Objective To investigate the effect of cyclin A antisense oligodeoxynucleotide (ASON) on mediating proliferation of K562 cell. Method After in vitro co-culture of cyclin A ASON with K562 cell, cyclin A protein expression levels were measured by flow cytometry. Cell growth was detected by trypan blue dye exclusion and colony-forming experiment. Results In cyclin A ASON group, cyclin A protein expression was significantly inhibited, compared to those in sense oligodeoxynucleotide (SON) group and blank group. Moreover, when the ASON concentration increased, the proliferation ratio of K562 cells and the CFU- K562 were significantly inhibited. Conclusion Cyclin A ASON can specifically inhibit cyclin A protein expression as well as inhibit the K562 cell proliferation and can lead to leukemic cell apoptosis. The effect of cyclin A ASON is concentration dependent.
3.Effect of laser induced fluorescence-electrial signal on guiding biopsy under endoscopy

Objective Laser induced fluorescence spectroscopy have been used in early diagnosis of GI cancer, but the device is expensive and inconvenient. The purpose of this study was to test the effectiveness and feasibility of a new device designed by the authors. Methods Two formulas were set up based on the analysis of OMA data and a new system was established. Fifteen samples of normal tissue and 40 cases of endoscopic diagnosed cancer were tested. Results (1) First formula: general energy of fluorescence was W 359~659 nm . (2) The second: the ratio of energy integral was W 359~460 nm /W 460~659 nm . (3) The sensitivity and specificity were 93.3% and 100.0% for 15 samples of gastro intestinal cancer in vitro and were 90.0% and 92.5% respectively for gastrointestinal cancer endoscopically. Conclusions (1) The deviec may help doctors to determine the characteristics of lesion in endoscopy and the efficacy of the instrument was the same as OMA system. (2) The cost was about one tenth of that for OMA system.
4.Clinical significance of urinary IL-8 in patients with lupus nephritis

Objective To evaluate the clinical value of detection of urinary interleukin-8(IL-8) in patients with lupus nephritis (LN).Methods Urinary levels of IL-8 were determined by using sandwich ELISA in 38 LN patients.Results In the LN patients during the stage of normal renal function and diminished renal reserve stage, urinary levels of IL-8 were significantly higher than those in healthy controls(P<0.001),and urinary levels of IL-8 in the patients during diminished renal reserve stage were significantly higher than those in the patients during normal renal function stage(P<0.01).High IL-8 levels in LN patients were associated with high levels of erythrocyte sedimentation rate and lower levels of C3 complement components. Urinary levels of IL-8 in LN patients with microscopic leukocyturia were significantly higher than those without microscopic leukocyturia. In LN patients treated with cyclophosphamide (CTX) and prednisone, urinary IL-8 levels of post-treatment were significantly lower than those of pre-treatment(P<0.01). Conclusions Urinary levels of IL-8 may be a useful indicator for monitoring disease activity and degree of renal damage in LN. The mechanism of noninfective leukocyturia may be associated with the elevation of urinary IL-8 levels in LN. CTX and prednisone may have a significant effect on inhibiting the production of IL-8 in the immunocytes and renal intrinsic cells.
5.Water-soluble constituents from Dioscorea zingiberensis

Objective To study water-soluble constituents of Dioscorea zingiberensis in order to seek active components. Methods Various chromatographic techniques were used to isolate the constituents. Structures of compounds were elucidated by spectroscopic analyses of 1H-NMR, 13C-NMR, 135DEPT, HMQC, HMBC, and TOCSY. Results One new steroidal saponin was isolated and identified as 26-O-(?-D-glucopyranosyl)-(25R)-furost-5-en-3?, 26-diol-22-OMe-3-O-{?-L-rhamnopyranosyl-(1→4)-[?-D-glucopyranosl-(1→3)-?-D-glucopyranosl-(1→2)]-?-D-glucopyranoside}. Conclusion The compound is a novel compound named as zingierenin E. The other two compounds are reported for the first time from D. zingiberensis.

AIM: AIM: To explore the relationship between apoptosis in the lung tissues and lung ischemia/reperfusion injury, and observe effects of panax notoginseng saponins (PNS) on apoptosis in lung ischemia/reperfusion injury. METHODS: Single lung in situ ischemia/reperfusion animal model was used. Eighty four Japanese white rabbits were randomly divided into control group (control), ischemia/reperfusion 1 h group (IR1h), IR3h, IR5h, Panax Notoginseng Saponins 1 h group (PNS1h), PNS3h and PNS5h. 7.Establishment of a mechanical experimental system of biologic tract tissue with auto-measurement based on measuring mark.

As the core of mechanical experimental system of biologic tract tissue, the automatic measuring program of serial images based on measuring mark could provide two basic parameters for mechanical analysis, namely tissue length and average external diameter. Hybrid programming between VC+ + and Matlab is conducted. Subprogram will be in charge of the processing of single image (color-gray transform, image segmentation, pick-up target area according to measuring mark), while the main program written by VC+ + will orderly call the above subprogram again and again when it is traversing through the image sequence which records the process of a tissue's expansion and constriction under force, so the automatic measure of the serial images and mechanical analysis is achieved. The results showed: the experimental system could avoid the contrived errors caused by naked eye identification and manual choosing measuring area; the measuring precision could satisfy the need of tract tissue mechanical analysis; the system could save time and energy dramatically. The fact of tract tissue accords with the applicable conditions of mechanical analysis theory used in our experimentations.

8.Effects of CANELIM capsule drug-contained serum on viscoelasticity and adhesive mechanical properties of cholangiocarcinoma cells.

