1.AFFERENT CONNECTIONS OF SUBSTANTIA NIGRA IN THE RAT

The substantia nigra(SN)has been considered to be related to motor function.The recent physiological study has shown that the SN is not only one of theelements of extrapyramidal system,but is also related to the sensory system and takepart in the modulation of pain.In order to provide morphological evidence for itsfunctional aspect,42 rats have been employed for study by retrograde transportmethod of horseradish peroxidase(HRP),using microinjective and microiontophoreticdelivery system through vertical and oblique approaches.The results showed that the main labeled nuclei and neuro-structures were thecaudoputamen nucleus,dorsal raphe nucleus,central amygdaloid nucleus,globuspallidus,cerebral cortex,median raphe nucleus,locus coeruleus,periaqueductal grayand parabrachium nuclei.Topographical organization of the projections from these labeled nuclei andneuro-structures to SN was studied in detail.Among the structures studied the labeled cells were found most numerous incaudoputamen nucleus(CP).The projection fibers start in a order from the antero-dorso-medial to postero-ventro-lateral part of CP and end in the medial to lateralof SN in succession.Topographical projection from dorsal raphe nucleus to SN was also found.Thedorsal raphe nucleus only project to the medial two-thirds of SN.The fibers from central amygdaloid nucleus and locus coeruleus terminate in thelateral two-thirds of SN.Only the medial one half of SN receives the projection fibers from globuspallidus.In cerebral cortex,the main regions where labeled cells could be seen wereinsula,frontal and parietal regions.The projection fibers coming from median raphe nucleus terminate to the medialone-third of SN.When HRP was injected into lateral or median one-third of SN,numerousand clear cut labeled cells can be seen in the ventro-lateral region of periaqueductalgray.The projection from parabrachium nuclei terminates to the lateral one half ofSN.
2.Orbicularis oculi muscle activities by analyzing dog's electroneurogram

Objective To monitor the activities of orbicularis oculi muscle by analyzing the electroneurogram (ENG) of its innervating nerve. Methods The ENG was recorded in vivo by a cuff electrode implanted around a dog’s zygomatic branch of facial nerve. The differences between the ENG during relaxing period of the orbicularis oculi muscle and during the constricting period were analyzed to explore a way to monitor the muscle activities. Results The ENG signals indicating occurrences of eye blinking events were extracted in the study. Time-frequency analysis showed that there was no ENG difference when the orbicularis oculi muscle was in relaxing periods no mater whether it was in the nature eye-opening period or in the interval of eye blinking. The ENG of orbicularis oculi muscle in constricting period was different from the ENG in relaxing period at amplitude and frequency. Conclusion It is feasible to monitor the orbicularis oculi muscle activities by ENG analysis.
3.Effects of Naloxone on plasma level ?-endorphinl in rats with alcoholic fatty liver

Aim To explore the effect of naloxone on th e plasma level of ?-endorphin (?-EP) in rats with alcoholic fatty liver (AFL) and its possible mechanism. Meth ods Forty-eight Wistar male rats were divided randomly into four grou ps: Model group (n 12), low doses treatment group (n12), high doses tre atment group (n12) and normal control group (n12). By the end of the f ourth week, all the rats were weighted, narcosised, sacrificed, and got the whol e liver. The level of ?-EP in plasma and the level of superoxidized dismutase (SOD) and malondialdehyde (MDA) in plasma and liver tissue were measured.The liv er samples were analyzed for histopathology with microscope. Results Rat models of AFL were established successfully. 4.Standard Probe Endoscopic Ultrasonography before Endoscopic Submucosal Dissection Avoids Misjudging the Size of Esophageal and Gastric Stromal Tumors by Miniprobe Sonography

Endoscopic ultrasonography (EUS)is routinely performed before endoscopic submucosal dissection (ESD)for treatment of upper gastrointestinal stromal tumors.However,when a miniprobe sonography (12,15 and 20 MHz)was used,the size of tumor revealed by EUS was often inconsistent with what it actually was,which might increase the difficulty of ESD and the risk of perforation and massive bleeding.Aims:To investigate the value of standard probe (5 and 7.5 MHz)EUS in detecting the size of upper gastrointestinal stromal tumors before ESD.Methods:Clinical data of patients who were suspicious of esophageal and gastric stromal tumors by gastroscopy and EUS from Jan.2012 to Oct.2014 at the Renmin Hospital of Wuhan University were collected.Of them,195 cases treated by ESD were retrospectively analyzed.Results:Of 195 cases treated by ESD,37 cases diagnosed by standard probe EUS and 108 cases diagnosed by miniprobe EUS were confirmed as stromal tumors by pathology.Fourteen cases were failure for ESD and then transferred to surgical treatment,one was due to misjudgement of the origin of tumor by standard probe EUS and 9 were due to misjudgement of the size of tumor by miniprobe EUS.The misjudgement rate of standard probe EUS was lower than that of miniprobe EUS with an insignificant difference (2.7%vs.8.3%,P>0.05).In 9 cases misjudged by miniprobe EUS, the size of tumor presented by miniprobe EUS was significantly smaller than its real size [(1.22 ±0.51)cm vs.(3.97 ±1.06)cm,P<0.01].ESD was avoided or terminated in 3 cases because of the accurate estimation of tumor origin, structure and blood flow by standard probe EUS.Conclusions:For patients who are going to receive ESD for suspected upper gastrointestinal stromal tumors,it would be best to select standard probe EUS to detect the size,origin and blood flow of the tumor before ESD.It will decrease the risk and improve the success rate of ESD.
5.Medication Compliance Investigation and Pharmaceutical Care for 50 Cases of Hypertension

