1.Effects of Astragalus Injection on Myocardial Cell Damages Due to Oxidative Stress

ObjectiveTo investigate the protective effects of Astragalus injection (AI) on hydrogen peroxide (H2O2 ) induced injury in cardiomyocytes. MethodsCultured neonatal rat cardiomyocytes were divided into: control group; H2O2 group, in which cells were treated with H2O2 0.15 mmol/L for 5 h; AI+H2O2 group, in which cells were pretreated with AI (with final concentration of 10, 30, 90 g/L) 30 min before H2O2 treatment; and AI (90 g/L)+L-NAME (20 μg/L). The cardiomyocyte viability was analysed by MTT assay, lactate dehydrogenase (LDH) activity and nitric oxide (NO) content were detected in culture media. Reactive oxygen species (ROS) were measured with laser-confocal-microscopy system. Mitochondrial memberane potential (ΔΨm) and apoptosis rate were measured with flowcytometry. ResultsCardiomyocyte viability in AI (10,30, 90 g/L) groups were higher than that in H2O2 group (P<0.05). Compared with H2O2 group, LDH activity and ROS content in AI (90 g/L) group decreased (P<0.01), NO content increased (P<0.01), ΔΨm of cardiomyocytes increased (P<0.05) and apoptosis rate decreased (P<0.05). Compared with AI (90 g/L) group, after treatment combining with L-NAME, LDH activity and ROS content increased (P<0.01), NO content decreased (P<0.01), ΔΨm of cardiomyocytes decreased (P<0.05) and apoptosis rate increased (P<0.05). ConclusionAI can protect cardiomyocytes from H2O2 injury by increasing NO content and inhibiting cardiomyocyte apoptosis induced by ROS.
2.Protective effects of Astragaloside Ⅳ on hydrogen peroxide-induced injury of cultured neonatal rat myocardial cells

Objective To investigate the protective effects of Astragaloside Ⅳ(ASⅣ)on cardiomyocytes against hydrogen peroxide(H2O2)-induced injury.Methods Cultured neonatal rat cardiomyocytes were divided into control group;0.1%DMSO solven group;H2O2 group,in which cells were treated with H2O2( 0.2 mmol?L-1) for 24 h;ASⅣ+H2O2 groups,in which cells were pretreated with ASⅣ(with final concentration of 3,10,30 mg?L-1) for 30 min before H2O2 treatment;Astragaloside injection group(150 g?L-1).Morphological changes of myocardial cells damaged for 24 h by 0.2 mmol?L-1 H2O2 under the inverted microscope were observed.The cardiomyocyte viability was analysed by MTT assay;lactate dehydrogenase(LDH) activity,superoxide dismutase(SOD) activity,malonaldehyde(MDA) content and nitric oxide(NO) content were detected in culture media.Results Most of cells in H2O2 group were crenulated,the nucleus were dim and the pseudopodia were disappeared.The degrees of lesion in ASⅣ(3,10,30 mg?L-1) groups were lessened obviously,the nucleus were bright and the the pseudopodia were thinned slightly.Cardiomyocyte viabilities in ASⅣ(3,10,30 mg?L-1) groups were higher than that in H2O2 group(P
3.Clinical characteristics and angio-architecture features of dural arteriovenous fistulas

To investigate the clinical characteristics and angio-architecture features of patients with dural arteriovenous fistulas (DAVF).The clinical data of 48 patients with DAVF were analyzed retrospectively.All patients were diagnosed by digital subtract angiography and 43 cases were also examined by MRI.Patients were divided into the bleeding group and non-bleeding group,whose clinical features and angio-architecture were compared.Of 48 cases,13 patients demonstrated intracranial bleeding,and men were more common in bleeding group (M/F:10/3) than in non-bleeding group (M/F:15/20) (P =0.036).The Cognard scores of bleeding group and nou-bleeding group were 3.77 ±0.28 and 2.49 ±0.21,respectively (P =0.002) ; however,there was no significant difference in age and the number of feeding artery between two groups.The results indicate that male patients with high Cognard scores tend to intracranial bleeding.
4.Double-row suture anchors under arthroscopy for avulsion-type greater tuberosity fractures (Mutch type I)

BACKGROUND: Greater tuberosity of humerus, as the attachment point of rotator cuff, plays a vital role in shoulder.Neer and AO classification for proximal humeral fractures have been accepted extensively. However, the classification for single greater tuberosity fractures of the proximal humerus is little reported, and its fixation method remains controversial.OBJECTIVE: To explore the curative efficacy of the double-row suture anchors under arthroscopy for avulsion-type greater tuberosity fractures (Mutch type Ⅰ) METHODS: Clinical data of 24 patients with avulsion-type greater tuberosity fractures (Mutch type Ⅰ) undergoing double-row suture anchors under arthroscopy were analyzed retrospectively. The displacement distance of the greater tuberosity of humerus was measured on CT before and after fixation. The shoulder joint was evaluated by Constant-Murley Score and the University of California, Los Angeles score at baseline, 1, 3, 6 and 12 months postoperatively. Besides, the visual analogue scale scores were detected at baseline and 12 months postoperatively.RESULTS AND CONCLUSION: (1) Double-row suture anchors effectively improved the displacement of greater tuberosity and rotator cuff injury. (2) The patients accompanied by rotator cuff injury accounted for 79.16%. (3) The Constant-Murley Score and the University of California, Los Angeles scores were significantly improved at each time point (P < 0.05). The visual analogue scale scores after fixation were significantly superior to those before fixation (P < 0.05). (4) These results suggest that the double-row suture anchors under arthroscopy can effectively improve the displacement of greater tuberosity and alleviate the pain. Moreover, it is conductive for early recovery of the shoulder function with little trauma, so it is a good choice for avulsion-type greater tuberosity fractures (Mutch type Ⅰ).
5.Anterior cruciate ligament reconstruction with autologous ipsilateral 1/2 peroneus longus tendon

