1.Down-regulated TCF3 expression inhibits growth and migratory abilities of non-small cell lung cancer cells

AIM:To investigate the effects of T-cell factor 3 (TCF3) expression on the growth and migratory abilities of the non-small cell lung cancer cells ( NSCLC) and the underlying mechanisms .METHODS: The non-small cell lung cancer cells A549 and H1299 were transfected with siTCF3 or negative control siRNA ( NCsiRNA) using Lipo-fectamine 2000.The expression of TCF3 at mRNA and protein levels was detected by real-time PCR and Western blot , re-spectively.The activity of TCF3 transcription was measured using luciferase reporter gene assay .The cell viability, colony formation ability, migratory ability and apoptotic rate were monitored by MTT assay , colony formation assay, Transwell method and flow cytometry with Annexin V-FITC/PI double staining , respectively .The protein expression levels of Wnt , c-Myc, matrix metalloproteinase (MMP)-9, MMP-13 and tissue inhibitor of metalloproteinase-1 (TIMP-1) were detected by Western blot.RESULTS:Compared with A549-NCsiRNA and H1299-NCsiRNA cells, the mRNA and protein levels of TCF3 were significantly inhibited in A549-siTCF3 and H1299-siTCF3 cells (P<0.01).The activity of TCF3 transcription and levels of c-Myc protein expression remarkably decreased as compared with NCsiRNA cells (P<0.05).The viability of A549-siTCF3 cells and H1299-siTCF3 cells cultured for 24 h, 48 h, 72 h and 96 h were notably lower than that of NCsi-RNA cells (P<0.05).The colony formation ability was significantly inhibited by siTCF 3 compared with NCsiRNA cells (P<0.01).The numbers of migratory A549-siTCF3 and H1299-siTCF3 cells were significantly lower than those of A549-NCsiRNA and H1299-NCsiRNA cells (P<0.05).The apoptotic rates of A549-siTCF3 cells and H1299-siTCF3 cells were notably higher than those of A549-NCsiRNA cells and H1299-NCsiRNA cells (P<0.01).Down-regulated TCF3 expres-sion inhibited the protein expression of Wnt , decreased the protein levels of MMP-9 and MMP-13 and enhanced the protein level of TCF3.CONCLUSION:siTCF3 suppresses the abilities of growth and migration , and induces apoptosis in A 549 cells and H1299 cells by down-regulating Wnt pathway and regulating expression of key MMP family genes .

At present,the combined interventional therapy that is based on transcatheter arterial chemoembolization (TACE) technique can control the progression of primary hepatocellular carcinoma (PHC)in different degrees,at the same time the clinical objectives,such as relieving portal hypertension,eliminating portal vein occlusion,controlling refractory ascites and lowering upper gastrointestinal bleeding probability,can be reliably achieved.Thus,the quality of life of the patients can be effectively improved and the survival time will be reliably prolonged.This paper aims to make a comprehensive review about the medical articles concerning TACE combined with other interventional therapies for PHC complicated by portal vein tumor thrombus,which have been published both at home and abroad in recent years,in order to provide practical help for clinical diagnosis and treatment of PHC.
3.Homeopathic closed leverage anatomical plate with compression bolt for treatment of displaced intra-articular calcaneal fractures

