1.Construction of recombinant plasmid pEGFP-NGB and its expression in cultured neuroglial cells

AIM: To establish recombination plasmid pEGFP-NGB and to investigate the expression of pEGFP-NGB in culture neuroglia cells. METHODS: The NGB cds was isolated by using RT-PCR method with total RNA extracted from fetal Kunming mouse brain, then the NGB cds was cloned into the eukaryotic expression vector pEGFP-C1 of EGFP reported green fluorescence protein. The expression vector of recombinant plasmid pEGFP-NGB was successfully constructed. GeneJamer transfection reagent was used to transfer recombinant plasmid pEGFP-NGB into culture neuroglial cells. The mRNA and protein expression of pEGFP-NGB in culture neuroglial cells were investigated. RESULTS: The positive clone sequencing was consistent with the sequence of Genbank. The NGB mRNA and protein expression of pEGFP-NGB in culture neuroglial cells were detected at high levels. The high expression of green fluorescence protein was observed by fluorescence microscope in culture neuroglial cells. CONCLUSION: The expression vector of recombinant plasmid pEGFP-NGB was successfully constructed and green fluorescence protein was expressed in cultured neuroglial cells.
2.Effect of intrathecal injection of morphine on patients with severe rectum cancer pain

Objective To investigate the effect of intrathecal injection of morphine on patients with severe rectum cancer pain.Methods According to random number table method,28 patients with severe rectum cancer pain,admitted to our hospital from April 2014 to April 2015,were divided into two groups:intrathecal injection of morphine group (group Ⅰ) and epidural injection of morphine group (group E,n=14).Patients in group Ⅰ were treated with intrathecal injection of morphine,and group E with epidural injection of morphine;initial dose of group Ⅰ was 1/300 of daily average dosage,and group E 1/30 of daily average dosage;the additional dose of two groups was 1/10 of initial dose,with a locked time of 30 min.Resting and moving visual analogue scale (VAS) scores,life scores of quality of life questionnaire (QLQ)-C30,and daily dose of morphine were observed on the day before analgesia (T0),first week after analgesia (T1),second week after analgesia (T2),and first month after analgesia (T3).Nausea and vomiting,skin itching,retention of urine,and headache were recorded at the four points.Results As compared with those at T0,the resting and moving VAS scores and daily dose of morphine were significantly lower,life scores of QLQ-C30 at the T1,T2 and T3 were significantly higher in both two groups (P<0.05).As compared with those in group Ⅰ,the resting and moving VAS scores and daily dose of morphine in group E were significantly higher,life scores of QLQ-C30 in group E were significantly lower at the T1,T2 and T3 (P<0.05).The incidence of complications in group Ⅰ (21.4%)was significantly lower than that in group E (71.4%).Conclusion The effect of analgesia with intrathecal injection of morphine is better than that of analgesia of epidural injection of morphine in the treatment of severe rectum cancer pain.
3.Correlation between blood pressure variability and different types of stroke

Objective To explore the relationship between blood pressure variability and different types of stroke.Methods One hundred and twenty-five ischemic stroke subjects,78 hemorrhagic stroke patients and 66 non-stroke individuals,collected in our hospital from January 1,2013 to December 31,2015,were enrolled according to the inclusion and exclusion criteria.All indicators of blood pressure variability were calculated from 24 h ambulatory blood pressure,including nighttime blood pressure fall (BPF),moming surge in blood pressure (MBPS),standard deviation (SD),weighted SD (wSD),and coefficient of variation (CV).Results As compared with non-stroke subjects,ischemic and hemorrhagic stroke individuals had significantly increased systolic MBPS (P<0.05),and the systolic MBPS of ischemic stroke group was significantly lower than those of hemorrhagic stroke group (P<0.05).CV,wSD and BPF of systolic blood pressure (SBP) in the ischemic and hemorrhagic groups were significantly higher than those in the non-stroke subjects (P<0.05);CV and SBPF of diastolic blood pressure in hemorrhagic group were significantly different as compared those in the non-stroke subjects (P<0.05).The factors which were significantly associated with ischemic stroke in Logistic regression,included high-density lipoprotein,nighttime SBP,wSD of SBP,CV of SBP,while smoking,nighttime SBP,wSD of SBP,MBPS,and CV of SBP were observed as important influence factors for hemorrhagic stoke patients.Conclusion Whether ischemic stroke,or hemorrhagic stroke are associated with short-term blood pressure variability.
4.Relationship between blood pressure variability and arterial elasticity in elderly patients with stroke

Objective To explore the correlation between blood pressure variability (BPV) and arterial elasticity in elderly patients with stroke.Methods One hundred and eighty-three stroke patients (121 with cerebral ischemic stroke and 62 with hemorrhagic stroke),admitted to our hospital from May 2014 to December 2016,and 61 non-stroke individuals were enrolled according to the inclusion and exclusion criterion.Carotid-radial pulse wave velocity (CrPWV) was calculated and carotid artery ultrasound was used to measure intima-media thickness (IMT);24 h ambulatory blood pressure monitoring was performed to calculate the BPV parameters.The arterial stiffness index β was calculated.Results As compared with those in the non-stroke subjects,morning blood pressure surge (MBPS),standard deviation of systolic blood pressure (SBP),variable coefficient of SBP,systolic blood pressure fall (SBPF),CrPWV,IMT and stiffness index β in stroke patients were significant increased (P<0.05).Correlation analysis indicated that MBPS,standard deviation of SBP,weighted standard deviation of SBP,variance of SBP,variable coefficient of SBP,SBPF,and diastolic blood pressure fall were signficantly correlated with CrPWV and stiffness index β (P<0.05);IMT was closely related to MBPS,standard deviation of SBP,variable coefficient of SBP,and SBPF (P<0.05).Multivariate linear regression analysis showed that MBPS,standard deviation of SBP and SBPF were the independent influencing factors of IMT,CrPWV and stiffness index β5.Conclusion BPV showed significant relationship with vascular elasticity in stroke patients,and reduction of BPV helps to delay the process of atherosclerosis.
