1.Expression of NF-κB and its relation with epithelial-mesenchymal transition in pancreatic cancer

Objective To investigate the expression of NF-κB and epithelial-mesenchymal transition (EMT) related markers E-cadherin and Vimentin proteins in human pancreatic cancer tissues and its relation with the malignant features. Methods The expression of NF-κB、E-cadherin and Vimentin proteins in 62 cases of pancreatic cancer tissues were detected by using immunohistochemistry and compared with the clinicopathological data of pancreatic cancer. Results The positive expression rate of NF-κB was 81% (50/62), Vimentin protein increased of expression was 61% (38/62), and E-cadherin protein loss of expression was 55 % (34/62) in pancreatic cancer. The positive expression rate of NF-κB was significantly related with the lymph node metastasis (x2=11.761, P<0.05), distant metastasis (x2=9.225, P<0.05), the absent expression of epithelial marker E-cadherin protein (r =0.352, P <0.05) and the positive expression of mesenchymal marker Vimentin protein (r=0.343, P <0.05), but there was no relation with the patients gender,age, tumor location, tumor type and tumor differentiation (P >0.05). In addition, the significant correlation of E-cadherin expression loss and Vimentin expression with tumor lymph node metastasis and distant metastasis was found (x 2= 6.914, 4.984, 7.753, 5.144, P <0.05). Conclusion The overexpression of NF-κB in pancreatic cancer may accelerate invasion and metastasis of pancreatic cancer through inducing EMT.
2.Application and research progress in stem cells therapy for sensorineural deafness

BACKGROUND: The sensorineural deafness occurs as a result of loss of inner ear hair cells in the cochlea or of their primary afferent the spiral ganglion neurons. Stem cells to restore hearing following inner ear cell death has become a focus in recent years.OBJECTIVE: To summarize research progress in stem cells differentiating into inner ear cells in vitro and in vivo and to review the achievement in stem cells replacing inner ear cells in treating sensorineural deafness.METHODS: With "inner ear, stem cells" as key words, a computer-based online search of Pubmed and CNKI was performed for articles published from January 2000 to August 2009. RESULTS AND CONCLUSION: A total of 170 articles were collected, and experimental studies and review articles on stem cells in sensorineural deafness were included, while repetitive articles were excluded. Finally, 32 articles were summarized and analyzed. Different types of stem cells have the capacity to differentiate into inner ear cells. They can differentiate into neural cell types. Stem cells can live and migrate, differentiating into cell types of the sites of injury. It provides a therapy strategy to restore hearing following sensorineural deafness by he capacity of stem cells differentiating into inner ear cells. However, it remains further investigation how to function following cell differentiation and how to form the appropriate neural pathways by stem cell transplantation in sensorineural deafness.
3.Modified surgical treatment for Arnold-Chiari Ⅰ malformation complicated with syringomyelia

Objective To evaluate the methods and effects of modified surgical treatment for Amold-Chiari Ⅰ malformation complicated with syringomyelia.Methods Twelve patients with ArnoldChiari Ⅰ malformation complicated with syringomyelia,underwent a modified surgical treatment,effects were observed.Results During the first follow-up,sensation and motion function were significantly improved in 10 cases,unchanged in 2 cases.During the second follow-up,2 cases previously unchanged were improved,while the other were with no change.During the first follow-up,lesion showed on MRI disappeared in 5 cases,reduced in 4 cases,no significant change in 3 cases,no case extend,3 cases with hydrocephalus ventricle was significantly reduced.The second follow-up,MRI showed that there was no change compared with the first follow-up.Conclusion The remove of the pia mater of tonsillar hernia + reconstruction the cisterna magna have good effect on surgical therapies for Arnold-Chiari I malformation complicated with syringomyelia.
4.Establishment of a new model of the blast injury to the dog brain

Objective To reproduce an experimental model of blast injury to dog brain simulating the mechanism of primary effect of the blast wave of explosive weapons.Methods A spherical explosive with explosive energy of 1gTNT was used to detonate at different distances(9mm,13mm)from the right temple to produce blast injury to the dog brain.The changes in physiological index before or after detonating and the surviving time after injury were recorded.The changes in the behavior of the wounded animals and characteristics of pathological changes in the brain were observed.Results Respiratory arrest,hypotension,and decreased heart rate were observed after wounding.All animals survived longer than 6 hours after the explosion injury.Obvious subdural hematoma and sub-arachnoid hemorrhage occurred frequently.Traumatic conditions of the brain were different in the two groups.Conclusions The animal model reproduced in this experiment simulates that produced by blast wave of explosive weapons.It is reproducible,easy to handle,and can be used in experimental study of blast injury to the brain.
5.Study on the Antihepatocarcinoma Activity and the Acute Toxicity of Aclacimomycin-A Solid Lipids Nanoparticle for Injection in Mice in Vivo

OBJECTIVE:To study the antihepatocarcinoma activity and toxicity of Aclacimomycin-A solid lipids nanoparticle for injection(ACM-SLN)in nude mice in vivo with ACM for injection as the reference preparation.METHODS:The nude mice were divided into control group,ACM group and ACM-SLN group after tumor transplantation,which were injected with the corresponding medicine before the tumor-inhibition rates of which were calculated,which were then injected with ACM and ACM-SLN,respectively before the LD50 of mice were calculated.RESULTS:Compared with the control group,the tumor inhibition rates of ACM-SLN group and ACM group were 78.4%and 38.8%,respectively,the LD50 of which were 16.3 mg/kg and 18.0mg/kg,respectively.CONCLUSION:The ACM-SLN is superior to ACM in terms of the anti-tumor effect while without the increase of toxicity.

