1.Establishment of a rat model of hyperuricemia associated with uric acid excretion disorder

Objective To explore a reliable method to establish a rat model of hyperuricemia associated with abnormal uric acid excretion, and to lay the foundation for the study of pathogenesis of uric acid excretion disorder and the optimization of the treatment plan.Methods The models were established respectively by potasium oxonate(300 mg/kg) with pyrazinamide (300 mg/kg) or ethambutol(250 mg/kg).Continuous dosing for 1, 3 and 5 weeks, to determine the content of uric acid in rat blood, urine, and stool, the function of liver and kidney was detected and pathological examination was performed.Results The blood uric acid in the potasium oxonate and ethambutol group was increased first and then decreased, while in the potasium oxonate and pyrazinamide group were increased steadily and the excretion of uric acid in urine was stable during the continuous administration.The two methods showed no harmful effect on the liver and kidney function.Conclusions A stable rat model of hyperuricemia associated with uric acid excretion disorder can be effectively established by potassium oxonate and pyrazinamide, exhibiting similar manifestations of clinical hyperuricemia and uric acid excretion disorder.

3.Evaluation of effects of simvastatin on vascular elasticity in fat-fed rabbits by echo-tracking technique

Objective To evaluate the effects of simvastatin on the vascular elasticitv in fat-fed rabbits by echo-tracking(ET)technique.Methods Twenty seven rabbits were divided randomly into 3 groups:normal control group(group A),high fat diet group(group B),simvastatin group(group C:high fat diet for the 1st 4 weeks and high fat diet+simvastatin for the 2nd 4 weeks).β,EP,AC,and PWVβof abdominal aorta (AO) and femoral artery(FA)of rabbits in group A、B、C were measured by ET technique at the end of 4th week and 8th week respectively.The expression of iNOS was measured at the end of 8th week.Results At the end of the 4th week,β,Ep,and PWVβ of FA were significantly inereased and AC was significantly decreased in group B compared to those in group A(P<0.05).At the end of the 8th week,β,Ep,PWVβof AO were significantly increased and AC was significantly decreased in group B compared to those in group A(P<0.05);β,Ep,PWVβ of FA were statistically decreased and AC was statistically increased in group C compared to those in group B(P<0.05),however there was no significant difference of AO between groups B and C.Conclusions Short-term administration of simvastatin can improve the elasticity of some muscular arteries such as FA but not AO.
4.The performance evaluation of the five inspection methods in the diagnosis of peripheral neuropathy in diabetes

Objective To evaluate the diagnostic value of the 10 g nylon silk,vibration threshold value,sense of temperature,pain sense and ankle reflex in the diagnosis of diabetic peripheral neuropathy (DPN)and to search an exact diagnostic method of DPN not only economical but also easy to handle.Methods To carry out the measurements of the NCV,10 g nylon silk,vibration threshold value,sense of temperature,pain sense and ankle reflex on the patients who have been diagnosed with diabetes (DM)between April 2014 and June 2014 in the endocrinology department of our hospital.The sensitivity,specificity,positive forecast value,negative forecast value,Youden index and K value serve as assessments of the diagnostic efficiency of clinically diagnostic criteria. Results The sensitivity,specificity,positive forecast value,negative forecast value,Youden index and K value of the ankle reflex are 66.04%,95.56%,97.22%,54.43%,61.60%,and 0.5 1 in mediate accordance with the diagnostic criteria and it is the best one among the five methods.While the sensitivity and accordance of the pressure sense are of the least accordance with the diagnostic criteria.The correspondent value of the combination of vibration threshold sense,temperature sense and ankle reflex are respectively 86.79%,91.11%,95.83%,74.55%, 77.90% and 0.73,in positive accordance with the diagnostic criteria.Conclusion Among the five methods the consistency between the combination of vibration threshold sense,temperature sense and ankle reflect and diagnostic criteria is of positive accordance and can be applied in screening DPN clinically.
5.Effect of Tangmoning on nerve conduction velocity and RBC sorbitol level in diabetic rats

AIM: To investigate the effects of Tangmoning (TMN) Granule on nerve conduction velocity, red blood cell sorbitol (RBC S) level in diabetic rats and probe the mechanism of prevention and treatment of diabetic peripheral neuropathy. METHODS: The model of diabetic rat was induced by strepotozotocin. The animals were randomly divided into six groups:TMN minimal dose group, TMN mild moderate dose group, TMN maximal dose group, methycobal group, model group and normal control group. All TMN groups were treated with TMN Granule through tube feeding (2.95 g/kg、5.90g/kg、11.80g/kg). Methycobal group was treated with methycobal tablets through tube feeding (0.14mg/kg). The duration of treatment was 3 weeks. The caudal nerve conduction velocity, RBC S content were investigated before and after treatment. 6.Treatment progress of chronic lymphocytic leukemia

Chronic lymphocytic leukemia (CLL) is a highly heterogeneous disease, and combined chemotherapies symbolized by rituximab + fludarabine + cyclophosphamide have made great achievements in the era of traditional immunochemotherapy. However, for the patients with high genetic risk, relapsed and refractory patients as well as the elderly patients who cannot tolerate it, the therapeutic effect of this regimen is not good. The new drugs represented by BTK inhibitors have solved the above problems to a certain extent, and it is hopeful to move into the chemotherapy-free age of CLL treatment and bring the possibility of drug withdrawal. This article reviews the progress of CLL treatment.
7.Identification of diagnostic biomarkers for sarcopenia based on bioinformatics and machine learning

