1.Observation of effects of the brain-located mild hypothermia therapy on acute progressive cerebral infarction

ObjectiveTo study effects of treating acute progressive cerebral infarction by the brain-located mild hypothermia therapy.Methods60 patients were randomly divided into the treatment group (30 cases) and the control group (30 cases). Drugs were used in both groups, and the located mild hypothermia therapy was only applied in the treatment group. After 72 hours and 7 days, the volume of cerebral infarction and the nerve function deficiency (NFD) scores of hemiplegic limbs were compared.ResultsThe volume of cerebral infarction of the treatment group was significantly smaller than that of the control group (P<0.01).There were significant differences on the neurological impairment scores between two groups (P<0.01).Conclusions The brain-located mild hypothermia therapy can reduce hydrocephalus of patients with acute progressive cerebral infarction, and have brain protection. These results provide an important theoretical foundation for the treatment of cerebral infraction with low-temperature therapy.
2.Protective effect of polysaccharides from Lycium ruthenicum Murray against ultraviolet B radiation-induced photodamage in HaCaT cells

Objective To evaluate preventive effect of polysaccharides from Lycium ruthenicum Murray against photodamage in HaCaT cells,and to explore its possible mechanism.Methods Ultrasoundassisted extraction was used to extract polysaccharides from Lycium ruthenicum Murray in Qaidam Basin.In vitro cultured HaCaT cells were randomly divided into 3 groups:control group receiving no treatment,ultraviolet B (UVB) group irradiated with 30 nJ/cm2 UVB alone for 1 hour,experimental group pretreated with 2 g/L Lyeium ruthenicum polysaccharide solution followed 6 hours later by 30 mJ/cm2 UVB radiation for 1 hour.At twelve hours after UVB radiation,an inverted microscope was used to observe cell morphology.Then,MTS assay was performed to estimate cell proliferation,flow cytometry to detect cell apoptosis,an enzyme-labeled antigen method to detect levels of malondialdehyde (MDA) and glutathione peroxidase (GSH-Px),as well as to evaluate the activity of superoxide dismutase (SOD) and catalase (CAT),and enzyme-linked immunosorbent assay (ELISA) to measure levels of intedeukin-1 (IL-1) and tumor necrosis factor-alpha (TNF-α) in HaCaT cells and their culture supernatant.Results Compared with the control group,the UVB group showed obscure cell morphology,cell death and floating phenomenon,while cells became swollen but renained morphologically distinct in the experimental group.MTS assay revealed that the cell proliferative activity significantly differed among the above 3 groups (F =48.88,P < 0.01),and the cell proliferative activity was significantly lower in the UVB group (1.72 ± 0.12) than in the control group (2.34 ± 0.11,P < 0.05) and experimental group (2.11 ± 0.10,P < 0.05).Moreover,the apoptosis rate was significantly higher in the UVB group (82.41% ± 2.49%) than in the control group (3.98% ± 0.19%,P < 0.05) and experimental group (22.79% ± 0.97%,P < 0.05),as well as higher in the experimental group than in the control group (P < 0.05).Compared with the control group,the UVB group showed significantly higher levels of MDA,supernatant levels of IL-1 and TNF-α,and intracellular levels of TNF-α,but significantly lower GSH-Px levels and activity of SOl and CAT (all P < 0.05).However,there was no signifi-cant difference in the intracellular level of TNF-α between the UVB group and control group (P > 0.05).Conclusion Lycium ruthenicum polysaccharide has protective effects against photodamage in HaCaT cells,likely by reducing the synthesis and secretion of inflammatory substances as well as free radicals.
3.Effects of vIL-10 on MHC-I antigen processing“the operon”in nasopharyngeal carcinoma cell lines

