1.Application of Multi-slice Spiral CT Three Dimensional Reconstruction in Diagnosis of Mechanical Intestinal Obstruction

Objective To investigate the diagnostic Value of multi-slice spiral CT 3-Dimensional reconstruction in intestinal obstruction.Methods 28 Patients with malignant lesions of colon underwent volume scanning using multislice helical CT.Four types of reconstruction included multiple planner reconstruction(MPR),CT virtual colonoscopy(CTVC),shaded surface display(SSD)and raysum.The results were compared with those of colonoscopy and patholog.Results CT diagnostic accurate rate was 100%,and the accurate rate of the cause was 96% in intestinal obstruction,including primary or secondary neoplasms(18/28),abhesions(2/28),bowel torsion(2/28)and external or internal hernias(3/28) and gallstone ileus(3/28).Conclusion There were advantages and disadvantages in diagnosis of mechanical intestinal obstruction with MPR,CTVC,SSD and Raysum respectively.The combined use of the four post-processing technigues can be more helpful to precise localizing and qualitative diagnosis of lesions.MPR should be the optimal post-processing technigues for the diagnosis of intestinal obstruction
2.CT in Combination with MRI in Diagnosis and Differential Diagnosis of Chondrosarcoma

Objective To investigate CT in combination with MRI in diagnosing and differential diagnosing chondrosarcoma.Methods CT and MRI findings pathologically in 19 cases of chondrosarcoma proved were analyzed retrospectively.Results On CT,the tumors appeared as bony destruction in 15 cases,soft tissue masses in 14 cases,and with multiple calcifications in different shape.Postcontrast CT scan demonstrated peripheral and septal enhancement in 2 cases,and inhomogeneous enhancement in 3 cases.On MRI,the chondrosarcomas were hypointense or isointense on T1WI,heterogeneous hyperintense on T2WI.Postcontrast MRI demonstrated peripheral and septal enhancement with wreath-or honeycomb-like appearance in 9 cases,heterogeneous enhancement in 2 cases,obvious enhancement in 2 cases.On fat-suppression T2WI,the boundary and size of lesions were demonstrated clearly.The phylloid and cauliflower-like shape of tumors were displayed better on sagital and coronal position.Conclusion CT and MRI are both important in diagnosis and differetial diagnosis of chondrosarcoma,the combination of the two methods may improve the diagnosing accuracy of this tumor and minimize misdiagnosis.
3.Expression levels of serum visfatin and leptin and their clinical significance in patients with type 2 diabetes mellitus

Objective To investigate the expression levels of serum visfatin ,leptin and other indexes and their correlation and clinical significance in the patients with type 2 diabetes mellitus(T2DM ) .Methods Eighty-two age-matched and gender-matched cases of T2DM (T2DM group)and 71 cases of healthy subjects(health control group)were tested serum visfatin ,leptin ,insulin ,lipid and glycemia levels .Results The serum visfatin ,leptin ,triglycerides(TG) ,total cholesterol(TC) ,high-density lipoprotein choles-terol(HDL-C) and low density lipoprotein cholesterol(LDC-C) ,fasting plasma glucose ,fasting insulin(FINS) ,homeostasis model assessment of insulin resistance(OMA-IR) ,2 h FPG ,2 h FINS had statistically significant differences between the T 2DM group and the health control group(P< 0 .05);the stepwise regression analysis showed that visfatin were positively correlated with BMI , FINS ,HOMA-IR ,TC ,TG ,LDL-C (P<0 .05)and negatively correlated with HDL-C(P<0 .05);leptin were positively correlated with BMI ,FINS ,HOMA-IR(P<0 .05) .Conclusion Serum visfatin and leptin in the T2DM patients are hither than those in the healthy subjects and closely related with blood lipid ,blood glucose and insulin ,which may become the new diagnostic indexes of T2DM .
4.CT Value in Examining Intestinal Tumor Obstruction

