1.Proliferation of vascular smooth muscle cells regulated by NO/PKG mediated via Ca~(2+)/calcineurin signaling pathway

Objective To study the regulation of VSMC proliferation by NO/PKG via modulating intracellular Ca~(2+)/calcineurin(CaN) signaling pathway.Methods Primary VSMCs from rat aorta were used as the experimental model.CaN protein and its activity were assayed using immunoblotting and free inorganic phosphate content analysis respectively.Growth and viability of cells were determined by MTT assay and acridine orange and ethidium bromide staining.Results The addition of SNAP and Sp-8-pCPT-cGMPS decrease absorbance of cells stimulated by phenylephrine(PE),whereas the addition of Rp-8-pCPT-cGMPS increases it.In SMCs p retreated with Ver,absorbance of cells stimulated by PE decreased and was further inhibited by the additional treatment of SNAP and Sp-8-pCPT-cGMPS.In SMCs pretreated with CsA,absorbance of cells stimulated by PE decrease,but it can not be further altered by the additional treatment of SNAP,Sp-8-pCPT-cGMPS and Rp-8-pCPT-cGMPS.Moreover,expression and activities of CaN induced by PE is inhibited by SNAP and Sp-8-pCPT-cGMPS.Conclusion NO/PKG inhibits the proliferation of vascular SMCs without decreasing cell survival rate,which is mediated via intracellular Ca~(2+)/CaN signaling pathway.

Objective:To prepare bifunction antibody of anti-human laryngocarcinoma/anti-CD 3 for the active immune therapy of laryngocarcinoma.Methods:To corss link monoclonal antibody of anti-human laryngocarcinoma and anti-CD 3 into bifunction antibody by chemical method,which can guide the killer function on carcinoma cells.Results:The killer rate of effector cells guided by bifunction antibody is higher than that of anti-CD 3 which can arise with the elevation of effect target ratio.Conclusion:Bifunction antibody of anti-human laryngocarcinoma/anti-CD 3 prepared by chemical cross linking method has the potential value for clinical application. 5 cm. Track record of average range of motion and KSS score. Results A total of 27 knees(10.23%) showed HO after TKA, which included type Ⅰ13 knees, typeⅡ8 knees, typeⅢ 6 knees. Average KSS score of typeⅢ was 83.1?1.5. It was lower than normal group(P0.05). Conclusion TypeⅢ HO after TKA impact clinic outcome. Ostephte increases the probability of HO, and NSAIDS decrease the probability. The incidence of HO have no relation with implants.
5.lncRNA-H19 induces epithelial-mesenchymal transition and enhances the invasion of non-small cell lung cancer cells

Background and purpose:In recent years, the studies have indicated that long non-coding RNAs (lncRNAs) may play important roles in the initial stage and development of tumors. H19 is one of those lncRNAs, which is already proved to overexpress in a variety of tumors such as bladder cancer, stomach cancer and hepatocellular carcinoma (HCC). And H19 also could promote tumor proliferation and increase tumor cell migration and invasion ability, but neither the expression nor the function of H19 in non-small cell lung cancer(NSCLC) are clariifed. This study aimed to observe the effects of H19 on the proliferation, epithelial-mesenchymal transition (EMT) and invasion ability of NSCLC cell line A549.Methods:H19 was overexpressed by plasmids transfection, then the effect of H19 on the proliferation of A549 was measured by cell counting kit-8(CCK-8), the invasion of A549 cells was detected by Transwell assay, and the changes of cell morphology were observed with an optical microscope, and the expression of EMT-related proteins was detected by Western blot, and the promoter activity of CDH1 was measured by luciferase assay.Results:The proliferation of A549 cells was increased under the overexpression of H19(D value in blank group was 1.64±0.02, in negative control group was 1.59±0.04, in overexpression of H19 group 1.89±0.02,P<0.05), the invasion ability of A549 cells was dramatically enhanced [negative control group (30±6)/vision, overexpression of H19 group (110±7)/vision,P<0.05], and the A549 cells developed longer pseudopodia and had wider intercellular spaces. All these morphology changes indicated that the cells were undergoing the process of EMT, and meantime, the expres-sion of CDH1 was decreased, along with the expression of VIM and SNAI2 elevated, which were also related to the progression of EMT, and H19 also could depress the promoter activity of CDH1 by 60% (P<0.05).Conclusion:The overexpression of H19 induces the EMT, and enhances the proliferation and invasion ability of A549 cells.
6.CT-guided percutaneous injection of bone cement for treating osteolysis pelvic disease in 9 cases

BACKGROUND: Osteolysis has always occurred in pelvis. Percutaneous injection of bone cement stabilized bone fracture, relieved pain or even treated tumor. However, leakage of bone cement might cause severe complications. OBJECTIVE: To explore the therapeutic effect of peroutaneous injection of bone cement on treating osteolysis pelvic disease in 9 cases by the CT guidance. METHODS: By the CT guidance, needing degree was determined firstly. Focal size and scanning layers were used to calculate focal volume and estimate injected dose of bone cement. Three-dimensional targeting device was used to introduce the puncturation. The bone cement which was 0.2-0.5 mL less than the calculated volume was injected into osteolysis site. The accuracy, injected dose, clinical efficacy, and complications were investigated. RESULTS AND CONCLUSION: The following-up ranged from 5 months to 4 years, with mean duration of 1.5 years. At 1-48 hours after operation, symptoms were recovered, including complete recovery (n=6), partial recovery (n=2), and light recovery (n=1). Leakage of bone cement was not detected out around focal region. This suggested that percutaneous injection of bone cement into the erosion site is an effective method to treat pelvic osteolysis disease, characterizing by security, effective, and less invasive.
7.Diagnostic performance of HCV antibody and RNA Determination