BACKGROUND:Anterior cruciate ligament (ACL) injury is a commonly sport-induced knee joint injury that does serious harm to the knee stability. ACL reconstruction is a commonly used treatment method, but researches on 1/2 peroneus longus tendon (PLT) graft are rarely reported. OBJECTIVE: To investigate the clinical outcomes of removing the autologous ipsilateral 1/2 PLT under arthroscopy for ACL reconstruction. METHODS:106 patients with complete ACL rupture in the Affiliated Hospital of Traditional Chinese Medicine, Southwest Medical University from December 2010 to December 2014 were enrolled, and autologous ipsilateral 1/2 PLT was removed under arthroscopy for ACL reconstruction. At baseline, 3, 6 and 12 months postoperatively, the knee stability was evaluated manually through the anterior drawer test, Lachman test, and pivot-shift test, and the knee function was evaluated by Tegner activity scale, Lysholm and International Knee Documentation Committee scores. RESULTS AND CONCLUSION: Postoperative anterior drawer test, Lachman test, and pivot-shift test tests were negative in all patients. In terms of Tegner activity scale, Lysholm and International Knee Documentation Committee scores, there were significant differences at baseline and postoperative 3 months as compared with postoperative 6 months (P < 0.05); the scores at baseline and postoperative 3 months showed significant differences compared with 12 months postoperatively (P < 0.05); the scores showed no significant difference between 6 and 12 months postoperatively (P > 0.05). These results indicate that autologous ipsilateral 1/2 PLT is a good choice for ACL reconstruction under arthroscopy, achieving rapid and satisfactory functional recovery of the knee joint, which is not only minimally invasive and easy to operate, but also exhibits good therapeutic efficacy.
6.Application of thermoplastic elastomer in hot-melt pressure sensitive adhesives for transtermal drug delivery.

Development of drug dosage forms to a great extent depends on the development of drug auxiliary materials. The development of a new type of polymeric drug auxiliary materials will bring on the developing of a novel dosage forms technology and a flood of new drug dosage forms. Thermoplastic elastomer is a new type of drug polymeric auxiliary materials, at present, which has a broad application in the field of hot-melt pressure sensitive adhesives. This review mainly discussed a new transtermal Chinese drug delivery system, including matrix composition of the formula, modified thermoplastic elastomer for hot-melt pressure sensitive adhesives and their development prospects in the traditional Chinese drug delivery system. It suggested that thermoplastic elastomer of hot-melt pressure sensitive adhesives has broad development prospects in the field of the transtermal drug delivery system for traditional Chinese medicine.

7.Percutaneous permeability study for patches Shangshi Zhitong on different kinds of bases.

OBJECTIVETo study the percutaneous permeability of patches Shangshi Zhitong on different kinds of bases with the permeation percentage of brucine, strychnine and atropine.
METHODUsing modified Franz difusion to investigate the penetration quantity of brucine, strychnine and atropine. The contents of brucine, strychnine and atropine were determined by HPLC.
RESULTThe average accumulative permeation percentage of brucine, strychnine and atropine on new base were 53.25%, 74.52% were 34.32%, respectively, and on old base are 54.90%, 50.24%, 46. 54%, respectively.
CONCLUSIONThe new base benefits the lipophilic drugs and releases more stably.

8.Erastin induces ferroptosis in lung fibroblasts through MAPK mediated oxidative stress signaling pathway

Objective :
To investigate the mechanism by which Erastin affects ferroptosis in lung fibroblasts.
Methods:
Mouse lung fibroblasts (C57/B6⁃L) were treated with varying concentrations of the iron death inducer Erastin, Cell viability was assessed using the cell counting Kit⁃8 (CCK⁃8) assay. Oxidative stress levels were visualize using a fluorescence microscope , and the expression of proteins related to the mitogen⁃activated protein kinase (MAPK) signaling pathway was analyzed using Western blot. Additionally , the p38 and extracellular regulated protein kinase (ERK) inhibitors SB203580 and U0126 were employed to further elucidate the mechanism by which Erastin induces iron death in lung fibroblasts.

At a concentration of 100 μmol/L , Erastin effectively in⁃duced ferroptosis in lung fibroblasts , leading to an upregulation of oxidative stress. Furthermore , the phosphoryla⁃tion levels of p38 and ERK proteins in the MAPK pathway were elevated (P < 0. 05) . The addition of SB203580 and U0126 inhibitors resulted in a significant reduction in oxidative stress levels and a notable increased in cell actiivity in lung fibroblasts (P < 0. 05) .
It can be concluded that Erastin induces ferroptosis in lung fibroblasts , potentially through the mediation of oxidative stress via the MAPK signaling pathway.