Objective To compare the treatment of displaced intra-articular calcaneal fractures by homeopathic closed leverage anatomical plate with compression bolt through small posterior lateral approach vs.traditional open reduction and internal fixation.Methods A retrospective case control study was made on 98 cases of displaced intra-articular calcaneal fractures admitted from September 2012 to May 2015.According to the random number table,the subjects were assigned to homeopathic closed leverage anatomical plating with compression bolt through small posterior lateral approach (experiment group,58 cases,66 sides) and open reduction and internal fixation through L-shape approach (control group,40 cases,45 sides).Experiment group consisted of 50 male and eight females cases aging from 27-56 years (mean,41.9 years),and the Sanders classification was 40 cases of type Ⅱ,24 type Ⅲ and two type Ⅳ.Control group consisted of 36 male and four female cases aging from 25-58 years (mean,43.7 years),and the Sanders classification was 25 cases of type Ⅱ,18 type Ⅱ and two type Ⅳ.Operation time,bone reduction,postoperative Bohler's angle,width of the calcaneum,and incision healing were recorded.Functional outcomes were evaluated with Maryland hindfoot scoring system at last follow-up.Results Operation time was (52.6 ± 11.2) min in experiment group,significantly shorter than that in control group [(86.4 ± 14.1) min] (P < 0.01).All cases were followed up from 18-50 months (mean,30.8 months).Reduction of the calcaneal posterior facet in 53 sides (80%) was graded as nearly anatomical in experiment group,and 38 sides (84%) in control group (P > 0.05).Postoperative Bohler's angle was (28.0 ± 6.2) ° in experiment group,and (26.8 ± 7.0) ° in control group (P > 0.05).Width of the calcaneum was (31.3 ±3.6)mm in experiment group and (34.9 ± 4.0)mm in control group (P < 0.01).All cases presented satisfactory shape of the calcaneus without lateral-side impact syndrome.No case had wound infection and incision-edge necrosis in experimental group,while two cases of superficial wound infection and three cases of incision-edge necrosis were found in control group (P < 0.01).At last follow-up,Maryland hindfoot score was (87.1 ± 7.6)points in experiment group and (84.9 ± 9.1)points in control group (P > 0.05).Conclusion Homeopathic percutaneous leverage and anatomical plate with compression bolt through small posterior lateral approach is an effective method for treatment of displaced intra-articular calcaneal fractures,for it has advantages of minimal invasion,less operation time,good reduction and function,and less wound complications.

High-risk human papillomavirus infection is the major cause of tumors such as cervical cancer and head and neck cancer.MicroRNAs (miRNAs) are 20-25nt small non-coding RNAs,which are involved in the proliferation,apoptosis,invasion and migration of tumors.HR-HPVs,which integrate into the host genome,encode three oncogenes:E6,E7 and E5 and enhance carcinogenesis by regulating downstream miRNAs and genes e.g.p53 and pRb.
5.Deficiency of DNA double-strand break repair and enhanced radiosensitivity in Tip60 silenced cells

Objective To investigate the effect of Tip60 on the cellular radiosensitivity,and to explore the related mechanism.Methods siRNA and anacardic acid (AA,an inhibitor of Tip60 acetyltransferase) were used to inhibit Tip60 expression and its acetyltransferase activity,respectively.Radiosensitivity was analyzed by colony-forming ability assay.γ-H2AX foci were detected to analyze the DNA double-strand break (DSB).Immunoprecipitation was used to determine the interaction of proteins.Results siRNA-mediated silencing of Tip60 led to enhanced sensitivity of U2OS cells at 1,2 Gy after γ-ray irradiation,but had no significant effect at 4 Gy post-irradiation ( t =3.364,3.979,P < 0.05 ).γ-H2AX foci detection indicated that Tip60 silencing resulted in a decreased capability of DNA doublestrand break repair at 1,4 and 8 h after irradiation( t =3.875,3.183 and 3.175,respectively,P < 0.05 ).The interaction of Tip60 and DNA-PKcs was prompted by ionizing radiation.Anacardic acid largely abrogated the phosphorylation of DNA-PKcs at T2609 site induced by irradiation.Conclusions Tip60plays a role in the cellular response to ionizing radiation-induced DNA damage through,at least in part,interacting with DNA-PKcs and regulating its phosphorylation.
6.The clinical characteristics and molecular genetics of infantile neuroaxonal dystrophy: The current research progress

Infantile neuroaxonal dystrophy (INAD) is a rare autosome-recessive disease characterized by progressive motor and cognitive regression.The PLA2G6 gene is its causative gene,which encodes calcium-independent phospholipase A2 enzyme (iPLA2-VIA).The diagnosis of INAD is difficult because of its clinical heterogeneity,and the rate of misdiagnosis is high.The purpose of this study is to describe the clinical characteristics,molecular genetics,treatment and prognosis of INAD to improve the acknowledgement of INAD in medical workers and to help make an early diagnosis of INAD.
7.Application of spectral CT in the differential diagnosis of benign and malignant mediastinal lymph nodes

