1.Risks and Legal Analysis of Gene Editing

With the application of gene editing in medical treatment,food engineering,environmental protection,agriculture and forestry,it gives rise to a wide range of potential social risks.Both the advanced aspects and defects of this technology bring complicated social conflicts.It is undoubtedly crucial to coordinate the conflict of interests of all parties,improve the legal system and avoid the harmfulness and blindness of gene editing.Meanwhile,only that the ethical,legal and social concerns are introduced will govern the technical risk as a necessary condition in order to attain a virtuous cycle of technological improvement and social prosperity.
2.Establishing of rat heart failure model in myocardial infarction:the changes of cytokines

Objective To establish rat heart failure(HF)model following myocardial infarction(MI)and then detect the possible cytokines expression levels at different HF groups,so to explore the development of cytokines levels during the progression of heart failure.Methods The model was established by ligature of the left descending coronary artery.The rats were assessed by echocardiography and cardiac catheterization before and after 4 and 16 weeks following coronary ligature.Serum levels of TNF-α and IL-6 were determined.Results ①Serum cytokines could not be detectable in sham group using ELISA Methods,while in operated group,the concentration of TNF-α and IL-6 were significantly higher,especially IL-6.②The diagnosis coincidence between echocardiography and hemodynamics was high.Echocardiography can be used as an invasive measure to assess cardiac function in rat heart failure models.Conclusion ①Cytokines content is higher in compensated compared with the other two groups,especially to the IL-6 expression.②Cardiac ultrasound can be used as a useful method in monitoring postinfarction heart failure rat model,especially as an invasive measure.It can provide us successive data of the model.
3.THE CHANGES IN DENSITY OF PLATELET GLYCOPROTEIN(GP)Ⅱb/Ⅲa RECEPTOR DURING THE PERIOPERATIVE PERIOD OF HEPATECTOMY IN RATS WITH MYOCARDIAL INFARCTION

To investigate changes in density of platelet glycoprotein (GP)Ⅱb/Ⅲa receptor during the perioperative period of hepatectomy in rats with myocardial infarction. 26 adult SD rats were randomly divided into 2 groups: the control group(1)( n =13) and the infarction group(2)( n =13). Myocardial infarction was produced by celiac injection (1 time/d) of isoproterenol (30mg/kg) for 3 days. All rats underwent hepatectomy after 3 weeks. Platelet surface density of GPⅡb/Ⅲ receptors was determined by flow cytometric analysis before and 6, 12, 24, 48, 72 hours after the operation. 12 hours after the operation, the density of GPⅡb/Ⅲa was transiently increased compared with preoperative period in control group ( P
4.MRI Analysis of Morphological Structure in Protrusion of cervical Disc under Different Directions of Traction

Objective The purpose of this study was to compare the functional and morphological changes in the cervical spine with protrusion of cervical disc (PCD) under different directions of traction. Methods MRI scanning was used to study the effect of different directions of traction on the movement of protrusive disc,compression index of spinal cord and changes of intervetebral space,as well as the cases with degeneration. Results Differences were found in the effect of different directions of traction. Most significant partial reduction of the protrusive disc was experienced in flexion traction. Both flexion and extension traction caused significant decrease in the index of compression of spinal cord. Extension traction caused marked narrowing of the posterior intervertebral space in the group with decreased disc size,while no difference between the groups with and without decreased disc size when given flexion traction. There was no difference in the anterior intervertebral space between the two directions of traction. No morphological change was observed in cases with degeneration using either direction of traction. Conclusion PCD could caused the change in the fine morphology of normal vertebral joints. The traction direction should be chosen according to the condition of the individual case. The compression index could better reflect the nature of spinal cord injury than the size of the disc protrusion. Traction would not give much benefit in cases with degeneration.
5.Relationship of lung infection and multiple organ failure in the elderly.

Objective The aim of the study is to explore the relationship of lung infection(LI)and multiple organ failure in the elderly(MOFE).Methods Consecutive patients ,who were admitted to the Institute of Geriatric Cardiovascular Diseases of the PLA General Hospital and the Endocardial Department of the Air Force General Hospital,withage≥65 years old were enrolled into 5 groups retrospectively by following criteria:acute LI alone,LI with the first presentation of acute lung edema,chronic bronchitis complicated with LI,chronic heart failure complicated with LI,and nic bronchitis and heart failure complicated with LI.Results Sixty-eight patients were selected of(72.5?7.6)years old ( 38 male).There were 4 cases of pure LI(4%),12 cases of LI firstly presented with the symptoms of acute lung edema(18%),16 cases of LI complicated with chronic bronchitis(24%),15 cases of LI based on chronic heart failure(22%)and 22 cases of LI complicated with chronic bronchitis and heart failure(32%).LI initiated MOFE in 25 cases(37%).Most of them were developed on the basis of chronic bronchitis and/or heart failure(34%).Mortality of secondary LI was higher than that of the primary LI(7.4% VS 0%,P

