1.STUDY ON INTERFERON-? GENE-MODIFIED HUMAN TUMOR CELLS

The whole length cDNA of human interferon-?(IFN-?) gene including signal peptide was constructed to retroviral vector pLXSN and then packaged with PA317. A Human Hepetoma Cell Line(HHCL) and a human gastric cancer cell line (MKN-45) were infected by using supernatant containing retroviral vector with IFN-?. the gene-modified tumor cell. were isolated by G418 selection. Some changes were found in cell cycle, expression of HLA class Ⅰ and classⅡ, and tumorigenecity. Elevated immunogenecity and abrogated rumerigenecity suggest a means for generating gene-modified tumor cell vaccine. for the treatment of cancer.
2.Study progress of antiviral therapy in children with chronic hepatitis B

The antiviral treatment for children with chronic hepatitis B virus(HBV) infection should be started in the immune active phase.Interferon (IFN) is the drug of first choice in most cases,however,the individual treatment and management of adverse effects need to be considered.Currently,Nucleos (t)ide analogues (NAs) can be used as the first choice only for those special children who can't use IFN.Efficacy will be seriously affected once the emergence of viral resistance mutants to NAs.However,there is high risk of relapse after the NAs are discontinued.Compared with the mono therapy,the therapeutic strategy of interferon combined with NAs have obtained better effect and safety in children with chronic HBV infection.

4.Studies on the Apoptotic Cell Death of Interleukin-2 Gene Modified Mouse Hepatoma Cells

Retroviral vector was used to introduce the interleukin - 2 (IL -2) gene into H22 cells, a BALB/c mouse hepatoma cell line. The IL-2 gene and marker gene (NeoR) were assessed by using PCR and RT-PCR methods. FACS analysis demonstrated that the cells with low DNA content in IL-2 gene modified H22 cells were more than these in control H22 cells. Electron microscopic morphological structure showed that some cells in IL-2 gene modified H22 went into apoptotic cell death. The study demonstrated that apoptotic cell death of H22 - IL-2 cells was induced by the IL-2 gene modification. It may be one of mechanisms of decreased tumorigenicity of IL - 2 gene modified tumor cells.
5.Study on Anti-Tumor Effect of Vaccination with Irradiated Tumor Cells Engineered to Secrete TNF-?

We compared the ability of tumorigenicity of TNF-? gene-modified and unmodified H22 tumor cells, and therapeutic functions of the irradiated cells. Results indicated that H22-TNF-? cells were less tumorigenic as compared to H22 and H22-LXSN cells. In case of treatment groups, injected with irradiated tumor vaccine in 3 or 6 days after inoculation of H22 cells, the tumor growth was suppressed.
6.Antiviral therapy of chronic hepatitis B in children

For children with chronic hepatitis B (CHB), the antiviral therapy should be started in the immune active phase. There are two categories of antiviral drugs used for CHB: interferons (IFNs) and nucleos(t)ide analogues (NAs). In this paper, the current status of antiviral therapy of CHB in children is reviewed. It is pointed out that IFNs are the drug of first choice in antiviral therapy for children with CHB, and the individualized treatment and management of adverse events need to be considered. Some NAs can be given to those special children who cannot accept IFNs. However, there is a high risk of relapse after drug withdrawal. And the subsequent treatment will be seriously affected once the drug resistance is developed. It needs further investigation to improve the efficacy of antiviral therapy and reduce the adverse events and the drug resistance in the future.
7.Enhanced expression of HLA class Ⅰ molecules in human hepatocellular carcinoma cells transduced with γ-interferon gene

Objective To investigate the expression of exogenous γ-interferon gene in human hepatocellular carcinoma cells following retroviral transduction and the effect on the expression of surface HLA class Ⅰ molecules.Methods Retroviral vector pLXSN was used to introduce human γ-interferon (IFN-γ) gene into four different human hepatocellular carcinoma cell lines (HCC). The G418-resistant colonies were isolated and cloned. The integration and expression of IFN-γ gene were determined by PCR and RT-PCR analysis. A bioassay method was used to test the amount of IFN-γ secreted by gene modified HCC cells. The expression of HLA class Ⅰ molecules in HCC cells were analyzed by flow cytometry using indirect fluorescence staining.Results Four different HCC cell lines were successfully transduced with human IFN-γ gene using retroviral vector. The integration and expression of IFN-γ gene were shown only in the transduced cells. All four genetically modified HCC cells can secrete varied amount of IFN-γ and demonstrate a significant up-regulation of surface HLA class Ⅰ antigens. One specific HLA class Ⅰ antigen, HLA-A2, has almost the same degree of increase as that of the total HLA class Ⅰ molecules after transduction with IFN-γ gene.Conclusions Gene modification with IFN-γ gene can significantly enhance the expression of HLA class Ⅰ molecules in HCC cells and may increase its immunogenicity. These gene modified tumor vaccines can be helpful in tumor biotherapy.
8.Clinical application of clinical nursing pathway management in gynecologic patients with laparoscopic surgery

Objective To study the clinical application effectiveness of clinical nursing path-way management in laparoscopic surgery patients.Methods A total of 120 patients with laparo-scopic surgery were randomly divided into observation group and control group according to random number table method.The control group was used conventional nursing interventions while the ob-servation group received clinical nursing pathway management model based on routine care interven-tions on.After surgery,the preoperative waiting time,length of hospital stay,time to ambulation, anal exhaust time and feeding time were assessed,postoperative complications,satisfaction and mas-tery of health education of the two groups were anlyzed.Results The waiting time before surgery and hospital stay time in the observation group were significantly shorter than that in the control group,hospitalization costs in the observation group was significantly less than the control group, which showed a statistically significant difference(P <0.05).Ambulation time,anal exhaust time and feeding time were significantly shorter than that in the control group,the difference was statis-tically significant (P <0.05).The excellent rate of health education knowledge mastery of patients in the observation group was significantly higher,there was statistically significant difference (P <0.05).Conclusion The clinical application of clinical nursing pathway management is effective in gynecological patients with laparoscopy,so it is worthy of widely application.
9.The level of TNF-α secretion of PBMC in patients with chronic hepatitis C and Nonalcoholic fatty liver

Objective To study the roles of TNF-α secretion of PBMC in patients with chronic hepatitis C and Nonalcoholic fatty liver. Methods The level of TNF-α secretion of PBMC in patients with chronic hepatitis C and Nonalcoholic fatty liver was detected by ELISA after culturing for 72 hours in vitro, as well as patients with chronic hepatitis C and the normal control. Results (1) Compared with the normal control, the level of TNF-α in group with chronic hepatitis C and in group with chronic hepatitis C and Nonalcoholic fatty liver notably increased. (2) Compared with the group with chronic hepatitis C, the level of TNF-α in group with chronic hepatitis C and Nonalcoholic fatty liver also notably increased. Conclusion It suggested TNF-α takes important roles in the infection course of chronic hepatitis C with Nonalcoholic fatty liver.