ABSTRACT:Objective To explore the effects of virus interleukin‐10 (vIL‐10 ) on different expressions of MHC‐I antigen processing “the operon” .Methods We collected nasopharyngeal carcinoma cells (CNE‐1 and CNE‐2) treated by vIL‐10 at different time points ,and detected the changes of MHC‐I antigen processing “the operons” (TAP‐1 ,TAP‐2 ,LMP‐2 ,LMP‐7 and HLA‐I) by RT‐PCR and Western blot .Results ① mRNA level :There was no difference in the expression of TAP‐1 in CNE‐1 and CNE‐2 cells at various time points .The expressions of TAP‐2 and LMP‐2 in CNE‐1 and CNE‐2 did not change at 1 ,4 ,6 ,12 h ,but downregulated and even disappeared at 24 h .The expression of LMP‐7 in CNE‐1 decreased 4 h after vIL‐10 was added ,and that in CNE‐2 decreased at 6 h .The expression of HLA‐I in CNE‐1 and CNE‐2 showed significant decrease at 24 h .② Protein expression :The expression of TAP‐1 in CNE‐1 and CNE‐2 showed significant decrease at 24 h .The expression of TAP‐2 in CNE‐1 and CNE‐2 was gradually downregulated at different time points .The expressions of LMP‐2 and LMP‐7 in CNE‐2 were gradually downregulated at different periods ,while that in CNE‐1 was only decreased at 12 h .The expression of HLA‐I in CNE‐1 and CNE‐2 was gradually downregulated ,but there was no significant difference at each period in CNE‐1 ,while the expression of HLA‐I in CNE‐2 at 24 h was significantly downregulated .Conclusion vIL‐10 can inhibit the expression of MHC‐I antigen processing “the operon” in NPC in the time‐dependent manner .
4.Diagnosis and treatment strategies on primary hyperparathyroidism.

OBJECTIVE:
Through analyzing the 13 cases with primary hyperparathyroidism (PHPT) retrospectively, We detected the reasons of the misdiagnosis and the strategy of diagnosis and treatment for the disease.
METHOD:
Thirteen cases of clinical materials on PHPT were collected from 2002 to 2012. Initial symptom, laboratory examination and imaging findings were analyzed. Thirteen cases of patients with PHPT all were performed surgery. The period of postoperative follow up was from 3 to 24 months, averaged for 12.2 months.
RESULT:
Surgeries on 13 patients were successful, including 12 cases of parathyroid adenoma and 1 case of parathyroid hyperplasia. After the operation, blood calcium went back to normal, and the symptoms of bone and urinary tract were relieved.
CONCLUSION
Initial symptoms of PHPT are predominantly manifested as bone and kidney types. Elevated PTH/blood calcium levels, B-ultrasound/CT and ECT localization are the important diagnosis basis of this disease. Surgical excision of the lesion parathyroid tissue is the most effective treatment.

5.Clinical study on cytokine induced killer cells therapy to laryngeal cancer after radiotherapy.

OBJECTIVE:
To observe the immunity changes of patients after CIK cells being transfused back, and then to discuss the effects of CIK cells on curing laryngeal cancers.
METHOD:
Forty eight laryngeal cancer patients with low immune function were collected. The immunity index in the peripheral blood of patients before/after radiotherapy and after CIK cells therapy were measured and compared with normal one.
RESULT:
After radiotherapy, the percentage of CD3+, CD4+ cells declined, the percentage of CD8+ cells increased; the rate of CD4+ /CD8+ declined and the rate of Th1/Th2 reversed. There were no significant difference between the immunity indexes before and after radiotherapy (P < 0.05). After CIK cell therapy, the above indexes were improved (P < 0.05), but the values didn't returned to normal. After radiotherapy and after CIK therapy, the value of B cell didn't changed obviously (P > 0.05), while the percentage of NK cells changed obviously (P < 0.05).
CONCLUSION
Radiotherapy can restrain the immune function of the patients with laryngeal cancers. CIK therapy is safe and might improve the recent immune function of the patients.

It is widely acknowledged that holistic processing is a key characteristic of face perception. Although holistic processing implies the automatic integration of face parts, it is unclear whether such processing requires the awareness of face parts. Here, we investigated the interactions between visible face parts and face parts rendered invisible using continuous flash suppression (CFS). In the first experiment with the upper half-face visible and the lower half-face invisible, the results showed that perceived face identity was influenced by the invisible lower half-face, suggesting that integration occurs between the visible and invisible face parts, a variant of the “composite face effect”. In the second experiment, we investigated the influence of visible face parts on the processing of invisible face parts, as measured by the time it took for the invisible parts to break out from CFS. The results showed a visible-to-invisible facilitation effect, that the aligned invisible face parts broke through CFS faster than when the visible and invisible face parts were misaligned. Visible eyes had a stronger influence on the invisible nose/mouth than the other way around. Such facilitation of processing from visible to invisible parts was also found when Chinese characters were used as stimuli. These results show that information integration occurs across the consciousness boundary.
