway in the stimulation of CXCL-8 on MSCs. METHODS:Under the high glucose environment, the MSCs supplemented with 100μg/L CXCL-8 were set as CXCL-8 group;the MSCs that were preprocessed with 5μmol/L octyl maleimide for 45 minutes and then stimulated with 100μg/L CXCL-8 were as Shh inhibitor group;the MSCs that were routinely cultured in a high-glucose medium were as control group. The cel supernatant of each group was extracted as conditioned medium (CM) to culture HUVECs, respectively, and these cel s were referred to as CXCL-8 CM group, Shh inhibitor CM group, and control CM group, respectively. Cel counting kit-8, cel scratch and Transwel chamber tests were used to observe the effect of each CM on HUVEC proliferation, apoptosis and chemotaxis. By establishment of a diabetic skin ulcer model in C57BL/6J mice, the CM of each group was used to treat the mouse model to confirm the effects of CXCL-8 stimulated MSCs CM on HUVEC homing and ulcer healing. RESULTS AND CONCLUSION:(1) The experimental results in vitro:compared with the control CM group, CXCL-8 CM group significantly promoted the proliferation of HUVECs, and decreased the apoptosis of HUVECs, the closure rate and migration rate of HUVECs were significantly increased (P<0.01 or P<0.01), and the levels of vascular endothelial growth factor and epidermal growth factor were significantly increased (P<0.01 or P<0.01). Compared with CXCL-8 CM group, however, the above results in the Shh inhibitor CM group showed reverse changes (P<0.01). (2) The experimental results in vivo:compared with the MSCs CM group and Shh inhibitor CM group, the healing effect of diabetic skin ulcer and the number of HUVECs labeled by green fluorescent protein in the CXCL-8 CM group were significantly increased (P<0.01). To conclude, these findings indicate that CXCL-8 stimulated MSCs secrete paracrine factors, vascular endothelial growth factor and epidermal growth factor, through the Sonic Hedgehog signaling pathway under the high glucose environment, which enhance the homing ability of HUVECs.

[Abstract ] Objective Liver cirrhosis modeling with carbon tetrachloride (CCL4) may be influenced by many factors, such as drug concentration and dosing methods. In this article, we explored the influences of different concentrations and different dosing methods and time of CCL4 on the induction of liver cirrhosis in rats. Methods We constructed rat models of liver cirrhosis with different con-centrations of CCL4(30%and 50%), using different dosing methods (subcutaneous injection, intraperitoneal injection, and intragastric administration) , and for different lengths of dosing time (8 wk, 10 wk, and 12 wk) .We collected blood and liver tissues from the rats at different time points for HE and MTC staining, biochemical and histomorphological scores based on the Scoring Model for Liver Cirrho-sis Disease (SLCD, expressed by R) and the Laennec Fibrosis Scoring System (LFSS, expressed by L), and analysis of the results by 3 ×2 ×3 factorial experiment design. Results The R value was lower in the intraperitoneal injection than in the subcutaneous injection and intragastric administration groups, and so was it in the 50% than in the 30%CCL4 group, decreasing with the extending of dosing time, with statistically significant differences in the main effects ( P<0.05) as well as a remarkable correlation among drug concentrations, dosing methods, and dosing time (P<0.05).The L value was higher in the intraperitoneal injection than in the subcutaneous injection and intra-gastric administration groups, and so was it in the 50% than in the 30% CCL4 group and in the 12 wk than in the 10 wk and 8 wk groups, with statistically significant differences in the main effects ( P<0.05) but no remarkable correlation among drug concentrations, dosing methods, and dosing time ( P>0.05) .The death rate showed an increasing trend in the intraperitoneal injection, subcutaneous injection and intragastric administration of 30% CCL4 (25.33%, 37.78%, and 38.37%) and 50% CCL4 (42.97%, 47.85%, and 51.88%), higher in the 50%than in the 30%CCL4 .However, no significant differences were found in the survival curves among differ-ent dosing methods or between different drug concentrations (P>0.05). Conclusion Intraperitoneal injection was better than subcu-taneous injection and intragastric administration of CCL4 in inducing liver cirrhosis, and the three dosing methods all showed progressively improved efficiency of modeling with the increase of drug concentration and dosing time.

