1.Detection of Interleukin 6 and Tumor Necrosis Factor alpha in Patients with Psoriasis Vulgaris

The activities of interleukin 6 (IL 6) and tumor necrosis factor alpha (TNF?) produced by peripheral blood mononuclear cells (PBMCs) in vitro and their relevant levels in sera were measured in 49 patients with psoriasis vulgaris (PV) and in 20 normal controls using bioassay. The relationship between them was also analysed. The results showed that in PV patients the activites of IL 6 and TNF? produced by PBMCs and their serum levels were higher than those in healthy controls. There was a positive linear correlation between the activites in vitro and relevant levels in sera, and a positive linear correlation between serum levels of IL 6 and TNF?. These results indicate that there is dysfunction of cellular immune of PBMCs in PV patients. The increase of serum levels of IL 6 and TNF? may be related to the increased activity of PBMCs, and there may be a synergic action between IL 6 and TNF? in the pathogenesis of psoriasis.
2.The Expression and clinical Significance of p53、p16 protein in advanced gastric carcinoma

Objective To investigate the expressions of p53,p16 protein and the clinical significance in advanced gastric carcinoma. Methods The expressions of p.53 ,p16 were detected by using immunohistochemistry in 24 specimens of normal gastric mucosa and 55 specimens of gastric carcinoma. Results The positive rate of p53 ,p16 in normal gastric mucosa and gastric carcinoma tissues were 0,100% ;67.3% ,30. 9%, respectively (P <0. 05). The positive rate of p53, p16 in non-differentiated、low- differentiated and moderate- differentiated 、high- differentiated gastric carcinoma tissues were 80. 0% ,56.7% and 48. 0% ,56. 0% ,respectively(P <0. 05). Conclusions The abnormal expressions of p53 and p16 play an important role in the course and prognosis of gastric carcinoma. The positive expression of p53 was correlated with the degree of defferentiation of gastric carcinoma. No correlation between the expression of p16 and grade of differentiation was observed.
3.Stem cell therapy for degenerative disc diseases:present status and prospects

BACKGROUND:None of the current treatment strategies has been focused on relieving or reversing the disk degeneration process after degenerative disc diseases. In recent years, more and more scientists try to treat degenerative disc diseases using stem cel therapy. OBJECTIVE:To explore the research status and prospects of stem cel therapy for degenerative disc diseases. METHODS: A computer-based online search of PubMed database between January 2004 and December 2014was performed to search related articles with the key words of “stem cel, intervertebral disk” in English. Literatures related to stem cel therapy for degenerative disc diseases were selected; in the same field, the articles published lately in authoritative journals were preferred. RESULTS AND CONCLUSION: A total of 342 articles were primarily selected, and 43 articles were involved in result analysis according to inclusion criteria. Stem cel therapy is a newly treatment for degenerative disc diseases. Cels appropriate for stem cel therapy include embryonic stem cels, induced pluripotent stem cels, mesenchymal stem cels, human umbilical cord mesenchymal stem cels and chondrocytes or nucleus pulposus cels. Although cel leakage, intervertebral disc infection and tumorigenesis are the main chalenges, stem cel therapy for degenerative disc diseases is promising in the future.
4.Lumbar disc degeneration:tissue engineering repair and biomechanical evaluation

BACKGROUND:At present, the clinical treatment of lumbar disc degeneration mainly includes conservative treatment, traditional surgery and minimal y invasive surgery. The therapeutic purpose is to relieve symptoms, but the long-term effect is not very satisfactory. Therapeutic methods focusing on biological functional recovery have been concerned gradual y, but the clinical application is far in sight. OBJECTIVE:To review the advances in the treatment of lumbar disc degeneration regarding tissue-engineered repair and biomechanics. METHODS:PubMed database was searched by the first author for relevant articles published before December 2014 using the keywords of“intervertebral disc degeneration, clinical treatment, biological treatment, tissue engineering, biomechanics, repair, progress”in English. A total of 100 articles were searched initial y and final y, 40 articles were included in result analysis. RESULTS AND CONCLUSION:Although the therapeutic schemes are varied, the treatment of intervertebral disc degeneration is a great chal enge for clinicians and basic researchers. Currently there is no perfect clinical treatment, and indications corresponding to various therapies should be paid attention as wel as long-term fol ow-up evaluation. For various reasons, the biological treatment for intervertebral disc degenerative disease is becoming more and more popular, providing a promising prospect for the treatment of intervertebral disc degeneration. So far, large amounts of data have been obtained from animal experiments, but there are stil many problems to be solved. Other chal enges also involve the al aspects of general tissue engineering methods, such as cel s, cytokines and scaffolds. In these studies, the nucleus pulposus tissue engineering based on the combination of heparin-functionalized chitosan hydrogel, cytokines and stem cel s exhibits a promising prospect.
5.Content Determination of AV45,a Labeled Precursor of PET Agent for AβPlaques,and Its Related Substanc-es by HPLC

