1.Bacterial Infection after Gunshot Injury in Chongqing and Lasa:A Comparative Study

OBJECTIVE To observe Staphylococcus aureus(SAU) infection at high altitude and plain after gunshot injury. METHODS Used 7.62mm bullets to injure posterior limbs of piglets with the same age.We got contusing tissues in different time after injury,to make bacteriologic examination,identify and make a statistic analysis of SAU. RESULTS The main infection was caused by SAU after gunshot injury in two regions.From contusing tissues in two regions,the rate of contamination caused by SAU in Chongqing and Lasa was 48% and 18%,respectively.The time of SAU colonization was 3 hours at high altitude,and less 3 hours at plain.The velocity of reproduction of SAU at high altitude for 12 hours was 5-6 passages,and at plain was 4-6 passages for 3-6h.The time of remaining relative steady state between tissues and germs was 48 hours at high aititude and 24 hours at plain.The difference of the number of SAU between two regions after injury 3-36 hours was significant,But after 36 hours the difference was unsignificant. CONCLUSIONS At high altitude region,SAU is the main germ that causes contamination after gunshot injury,the velocity of reproduction of SAU is slower than that at plain,the time of infection is delayed obviously.
2.Losartan attenuates vascular remodeling of the aorta in spontaneously hypertensive rats and the underlying mechanism

Objective To determine the effect of losartan on vascular remodeling and the underlying mechanism in spontaneously hypertensive rats(SHR). Methods SHR of 12 weeks old were given losartan orally [0,15,30 mg/(kg·d),n=12]. The tail arterial pressure was measured every week.Eight weeks later, the pathological changes and p22phox expression in the thoracic aorta, the activity of catalase (CAT), the contents of H2O2 and AngⅡ in the plasma were evaluated. Results Blood pressure was increased in the SHR accompanied by the thickened wall and increased p22phox expression in the thoracic aorta. The plasma levels of H2O2 and AngⅡwere elevated while the CAT level was decreased in the SHR. Administration of losartan reversed the thickened wall and increased the CAT activity concomitantly with the decreased plasma levels of H2O2 and p22phox expression in the SHR. The plasma level of AngⅡincreased after the losartan treatment. Conclusion Oxidative stress induces the vascular remodeling of the aorta in the SHR. Losartan can reverse the vascular remodeling through down-regulating p22phox expression and inhibiting the oxidative stress.
3.Expressions and significance of erythropoietin producing hepatocellular cell line receptor A2 in pediatric brain glioma

Objective To investigate the expressions and significance of erythropoietin producing hepatocellular cell line receptor A2 (EphA2) in pediatric brain glioma.Methods Seventy-eight fresh pediatric glioma specimens with a defined histological grade were collected in the Fifth Affiliated Hospital of Zhengzhou University from Jan.2009to Mar.2013,which included 36 of low grade glioma(Ⅰ-Ⅱ grade),42 of high grade glioma(Ⅲ-grade),another 33 cases with brain trauma line pressure to remove children brain tissues were collected as control group.The expressions of EphA2 mRNA and protein were detected by reverse transcription-polymerase chain reaction (RT-PCR),Western blot and immunohistochemistry.Results 1.RT-PCR and Western blot showed that EphA2 did not express in control brain tissue,but the expression levels of EphA2 mRNA were over-expressed in pediatric brain glioma,and the difference was statistically significant(F =36.271,P < 0.05) ;the expression levels of EphA2 protein were significantly higher in high-grade pediatric glioma group than in low-grade pediatric glioma group,and the difference was statistically significant(F =42.839,P < 0.05).2.Immunohistochemistry showed that EphA2 expression was negative in control group,the positive expression in low-grade glioma group was 88.57%,and the positive expression in high-grade glioma group was 100.00%.Moreover,the higher the grade glioma,distribution of EphA2 expression was stronger,and the difference was statistically significant(Z =4.962,P < 0.05).Conclusions The mRNA and protein expression levels of EphA2 were significantly high in pediatric brain glioma which were associated with the grade of glioma.Therefore,EphA2 may participate in the development and progression of pediatric brain glioma.
4.Proposal on the cultivation of hearing aids fitting practitioner in China

