1.Effect of Wendan Tang adjuvant therapy on serum immune antibodies and efficacy in patients with phlegm dampness infertility

Objective To analyse effect of Wendan Tang adjuvant therapy on serum immune antibodies and efficacy in patients with phlegm dampness infertility.Methods 52 patients who were diagnosed with phlegm dampness infertility were collected.All patients were randomly divided into experimental group and control group, 26 cases in each group.Patients in control group received treatment with clomifene citrate capsules, patients in experimental group were given Wendan Tang treatment on the basis of control group treatment, after the treatment, the serum levels of Sp17Ab, AcrAb and treatment effect were detected in all patients.Results After treatment, compared with control group, the serum level of Sp17Ab was lower in experimental group(P<0.05); the serum level of AcrAb was lower in experimental group(P<0.05); the pregnancy rate was higher in experimental group(P<0.05).Conclusion The Wendan Tang adjuvant therapy can significantly reduce the serum levels of Sp17Ab and AcrAb in patients with phlegm dampness infertility,improve the pregnancy rate, improve the treatment effect.
2.Influence on magnetic resonance image of posteiror titanium cervical internal fixation system

[Objective]To discuss the influence of posterior cervical titanium internal fixation system on magnetic resonance image(MRI).[Method]Six cases that had operated in Changhai hospital with posterior cervical titanium internal fixation system performed MRI scan.The MRI system were 1.0-T or 1.5-T scanner.Patients all performed sagital T1,T2 and axial T1-weighted image scan under turbo spin echo(TSE)sequence,then the artifacts and its influence on the MRI image were analyzed.[Result]Mean artifact size ranged from 2 to 3 times of actual screw size.The artifact was similar to the primary shape of internal fixation system with low intensity signal on center and high intensity signal on the margin.The details in the spinal canal can be distinguished.The rod had small artifact,but the laminar hook had large artifact which obscured the posterior structure of spinal canal,and the artifact of pedicle screw had influence on lateral spinal canal,nerve root and vertebral artery.[Conclusion]Patients with posterior cervical titanium internal fixation system should choose Turbo spin echo(TSE)sequence for MRI scan,if the spin echo is performed,the TE should be minimized.
3.The vitro research of effects of Beclin1 on paclitaxel-sensitivity in laryngeal carcinoma cell Hep-2.

OBJECTIVE:
Background: We detect the effects of Beclinl on paclitaxel-sensitivity in laryngeal carcinoma cell.
METHOD:
This study used Hep-2, Hep-2-pcDNA3. 1, Hep-2-Beclinl as invitro model. The effect of paclitaxel on the proliferation and cell apoptosis of laryngeal cancer cell lines was evaluated by MTT assay and flow cytometry. The protein expression level of Akt and p-Akt was detected by Western blot. Result: After treated by paclitaxel, the inhibition rate was significantly higher in Hep-2-Beclin cells than in Hep-2-pcDNA3. 1 cells and Hep-2 cells (P<. 05). After dealing with 10 tg/L paclitaxel, the apoptosis rate in Hep-2, Hep-2-pcDNA3. 1, Hep-2-Beclinl were (23. 75 ± 2 3. 77) %, (21. 25 ± 4. 92) %, (32. 50 ± 5. 97) %, respectively. After dealing with 20µg/L paclitaxel, the apoptosis rate in Hep-2, Hep-2-pcDNA3. 1, Hep-2-Beclinl were (38. 75 ± 4. 79) %, (38. 75±6. 55) %, (50. 00±7. 26) %, respectively. Paclitaxel-induced apoptosis was higher in Hep-2-Beclin cells than in Hep-2-pcDNA3. 1 cells and Hep-2 cells (P<. 05). The result of western blot showed that the protein expression level of p-Akt in Hep-2-Beclin cells was lower than in Hep-2-pcDNA3. 1 cells and Hep-2 cells (P<0. 05) and the protein expression level of Akt was similar in three cell lines (P>0. 05).
CONCLUSION
Beclinl enhances paclitaxel-sensitivity by inhibition of PI3K/Akt pathway.

4.Knockdown of ClC-2 Gene Expression Inhibits the Growth of BT-325 Human Glioma Cells

ObjectiveTo observe the growth of BT-325 human glioma cells after interfering volume-regulated chloride channel ClC-2 gene.MethodsTwo expression recombinant vectors of ClC-2 gene were designed and constructed. The primary plasmid, pSUPER.puro-shRNA, and the two recombinant plasmids, pSUPER.puro-shRNA-ClC-21 and pSUPER.puro-shRNA-ClC-22, were transfected into BT-325 cells by LipofectamineTM2000 (Groups: control, PP1 and PP2, respectively). The mRNA expression of ClC-2 gene was detected with reverse transcription polymerasse chain reaction (RT-PCR), the cellular survival was determined with MTT assay, and the cell cycle was measured with flow cytometry (FCM). ResultsClC-2 mRNA expression and the growth of the cells in PP1 and PP2 groups were significantly lower than that of control group. The cell cycle progression was blocked in G1 phase (PP1 and PP2 vs control,P<0.01). ConclusionThe growth of BT-325 human glioma cells is prevented by knockdown of ClC-2 gene expression, which may be one of the novel targets to inhibit growth of human malignant glioma cells.
5.The study on clinical significance of the expression of EGFL7 in laryngeal squamous cell carcinoma.