Protein ubiquitin-like modification of Neddylation is an important post-translational modification mode of protein to widely regulate cell cycle, growth, development and other biological processes. Recent studies have found that MLN4924, small molecule inhibitor of Neddylation modification, has a significant anti-tumor effect, which can inhibit the tumor growth by inducing cell cycle arrest, senescence and apoptosis, and inhibiting tumor angiogenesis. This paper reviews the related mechanism research of cell programmed death induced by Neddylation modification pathway inhibitor MLN4924.
7.Effect of Aerobic Exercise on Platelet Function in Elderly Patients with Hypertension and Type 2 Diabetes

Objective To explore the effect of aerobic exercise on platelet function in elderly patients with hypertension and type 2 diabetes.Methods81 elderly patients with hypertension and type 2 diabetes were divided into the routine treatment group (group A, n=41) and regular exercise group (group B, n=40). All patients of two groups were treated with routine treatment. The exercise with moderate intensity was performed in the group B for 3 months besides routine treatment. The platelet membrane glycoprotein CD62P, CD61 (Ⅲa) were detected by flow cytometric analysis and platelet aggregating ratio was measured before and after regular aerobic exercise.ResultsAfter three months, the systolic blood pressures were 166±3.6 mm Hg (group A) and 152±3.5 mm Hg (group B); the diastolic blood pressures were 93±4.2 mm Hg (group A) and 83±4.3 mm Hg (group B); the contents of blood glucose were 7.4±2.4 mmol/L (group A) and 6.3±1.9 mmol/L (group B); the positive percents of platelet membrane glycoproten CD62P were 27.3±2.2% (group A) and 21.5±3.3% (group B), CD61(Ⅲa) were 26.3±2.3% (group A) and 20.2±2.9% (group B) and platelet aggregation rates were 78.4±4.5% (group A) and 69.7±5.4% (group B), there was a significant difference between two groups ( P<0.05~0.01).ConclusionRegular aerobic exercise can decrease the positive percent of platelet membrane glycoprotein and platelet aggregation rate in elderly patients with hypertension and type 2 diabetes.
8.Involvement of Sympathetic Nerve System in Vascular Headache

Objective To investigate the involvement of sympathetic nerve system in the transmission of nociceptive information related to vascular headache. Methods Fos expression in the midbrain periaqueductal grey (PAG) induced by electrical stimulation of dural matter near the superior sagittal sinus (SSS) of the rat was determined before and after the extirpation of the superior cervical ganglion (SCG) using standard avidin-biotin immunohistochemistry. Results Fos immunoreactive positive neurons were detected in the ventrolateral PAG, and were bilateral symmetry. Fos expression was (133.3±12.5) in SCG extirpated group, and higher than that in sham-operated group (76.0±11.3). Conclusion The sympathetic system may be involved in the transmission of nociceptive information through PAG, associated with vascular headache such as migraine.
9.Effect of disequilibrium of calcium homeostasis on apoptosis of MDCC MSB1 cells induced by lanthanum chloride

To investigate the effect of the disequilibrium of calcium homeostasis on the apoptosis of MDCC-MSB1 cells induced by lanthanum chloride(LaCl3 ), MDCC-MSB1 cells was conventionally cultured in RPMI1640, which was tured for 24 h, MTT was utilized to detect the cell multiplication inhibition ratio,DNA ladder and TUNEL to detect apoptosis,the Fura-2/AM as the probe to detect the [Ca2+]i within cells. The results indicated when LaCl3 concen and [Ca2+]i also increased,and presented a dose-effectiveness relationship. LaCl3 could restrain the proliferation of MDCC-MSB1 cells,suggesting their apoptosis induced by changing [Ca2+]i.
10.Mesenchymal stem cell-conditioned medium could ameliorated triptolide induced damage in KGN cells

Objective:To explore the ameliorative effect and machanism of MSCs conditioned medium on the ovarian granulosa cells damage induced by triptolide.Methods: Cell Counting Kit 8 assay was used to examine the cell vitality of KGNs with the treatment of triptolide.The mixed enzyme digestion method were used for the isolation of human umbilical cord mesenchymal stem cells (MSCs),and flow cytometry was used for the subsequent immunotype identification.MSCs conditioned medium was collected ,and Cell Counting Kit 8 assay and PI staining was used to analyse the effect of MSCs conditioned medium on the cell vitality and cell cycle distri -bution of triptolide-damaged KGN.Real-time PCR method was used to examine the expression of cell cycle related gene CDKN1A.Results:Triptolide can inhibit KGN cell growth with the inhibition of cell vitality and cell cycle of KGN.MSCs conditioned medium did not influence the proliferation and cell cycle of normal KGN , but improved the triptolide-induced vitality inhibition and extent of S-phase arrest, and inhibit the abnormal up-regulation of CDKN1A in KGN.Conclusion: MSCs conditioned medium ameliorated the KGN cell damage induced by triptolide.
