1.THE EXPRESSION AND ASSOCIATION OF CONNEXIN 36 WITH ZONULA OCCLUDENS PROTEIN 1 IN HeLa CELLS

Objective To investigate the expression and association of Cx36 with ZO-1 in Cx36 transfected HeLa cells. Methods RT-PCR,PCR products cloning into PCR2.1TOPO vector,Cx36-pcDNA3 expression vector construction,cell culture,transient transfection using lipofectamine 2000,stable clone screening with G418,Western blotting,double immunofluorescence,and immunoprecipitation(IP) were used. Results Cx36-pcDNA3 expression vector was constructed and transfected into HeLa cells.Using homogenates from Cx36 transfected HeLa cells,Western blotting showed Cx36 band.By immunofluorescence microscopy,Cx36 transfected HeLa cells displayed punctate immunolabeling for Cx36 between cells.Irmmunolabeling for ZO-1 in these cells exhibited similar distribution to Cx36.By laser scanning confocal microscopy after double labeling with Cx36 and ZO-1 antibody revealed a high degree of Cx36 and ZO-1 colocalization at sites of cell-cell contact.Cx36 transfected HeLa cells were taken for IP with ZO-1 antibody,blots of IP protein probed with Cx36 antibody showed detectin of Cx36.Blots showed detection of ZO-1 after IP with Cx36 antibody from Cx36 transfected HeLa cells.Conclusion Cx36 was expressed in Cx36 transfected HeLa cells,Cx36 colocalizated and associated with ZO-1 in Cx36 transfected HeLa cells.
2.Clinical characteristics of 16 fatality cases of hand foot and mouth disease caused by enterovirus 71

ObjectiveTo explore clinical characteristics of fatality cases of hand foot and mouth disease caused by enterovirus 71 (EV71),and to improve the diagnosis and treatment of the disease.Methods Sixteen children infected with EV71 were retrospectively reviewed.Clinical manifestation,laboratory data and death causes were analyzed.ResultsAll cases were younger than four years,and the mean age was 18.9 months.Seasonal variations in incidence were observed,with a peak in incidence during the summer season.These cases typically presented with a brief duration of febrile illness,nonspecific neurological signs including headache,vomiting,limb tremor and convulsions.After two to four days,they all got sudden deterioration,manifested with tachycardia,poor peripheral perfusion,tachypnea and transient hypertension.After intubation,they all had pink frothy or blood from the endotracheal.Tweleve of them died from pulmonary edema or pulmonary hemorrhage,and acute refractory cardiac dysfunction.The other four of cases died from brain stem encephalitis.Atypical presentation such as absence of skin rash should alert the physician.Total white cell count,blood glucose and cardiac troponin I were raised significantly.The protein raised in cerebrospinal fluid.ConclusionHand foot and mouth disease caused by EV71 progresses rapidly.The primary site of attack is the central nervous system,particularly the brainstem,and the cause of death are neurogenic pulmonary edema and/or hemorrhage,acute refractory cardiac dysfunction.
3.Treatment of 116 Cases of Facial Pigmented Spots by Auricular Needle-embedding Method

116 cases of facial pigmented spots were treated by the needle-embedding method in Uterus (MA-TF). EarShenmen (MA-TF1), Endocrine (MA-IC3) and Lung (MA-IC1) as the main points and the additional points selected based upon pattern identification. All cases were cured.
4.Study on Testing of the Indoor Particle Removal Efficiency of Plasma Air Cleaner

Objective By testing the particle removal efficiency of LG Electronics Company's two types of plasma air cleaners,to explore more scientific and reasonable methods for evaluating the purifying effectiveness of indoor air cleaners so as to obtain scientific and contrastable experimental data and calculating results. Methods This evaluating method referred to Standard Examination Method of Portable Household Electric Cord-Connected Room Air Cleaners of America (ANSI/AHAM AC-1-2000) and used one of indexes regu lated in standard Clean Air Delivery Rate (CADR) as the main evaluating index of the effectiveness of indoor air cleaners,meantime several kinds of domestic and international current evaluating indexes were adopted to synthetically evaluate decontaminating function of the air cleaners. Results The index of CADR could accurately and objectively manifest the true decontaminating function of indoor air cleaner. But if using the index of purifying efficiency to reflect the purifying effectiveness of air cleaners,pollutant's natural decay rate and initial concentration should be considered and at the same time other experimental conditions should be given out. Conclusion The establishments of accurate testing methods and accurate expressions of testing results were the sufficient and essential conditions for accurate evaluation on testing objectives.
5.Arthroscopic treatment and high tibial osteotomy with fixation of staple made of retention alligation for the treatment of osteoarthritis of the knee