OBJECTIVE:
To investigate the association and mechanism of EGFL7 expression level with the invasion and metastasis of laryngeal squamous cell carcinoma.
METHOD:
RT-PCR and Western blotting were used respectively to detect the level of EGFL7 mRNA and protein form 33 fresh laryngeal carcinoma tissues and matched para-neoplastic non-tumor tissues. The immunohistochemistry technique was performed to determine microvessel density (MVD) in 33 tumor tissues. The association of EGFL7 expression and MVD with clinicopathological characteristics was analyzed.
RESULT:
EGFL7 mRNA and protein expression were both significantly higher in the tumor tissues than in the paraneoplastic non-tumor tissues (P < 0.01). Furthermore, the expression of EGFL7 mRNA was highly correlated with the expression of EGFL7 protein (r = 0.786, P < 0.01). EGFL7 expression and MVD were highly correlated with clinical stage, tumor size and the presence of lymph node metastasis (P < 0.05), but was not correlated with the patients gender, age, tumor sit and tumor site differentiation (P > 0.05).
CONCLUSION
EGFL7 may have a close correlation with the development of laryngeal carcinoma via its impact on tubulogenesis and vessel shape. EGFL7 might serve as a tumor marker for assessing the progression of laryngeal carcinoma and a guide of clinical therapeutic decisions.

6.Phospho-proteomic analysis of antibacterial peptide merecidin act on human lung adenocarcinomaA549 cell line

Objective: To investigate the effects of antimicrobial peptides merecidin on the biological functions of human lung adenocarcinomaA549 cells and the potential signaling pathways and targets that involved in promoting apoptosis, by studying the changes of phosphorylation levels of proteins in A549 cells after merecidin treatment. Methods: The antibacterial peptide mericidin (9 μmol/L) was applied to treat A549 cells for 6 h, and the total protein was collected and extracted. The peptide was digested by trypsin and labeled with TMT, and then fractionated by HPLC. The phosphorylated peptides were enriched with IMAC-Fe, and finally subjected to mass spectrometry analysis. Library identification and quantification of phosphorylated peptides obtained by mass spectrometry were processed using MaxQuant software, to further analyze the functions and pathways of differentially expressed phosphorylated proteins by combining with bioinformatic analysis. Results: Through IPA analysis of phosphorylated proteins in the normal control group and the antibacterial peptide merecidin treatment group, 753 differentially phosphorylated proteins in mericidin treatment group were screened out under the conditions of |Fold Change|≥2 and P<0.05, including 229 significantly up-regulated genes and 417 down-regulated genes. Among them, the differentially expressed proteins related to apoptosis included RB1, MAPK1, ARAF, PTK2, FOXO, MARCKSandsoon.Theresultsofbiologicalprocessanalysisshowedthatdifferentiallyexpressed phosphorylated proteins were mainly concentrated in cell signal transduction, degradation and transport of nucleic acid, and cellular energy metabolism, protein translation and synthesis, and cytoskeleton formation etc. The enrichment results showed that the differentially expressed phosphorylated proteins were mainly involved in apoptosis-related MAPK, ErbB, PI3K-Akt, and Ras signaling pathways. Protein-protein interaction analysis indicated the associations among apoptosis-related proteins PTK2, PRKCA, MA2PK2, MAPK1, and LMNA. Conclusion: The antibacterial peptide merecidin may induce apoptosis and alteration of other cell functions by affecting a variety of genes and signaling pathways such as RB1, MAPK1,ARAF, PTK2, FOXO and MARCKS etc.
7.CD44 regulates epithelial-mesenchymal transition and metastasis in nasopharyngeal cancer cells.

OBJECTIVE:
To study the correlation of CD44 with epithelial-mesenchymal transition(EMT) and metastasis in nasopharyngeal cancer cells, and explore the possible mechanism of CD44 regulates EMT and metastasis in nasopharyngeal cancer cells.
METHOD:
The CD44 and EMT-associated proteins in 5-8F and 6-10B nasopharyngeal cancer cell lines were assayed by Western blotting. The erasion trace test was performed to observe the migratory ability of 5-8F and 6-10B nasopharyngeal cancer cells. Using lipid-mediated DNA transfection technique, the low metastatic nasopharyngeal cancer cells 6-10B were transfected in vitro with plasmid which contained CD44 gene, and then new nasopharyngeal cancer cells were obtained. The CD44 and EMT-associated proteins in 6-10B, empty vector transfected and CD44-transfected cells were assayed by Western blotting. The erasion trace test was performed to observe the alteration of migratory ability of nasopharyngeal cancer cells before and after CD44 transfection.
RESULT:
The expression of CD44 and EMT-associated protein MMP-9 in 5-8F was higher than that in 6-10B, but EMT-associated protein E-Cadherin in 5-8F was lower than that in 6-10B. The migratory ability of 5-8F was higher than that of 6-10B. The expression of CD44 and MMP-9 were significantly higher in the CD44-transfected nasopharyngeal cancer cells than in the control groups. Compared with control groups, the migratory ability of CD44-transfected nasopharyngeal cancer cells was significantly increased.
CONCLUSION
CD44 positively regulates the metastatic ability of nasopharyngeal cancer cells, which is relevant to the process of EMT.

