AIM　A series of 4-styrylcoumarin derivatives had been designed and synthesized in order to find compounds of antitumor activities by screening. METHODS　Title compounds (1-20) were synthesized by Phase-Transfer Wittig reaction or Wittig-Horner reaction, and screened by several antitumor models in vitro. Their structures were determined by 1HNMR, MS and elemental analysis. RESULTS　Twenty compounds (1-20) are new compounds. Compound 18 had effects on KB cell lines in vitro. CONCLUSION It was seen that compound 18 had certain antitumor activities, and it was worth further studying.

Objective To study the radiation effects on the serum concentration of urea nitrogen (BUN), creatinine (Cre), ?2-microglobulin (?2-MG) and uric acid (UA), and to evaluate the injuries to renal function induced by a 60 Co irradiation accident in Shandong province. Methods The concentrations of urea nitrogen, serum creatinine and serum uric acid were determined by enzymatic methods. The concentrations of serum ?2-microglobulin were determined by the immunoturbidity methods using HITACHI 7170s automatic biochemical analyzer. Results The serum concentrations of BUN and Cre of 2 patients increased significantly after irradiation, and the serum concentration of UA in the two cases elevated too after irradiation, but didn't reach the normal value. The serum concentrations of ?2-microglobulin also increased significantly after irradiation. In case A which received 20~25Gy irradiation, the concentrations of serum BUN, creatinine and ?2-microglobulin increased to 41.63mmol/L, 364?mol/L and 9.2mg/L, respectively. In case B which received 9~15Gy irradiation, the concentration of serum BUN, creatinine and ?2-microglobulin increased to 52.2mmol/L, 199?mol/L and 11.3mg/L, respectively. There seemed a dosage-dependent effect on the BUN, Cre and ?2-microglobulin concentration. Conclusion 60 Co may irradiate the tissues of kidney directly, and irradiate the cells of other tissues and organs, lead to necrosis and degenerations accordingly on the tissues of kidneys and other organs. In the process of necrosis and degenerations, a large amount of metabolic products was released which is poisonous to the kidney. The present results showed the concentrations increased of all the four renal serum markers mentioned above. It is supposed that the renal cells might be damaged by the Cobalt ? ray irradiation.

OBJECTIVE: To optimize the preparation technology of phloroglucinol phospholipids complex and study its physicochemical properties.METHODS: The preparation technology for phloroglucinol phospholipids complex was optimized taking the molar ratio of phloroglucinol to phospholipids(A),reaction temperature(B) and reaction time(C) as factors and complex ratio of phloroglucinol and phospholipid as index.The dissolubility and oil/water partition coefficient of the complex in different solvents were determined and compared with crude drug,and the new complex's physicochemical properties were validated by UV and IR(infrared spectra).RESULTS: The optimized preparation conditions for phloroglucinol phospholipids complex were obtained as follows: A was 1∶2;B was 60 ℃ and C was 2 h.The complex ratio remained at above 98%.Compared with phloroglucinol crude drug,the phloroglucinol phospholipids complex in water or chloroform showed increase in both dissolubility and oil/water partition coefficient.UV and IR confirmed that no new complex had been formed from phloroglucinol and phospholipids.CONCLUSION: The optimized technology greatly enhanced the dissolubility and lipotropy of the phloroglucinol phospholipids complex.

Nano-biosensors have elicited considerable scientific interest in various research areas, including nanotechnology, bio-technology, microelectronics, and analytical techniques. The development of nanomaterials and nanotechnology has produced nano-bio-sensors with increasing potential applications in disease diagnosis. Prostate cancer is a malignant tumor that seriously threatens the health human males worldwide. Detecting the low-abundance biomarkers of prostate cancer is critical for its early diagnosis, treatment monitoring, and evaluation of its postsurgical recurrence. This review focuses on the research progress in nano-biosensing technology and its application in the detection of prostate cancer biomarkers.

Objective: To analyze the role of frequency complemcntarities of vestibular tests including caloric test (CT),head shaking test(HST),and vibration test(VT) in the evaluation of vestibular function. Methods: Five hundreds and eightyfour patients with unilateral peripheral lesions were tested with CT, HST and VT in order to compare the frequency characteristic of abnormal vestibular function. Results: Of the 584 patients, 189 (32.36%), 283 (48.46%) and 368 (63.01%)cases showed vibration-induced nystagmus (VIN), head shaking nystagnms (HSN) and abnormal unilateral weakness (UW)respectively. There were 22 isolated VIN, 52 HSN and 145 cases of abnormal UW respectively. One hundred and fifty-nine (27.23%) cases had combination damage of two frequency bands, 101 (17.29%) had vestibular damage at all frequency bands,479 (82.02%) had abnormal results in any of the three tests, and 105 (17.98%) had no abnormality in all those three tests.Through consistency test, CT and HST (Kappa=0.106, P＜ 0.05), CT and VT(Kappa=0.068, P＜ 0.05), VT and HST (Kappa=0.321, P＜0.05) showed low consistency among them. VIN and HSN were more hkely to be evoked with the increasing of the UW (X'2VIN=22.686,X2HSN=23.023, P＜ 0.05). Conclusion: The vestibular damage in the patients with vertigo could reflect at isolated low, middle, high frequency or multi frequency bands. Thus, CT, HST and VT all make significant contributions to multiple-frequency analysis of vestibular function and show a well complementarities. So they can be used in evaluating the overall function of the vestibular and indicating a serious vestibular lesion if the damage affected multi frequency bands.