7.Comparison of Clinical Outcomes Between Two Methods of Posterior Lumbar Interbody Fustion in Adult Spondylolisthesis

Objective To compare the clinical outcomes of posterior lumbar interbody fusion(PLIF) using simple cage alone fusion with pedicle screw fixation and autogenous bone grafting and cage fusion with pedicle screw fixation in adult spondylolisthesis. Methods From March 2003 to March 2004,Twenty-seven patients with lumbar spondylolisthesis were divided in two groups. In group A, 15 patients were treated by PLIF using simple cage alone fusion with pedicle screw fixation, including 4 males and 11 females, aging 53-68 years. Isthmic defects were located at L4 in 9 cases, at L5 in 6 cases. Four patients were smokers. The preoperative mean disc space height was 5.4±2.3 mm, the mean percentage of slip was 36.8%±7.2%. In group B, 12 patients were treated by PLIF using autogenous bone grafting and cage fusion with pedicle screw fixation, including 3 males and 9 females, aging 56 years. Isthmic defects were located at L4 in 8 cases, at L5 in 4 cases. Five patients were smokers. The preoperative mean disc space height was 5.7±2.5 mm, the mean percentage of slip was 37.8%±6.2%. Two groups were compared in the amount of blood loss, duration of hospitalization, back pain, radiating pain, fusion rate, the intervertebral disc space height, the postoperative degree of slip and the fusion rate. Results All patients were followed up for 24-38 months. The mean follow-up was 29(24-36) months in group A and 26(24-38) months in group B. There were no statistically significant differences in follow-up period, age,sex, the location of isthmic defects, smoking, the preoperative disc space height and the percentage of slip between two groups (P>0.05). There were no statistically significant differences in the amount of blood loss, the duration of hospitalization, the fusion time between two groups(P>0.05). But there were statistically significant differences in the back pain score, the radiating pain score and the fusion rate between two groups(P<0.05).The postoperative disc space height and the degree of slip of the last follow-up were 5.8±2.2 mm and 25.6%±7.2% in group A, 6.2±2.5 mm and 24.1%±7.4 % in group B, showing statistically significant difference (P<0.05). Conclusion The PLIF using autogenous bone grafting and cage fusion with pedicle screw fixations is more beneficial to improving the fusion rate and preventing long-term instabilities than simple cage alone fusion with pedicle screw fixation in adult spondylolisthesis.
8.Comparison between auripuncture or tympanostomy tubes combined with adenoidectomy in children with otitis media with effusion

OBJECTIVE To compare the clinical efficacy between adenoidectomy combined with auripuncture (AT+A) and adenoidectomy combined with tympanostomy tubes (AT+T) in children wiht otitis media with effusion. METHODS 92 cases of children with otitis media with effusion were selected from September 2010 to September 2014 in our hospital which was divided into AT+T group and AT+A group, depending on therapy methods.There were 48 in AT+T group and 44 in AT+A group. Retrospective analysis were made on a series of clinical data ,including the therapeutic effect, hearing threshold before and after treatment, infection rate and recurrence rate. RESULTS The hearing threshold has been significantly reduced within one year of follow-up after treatment,however,there was no significant difference (P>0.05). Total efficiency rate was 97.9% in AT+T group and 95.4% in AT+A group, no significant difference was found (P>0.05). The average middle ear fluid duration, infection and recurrence rate of children in AT+T group and AT+A group was (7.3±0.8) days, 6.3%, 4.2% and (11.7±0.4) days, 15.9%, 11.4% respectively, there was significant differences (P<0.05). CONCLUSION Both AT+T and AT +A have a good therapeutic effect for children with otitis media with effusion, which can significantly improve the hearing. Compared to AT+A, AT+T have a higher recovery rate,significantly reduce the recurrence and infection rate,and may be more suitable for clinical treatment of children with otitis media with effusion.
9.Effects of inhibiting Kit signal pathway on proliferation and spontaneous rhythmic motility of interstitial cells in bladder of guinea pigs

Objective To investigate the role of Kit signal pathway in the proliferation and spontaneous rhythmic motility of interstitial cells(ICs)in the bladder of guinea pigs.Methods Fourteen Guinea pigs aged 45-60 days were administered with Imatinib intragastrically for 20 and 40 days.Three were fed on normal saline as control.The morph and number of ICs were observed by immunofluorescence,Kit protein expression was detected by Western blot and spontaneous rhythmic motility of bladder were investigated in in vitro organ bath.Results Quite a few ICs were observed in the bladder of controls.After administration of Imatinib,the amount of ICs and the expression of Kit protein decreased gradually,and the spontaneous rhythmic contraction weakened.Conclusion ICs might be the pacemaker of bladder in guinea pigs.Kit protein might play an important role in the survival and functional maintenance of ICs.
10.Determination of the Concentration of Valproic Acid in Serum With RP-HPLC

OBJECTIVE:To determine the concentration of valproic acid in serum.METHODS:Determination was performed with RP-HPLC with methanol:water(70∶30) as mobile phase,?-bromoacetophenone as deriving agent and cyclohexanecarboxylic acid as internal standard,and detected at wavelength 248nm.RESULTS:The calibrating curve of valproic acid was linear in the range of 14.47~248.0?g/ml.CONCLUSION:The method was convenient,rapid,accurate and suitable for TDM.
