Medical Journal of Chinese People's Liberation Army 1982;0(01):-
Immediate patency assessment of a completed microvascular anastomosis is considered an essential step in a microsurgical procedure. The most frequently used patency test is the milking patency test, which is too traumatic for use in clinical microsurgery. An atraumatic patency test remains to be devised. According to the principle of mechanics that rolling friction is smaller than sliding friction, we designed the so-called rolling patency test. The femoral arteries of Wistar rats were used in the experimental study to make a comparison between the two patency tests. The result showed that intact endothelial cells covered more than 90% of the inner wall of the artery in the rolling patency test, but less than 25% in the milking patency test. The study suggests that the rolling patency test is a reliable and atraumatic immediate patency test for use in clinical microsurgery.

Summary: The expression of CH50 polypoptide in bladder cancer cell line BIU-87 and the effects on the invasion ability of BIU-87 were investigated. The eukaryotic expressing vector pCH510 of polypeptide CH50 was introduced into BIU-87 cells by gene transfection in vitro. The expression of CH50 polypeptide was detected by using immunohistochemical S-P method. The expression of the transfected gene was identified by RT-PCR. Cell invasion assay kit was applied to detect the effect of CH50 polypeptide on the invasion ability of BIU-87. The results showed that the BIU-87 cells transfected with pCH510 could express the CH50 polypeptide, while in the control group, no CH50 polypeptide was detectable. In the transfection group, the invasion ability of BIU-87 in vitro was lower than in control group (P<0.05). It was concluded that CH50 polypeptide was successfully expressed in BIU-87 cells by gene transfection, by which the in vitro invasion ability of BIU-87 was inhibited.

In this study, the effects of pirrolidine dithiocarbamate (PDTC) plus leflunomide (Lef) models were investigated.NIH mice and Wistar rats served as donors and recipients respectively.Mouse-to-rat cardiac xenotransplantation was performed. The recipients were divided into 5 groups:group A (the control group),group B (PDTC group),group C (PDTC plus CsA group),group D (PDTC plus Lef group) and group E (PDTC plus Lef and CsA group). The expressions of IKKα/β,by immunohistochemistry and Western blot as well as electrophoretic mobility shift assay (EMSA).The median survival time of cardiac xenografts in the control group,PDTC group,PDTC plus CsA group,PDTC plus Lef group and PDTC plus Lef and CsA group was (2.17±0.41),(2.33±0.52),(4.67±1.21),(7.00±1.79) and (9.00±1.41) days respectively. The survival time of xenografts in the PDTC plus Lef and CsA group was significantly longer than that in other four groups (P<0.05 for all),that in the PDTC plus Lef group longer than that in the control group,PDTC group and PDTC plus CsA group (P<0.05 for all),that in PDTC plus CsA group longer than the control group and PDTC binding activity were notably increased in mouse-to-rat cardiac xenografts. The expressions were decreased in the control group,PDTC group,PDTC plus CsA group,PDTC plus Lef and PDTC plus Lef and CsA group in turn.It was concluded that PDTC plus Lef and CsA can significantly suppress prolonging the survival of the cardiac xenografts.

Objective To explore the effects of monocular deprivation(MD)on the expression of astrocytes in the superior colliculus, hippocampus, and visual cortex in mice during the critical period of visual development. Methods Eighteen C57BL/6J mice were randomly divided into the normal control group(CON group)and the MD group, with 9 mice in each group.Mice were bred under the 12 h/12 h dark/light conditions.Mice in the CON group received no treat-ment,while mice in the MD group underwent MD of the right eye on postnatal day 27,and the tissue was removed after 7 days. The mRNA and protein expression levels of glial fibrillary acidic protein(GFAP)in the superior colliculus,hippo-campus and visual cortex of mice in the two groups were detected using the real-time reverse transcription quantitative pol-ymerase chain reaction(RT-qPCR)and Western blot, respectively. The number of astrocytes labeled by GFAP and central nervous system specific protein β(S100β)in the superior colliculus, hippocampus and visual cortex of mice in the two groups was detected using the immunofluorescence staining. Results RT-qPCR and Western blot results showed that compared with the CON group, the mRNA and protein expression levels of GFAP in the superior colliculus,hippocampus(CA1,CA3 and dentate gyrus)and visual cortex of mice in the MD group decreased, and the differences were statistically significant(all P<0.05).The immunofluorescence staining results showed that compared with the CON group, the number of GFAP and S1OOβ co-labeled astrocytes in the superior colliculus,hippocampus(CA1,CA3 and dentate gyrus)and visual cortex of mice in the MD group decreased, and the differences were statistically significant(all P<0.05).Conclusion MD of mice during the critical period of visual development can result in a decrease in the number of astrocytes in the supe-rior colliculus, hippocampus and visual cortex.