This paper discussed the current status of hearing aids fitting practitioners in China and proposed some proposals on how to cultivate qualified practitioners.The proposals includes setting up short-term training program,launching hearing aids fitting major in occupational schools and cultivating the hearing aids fitting practitioners from the would-be audiologists or otologists in an aim to meet the urgent hearing aids fitting need of Chinese deafness.
5.Analysis of methyl tert-butyl ether in urine of primary school students in Beijing

Objective:
To investigate the urine methyl tert-butyl ether (MTBE) level of a primary school students in Changping District, Beijing, and analysis the effect factors, give the advice if the MTBE is eligibility as an exposure biomarker of automobile exhaust.
Methods:
Using the panel study, randomly selected 20 sixthgrade students (10 male and 10 female) from Zhixing School in Changping district, Beijing, from June 7th to 13th, 2018. All participants completed a questionnaire for collecting demographic and geographic information and daily activities. All participants were given guidance to repeatedly collected urine samples 3 times (8:00, 11:00 and 16:00 instant urien) a day for 7 days, totally 420 urine samples were collected. Urine MTBE were measured and the statistic significant of the Urine MTBE at different groups were evaluated.
Results:
MTBE were detected in all 420 urine samples of primary school students, the geometric mean of creatinine-corrected MTBE exposure level in 8:00, 11:00 and 16:00 instant urine were 0.12, 0.12 and 0.12 mg/g creatinine, respectively. Urine MTBE concentration was statistical significant in different daily outdoor activity time, the distance of home address to the main road, and daily use time of air purification equipment in class(P=0.00, 0.04, 0.00).
Conclusion
There was a universal MTBE exposure among students in a primary school in Beijing, and the impact factors to exposure level were daily outdoor activity time, distance from home address to the closest main road, and daily use time of air purification equipment in class. This result indicate MTBE may be a valuable candidate for exposure biomarker of automobile exhaust, and more research should be done.
6.Influence of relapse of medically intractable temporal lobe epilepsy shortly after surgery in its long-term efficacy

Objective To investigate the influence of drug refractory temporal lobe epilepsy relapse shortly after surgery in its long-term efficacy.Methods The clinical data of 57 patients with medically intractable temporal lobe epilepsy,underwent surgery from August 2010 to August 2011 in our hospital,were retrospectively analyzed; all the patients were performed surgery,and postoperative treatment of epilepsy (adjustment antiepileptic drugs) in our hospital and rehabilitation center was chosen;the parallel regular follow-up clinic or telephone was also performed.According to the relapse situation one year after surgery,these patients were divided into control group (no epilepsy,n=18) and experimental group (relapse,n=39).Follow-up data,long-term efficacy and overall efficacy between the two groups were analyzed.Results (1) Postoperative medial temporal lobe sclerosis,neoplastic lesions,mixed lesions and normal patients were 27,7,10 and 13,respectively,counting for 47.37%,12.28%,17.54% and 22.81% of the patients; (2) on the second year of surgery,patients in the experimental group had 44.44% satisfactory seizure control,which was significantly lower than those in the control group (74.36%,x2=4.839,P=0.028); (3) Kapan-Meier survival curve indicated that epilepsy remission rate in the control group was significantly higher than that in the experimental group on the second year of surgery (x2=5.928,P=0.017); (4) multivariate Logistic regression analysis showed that risk factors affecting the long-term efficacy included incentives recurrence (OR=4.511,95%CI:3.195-7.569),relapse frequency≥ 3 times (OR=4.281,95%CI:0.040-0.712) and recurrence of epilepsy type to complex partial seizure/generalized tonic-clonic seizures (OR=3.344,95%CI:3.186-8.400).Conclusion Drug refractory temporal lobe epilepsy relapse shortly after surgery will affect its long-term efficacy,and the determinate factors included incentives recurrence,relapse frequency≥ 3 times and recurrence of epilepsy type.
7.Effect of colon cancer cell-derived IL-1α on the migration and proliferation of vascular endothelial cells.

