1.A review of vascular regeneration in the invasive growth process of Echinococcus multilocularis

Alveolar echinococcosis (AE) is a potentially fatal zoonosis caused by Echinococcus multilocularis (E .m) , primary located in the liver and showed malignant invasion .AE can develop to the lungs and brain metastases .Operation treat-ment and medication therapeutic efficacy show poor effect .The mechanism of AE invasive growth and metastasis is unclear . Angiogenesis is a highly regulated behavior in normal tissues .Abnormal angiogenesis is one of the key signs of malignant tumor ,and VEGF is one of the key mediators of angiogenesis .Numerous studies have shown that VEGF and CD34 plays an important role in the formation of tumor angiogenesis and invasive growth process .This article discussed the role of angiogene-sis in AE aggressive ,aimed to provide a new way for the clinic conservative treatment for patients with advanced stage ,preven-ting recurrence after operation ,and adjunctive therapy of operation and drug .
2.Clinical Study onXiaozhikang Formula Granules for the Treatment of Nonalcoholic Fatty Liver Disease

Objective To observe the clinical efficacy ofXiaozhikang Formula Granules for the treatment of nonalcoholic fatty liver disease.Methods Totally 90 patients with nonalcoholic fatty liver disease were randomly divided into control group and treatment group, 45 cases in each group. Patients in the control group were treated with conventional western treatment and oral metformin enteric-coated metformin hydrochloride, while patients in the treatment group were treated withXiaozhikang Formula Granules on the basis of conventional western treatment for 3 months. Blood lipid and insulin resistance in the two groups before and after treatment were observed, and the clinical efficacy between the two groups was compared.Results Clinical excellence (35.5%), total effective rate (84.5%) and TCM syndrome score (13.78 ± 3.6) after treatment of treatment group were better than those of control group (P<0.05). GPT, GOT, TG, and TC in both groups were lower after the treatment (P<0.05,P<0.01). There existed statistical significance in TC and TG between the two groups after treatment (P<0.05). There existed statistical significance in B ultrasound index between the two groups after treatment (P<0.05), and the treatment group was superior. FBG and HOMA-IR in both groups significantly decreased after treatment (P<0.05), and the FBG in the treatment group after treatment was better than control group (P<0.05).Conclusion The efficacy ofXiaozhikang Formula Granules in the treatment of nonalcoholic fatty liver disease is remarkable.

Aim To investigate the effect of taurine(Tau)on the cardiac fibroblast(CFb)cell cycle and its regulatory protein expression,and to explore the underlying mechanism of Tau-inhibiting the proliferation of CFb.Methods The proliferation of cultured neonatal rat CFb was induced by Angiotensin Ⅱ and detected by the thiazole blue(MTT)colorimetric assay.Hydroxyproline measurement was carried out to detect the collagen quantification.The expression of cell cycle regulatory protein cyclin D and p27 were determined by the combination of immunocytochemical staining and image analysis software.Cell cycle and p27 were assessed via flow cytometry.Results CFb proliferation and hydroxyproline content in culture medium were markedly inhibited when CFb were treated with taurine at concentrations of 40,80 and 160 mmol?L-1 for 24 hours(P

Aim To investigate the molecular mechanism of anti-proliferation effect of Ginsenoside Rh2(G-Rh2)on mouse MFC gastric cells.Methods The cell viability was determined by MTT method and the state of cell proliferation was analyzed.The morphologic changes were observed by invert microscope and fluorescence microscope(Hoechst 33258).Early apoptotic rate was detected by Annexin Ⅴ-FITC through flow cytometry.Western blot was used to detect the p-JNK and p-c-jun activity in MFC cells.Immunocytochemistry staining was used to detect caspase-3 positive cells.Results G-Rh2 could inhibit proliferation of MFC cells in a dose-and-time-dependent manner.The morphological changes of apoptosis were observed in MFC cells after G-Rh2 treatment.The apoptotic ratio was increased.The phosphorylation of c-Jun and c-Jun NH2-terminal kinase(JNK)activity increased with time within 4 h compared with that of control.The expression of caspase-3 positive cells was increased.But all the above could be partly inhibited by pre-treatment with SP600125(5 ?mol?L-1)for 2 h.Conclusion G-Rh2 can decrease cell viability by inducing apoptosis and the process may be associated with the activation of JNK transduction pathway and expression of caspase-3.
7.Enalaprilat inhibited the proliferation of Neonatal rat Cardiac Fibroblasts via modulation of endothelin-1 and nitric oxide production

Aim To investigate the effects of angiotensinI-converting enzyme inhibitor enalaprilat on proliferation,Endothelin-1 and Nitric oxide System of neonatal cardiac fibroblasts and to probe its mechanism of inhibiting cardiac fibrosis.Methods CFb was isolated by trypsin digestion method.MTT colorimetric assay was adopted to evaluate cell proliferation,HYP method was used to test the collagen synthesis;FCM for cell cycle;Nitric acid reductase method,radio-immunological method was used to detect NO content and ET-1 activity respectively.Results Ena decreased contents of MTT and HYP dramatically.The percentage ratio of phase G0/G1 was increaseded and the percentage radio of phase S was decreased.NO content was increased and ET-1 activity was dropped in a dose and time-dependent manner.Conclusion The anti-proliferative effects of Ena on CFb can be attributed to dropping endothelin-1 activity and enhancing Nitric oxide.
8.Protective effects of total saponins of tribulus on cerebral ischemia reperfusion injury in rats

Aim To observe the protective effects of total saponi ns of tribulus (TST) on cerebral ischemia reperfusion injury in rats. Meth ods Rat cerebral ischemia-reperfusion injury was induced by middle cer ebral artery occlusion .The neurological outcome was evaluated. CK, LDH and NO a ctivity in serum,SOD and MDA levels of cerebral tissue were measured. Re sults The levels of CK,LDH,NO,SOD and MDA in TST(30 mg?kg -1,1 0 mg?kg -1) groups were significantly lower than those in model group. The level of cellular and intercellular edema was reduced markly in the pathomorpho logy examination. The neurological deficits was decreased by TST.Conclus ion TST has protective effects on cerebral ischemia reperfusion injur y in rats.

Aim To study the anti-tumor activity of PQEP and it s immunomodulation effect.Methods The pharmacological activity of PQS was measured using mice transplanted H22 and MFC tumor. Tumor-inhibiting a ctivities, weight of immune organ, nature killer cells activity and lymphocyte p roliferation ratio were investigated.Results PQEP showed antitumo r and immunomodulation activities in different degrees.Conclusion The anti-tumor activity and immunomodulation are closely linked. The anti- tumor effect of PQEP due to activation of the immunocompetence of the body.
10.Induction of apoptosis by Panax quinquefolium effective parts (PQEP) on K562 cells

Aim This research was performed to study the anticancer effects of Panax quinquefolium effective parts (PQEP) in human chronic myeloid leukemia (K562). Method After K562 cells were routinely cultured,MTT assay was performed for cytotoxicity test. Cytotoxicity of PQEP (6.25~400 mg?L -1) in K562 cells was increased in a dose-and time-dependent manner. To explore the mechanism of cytotoxicity, we used several measures of apoptosis to determine whether these processes were involved in PQEP-induced leukemic cell death.Results PQEP induced the cell shrinkage, cell membrane blebbing, chromosomes condensation and DNA fragment. In addition, the flow cytometric analysis revealed PQEP (10~400 mg?L -1) dose-dependently increased apoptotic cells with hypodiploid DNA contents. Conclusion These results indicate that PQEP can control leukemic K562 cells through apoptosis and may have a possibility of potential anticancer activities.