[Objective]To evaluate the results of treatment with arthroscope and high tibial osteotomy with fixation of staple made of retention alligation for osteoarthritis of the knee,and to discuss the indication for this technique.[Method]From March 1999 to May 2003,42 arthroscopic treatment and high tibial osteotomies were performed in 34 patients.There were 6 men(8 knees)and 28 women(34 knees,with a mean age of 54.2 years(ranged,42~67 years.The average postoperative follow-up was 36 months(ranged,16~44 months.The indications for high tibial osteteotomy were unicompartmental osteoarthritis and varus malalignment.After arthroscopic treatment,a lateral closing-wedge osteotomy was performed with fixation of staple made of retention alligation.The arthroscopic examination showed:plica synovialis in l0 knees,medial meniscus injuries in 12 knees,lateral meniscus injuries in 6 knees,articular cartilage injuries in 20 knees,body loose in joint in 7 knees,intercondylar fossa stenosis in 9 knees.Common peroneal nerve was not exposed and the periosteum opposite to the insicion position was left intact for sake of stability and acted as a hinge around which the wedge osteotomy was closed.The patients were reexamined to obtain a knee score,to make lateral andanteroposterior radiograph of the involved knee with the patient standing.[Result]During the follow-up,the patients showed satifying pain relief,improvement of joint function,correction of yams deformity.The average preoperative knee score was 53.714?6.7,The average postoperative knee score at the time of the latest follow-up was 91.02?7.7.Comparing preoperative with postoperative,there were significant differences(P

7.Effect on physiological and blood gas indicators of beagles by combined anesthesia with tiletamine and zolazepam

Objective To study the physiological indicators and arterial blood gas values,and create a reliable data for researchers using beagle dogs.Methods Beagle were sampled before and after Zoletil anesthesia (10 mg/kg) via intramuscular injection.Then aterial blood gas was detected by blood gas analyzer.Then the physiological indicators,including heart rate,blood pressure and body temperature were measured via large animal ECG.Results For blood gas values,the pH value decreased significantly (P < 0.01).pCO2 increased significantly and pO2 decreased significantly,the differences were significantly (P < 0.05).In contrast,levels of Lac and HCO3-remained unaltered,indicating a typical respiratory acidosis.For physiological indicators,blood pressure increased slightly,but not significant.The heart rate increased significantly (P < 0.01) and body temperature decreased significantly (P < 0.01).There was no difference between sexes.Conclusion Zoletil anesthesia causes respiratory arrest and respiratory congestion,which further induced respiratory acidosis.Meanwhile,anesthesia also results in elevated heart rhythm and lower body temperature.These data indicate that warming and airway clear are necessary during anesthesia in Beagles.Moreover,it should be considered about the effects of anaesthetics when comparing differents in animals before and after treatment.
8.Decorative porcelain temperature firing affects the shear bond strength between zirconia ceramics and resin binder