OBJECTIVETo explore the effect of colon cancer cell-derived interleukin-1α on the migration and proliferation of human umbilical vein endothelial cells as well as the role of IL-1α and IL-1ra in the angiogenesis process.
METHODSWestern blot was used to detect the expression of IL-1α and IL-1R1 protein in the colon cancer cell lines with different liver metastatic potential. We also examined how IL-1α and IL-1ra influence the proliferation and migration of umbilical vascular endothelial cells assessed by PreMix WST-1 assay and migration assay, respectively. Double layer culture technique was used to detect the effect of IL-1α on the proliferation and migration of vascular endothelial cells and the effect of IL-1ra on the vascular endothelial cells.
RESULTSWestern blot analysis showed that IL-1α protein was only detected in highly metastatic colon cancer HT-29 and WiDr cells, but not in the lowly metastatic CaCo-2 and CoLo320 cells.Migration assay showed that there were significant differences in the number of penetrated cells between the control (17.9±3.6) and 1 ng/ml rIL-1α group (23.2±4.2), 10 ng/ml rIL-1α group (31.7±4.5), and 100 ng/ml rIL-1α group (38.6±4.9), showing that it was positively correlated with the increasing concentration of rIL-1α (P<0.01 for all). The proliferation assay showed that the absorbance values were 1.37±0.18 in the control group, and 1.79±0.14 in the 1 ng/ml rIL-1α group, 2.14±0.17 in the 10 ng/ml rIL-1α group, and 2.21±0.23 in the 100 ng/ml rIL-1α group, showing a positive correlation with the increasing concentration of rIL-1α(P<0.01 for all). IL-1ra significantly inhibited the proliferation and migration of vascular endothelial cells (P<0.01). The levels of VEGF protein were (1.697±0.072) ng/ml, (3.507±0.064)ng/ml and (4.139±0.039)ng/ml in the control, HUVECs+ IL-1α and HUVECs+ HT-29 co-culture system groups, respectively, showing a significant difference between the control and HUVECs+ 10 pg/ml rIL-1α groups and between the control and HUVECs+ HT-29 groups (P<0.01 for both).
CONCLUSIONSOur findings indicate that colon cancer cell-derived IL-1α plays an important role in the liver metastasis of colon cancer through increased VEGF level of the colon cancer cells and enhanced vascular endothelial cells proliferation, migration and angiogenesis, while IL-1ra can suppress the effect of IL-1α and inhibit the angiogenesis in colon cancer.

8.Correlation of IL-6-572GC mutation with incidence and severity of coronary heart disease in elderly Han people in Henan Province

Objective To investigate the correlation of IL-6-572GC mutation with the incidence and severity of CHD in the Han elderly in Henan Province.Methods A total of 446 elderly CHD pa-tients of Han majority in Henan Province who were admitted to Department of Cardiovascular Diseases of our hospital from January 2020 to December 2022 were enrolled as the study group,and 218 healthy elderly patients taking physical examination in the same period were selected as the control group.The genotypes of IL-6-174GC,IL-6-572GC and IL-6-597GA were detected in the two groups.Logistic regression analysis was used to analyze the relationship between IL-6-572G/C gene polymorphism and CHD in the elderly,as well as the correlation between IL-6-572GC genotype and the severity of CHD.Results Larger proportions of hypertension and diabetes and higher LDL-C level,but lower HDL-C level were observed in the study group than the control group(P<0.05,P<0.01).There were three genotypes at IL-6-174GC locus(GG,GC and CC),three genotypes at IL-6-572GC locus(GG,GC and CC),and three genotypes at IL-6-597GA locus(GG,GA and AA).There were no significant differences in the genotype fre-quencies of IL-6-174GC and IL-6-597GA between the two groups(P>0.05),but there was a sig-nificant difference in the genotype frequency of IL-6-572GC(P<0.01)and the genotype frequen-cy of GG was significantly lower in the study group than the control group(33.18%vs 46.82%,P<0.01).Logistic regression analysis showed that IL-6-572GC(OR=1.534,95%CI:1.180-1.995),hypertension(OR=1.442,95%CI:1.033-1.957),diabetes(OR=1.610,95%CI:1.083-2.391),HDL-C(OR=0.467,95%CI:0.266-0.818)and LDL-C(OR=2.004,95%CI:1.104-3.636)were all influencing factors of CHD(P<0.05,P<0.01).At IL-6-572GC locus,the inci-dence of single branch lesions was higher in the carriers of GG genotype than those of GC and CC genotypes,and the incidence of 3-branch lesions was lower than that of CC genotype(P<0.05).Conclusion The mutation of IL-6-572GC gene is correlated with the incidence and severity of CHD in the elderly Henan Han people,and its mutation also increases the risk and severity of the disease.