Objective: To understand the medication compliance of inpatients with essential hypertension, analyze the influencing factors in medication compliance of the patients and perform the targeted medication education and pharmaceutical care.Methods: Medication education and guide were conducted in 50 cases of patients with essential hypertension based on the main contents in guidelines for prevention of hypertension.Every patient completed regular follow-ups in two months after being recruited.The relevant information about the treatment and medication were collected and analyzed.Results: Through the medication education and pharmaceutical care, the patients had a better understanding on the relevant knowledge of hypertension, rational drug use and treatment.Among the 50 patients, the percentage of completely following the doctor's advice on the hypertension medication was 86%, that of quitting smoking and limiting alcohol was 82%, and that of optimized life way was 78%, and totally 82% of the patients effectively controlled blood pressure or improved markedly when compared with the situation on admission.Conclusion: Medication education and pharmaceutical care conducted by clinical pharmacists is very important for the patients with high blood pressure.It can help patients build a good lifestyle or improve the quality of life by improving the medication compliance of patients, alleviating complications and reducing adverse drug reactions.
6.The clinical characteristics of primary gastrointestinal lymphoma on the different site of origin

Objective To investigate the clinical characteristics of primary gastrointestinal lymphoma (PGIL) on different origin site in order to improve its diagnosis.Methods The clinical data from 202 patients with PGIL diagnosed by histology from January 1999 to June 2007 were identified from the clinical databases of 8 hospitals in Wuhan area and retrospectively analyzed.The patients were divided into gastric,small intestinal and large intestinal lymphoma groups according to the site of origin and there clinical characteristics were compared.Results The PGIL localization was gastric in 113 (56.0%) cases, small intestine in 37(18.3%) cases and large intestine in 52 (25.7%) cases.One hundred and thirty (64.4%) were males and 72 (35.6%) were females.The male patients were predominant.The median duration of symptoms in gastric lymphoma group was longer than small intestinal lymphoma group (3.0 months vs.1.0 month,P=0.013).The most common symptoms were abdominal pain and anemia. The clinical stage was Ⅰ E and Ⅱ E in 71.3% of cases.The large intestinal lymphoma group presented more advanced-stage disease compared with gastric lymphoma group (P = 0.014).The frequent histological type was mucosa-associated lymphoid tissue lymphoma (MALT),diffuse large B-cell lymphoma and T-cell lymphoma.Gastric,small intestinal and large intestinal lymphomas presented more frequently as low-grade MALT lymphoma (56.9%),T-cell lymphoma (34.4%) and high-grade B-cell lymphoma (51.1%),respectively (all P value <0.05).The common macroscopic type of PGIL were nodular protruding and ulcerative type.Compared with gastric lymphoma,nodular protruding type was more common and ulcerative type was less common in large intestinal lymphoma (P = 0.000).The diagnosis confirmed by endoscopic biopsy were 58.7% (61/104),25.0% (4/16),48.2% (13/27) in gastric,small intestinal and large intestinal lymphoma groups,respectively.Conclusions The clinical characteristics are different in patients with different localization of PGIL including patient characters, initial symptoms,histological classification,clinical stage,macroscopic feature,endoscopic findings. Analysis of these clinical characteristics is helpful to improve its diagnosis.
7.An experimental study on the influence of inhibition of postburn stress on inflammatory reaction in severely scalded rats.

OBJECTIVETo investigate the influence of hibernation drugs on postburn stress and inflammatory reaction in severely scalded rats.
METHODSSprague-Dawley rats inflicted with 30% TBSA deep partial thickness scalding were employed as the model. The rats were divided into A (scalding with immediate resuscitation), B (scalding with immediate resuscitation and lytic cocktail), C (scalding with delayed resuscitation), D (scalding with delayed resuscitation and lytic cocktail) and E (sham injury) groups. The rat plasma levels of NE (norepinephrine), E (epinephrine) and DA (dopamine) were determined by HPLC (high performance liquid chromatography) at 3, 6, 12, 24 and 48 postburn hours (PBHs), and the plasma IL-1alpha and PGE(2) levels were detected by ELISA (enzyme-linked immunosorbent assay) and RIA (radioimmunoassay) methods. The NF-kappaB activity in PBMCs (peripheral blood mononuclear cells) was determined by laser scanning confocal microscope.
RESULTSThe plasma NE and E levels reached summit at 6 PBH, while those in B group were lowest. But the plasma DA level was similar among all groups at all time points. The plasma IL-1alpha and PGE(2) levels increased continuously, however, the levels were were lower in B than A groups at the same time points, and also that of D were lower than C groups. At the same time points, the levels in B group were lower than those in D group. The NF-kappaB was located in the cytoplasma of PBMCs in E group and in the nucleus in A group at 6 PBH. Furthermore, the NF-kappaB was concentrated more in the cytoplasm than that in the nucleus in B group, while it was more concentrated in the nucleus in C and D groups.
CONCLUSIONThe secretion of stress hormones could be attenuated by hibernation drugs. The plasma cytokine levels and the nuclear translocation of NF-kappaB in PBMCs could also be modulated by the drugs.
