2.Effects of group cognitive behavioral therapy on clinical efficacy and executive function in patients with generalized anxiety disorder

BackgroundPrevious studies have found that patients with generalized anxiety disorder (GAD) have impaired performance in executive function, and group cognitive behavioral therapy (CBT) has been shown to be effective in alleviating negative affect in patients with GAD, while its efficacy on executive function remains unclear. ObjectiveTo explore the efficacy of group CBT on anxiety symptom and executive function in GAD patients, so as to provide references for the rehabilitation program for GAD. MethodsA total of 80 consecutive patients with GAD who were hospitalized in Sleep and Psychosomatic Medical Center of Shiyan Taihe Hospital from March 2021 to August 2022 and met the Diagnostic and Statistical Manual of Mental Disorders, fifth edition (DSM-5) diagnostic criteria for GAD were enrolled, and they were assigned into study group (n=40) and control group (n=40) using random number table methods. All patients were subjected to routine medication treatment and regular health education, based on this, study group received group CBT once a week (6 weeks, 60 to 90 minutes per session). At the enrollment and after 6 weeks of treatment, patients were assessed using Hamilton Anxiety Scale (HAMA) and Frontal Assessment Battery (FAB). ResultsANOVA with repeated measures on HAMA score revealed a significant time effect (F=1 870.320, P<0.01), no significant group effect and no significant time×group interaction effect (F=1.254, 0.293, P>0.05). Significant time effect, group effect and time×group interaction effect were reported on FAB scores (F=311.190, 4.399, 7.021, P<0.05 or 0.01). Further analysis indicated that FAB scores of both groups after treatment were higher than those at baseline (t=200.569, 115.401, P<0.01).And the FAB score of study group was higher than that of control group after treatment (t=-3.211, P<0.01). ConclusionGroup CBT combined with medication treatment for GAD may alleviate the anxiety symptoms and improve executive function in GAD patients. 3.Comparison of the Legiolert enzyme-substrate method and the conventional cultivation for the detection of Legionella pneumophila in water samples from public places

Objective To compare the detection effects of Legionella pneumophila in water samples from public places by the Legiolert enzyme-substrate method and the conventional cultivation. Methods The Legiolert enzyme-substrate method and the conventional cultivation method were used to detect Legionella pneumophila in cooling water and shower water samples collected in public places. Isolated strains were verified and serotyped. Results A total of 68 samples were collected and tested. The positive rate of the conventional cultivation and the Legiolert enzyme-substrate method were 5.88%(4/68)and 35.29%(24/68), respectively, with a significant difference (χ2=16.41,P<0.01). The coincidence rate of the two methods was 64.71% (44/68), and the difference of the detection effects of the two methods was statistically significant (χ2=16.41,P=0.000). A total of 25 strains of Legionella pneumophila were isolated,and the serum types were mainly LP1(14/25). Conclusion The Legiolert enzyme-substrate method represente a higher detection rate of Legionella pneumophila in water samples from public places than the conventional cultivation.
4.Different emotion faces stimulation in patients with major depressive disorder by Event-related Potential N170

Objective To investigate effects of three kinds of neutral,happy,sad emotional faces stimulus on sustained attention task using N170 in depression patients to analysis the correlation of depression in patients with depression,anxiety severity and the amplitude and latency of N170.Methods Twenty-eight patients with depression aged from 22 to 69 years (case group),and 31 healthy controls (control group) aged from 20 to 61 years were recruited for the study.Subjects were asked to perform tasks in the selection of attention following randomly presented three kinds of happy,neutral and sad emotional faces stimuli.Brain auditory evoked potential including N170 amplitude and latency were recorded during the tasks.HAMD and HAMA were used to assess the severity of depression and anxiety.Results There was significant difference between case group and control group in N170 latency by three kinds of happy,neutral and sad emotional faces stimulation in the local skull (T5,T6,O1,O2) (P<0.05).But the difference was not significant in N170 amplitude (P>0.05).The total score of HAMD in case group had a positive correlation with N170 amplitude by neutral facial emotion stimulation in part of T5 (r=0.443,P=0.018).Conclusion There is significant change in the initial cognitive processing of different emotional faces in the patients with depression.And the severity of depression is related to the N170 amplitude induced by the neutral emotional faces stimulation in some parts of brain.
7.Study on hypovascular tumor' s treatment with targeted mierobubbles combined with ultrasonic abrupted destroy