Objective To explore changes of anti-HCV antibody and HCV-RNA in patients with HCV infection and its clinical significance in diagnosis and treatment of hepatitis C .Methods Serum samples from patients with HCV infection were collected . HCV antibodies were analyzed with a chemiluminescence micro-particle immunoassay method ,while a PCR-fluorescent probe meth-od was used to detect HCV-RNA .Concentrations of ALT and AST were also determined .Based on the concentrations of ALT , AST and HCV-RNA ,samples were divided into two groups respectively and the changes of different indicators were analyzed and compared .Meanwhile samples from 37 HCV-infected patients were collected continuously .Different indicators after treatment were compared with those before treatment .Results HCV-Ab and logarithm values of RNA load in the group with abnormal concentra-tions of ALT and AST were significantly higher than those in the normal group (P<0 .05) .HCV-Ab ,ALT and AST concentrations in HCV-RNA positive group were significantly higher than those in HCV-RNA negtive group(P<0 .05) .The areas under receiver operating characteristic curve of HCV-Ab and logarithm values of RNA load were 0 .990 and 0 .838 respectively .Concentrations of ALT ,AST and logarithm values of RNA load after treatment were significantly lower than those before treatment (P<0 .05) while there was no significance between HCV-Ab level after treatment and that before treatment (P>0 .05) .Conclusion The diagnostic performance of HCV-Ab is better than that of logarithm values of RNA load .Determination of ALT ,AST and HCV-RNA is of clinical importance in monitoring the effect of hepatitis C treatment .
8.Study on the effect and safety of sorafenib in treatment of patients with metastatic renal cell carcinoma

ObjectiveTo evaluate the effect and safety d sorafenib in treatment of patients with metastatic renal cell carcinoma.Methods40 patients with metastatic renal cell carcinoma,were given sorafenib tablets methanesulfonate treatment,the initial dose of 800mg/d,bid,continuous administration of 21d,stopping 7d,The adverse reactions,efficacy and immunohistochemistry results were observed.Results40 cases with metastatic renal cell carcinoma were not found complete remission (CR) and partial remission (PR) ; The 32 cases in stable disease (SD)(80.0% ) and 8 cases ( 20.0% ) in PD; The incidence of 28 cases ( 70.0% ) in digestive system; The expression of positive rate 30.5% of local in connexin 32,Cx32 was significantly lower than that 1.2% in metastatic renal cell carcinoma( x2 =8.123,P <0.01 ),The expression of Cx32 was negatively correlated with the clinical stage( r =-0.419,P <0.05 ) ;The expression positive rate of vascular endothelial growth factor(VEGF) in renal cell carcinoma was significantly higher than 18.5% in normal renal tissue( x2 =8.723,P < 0.01 ) ; The expression positive rate 72.0% in the limitations of renal cell carcinoma and metastatic renal cell carcinoma 89.1% was not statistically significant( x2 =1.978,P > 0.05 ).ConclusionSorafenibin for advanced kidney cancer had better disease control effect,and was new choice for treatment of metastatic renal cell carcinoma.
9.Effects of different resin core materials on the overall flexural strength of fiber posts

BACKGROUND:Selection of resin core materials may affect the overal strength of the fiber posts.
OBJECTIVE:To compare the overal flexural strength of five kinds of resin core materials combined with glass fiber posts.
METHODS:Fifty viva glass fiber posts were randomly divided into five groups respectively binding to five different resin materials for repair:group A, MEDENTAL dual curing resin cement+glass fiber post;Group B, Tina dual curing resin cement+glass fiber post;group C, Bisco BisCem+glass fiber post;group D, 3M nano composite resin curing light P60+glass fiber post;group E, PULPDENT dual curing resin cement+glass fiber post. The root canals were embedded with self-curing plastic, and fixed in the universal testing machine. The load in tooth length axis was added onto the core at a 135° angle with a loading speed of 1.0 mm/min, until the fracture. Then, the stress at fracture and the fracture mode were measured.
RESULTS AND CONCLUSION:The flexural strength was (83.248±7.857) N in group, (89.230±4.326) N in group B, (95.188±5.147) N in group C, (76.646±6.463) N in group D, and (83.064±3.964) N in group E. Except groups A and E, there were significant differences between every two groups (P<0.05). These findings indicate that Bisco BisCem resin cement binding to the fiber post can obtain a higher flexural strength.
10.Histological changes of bone marrow mesenchymal stem cells combined with Bio-oss in repairing rabbit skull defects

BACKGROUND:Some studies have focused on bone marrow mesenchymalstem cells (BMSCs) combined with allograft bone or artificial bone substitute materials for bonedefect repair. But there is no report on BMSCs combined with Bio-oss for repair of rabbit skull defects as yet.OBJECTIVE:To observe the effect ofBMSCs combined with Bio-oss in repairing skull defects in rabbits.METHODS:BMSCs from male rabbits were isolated, cultured, and used as seed cells. In the skull of the female rabbits,three full-thickness bone defects with the same external diameter of 6 mm were made by a ring bone drill. Ninety-six female rabbits were randomly divided into four groups, and given Bio-oss/BMSCs in combination group, Bio-oss alone in Bio-oss group, BMSCs implantation in BMSCs group, and no intervention in blank group. All the implant surfaces were covered with guided tissue regeneration membrane.RESULTS AND CONCLUSION:The osteogenic effect in the combination group was better than that in the other three groups, and the Bio-oss group showed better osteogenesis in comparison with BMSCs and blank groups. But there was no significant difference between the BMSCs and blank groups. These findings indicate that the combined use of BMSCs as seed cells and Bio-oss as a scaffold material exerts overt osteogenic effects in rabbit skull defect area, which provides a new idea for the clinical treatment of bone defects.