Objective To compare the differences of the spectral CT parameters between benign and malignant mediastina lymph nodes and to investigate the value of spectral imaging in the differential diagnosis. Methods According to the follow-up and pathology results, a total of 73 patients were enrolled in this study. One experienced radiologist determined the locations of lymph nodes on CT images corresponding to the surgical pathological ones. A total of 47 malignant nodes and 65 benign nodes were found. The longest and shortest diameters of lymph nodes in mediastina window were measured .The vHU, IC, WC, Zef and the CT values of normal lung, normal muscle and lymph nodes at 40, 70 and 140 keV were measured. The indices were compared between the benign nodes and malignant nodes by independent-samples t test. The Spectral HU curves were drawn. Results The long and short diameter of malignant lymph nodes were (28.8 ± 10.5),(20.3 ± 9.1)cm , while they were were (14.0 ± 4.6),(7.5 ± 1.8)cm for benign lymph nodes. Malignant nodes were significantly larger than benign nodes (t= 6.54, 7.13, respectively, P<0.05), The differences of the CT values at 40, 70 and 140 keV and the vHU, IC, WC, Zef for the normal lung, normal muscle and lymph nodes between the two groups had no statistical significance (P>0.05). The two groups showed similar spectral HU curve slopes (1.86 values 1.73). Conclusion Spectral imaging may not be helpful in the differential diagnosis of malignant and benign lymph nodes.
8.Protection of vanillin derivative VND3207 on plasmid DNA damage induced by different LET ionizing radiation

Objective To evaluate the radioprotective effect of vanillin derivative VND3207 on DNA damage induced by different LET ionizing radiation.Methods The plasmid DNA in liquid was irradiated by 60Co γ-rays, proton or 7Li heavy ion with or without VND3207.The conformation changes of plasmid DNA were assessed by agarose gel electrophoresis and the quantification was done using gel imaging system.Results The DNA damage induced by proton and 7Li heavy ion was much more serious as compared with that by 60Co γ-rays, and the vanillin derivative VND3207 could efficiently decrease the DNA damage induced by all three types of irradiation sources, which was expressed as a significantly reduced ratio of open circular form (OC) of plasmid DNA.The radioprotective effect of VND3207 increased with the increasing of drug concentration.The protective efficiencies of 200 μmol/L VND3207 were 85.3% (t =3.70,P =0.033), 73.3% (t = 10.58, P =0.017)and 80.4% (t =8.57,P =0.008)on DNA damage induction by 50 Gy of γ-rays, proton and 7Li heavy ion, respectively.It seemed that the radioprotection of VND3207 was more effective on DNA damage induced by high LET heavy ion than that by proton.Conclusions VND3207 has a protective effect against the genotoxicity of different LET ionizing radiation, especially for γ-rays and 7 Li heavy ion.
9. The research progress of skin anti-aging effects of adipose derived stem cells

Adipose-derived stem cells (ADSCs) are adult mesenchymal stem cells, which are derived from adipose tissue. ADSCs have broad applications, due to the potential of multiple differentiation, easy sampling, wide sources and high proliferation capacity in vitro. Many researches demonstrated that ADSCs have significant effects on anti-aging. Its mechanism and therapeutic effects also become a hot topic in recent years. This review summarized the biological characteristics of ADSCs, the mechanism of skin aging, the anti-aging mechanism of ADSCs, so as to provide reference for the use of ADSCs for skin anti-aging in the future.
10.Developing turns-amplitude clouds for healthy Chinese: reference values and influence factors

Objective To establish the reference values and evaluate influence factors of turns-amplitude clouds for healthy Chinese.Methods We recruited 59 healthy subjects From July 2017 to December 2017 in Nanfang Hospital,Southern Medical University,including 34 males and 25 females,who were divided into young group (15-39 years old),middle-aged group (40-64 years old) and the elderly group (over 65 years),respectively.The number of turns (NT) and mean amplitude (MA) normative data of sternocleidomastoid muscle,deltoid muscle,biceps brachii muscle,extensor digitorum muscle,abductor digiti minimi muscle,quadriceps femoris muscle,tibialis anterior muscle,and gastrocnemius muscle were obtained by using concentric needle electrodes.A total of 20-30 records were collected from each muscle in different degree of contraction.A linear regression of lg(MA-100) versus lgNT was made and the slope,intercept,the 95% confidence interval of intercept,the largest MA and largest NT were calculated for each muscle.The influence factors,such as gender,age,muscles,and dominant limbs on the shape of cloud were evaluated by using the Chow breakpoint test.Results Normal turns-amplitude clouds were obtained and presented in this article.Muscle and age were found to be the main influence factors affecting the shape of cloud,and gender may affect some clouds,while the dominant limbs had little influence on the shape of the cloud.Conclusion The reference values of turns-amplitude cloud based on muscle,gender and age are of practical significance in clinic.