BACKGROUND:Studies have confirmed that the surface treatment methods such as silane coupling agent and sand blasting could improve bonding strength of zirconia ceramics and resin binder. In addition, the increase of micro cracks on the surface of zirconia ceramics can also improve the bonding strength of zirconia ceramics and resin binder. But there is a lack of related studies addressing whether repeatedly sintering would have an impact on shear bond strength of zirconia ceramics.
OBJECTIVE:To determine the effect of decorative porcelain temperature firing on the shear bond strength between dental zirconia ceramics and resin binder.
METHODS:Twenty pieces of zirconia ceramics specimens were selected from 40 tablets, and then randomly divided into five groups according to the number of sintered times:control group (without sintering), sintering groups for 2, 4, 6, 8 times. Heat starting temperature was 500 ℃, and final temperature was 1 000 ℃ with a heating rate of 55 ℃/min. The vacuum time was 7 minutes. During the sintering, the final temperature was constant. Unsintered ceramic pieces were bonded using resin binder. Universal testing machine was used to test the shear strength of the interface between the two pieces of zirconia ceramics. The interface after shearing was characterized by scanning electron microscope.
RESULTS AND CONCLUSION:The shear strength of sintering groups for 4, 6, 8 times was significantly higher than that of the control group (P<0.05). Shear strength of specimens sintered twice was slightly higher than that of specimens without sintering, but the difference was not significant (P>0.05). Shear strength of specimens sintered 8 times was slightly higher than that sintered 4 and 6 times (P<0.05). Under the scanning electron microscope, there was no crack on the surface of non-sintered zirconia ceramics. After 2 times sintering, the surface appeared to have fine cracks. After sintering 4 times, visible cracks were increased. After six rounds of sintering, the surface began to have significant changes;cracks rose along with slight gaps and a smal amount of adhesive residues were found. After sintering for 8 times, cracks and voids were obviously increased on the sintered surface and adhesive residues were found. It indicated that after sintering for 4, 6 and 8 times, zirconia ceramics could have a good shear strength binding to resin adhesives;with the increasing of sintering times, the shear bond strength could be increased.
9.3D Nonlinear finite element analysis of suitable loading force during mandibular anterior teeth intrusing through improved segment arch

Objective The Improved segmental arch technique can effectively intrude incisors and correct deep overbites. To research its biomechanical characteristics, we reconstruct 3D nonlinear finite element model of mandibular teeth with improved segmen-tal arch, study the biomechanical characteristics and the suitable loading force of intrusion arch. Methods Combined with the re-sults of previous research, we complete the reconstruction of 3D nonlinear finite element model of mandibular teeth with improved seg-mental arch.Then, we set loading force of intrusion arch five conditions from 0.2 N~0.6 N, calculate the movement trend and stress distribution of improved segmental arch. Results In the five conditions of improved segmental arch, lateral incisors and canines in-truded and tipped labially, first molars tipped distally and rotated. In addition, the moment the initial loading force of intrusive arch in-creased, the movement of these teeth increased. However, other teeth did not move clearly. When the initial loading force of improved segment arch was 0.5 N, it can achieve appropriate intrusion of anterior teeth. Conclusion Under an appropriate intrusive force, improved segment arch can effectively intrude incisors and control the extrusion of posterior teeth. It can be used to correct the deep overbites, especially with high mandibular planes, gummy smile or adult stage.
10.The primary experimental study of self-made percutaneous catheterized thrombectomy device for acute massive pulmonary embolism

Objective To evaluate efficacy,feasibility and safety of the self-made percutaneous catheterized thrombectomy divice in animal model for thrombus removal. Methods Seven dogs were selected,with acute massive pulmonary embolism animal models created by injecting thrombi into the pulmonary arterial trunk via percutaneous femoral vein approach. After half an hours the catheter sheath was inserted into the occluded pulmonary artery through right femoral vein in 5 dogs,left femoral vein in 1 dog and right internal jugular vein in another one. The procedure began to remove the thrombi with simultaneous recording the thrombectomy time and the blood volume drainage. Blood gass was tested before and after embolization together with those of thrombi removement,continuously monitored pulmonary arterial pressure and intermittently performed angiography. The mean time form vascular recanalization to euthanasia was 2 hours,and then the lung specimens were resected for histological examination. Results One animal died of pulmonary arterial penetration during thrombi removal,but others were all alive by the end of the test. Mean time of removing thrombi was 2.4 minutes with mean volume blood drainage of 84 ml. Angiograms showed the approximately complete patency of the pulmonary arterial trunk after reopenning of occlusion but still with remnont thrombi within distal branches and arterial pressure with blood gas returned to normal level. Pathology revealed the recanalization of pulmonary arterial trunk but with thromi still staying in the distal branches,and effusion around the arteries. Conclusions The self-made percutaneous catheterized thrombectomy device is effective,feasible and comparatively safe in the treatment of acute massive pulmonary embolism in this primary test.