Objective To assess the therapeutic efficacy of colonic tumor with targeted microbubbles encapsulated VEGFR2 monoantibody (mAb) combined with ultrasonic abrupted destroy.Methods Seventeen Balb/C nude mice with subcutaneous colonic carcinoma xenografts were divided into three groups:group A (5 mice) underwent contrast-enhanced ultrasonography (CEUS) examination and sham ultrasonic abrupted destroy;group B (6 mice) underwent lipid-microbubbles administration combined with ultrasonic irradiation;group C (6 mice) underwent VEGFR2 monoantibody-loaded microbubbles injection combined with ultrasonic irradiation.Red fluorescent protein(RFP) was labeled to all the nude mice model.Both CEUS and flurography were performed before and one week after abrupted destroy.The size,fluroscent area and fluroscent intensity(FI) and vessel density (VD) of each tumor were measured and compared.Results The parameters of length,fluroseent area,FI and VD of each tumor before abrupted destroy were no significant difference among three groups ( P >0.05).Parameters of post-sham ultrasonic abrupted destroy in group A were higher than those before sham ultrasonic abrupted destroy ( P <0.05).FI and VD in group B were significantly lower than those after abrupted destroy( P <0.05).There were no difference of length,fluroscent area of tumor in group B between pre- and post- ultrasonic abrupted destroy (P >0.05).Length,fluroscent area,FI and VD of each tumor in groups C were decreased significantly compared with post ultrasonic abrupted destroy ( P <0.01 ).There were significant difference of length,fluroscent area,FI and VD of each tumor among groups after ultrasonic abrupted destroy( P <0.05).Conclusions VEGFR2 mAb-loaded lipid microbubble combined with ultrasonic abrupted destroy can improve the therapeutic efficacy of colonic tumor.
8.Mechanism of polyphyllin Ⅰ targeting EGFR to affect proliferation and apoptosis of human breast cancer cells.

This study aims to investigate the molecular mechanism of polyphyllin Ⅰ(PPⅠ) inhibiting proliferation of human breast cancer cells. Human breast cancer BT474 and MDA-MB-436 cells were treated with different concentrations of PPⅠ, and then the effect of PPⅠ on cell proliferation was detected by MTT assay, trypan blue dye exclusion assay, real-time cell analysis, and clone forming assay, respectively. The apoptosis was detected by Annexin V-FITC/PI staining and then analyzed by flow cytometry. The change of mitochondrial membrane potential was detected by flow cytometry after fluorescent probe JC-1 staining. Western blot was used to detect protein expression and phosphorylation. Molecular docking was performed to detect the binding between PPⅠ and EGFR. The affinity between PPⅠ and EGFR was determined by drug affinity responsive target stability assay. The results indicated that PPⅠ inhibited the proliferation and colony formation of BT474 and MDA-MB-436 cells in a time-and concentration-dependent manner. The PPⅠ treatment group showed significantly increased apoptosis rate and significantly decreased mitochondrial membrane potential. PPⅠ down-regulated the expression of pro-caspase-3 protein, promoted the cleavage of PARP, and significantly reduced the phosphorylation levels of EGFR, Akt, and ERK. Molecular docking showed that PPⅠ bound to the extracellular domain of EGFR and formed hydrogen bond with Gln366 residue. Drug affinity responsive target stability assay confirmed that PPⅠ significantly prevented pronase from hydrolyzing EGFR, indicating that PPⅠ and EGFR have a direct binding effect. In conclusion, PPⅠ inhibited the proliferation and induced apoptosis of breast cancer cells by targeting EGFR to block its downstream signaling pathway. This study lays a foundation for the further development of PPⅠ-targeted drugs against breast cancer.