8.Analysis of mutations in ARVC desmosomal protein gene in relatives of Yunnan unexplained sudden death cases in Nanjian County, Dali Prefecture

Objective:To analyze the mutations in desmosomal protein genes of arrhythmogenic right ventricular cardiomyopathy (ARVC) in relatives of Yunnan unexplained sudden death (YUSD) cases in Nanjian County, Dali Prefecture, Yunnan Province, and provide a basis for etiological hypothesis and control measures.Methods:The blood samples of YUSD case relatives ( n = 7) and control villagers ( n = 7) were collected, and basic situation investigation and electrocardiography (ECG) examination were performed at the same time. Blood DNA was extracted as a template for PCR amplification, and Sanger method was used to perform plakophilin 2 (PKP2), desmoglein 2 (DSG2), desmocollin 2 (DSC2), desmoplakin (DSP), and junction plakoglobin (JUP) five ARVC desmosomal protein genes sequencing of a total of 97 exons, and comprehensive analysis of gene mutations was carried out. Results:Five of YUSD case relatives carried genetic mutation sites, including DSP gene heterozygous synonymous mutations about exon 20 c.2862 C>T (p.Cys954Cys) and exon 24F c.7122 C>T (p.Thr2374Thr), DSC2 gene heterozygous missense mutation about exon 15 c.2326 A>G (p.Ile776Val), and all the five people were single heterozygous mutation carriers. Among them, two case relatives of the father-son carried the same site mutation of the DSC2 gene; the abnormal ECGs of three YUSD case relatives were ST-T change or clockwise rotation. However, the mutation sites of PKP2, DSG2, DSC2, DSP and JUP genes in control villagers were not detected.Conclusions:YUSD case relatives in Nanjian County carry ARVC desmosomal protein genes DSP and DSC2 mutations. Pathogenic mutation of DSC2 gene c.2326 A>G (p.Ile776Val) is may related to the incidence of some YUSD cases.
9. Effects of chronic intermittent hypoxia on blood pressure and vascular remodeling

Objective:
To investigate blood pressure and vascular remodeling of OSAS by establishing the chronic-intermittent hypoxia model in rat.
Methods:
Experiments were performed on 35 adult male Sprague-Dawley rats. Animals were randomly divided into four groups: unhandled control group (with 5 rats in it), CIH group at 9/6/3 weeks (with 10 ratsin each group). Rats in CIH group went through 8-hour intermittent hypoxia everyday, and those in control group were raising normally. After 9-week experiment, blood pressure was measured. The changes of the following indexes were observed: pathological changes of aorta and the middle aorta thickness (HE staining), the collagen of aorta wall (Masson staining). The experimental data were analyzed by SPSS 24.0 statistical software. The variance was analyzed by one-way analysis of variance, and the irregularity was selected using the calibration
10.Management and prognosis of differentiated thyroid carcinoma with tracheal invasion.

OBJECTIVETo evaluate treatment modalities and prognosis differentiated thyroid carcinoma (DTC) with tracheal invasion.
METHODSClinical data were reviewed and analyzed in 50 patients treated for DTC with tracheal invasion between January 1990 and June 1998. The different surgical modalities were applied according to the extent of tracheal invasion: shave excision (20 cases), tracheal sleeve resection or tracheal partial resection (23 cases), total laryngectomy or laryngeal closure surgery (7 cases). Thirty-eight cases received postoperative (131)I therapy. Survival rate was evaluated using the Kaplan-Meier analysis.
RESULTSThe 5-, 10- and 15-year survival rates of all the cases were 90.0%, 74.0% and 56.0%, respectively. The 5-, 10- and 15-year survival rates were 94.7%, 81.6% and 65.8% respectively in 38 cases with postoperative (131)I therapy and were 75.0%, 50.0% and 25.0% respectively in 12 cases without postoperative (131)I therapy, with statistically significant differences in 5-, 10- or 15-year survival rates between the patients of two groups.
CONCLUSIONSThe tumors can be resected radically by corresponding surgery based on the extent of tracheal invasion. Postoperative (131)I therapy can enhance the survival rate of the patients with differentiated thyroid carcinoma involving in trachea.
