Objective:To evaluate the validity and reliability of adapted simplified Chinese Version of the Scoliosis Research Society-22(SRS-22)questionnaire.Methods:The English version of the SRS-22 was translated into simplified Chinese and was subjected to cross-culture adaptation;the final version of the simplified Chinese SRS-22 questionnaire was a consensus reached by an expert committee. Then the simplified Chinese version SRS-22 questionnaire and 36-Item Short Form Health Survey(MOS SF-36)questionnaire were used to survey 87 patients with adolescent idiopathic scoliosis who had been surgically treated;63 patients(72.4%)responded to the SRS-22 questionnaire. The average age of these patients(6 male,57 female)was(17.7?3.1)years(range,14.3-23.8 years).Fifty-six of the 63 patients returned the MOS SF-36 questionnaire. The 2 indicators of reliability were Cronbach's ? of internal consistency and intraclass correlation coefficient(ICC).Concurrent validity of the SRS-22 questionnaire was measured by comparing with domains of the SF-36 questionnaire, which was reflected by Pearson correlation coefficient(r).Results:The Cronbach ? values for 4 domains(function/activity,pain,self-image/appearance,and mental health)were all higher than 0.7;and the Cronbach ? value for satisfaction of management domain was acceptable. The intraclass correlation coefficients for function/activity,pain,self-image/appearance,mental health, and satisfaction domain were 0.74,0.78,0.86,0.81 and 0.84,respectively,demonstrating a satisfactory reproducibility. For the concurrent validity,3 domains had excellent correlation,10 had good correlation, and 21 had moderate correlation. Conclusion:The adapted simplified Chinese Version of the SRS-22 questionnaire has satisfactory reliability and concurrent validity, and might be suitable for post-operation clinical evaluation of Chinese adolescent patients with scoliosis.

Objective To discuss the characteristics of knee injury in traffic accidents so as to provide an objective basis for its forensic identification. Methods Retrospective analysis was carried out on 201 cases of knee injury in traffic accidents identified by our department from 2006 to 2009.Results According to the types of knee injury, all cases were divided into bone injury group, soft tissue injury group and bone combined soft tissue injury group.Analysis of the knee joint function in the three groups revealed that there were significant differences between bone injury or bone combined soft tissues injury groups and soft tissue injury group in weight bearing including the normal bearing rate and nonbearing rate( P < 0.05,respectively). ConclusionsIf patients' weight loading, walking functions as well as their gait conditions are introduced into the process of forensic identification for the knee injury in traffic accidents, the identification results will be more reasonable.

To study the effect of genistein on the proliferation of cardiac fibroblasts(CF), CFs were cultured from neonatal rat hearts, DNA synthesis of the cells was determined by incorporation of [3H]TdR into DNA, the cell cycle was measured by flow cytometric analysis. Genistein(0.5-50 μmol*L-1) attenuated 2.5% fetal calf serum-induced proliferation of CF in concentration-dependent manner. Genistein(50 μmol*L-1) arrested CF cell progression at G2/M phase. The results suggest that genistein be a potential substance for treatment of cardiac fibrosis.

Objective:To screen differentially expressed proteins(DEPs)in the hippocampus of epileptic rats,pro-viding ideas for further exploring the pathogenesis and drug therapeutic targets of epilepsy. Methods:The epileptic rat model(PTZ group)induced by pentylenetetrazol(PTZ).The protein profiles of hippocampal tissues of the PTZ group and control groups were detected by isobaric tag for relative and absolute quantitation(iTRAQ)combined with LC-MS/MS technology, and the fold changes in the protein expression of the PTZ group versus the control group was considered to be more than 1.5 or less than 0.67,and P<0.05 was taken as the criteria for screening DEPs.Then DEPs were subjected to bioinformatics analyses such as Gene Ontology(GO)functional annotation and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analysis. Results:A total of 80 proteins showed significantly different expression, which included 39 up-regulated and 41 down-regulated.GO analysis showed that the up-regulated DEPs were mainly involved in biological processes such as cellular response to nerve growth factor stimulus, axon develop-ment,cell surface receptor signaling pathway, neuron migration, actin filament depolymerization, and signal transduc-tion,while down-regulated DEPs were mainly involved in biological processes such as tricarboxylic acid cycle, citrate metabolism process,acetyl-CoA biosynthetic process from pyruvate, oxaloacetate metabolism process, and regulation of focal adhesion assembly.KEGG pathway enrichment analysis showed that up-regulated DEPs were mainly involved in five signaling pathways, including AMPK signaling pathway, regulation of actin cytoskeleton, sphingolipid signaling pathway, phenylalanine metabolism, and insulin signaling pathway; The down-regulated DEPs were mainly involved in six signaling pathways, including the citric acid cycle, carbon metabolism, acetate and dicarboxylate metabolism,meta-bolic pathways, amino acid biosynthesis and amyotrophic lateral sclerosis signaling pathways. Conclusion:DEPs from epileptic hippocampal tissues were screened by iTRAQ proteomics, and the metabolic pathways enriched by bioinformat-ics methods for DEPs may be closely related to the pathogenesis of epilepsy.

The structure of N-glycans on specific proteins can regulate innate and adaptive immunity via sensing environmental signals. Meanwhile, the structural diversity of N-glycans poses analytical challenges that limit the exploration of specific glycosylation functions. In this work, we used THP-1-derived macrophages as examples to show the vast potential of a N-glycan structural interpretation tool StrucGP in N-glycoproteomic analysis. The intact glycopeptides of macrophages were enriched and analyzed using mass spectrometry (MS)-based glycoproteomic approaches, followed by the large-scale mapping of site-specific glycan structures via StrucGP. Results revealed that bisected GlcNAc, core fucosylated, and sialylated glycans (e.g., HexNAc4Hex5Fuc1Neu5Ac1, N4H5F1S1) were increased in M1 and M2 macrophages, especially in the latter. The findings indicated that these structures may be closely related to macrophage polarization. In addition, a high level of glycosylated PD-L1 was observed in M1 macrophages, and the LacNAc moiety was detected at Asn-192 and Asn-200 of PD-L1, and Asn-200 contained Lewis epitopes. The precision structural interpretation of site-specific glycans and subsequent intervention of target glycoproteins and related glycosyltransferases are of great value for the development of new diagnostic and therapeutic approaches for different diseases.

The emergence of regular short repetitive palindromic sequence clusters (CRISPR) and CRISPR- associated proteins 9 (Cas9) gene editing technology has greatly promoted the wide application of genetically modified pigs. Efficient single guide RNA (sgRNA) is the key to the success of gene editing using CRISPR/Cas9 technology. For large animals with a long reproductive cycle, such as pigs, it is necessary to screen out efficient sgRNA