Objective:To identify characteristic genes in sarcopenia patients through bioinformatics and machine learning, and to explore the clinical relevance of characteristic genes in the diagnosis of sarcopenia.Methods:The microarray data of GSE25941, GSE38718 and GSE9103 associated with sarcopenia were downloaded from the GEO database, followed by identification of differentially expressed genes (DEGs) associated with sarcopenia. Subsequently, functional analysis of the DEGs was performed using gene ontology (GO) functional annotation and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis. The protein-protein interaction (PPI) network was constructed using STRING and Cytoscape, while biomarkers of sarcopenia were identified using LASSO regression and random forest analysis. The diagnostic performance of the characteristic gene was assessed by employing receiver operating characteristic (ROC) curve analysis. Furthermore, the expression levels of biomarkers for sarcopenia were validated using the external validation dataset of GSE28422. Finally, CIBERSORT was employed to analyze the infiltration of immune cells.Results:124 DEGs were identified between control and sarcopenia populations, which were primarily involved in growth factor receptor binding and cytokine activity. KEGG analysis revealed that the DEGs were predominantly associated with signaling pathways such as peroxisome proliferator-activated receptor signaling pathway, adipokine signaling pathway, Jak-STAT signaling pathway, and adenosine 5'-monophosphate (AMP)-activated protein kinase signaling pathway. Through machine learning techniques validated by ROC curve analysis and external datasets, three characteristic genes, namely DMRT2, FAM171A1, and ARHGAP36, were discovered. The infiltration analysis of immune cells revealed the potential involvement of mast cells, CD4 memory T cells, CD8 cells, γδT cells, and neutrophils in the pathophysiology of sarcopenia.Conclusion:DMRT2, FAM171A1 and ARHGAP36 can serve as diagnostic biomarkers of sarcopenia, and are closely related to the pathophysiological process of sarcopenia.
8.Determination of antioxidant activity of Polygoni Multiflori Radix Praeparata by HPLC-DPPH

An on-line HPLC-DPPH system was developed to determine the antioxidant activity of 16 batches of Polygoni Multiflori Radix Praeparata. By analyzing the chromatographic and biological activity fingerprints of 16 batches of Polygoni Multiflori Radix Praeparata, the dose-effect relationship was established and the total antioxidant activity was quantified by activity addition.The results suggested that the online HPLC-DPPH method can evaluate the antioxidant activity of different bathches of Polygoni Multiflori Radix Praeparata, with different processing methods, aiming to provide datasupport and scientific basis forquality evaluation of Polygoni Multiflori Radix Praeparata.
9.Down-regulation of p110β expression increases chemosensitivity of colon cancer cell lines to oxaliplatin.

This study examined the synergetic effect of class IA Phosphoinositide 3-kinases catalytic subunit p110β knockdown in conjunction with oxaliplatin treatment on colon cancer cells. Down-regulation of p110β by siRNA interference and oxaliplatin treatment were applied in colon cancer cell lines HT29, SW620 and HCT116. MTT assay was used to measure the inhibitory effect of p110β knockdown on the proliferation of colon cancer cell lines. SubG1 assay and Annexin-V FITC/PI double-labeling cytometry were applied to detect cell apoptosis. And cell cycle was evaluated by using PI staining and flow cytometry. The expression of caspase 3, cleaved PARP, p-Akt, T-Akt and p110β was determined by western blotting. The results suggested that down-regulation of p110β expression by siRNA obviously reduced cell number via accumulation in G(0)-G(1) phase of the cell cycle in the absence of notablely increased apoptosis in colon cancer cell lines HT29 and SW620 (S phase arrest in HCT116). Moreover, inhibition of p110β expression increased oxaliplatin-induced cell apoptosis and cell cycle arrest in HT29, HCT116 and SW620 cell lines. In addition, increases of cleaved caspase-3 and cleaved PARP induced by oxaliplatin treatment were determined by immunoblotting in p110β knockdown group compared with normal control group and wild-type group. It is concluded that down-regulated expression of p110β could inhibit colon cancer cells proliferation and result in increased chemosensitivity of colorectal cancer cells to oxaliplatin through augmentation of oxaliplatin-induced cell apoptosis and cell cycle arrest.
10.Clinical evaluation of the consistency between two diagnostic criteria for diabetic peripheral neuropathy.

OBJECTIVETo evaluate the consistency between the clinical diagnostic criteria and the ascertained diagnostic criteria for diabetic peripheral neuropathy (DPN) in the Preventive and Treatment Guidelines of Diabetes in China (2013) and explore an economic, convenient, and accurate approach to DPN diagnosis.
METHODSThe patients with type 2 diabetes admitted in our department from April to June, 2014 were examined for nerve conduction velocity, 10 g nylon silk, vibration threshold value, sense of temperature and pain, and ankle reflex. The sensitivity, specificity, positive predictive value, negative predictive value, Youden index, and Kappa value were calculated to assess the diagnostic power of the two diagnostic criteria.
RESULTSOf the 151 patients enrolled, 106 (70.2%) had a diagnosis of DPN consistent with the ascertained diagnostic criteria, as compared to 86 (56.95%) who were diagnosed according to the clinical diagnostic criteria; the latter patients accounted for 81.13% of former cases. The sensitivity, specificity, positive predictive value, negative predictive value, Youden index, and Kappa value of the clinical diagnostic criteria were 80.19%, 97.78%, 98.84%, 67.69%, 77.97%, and 0.69, respectively, which were highly consistent with those of the ascertained diagnostic criteria; the sensitivity to compression showed a poor consistency between the two diagnostic criteria. In the 5 screening tests, the combined test of temperature sensation, vibration perception, and ankle reflex showed the highest AUC value among their different combinations.
CONCLUSIONThe clinical diagnostic criteria for DPN show good consistency with the ascertained diagnostic criteria, and for patients with clinical symptoms or with only one positive sign, combination of the two diagnostic criteria can achieve the maximum diagnostic power.