By way of gastrogavage, we administered CANELIM capsules to rat for prepering the drug-contained serums. And then the serums obtained were used to plant cholangiocarcinoma cells. Lastly, using the micropipette aspiration technique, we investigated the effects which the drug-contained serums of different doses have on the viscoelasticity and adhesive mechanical properties of cholangiocarcinoma cells. The results showed that cholangiocarcinoma cells presented a characteristic of high elastic coefficient and low viscous coefficient. After being treated by the high dose and middle dose drug-contained serums, the viscoelastical properties of cholangiocarcinoma cells K1, K2 and micro evidently decreased (P < 0.01). But the properties of low dose did not evidently change. The adhesive force between cholangiocarcinoma cells and CD44v6 protein significantly reduced with the increasing of the dose of CANELIM capsules (P < 0.01). It is suggested that CANELIM capsules would destroy the cytoskeleton of cholangiocarcinoma cells, restrain the adhesion molecule CD44v6 on membrane from expressing, reduce the adhesion probability between cholangiocarcinoma cells and vasal endothelial cells, and finally, prevent the metastasis of cholangiocarcinoma cells.

9.In vitro induction, differentiation and seeding of source cells for the endothelialization of small-diameter artificial blood vessel of polyurethane

BACKGROUND: At present, after transplantation of small diameter artificial blood vessel, long-term patency rate is low due to being lacking of endothelial cells for lining and anti-thrombus characters. In some studies,mature endothelial cells were tried to be seeded in the artificial vessel to boost up its anti-thrombus capability so as to improve the long-term patency rate, but we got unsatisfied effect due to the defects of seed cells and scaffolds. Therefore, in clinic, proper seed cells and vascular scaffolds have been searched for improving the long-term low pateney rate in transplantation of small diameter artificial blood vessel.OBJECTIVE: To investigate the feasibility that differentiation of bone marrow mononuclear cells induced in vitro into endothelial-progenitor cells (EPCs) and seed polyurethane small diameter artificial blood vessel so as to provide proper seed cells for endotheliazation of polyurethane small diameter artificial blood vessel.DESIGN: Observation experiment SETTING: Cardivascular Medical Department and Staff Room of Immunology, First Hospital Affiliated to Sun Yat-sen University MATERILAS: This experiment was carried out at the First Hospital Affiliated to Sun Yat-sen University from September 2004 to May 2005. About 10 mL of bone marrow from healthy adult volunteers (n=7) was used in this experiment.METHODS: Bone marrow mononuclear cells of healthy adult were collected and put in the fibronectin pre-coated DMEM culture medium, then induced by vascular endothelial growth factor and basic fibroblast growth factor. Induced cells were observed under fluorescence microscope and identified with immunohistochemical staining. The induced and proliferated EPCs were seeded onto the surface of polyurethane small diameter artificial blood vessel. Morphological change was observed under scanning electron microscope.MAIN OUTCOME MEASURES: ① Cellular morphological change.② Staining results of immunohistochemical VWF and CD 34 antibody . ③ Adhesive growth status of EPCs on the polyurethane small diameter artificial blood vessel RESULTS: ① In the vascular endothelial growth factor and basic fibroblast growth factor and other inducers , bone marrow mononuclear cells differentiated into EPCs , presenting typical "spindle-shaped" appearance under an inverted fluorescence microscope and became to form a monolayer that arrayed in "cobblestone-like" ② Immunohistochemical staining showed von willebrand factor(VWF) and CD34 antigen stained positive. ③ Under the scanning electron microscope, surface of polyurethane small diameter artificial blood vessel without seeded cells presented typical polyporous honeycomb-like structure , and the size of hole suited the crawling of EPCs. After seeding the cells, we observed the adhesion, crawling and spreading of the EPCs on the surface of polyurethane small diameter artificial blood vessel. Some EPCs grew into the honeycomb-like holes were seen occasionally.CONCLUSION: Bone marrow mononuclear cells can be induced and differentiated into EPCs, while induced and differentiated EPCs well grow adhesively in the polyurethane small diameter artificial vessels, suggesting that differentiation of bone marrow mononuclear cells induced in vitro into EPCS, which can be used as seed cells for endothelialization of polyurethane small diameter artificial blood vessels.
10.Correction of prominent mandibular angle with aid of rapid prototyping technique

Objective To investigate the application of computer-aided 3D reconstruction and rapid prototyping(RP) technique in the correction of prominent mandibular angle.Methods Computer tomography scanning and 3D reconstruction were performed on 15 square face patients with prominent mandibular angles,then their actual mandible models were made by RP techniques.Surgical programs were made according to the model,including partial mandibular angle osteotomy,outer mandible table sagittal splitting osteotomy,chin augmentation with autogenous mandibule bones,and so on.In 15 cases,mandibular angle partial cutting was performed in 5 cases,sagittal splitting osteotomy in 6 cases,and mandibular angle partial cutting combined with splitting osteotomy in 4 cases.The autogenous mandibule bones were transplanted for chin augmentation in 3 chin microsomia patients.All the cases were treated according to the position and range set by the RP model.Results All the mandibular models produced by RP techniques were real and complete,which could directly and precisely show the state of the mandible.The operations completed smoothly and accomplished with the expected outcomes designed before operation.In all cases,the width of lower face was efficiently reduced and the face was symmetrical after operation.The follow-up period ranged from 3 months to 1 year in 12 patients,during which their facial appearances were in good condition and the results were satisfactory.Conclusion RP techniques is helpful in precise representation of the state of mandible,which providing ideal surgical models for accurate evaluation of prominent mandibular angle,design of surgical procedures as well as surgery instruction.It can provide good assistance to facial contour plastic surgery.