ObjectiveTo explore clinic features of large cerebral infarction and the relationship between infarcted area and prognosis.MethodsClinic materials such as dangerous factors, onset, course, treatment and prognosis, brain CT changes and internal carotid artery ultrasonic examinations of 68 patients with large cerebral infarction were analyzed retrospectively.ResultsThere were 28 cases caused by cerebral embolism, and 40 cases caused by cerebral thrombosis. 6 cases got recovery, and 34 cases, progress, and 11 cases,no progress, and 17 cases, death. ConclusionThe infarcted area was significantly related to prognosis (P<0.05). Internal carotid artery frequently occurring atheromas and senile non-valve atrial fibrillation were major embolic origins for large cerebral infarction.11 out of the 17 deaths could be attributed to brain herniation and secondary infection. The early abnormal brain CT showed that the rate of mortality was 62.5%, and the rate of mid-line dislocation was 42.9%. Either of them indicated that prognosis was bad.

Objective To observe the expression of hypoxia inducible faetor-1α (HIF-1α) in the retina of rabbits with acute high intraocular pressure and to investigate the mechanism of systemic domestic recombinant human erythropoietin (rhEPO) protecting the retina from ischemia-reperfusion injury. Methods First, control group and model group were established in rabbit eyes. The acute high intraocular pressure model was established by saline perfusion into anterior chamber, and then hypodermic injection of domestic rhEPO was made. HIF-1α protein in the retina was observed by immunohistochemical staining method on days 1, 3, 7 and 14 after retinal ischemla-reperfusion, respectively. Results No cells with HIF-la positive expression were observed in the retina of the control group. Ceils with HIF-1α positive expression in the model group outnumbered those in the control group (P < 0. 01). The resemblance pattern occurred in EPO group but its degree was slightly greater than that in the model group from day 3 after ischemia-reperfusion (P<0.05). Conclusion Domestic rhEPO can down-regulate the expression of HIF-1α in the retina with acute high intraocular pressure, which may be one of the mechanisms that rhEPO protects the retina from ischemia-reperfusion injury.

OBJECTIVETo establish a HPLC-ELSD fingerprint of Marsdenia tenacissima from different habitats, in order to provide a reliable method for scientific assessment and effective quality control.

METHODHPLC-ELSD was adopted to determine 25 baches of M. tenacissima herbs from different habitats. Traditional Chinese medicine chromatographic fingerprint similarity software assessment system 2004 developed by China Pharmacopoeia Committee was adopted to establish a common mode chart and assess chromatographic similarity based on the degree of correlation.

RESULTThe common mode for M. tenacissima herb C21 steroidal fingerprint was established, including 11 common characteristic peaks. Among them, 10 were identified. According to the assessment on the similarity of 25 batches of samples, 80% of them showed a similarity of over 0.80 in steroidal HPLC-ELSD fingerprint.

CONCLUSIONThe method can be used to assess the quality of M. tenacissima.

Many researchers employed mammalian expression system to artificially express cannabinoid receptors, but immunoblot data that directly prove efficient protein expression can hardly be seen in related research reports. In present study, we demonstrated cannabinoid receptor protein was not able to be properly expressed with routine mammalian expression system. This inefficient expression was rescued by endowing an exogenous signal peptide ahead of cannabinoid receptor peptide. In addition, the artificially synthesized cannabinoid receptor was found to aggregate under routine sample denaturing temperatures (i.e., [Symbol: see text]95°C), forming a large molecular weight band when analyzed by immuno-blotting. Only denaturing temperatures [Symbol: see text]75°C yielded a clear band at the predicted molecular weight. Collectively, we showed that efficient mammalian expression of cannabinoid receptors need a signal peptide sequence, and described the requirement for a low sample denaturing temperature in immuno-blot analysis. These findings provide very useful information for efficient mammalian expression and immuno-blotting of membrane receptors.

Objective To analyze the rule of lymph node metastasis in thoracic esophageal carcinoma,and to study the proper radiation target. Methods From September 1986 to December 1997,549 patients with esophageal carcinoma who had undergone radical reseetion were divided into surgery alone group (S,275 patients) or surgery plus radiotherapy group( S + R,274 patients). Radiotherapy was begun 3 to 4 weeks after operation. The radiation target included beth supra-clavicular areas and the entire mediastinum. The total dose was 50 Gy in 25 fractions over 5 weeks for the supra-clavicular areas and 60 Gy in 30 fractions over 6 weeks for the entire mediastinum. Results The 5-year overall survival of patients with lymph node metastasis in one anatomic site and two anatomic sites was 31.5% and 13.9% (P=0.013), respectively. For patients with > 2 positive nodes metastasis receiving surgery alone, the corresponding 5-year survival was 24.8% and 4.9% (P=0.046) ,respectively. The median number of dissected lymph nodes of the upper-,middle-and lower-segment esophageal carcinoma was 13,17 and 20, respectively. The rate of metastatic lymph node in the para-esophagus region was the highest(61.5%-64.9%) ,which was not different among the different primary sites(P=0.922). The anastomotic stoma recurrence rate of the upper-segment esophageal carcinoma was higher than that of the middle- or lower-segment carcinomas(16.7% ,3.1% ,and 7.7%, χ2=9.02,P<0.05). Conclusions For the thoracic esophageal carcinoma,the number of anatomic sites of lymph node metastasis is an important factor affecting the survival. The lower rate of lymph node metastasis of the upper segment esophageal carcinoma may be corrected with the less lymph node dissected. The rate of lymph node metastasis in para-esophageal region is not related with the lesion segment. The anastemotie stoma is an important radiotherapy target for upper segment esophageal carcinoma.