Objective To analyze the investigation data of the national schistosomiasis surveillance sites in 2015,so as to provide scientific evidences for schistosomiasis control, elimination and surveillance. Methods According to National Schisto-somiasis Surveillance Programme(version 2014),457 surveillance sites were selected, and the investigation data in residents, floating population, domestic animals and Oncomelania hupensis snails were collected and analyzed from four types of endemic counties. Results A total of 4468 seropositive cases were detected from 133350 residents, among which 4457 residents with seropositive results received the etiological tests, and 71 of them were identified with positive results. Most of them were fisher-men and farmers in the middle and old-aged group. The schistosomiasis infection rate was 0.05% in local population. Totally 977 seropositive cases were examined from 85047 migrant individuals, and 16 positive cases were found out from 966 individuals who took etiological tests, which showed the schistosomiasis infection rate was 0.02% in floating population. Imported cases were found among floating people in four provinces, namely Zhejiang,Hunan,Hubei and Anhui provinces. No acute schistoso-miasis cases were reported. A total of 13406 head of cattle received examinations and only 5 were determined as stool positives. The cattle infection rate was 0.04%. The snail survey covered an area of 22295.13 hm2 and snails were found in an area of 7426.63 hm2,including 3.47 hm2 newly detected area with snails. No schistosome-infected snails were found. Conclusions Based on the collected data from 457 national schistosomiasis surveillance sites of China, the Schistosoma japonicum infection rate is 0.05% in local population which maintains a stably descending trend. In floating population, there are imported schistosome-in-fected persons. Cattle are still a vulnerable species infected with schistosome. Although no infected snails are found, snails are widely distributed in endemic areas. Some provinces detect areas with snails for the first time or the reproduction of snails. The staff in endemic provinces should carry out the surveillance work according to National Schistosomiasis Surveillance Programme (version2014)to improve the surveillance system, and enhance the sensibility and effectiveness of surveillance work.

[Objective]To investigate the efficacy of a blended instruction model which incorporated preceptorship into the clinical anatomy course at Sun Yat-sen University and improve satisfaction and performance of medical students.[Methods]The study recruited participants including 375 Chinese students aged 20-24 years old,2 anatomy professors,49 surgeons and 7 lab technicians. The students were divided into 10 teams and each team was tutored by 1 anatomy professor,2 surgeons and 1 lab technician. After the course ended, the Course Experience Questionnaire (CEQ) and Student Experience in Education Questionnaire (SEEQ) were used in the survey to measure the quality of teaching and students' overall educational experience. Students' performance scores were collected and analyzed.[Results]The response rates for CEQ and SEEQ were 60.3% (226/375) and 54.6% (200/375),respectively. CEQ indicated a moderate level of overall satisfaction with 7.12 out of 10. Subscales like Clear Goals,Good Teaching and Generic Skills showed moderately positive reception of the course, while subscales like Appropriate Workload and Appropriate Assessment revealed students' concerns about heavy workload and difficult exam. There was no statistically significant difference in perceptions between male and female students. The students who are more satisfied with the quality of the course are also more satisfied with the way it is taught (P<0.01). SEEQ further confirmed the universal appeal of the course, with high scores in dimensions like Learning,Enthusiasm and Individual Rapport among the students. Qualitative responses highlighted areas of improvement, such as consistent teaching methods, practical hands-on experience for the students and alignment between what was taught and tested. The students after the preceptorship introduction achieved better academic performance than before.[Conclusions]The blended instruction model incorporating preceptorship enhances the student satisfaction and performance in clinical anatomy course, and also stimulates students' learning enthusiasm and group interaction. We need further improvement in the teaching quality control, syllabus contents and course assessment for teaching of clinical anatomy.

Objective To analyze the spatial clustering of human schistosomiasis at the village level in key counties in Hubei Province, to provide scientific evidence for formulating strategies for human schistosomiasis prevention and control in the next stage. Methods Gong'an County and Jiangling County in Hubei Province were selected as representative counties for this study. A town or village was set up as a research unit. Schistosomiasis cases of a positive fecal examination in 2015 and cases with positive detection for schistosomiasis serological antibody titer equal or above 80 in 2016-2018 were selected as research subjects in these two counties. The Kulldorff circular scan statistic was used for the spatial clustering analysis of human schistosomiasis infection status in the population. Results There was spatial clustering of positive schistosomiasis cases of fecal examination, at the level of a town or village in both counties in 2015. There was spatial clustering of positive human serological antibody detection at the level of town or village from 2016-2018. Eighty-six endemic villages in five towns in the northeast of Gong'an County, along the Yangtze river, including Mahaokou Town, Zhakou Town, Yangjiachang Town, Jiazhuyuan Town and Douhuti Town, were the most prominent. There was no spatial clustering of positive results of human serological antibody detection at the town and village level in Jiangling County, in2017, while there was spatial clustering of human serological antibody detection in 2016 and 2018,respectively. Fifty-seven endemic villages in two towns (Puji Town and Xionghe Town) in the southwest of Jiangling County, along the Yangtze river, were the most prominent. Conclusion There were spatial clustering of human schistosomiasis epidemic at village level both in Gong'an County and Jiangling County, Hubei Province. Compared with the previous studies, there was a trend of shrink and decline of clustering areas. Therefore, the current situation of the epidemic has put forward higher requirements for the implementation of precise prevention and control in the progress of schistosomiasis elimination work in various epidemic areas.