Objective To analyze the CT features of intestinal tumor obstruction, and explore its CT value.Methods CT manifestation and clinical materials of intestinal tumor obstruction proved by surgical findings or endoscopy biopsy in 35 cases were analyzed restrospectively, and compared with the results of surgery-pathology and endoscopy. Results CT findings were consistence with the results of surgery-pathology and endoscopy in 33 of 35 cases. The CT diagnostic accuracy of intestinal obstruction was 100%. And the accuracy of the cause was 94%, including colon carcinoma in 25, lyphoma in 5, gastrointestinal stromal tumor in 2, and lipoma in 3.Conclusion CT has unique advantage in examining intestinal tumor obstruction, not only for definiting the existence of the obstruction, but also locating the site of obstruction diagnosing the cause and chosing the appropriate treatment.
5.Effects of physical and chemical factors in environment for dried blood spot preparation of neonatal screening assay

Objective To investigate the effects of physical and chemical factors in the environment for dried blood sample (DBS) preparation of neonatal screening assay.Methods A total of 60 normal and 120 positive DBS were prepared under control and 10 different conditions.Another 30 normal and 80 positive DBS were prepared under control and 7 different concentration gradients of formaldehyde.The levels of phenylalanine (Phe),glucose-6-phosphate dehydrogenease (G6PD),thyroid stimulating hormone (TSH) and 17α-hydoxyprogesterone (17α-OHP) were tested by time-resolved fluorescence immunoassay or fluorescence assay.Statistical analysis was performed using SPSS 22.0 software.Results Compared with the control group,the results of Phe were not significantly different (P > 0.05) when the samples were dried under the formaldehyde sensitive threshold (4.62 to 6.95 ppm for 18 hours).G6PD levels were significantly lowered when the samples were dried under all the conditions except for fast cold drying (2 to 8 ℃ overnight and formaldehyde condition,0.30 to 0.38 ppm for 4 hours or 0.21 to 0.24 ppm for 18 hours).TSH and 17α-OHP levels were lowered obviously when the samples were dried under the conditions of humidity,UV and formaldehyde condition (TSH:0.32 to 0.52 ppm for 4 hours,0.38 to 0.45 ppm for 18 hours,17α-OHP:4.37 to 4.62 ppm for 4 hours,0.38 to 0.45 ppm for 18 hours).The results of Phe,G6PD,TSH and 17α-OHP were not statistically different with the control group when the samples were dried under the fast cold drying and 2 to 8 ℃ overnight.Conclusion The physical and chemical factors in the environment of DBS preparation should be related to the accuracy of neonatal disease screening closely.The necessary control factors including formaldehyde,ethanol,glacial acetic acid,ultraviolet irradiation,heat,humidity and decoration pollution may exhibit significant effects on the preparation of DBS.Fast cold drying and overnight at 2 to 8 ℃ could be available for DBS preparation.
6.Effects of calcitonin gene related peptide on cultured human umbilical vein endothelial cells following anoxia and reoxygenation

AIM: To investigate the effects of calcitonin gene-related peptide (CGRP) on cultured anoxic-reoxygenation human umbilical vein endothelial cells. METHODS:In the present experiment, an anoxic-reoxygenation model was established by using cultured human umbilical vein endothelial cells. The uptake rate of trypan-blue and calcium and magnesium contents of endothelial cells and lactic dehydrogenase (LDH) activity in medium and malondialdhyde (MDA) content of endothelial cells were measured 60,120 and 180 min after anoxia and 30 and 60 min after reoxygenation, and the effects of 1?10 -8 mol/L CGRP on anoxic-reoxygenation endothelial cells were studied. RESULTS: The findings showed that, as anoxia prolonged, the uptake rate of trypan-blue and LDH activity and MDA content gradually elevated and, during reoxygenation, these parameters sharply increased. Calcium and magnesium levels gradually declined as anoxia prolonged, and during reoxygenation calcium content significantly increased. Meanwhile, 1?10 -8 mol/L CGRP might significantly reduce the uptake rate of trypan-blue and LDH activity and MDA content during anoxia and reoxygenation and lessen the increase in calcium content and the loss of magnesium during reoxygenation. CONCLUSION: These results showed that CGRP might have a direct protective function to endothelial cells afflicted with anoxic-reoxygenation injury by inhibiting lipid peroxidation, attenuating calcium overload and loss of magnesium and enzyme.
7.High performance liquid chromatography analysis of porcine platelet-derived gro wth factor and its effect on DNA synthesis of human vascular endothelial cells