9.Mechanism of Polyphyllin Ⅰ Extract in Activating Hippo Signal to Induce Apoptosis and Autophagy of Colorectal Cancer Cells

ObjectiveTo study the inhibitory effect of polyphyllin Ⅰ (PPI) on the growth of colorectal cancer cells and its molecular mechanism. MethodRKO cells were cultured and divided into a blank group and PPI treatment groups with concentrations of 0.6, 0.8, 1.0 μmol·L-1, respectively. HRT18 cells were cultured and divided into a blank group and PPI treatment groups with concentrations of 1.2, 1.4, 1.6 μmol·L-1, respectively. The effects of PPI on the proliferation and morphology of colorectal cancer were detected by cell proliferation toxicity assay, trypan blue exclusion assay, plate clone formation assay, and confocal high-intension cell imaging analysis system. Flow cytometry was used to detect the apoptosis rate of colorectal cancer cells. The pQCXIP-GFP-LC3 plasmid transfection assay was used to detect the formation of autophagosomes in colorectal cancer cells after PPI treatment. Western blot was used to detect the expression of apoptosis-related proteins Caspase-3, Caspase-8, and poly ADP ribose polymerase (PARP), the expression of autophagy related protein LC3Ⅱ, and the expression and phosphorylation of Hippo signaling pathway proteins LATS1 and YAP. In the plvx-Flag-YAP plasmid transfection assay, YAP was overexpressed and treated with PPI, and the proliferation of colorectal cancer cells was detected by cytotoxicity assay. The expression of LC3Ⅱ and PARP in colorectal cancer cells was detected by Western blot. SwissADME predicted pharmacokinetic parameters of PPI. ResultAs compared with the blank group, the survival rate and clone formation ability of colorectal cancer cells in the PPI group were significantly decreased (P<0.01), the cell area of colorectal cancer cells in the PPI group was significantly decreased, and the roundness of colorectal cancer cells was significantly increased (P<0.01). As compared with the blank group, the apoptosis rate of colorectal cancer cells in PPI treatment groupw was significantly increased (P<0.01), the expression of apoptotic proteins Caspase-3 and Caspase-8 protein precursor in PPI treatment groups was decreased, and the cleavage of PARP was increased (P<0.01). As compared with the blank group, the expression level of autophagy-related protein LC3Ⅱ in colorectal cancer cells in PPI treatment groups was significantly increased, and the formation of autophagosomes was promoted (P<0.01). As compared with the blank group, the expression of YAP protein in colorectal cancer cells in PPI treatment groups was significantly decreased, and the expressions of phosphorylated LATS1 and YAP were significantly increased (P<0.01). As compared with the blank group, overexpression of YAP could significantly antagonize the effect of PPI on apoptosis, autophagy activation, and proliferation inhibition of colorectal cancer cells. SwissADME simulation results showed that PPI had good drug like activity. ConclusionPPI can induce apoptosis and autophagy of colorectal cancer cells through targeted activation of Hippo signaling pathway, thereby inhibiting their proliferation.
10.Research Progress in Hospice Communication between Cancer Patients and Doctors

With the increasing number of cancer patients in China, the lack of hospice communication between medical staff and cancer patients can easily cause doctor-patient conflicts. Facing the special group of cancer patients, by introducing the concept of hospice communication and comparing the current situation of hospice communication of cancer patients at home and abroad, this paper found the shortcomings of hospice communication between medical staff and cancer patients in China. This paper aimed to analyze the influencing factors of cancer patients’ hospice communication from three aspects of medical staff, cancer patients and social and cultural background, summarized the assessment tools and matters needing attention related to hospice communication, so as to provide reference for domestic medical staff to develop relevant tools for hospice communication with cancer patients, and help medical staff to implement more effective hospice communication with cancer patients in the context of tranquil care. It is also conducive to help patients open the topic of death from the perspective of doctors and build an open hospice communication environment that is more in line with national conditions of China.