OBJECTIVE:To establish a method for the content determination of AV45(the labeled precursor of PET agent for Aβ plaques)and its related substances. METHODS:HPLC method was performed on the column of Inertsil C18 with mobile phase of 50 mmol/L Disodium hydrogen phosphate solution- acetonitrile(containing 0.01% triethylamine)(15∶85,V/V)at a flow rate of 1.0 ml/min,the detection wavelength was 325 nm,and column temperature was 30 ℃,and volume injection was 20 μl. RESULTS:The AV45 and reaction intermediate as well as individual impurity peaks were well separated;the linear range was 200-1 000 μg/ml for AV45(r=0.999 2)and 20-100 μg/ml for intermediate(r=0.999 6);the detection limit and quantification limit of reaction in-termediate AV45 were 0.1 ng and 0.5 ng,respectively;RSDs of precision,stability and reproducibility tests were lower than 0.5%;recovery of AV45 was 98.93%-101.58%(RSD=0.69%,n=9). CONCLUSIONS:The method is simple and accurate with high specificity and good reproducibility,and suitable for content determination of AV45(the labeled precursor of PET agent for Aβplaques)and its related substances.
6.Expression of CDK5 and its correlation with peripheral inflammation-related cells and prognosis in colon cancer patients

Objective:To identify the clinical significance of CDK5 in colon cancer tissues.Methods:Two hundred colon cancer tissues were tested for CDK5 expression by immunohistochemistry on tissue microarrays. The correlation between CDK5 expression and clinicopathological features, prognosis and peripheral inflammation-related cells was analyzed.Results:CDK5 was low expressed in 100 cases (50.0%), and high in another 100 cases (50.0%). Longer time to tumor progression ( P=0.026) and overall survival ( P=0.035) were observed in patients with high CDK5 expression. By multivariate analysis , the expression of CDK5 was an independent risk factor for poor prognosis ( HR=0.45,95% CI: 0.21-0.99, P=0.049). The expression of CDK5 was not related to the counts of white blood cells and neutrophils ( P>0.05). Prognosis of patients with a positive lymph node ratio less than 0.15 was significantly better than that of patients with a higher lymph node ratio ( P<0.001). Conclusions:Patients with low CDK5 expression have poor prognosis, and CDK5 expression is not related to the counts of peripheral white blood cells and neutrophils.
7.The efficacy of postoperative radiotherapy in stage Ⅲ(N 2) non-small cell lung cancer: a meta analysis

Objective:To conduct meta analysis to compare the effect of complete resection with or without postoperative radiotherapy (PORT) on survival in stage Ⅲ(N 2) non-small cell lung cancer (NSCLC). Methods:Relevant studies of the efficacy of PORT for stage Ⅲ(N 2) NSCLC were searched from Wanfang Data, PubMed, and Cochrane Library from January 2006 to January 2022. Literature screening, extraction of information and assessment of the risk of bias of the included literature was carried out by two independent researchers. Meta analysis was performed using R4.0.3 software. Results:A total of 12 publications consisting of 2992 patients were included, 1479 cases in the PORT group and 1513 cases in the control group. PORT improved the overall survival (OS) and disease free survival (DFS) compared to the control group. Fixed-effects model meta analysis of 6 randomized controlled trials showed that PORT did not significantly reduce the risk of death ( HR=0.98, 95% CI: 0.80-1.20). Fixed-effects model meta analysis of 6 retrospective studies showed that PORT improved prognosis ( HR=0.68, 95% CI: 0.59-0.79). PORT could improve OS of patients with multiple (station) metastasis of ipsilateral mediastinum and / or submandibular lymph nodes ( HR=0.89, 95% CI: 0.80-0.99). Conclusions:PORT could improve OS and DFS in stage Ⅲ(N 2) NSCLC. A trend towards benefit can be observed in the subgroup with multiple/multi-station N2 metastasis.
8.Trend research on disease burden among the elderly in China based on GBD big data

Objective To understand the prevalence of diseases among the elderly in China and the main influencing factors on their ability to live independently in the community, so as to carry out comprehensive assessment and screening of the elderly in the community and achieve healthy aging. Methods The impact of diseases on the health of the elderly in China was quantified through indicators such as disability-adjusted life years, thereby assessing the changing trend of disease burden. Results The disease burden of chronic diseases and injuries is increasing year by year and has become the main disease burden for the elderly. Cardiovascular disease and cancer have the highest proportion of disability-adjusted life years year-round. Disability-adjusted life years for musculoskeletal diseases, traffic injuries, and accidental injuries increased significantly. The incidence of falls and road injuries in the elderly is increasing year by year. The prevalence of oral diseases has always been high and the disability-adjusted life years caused by them have been increasing year by year. At the same time, there are gender differences in prevalence rates and disability-adjusted life years among the elderly. Conclusion Comprehensive assessment of the elderly and timely intervention should be carried out extensively in the community, so as to detect potential problems and functional defects of the elderly early, reduce falls, disability, cognitive impairment, depression and other problems, and at the same time pay attention to the oral health of the elderly and improve their health. People's oral health care knowledge, publicize and popularize correct oral health care habits. Actively promote geriatric medicine education and encourage the whole society to participate in maximizing the maintenance of the functional status and quality of life of the elderly.