Objective: To determine the molecular weight and p urity of porcine platelet-derived growth factor (pPDGF) and to investigate its effect on DNA synthesis of human umbilical vein endothelial cells. Metho ds: In the present experiment, the high performance liquid chromatograph y was used and the molecular weight and purity of pPDGF were studied. Human umbi lical vein endothelial cells was cultured and effects of pPDGF on DNA synthesis of endothelial cells was observed by 3H-TdR incorporation in vitro. Results: The findings of high performance liquid chromatography showed that the molecular weight of pPDGF was 29 120 and the purity was 89.46%, a nd pPDGF significantly promoted DNA synthesis of quiescent endothelial cells wit h a maximal response at a concentration of 40 ng/ml at 48 h. Conclusion: The molecular weight of pPDGF is 29 120, and it can promote DNA synthes is of cultured human umbilical vein endothelial cells.

Objective:
To observe the clinical efficacy and safety of the patients of myelodysplastic syndromes-refractory anemia with excess blasts (MDS-REAB) treated with decitabine alone or based on low dose cytarabine (Ara-C) regimen CAG/HAG [aclarubrci (ACR) /homoharring-tonine (HHT) +cytarabine+granulocyte colony stimulating factor (G-CSF) ].
Methods:
Totally 121 patients with MDS-REAB were retrospectively analyzed, including 59 patients treated with decitabine alone (20 mg·m-2·d-1 for 5 days) , the rest 62 ones treated with low-dose Ara-C-based regimen CAG/HAG. Overall response rate (ORR) , overall survival (OS) and adverse events of the two groups were analyzed and compared retrospectively.
Results:
The ORR of decitabine alone or CAG/HAG were 66.2% and 56.4% respectively, with no statistically significant differences (
10. Ruxolitinib combined with prednisone, thalidomide and danazol for treatment of myelofibrosis: a pilot study

Objective:
To evaluate the efficacy and tolerability of ruxolitinib combined with prednisone, thalidomide and danazol for treatment of in myelofibrosis (MF).
Methods:
Patients of MF according to the WHO 2016 criteria, received ruxolitinib (RUX) combined with prednisone, thalidomide and danazol (PTD). The response, changes of blood counts and adverse events were evaluated.
Results:
Six PMF and one post-ET MF patients were enrolled. Four patients presented JAK2V617F mutation, one CALR mutation, one MPL mutation, one triple-negative. Responses per IWG-MRT criteria were clinical improvement in 5 patients, stable disease in 2 ones, spleen response in 6 ones. All of 7 patients were symptomatic responses, four patients achieved at least 50% improvement from baseline on MPN-SAF TSS. Three patients initially treated with RUX alone, all of 3 patients experienced treatment-associated anemia and thrombocytopenia. Then these 3 patients received RUX combined with PTD, both hemoglobin and platelet increased significantly. Four patients initially treated with RUX combined with PTD. Increased levels of hemoglobin and platelet were seen in all of 7 patients received RUX combined with PTD with maximum increased hemoglobin of 30(18-54) g/L and maximum increased platelets of 116(13-369)×109/L, respectively from baseline. The treatment dose of RUX increased due to improved platelet count in 3 patients. The frequent non-hematologic adverse events grade 1-2 were constipation, abdominal distension, crura edema and increased ALT.
Conclusions
RUX combined with PTD for treatment of MF may modulate initial hematologic toxicity observed when RUX alone, and may increase response due to improved levels of hemoglobin or platelet.
