1.Preparation of Tetrandrine Solid Dispersion Tablets and Formulation Optimization

OBJECTIVE:To optimize preparation formulation of Tetrandrine solid dispersion tablets. METHODS:Tetrandrine solid dispersion tablets were prepared by direct compression method. Excipients were screened with single factor test. Taking disinte-gration time as index,the formulation of Tetrandrine solid dispersion tablets was optimized by orthogonal design using the amount of PVPP,lactose-microcrystalline cellulose proportion and the amount of gum acacia as factors. The optimized formulation was vali-dated. Prepared tablets were compared with Tetrandrine common tablet in dissolution rate,and the contents of the tablets prepared by optimized formulation were also determined. RESULTS:Optimal formulation was as follows as 9.5% PVPP as disintegrating agent,lactose-microcrystalline cellulose(1∶2)as filler,mixing,1% aerosil as lubricant,direct compression. For 3 batches of tab-lets,disintegration time were 79,81 and 78 s;contents were 98.66%,99.24%,99.85%;RSDs were 0.72%,1.16%,1.33%,re-spectively. Combined with Tetrandrine common tablets,the dissolution rate of prepared tablets had been improved significantly. CONCLUSIONS:Tetrandrine solid dispersion tablets are prepared successfully with rational reproducible formulation.

3.Application percutaneous transluminal angioplasty and stent in the treatment of subclavian artery stenosis

Objective To evaluate and compare the therapeutic results of subclavian artery angioplasty and stenting in patients with aortoarerteritis and atherosclerosis. Methods 13 cases (9 males and 4 females; age, 28-69 years) of subclavian artery stenotic occlusive disease were performed with PTA and stent. Results Six patients had aortoarerteritis, 7 had atherosclerosis lesions. Twelve of 13 stenoses could be successfully angioplasty ( n =6) and stenting ( n =6) . The one failure was due to the inability to cross the lesion with the guidewrie. Compared with atherosclerosis, higher balloon inflation pressure was required to dilate the lesions of aortoarerteritis. 12 patients were asymptomatic after PTA and stent implatation. Patients were follow up for 3-24 months and clinical examination demonstrated patency of the vessels with PTA and stents. Conclusion Subclavian PTA and stent implatation is safe and good clinical result as in aortoarerteritis as in atherosclerosis.

5.Differential expression of microRNA-125b in the neuronal differentiation of adipose-derived Flk1~+ mesenchymal stem cells

BACKGROUND:The differentiation of mesenchymal stem cells(MSCs)is strictly regulated by both genetic and epigenetic Factors .Emerging evidences have demonstrated that microRNA also plays an important role in this process.OBJECTIVE:To explore the differential expression of microRNA-125b during neuronal dliferentiation of Flk1~+ adipose-derived MSCs(AD-MSCs).MATERIALS:The fat samples were provided bv healthy female volunteers aged 15-35 years and recruited from the Plastic Surgery Hospital affiliated to the Chinese Academy of Medical Sciences.METHODS:Adult adipose tissues were obtained from healthy voluntears with age of 15-35 years .Using adherence method,Flk1~+ MSCs were obtained and the 3~(rd) passage cells were taken in the experiment.Cultured in neuronalinduction medium.these MSC were induced to differentiate towards neuronal lineage.The expression of microRNA-125b was examined at days 0,4,8 and 12.To explore its role in neuronal differentiation,we need to change its expression.RT-PCR and Taqman real-time PCR were carried out to explore the difierentially expression of microRNA-125b during neuronal differentiation of AD-MSCs.The effect of inhibitor on the expression of microRNA-125b was detected.RESULTS AND CONCLUTION:①The Flk-1~+MSCs were successfully induced into neuronal differentiation and displayed typical morphological changes 12 days after induction:Most cells retracted their cytoplasm ,fomling spherical cell body and emitted cellular protrusions.RT-PCR and immunocytochemistry studies confirmed their phenotype with expression of known neuronal cell markers including neurofilament,glial fibrillary acidic protein.②The expression of microRNA-125b was significant up-regulated during neuronal differentiation .Results of RT-PCR and Taqman real-time PCR were concordarlce with that of microRNA chip technology.③Inhibitor could down-regulate microRNA-125b.The results implied that microRNA-125b may play an important role in neuronal differentiation.
6.Interaction between water-soluble nano-CdS and gelatin

The interaction between water-soluble CdS and gelatin 5n aqueous solution was explored using fluorescence,infrared (IR)and UV-Vis spectra at pH 12.0 and different temperatures.Results show that the formed complex had a strong ability to quench the fluorescence launched from gelatin.The fluorescence quenching data were analvzed according to Lineweave-Burk double-reciprocal equation and the gelatin had reacted with CdS to form a complex.This kind of static fluorescence quenching goes with non-radiation energy transfer happening within single molecule According to Lineweave-Burk equation,K(285 K:1.07×10~4 L/mol;292 K:9 69×10~3 L/mol;299 K:8.06×10~3 L/mol),the formation constants of the compound at different temperatures and the thermodynamic parameters(△H=-14.18 kJ/mol;△G=-21.98/-22.28/-22.36 kJ/mol;△S=27.36/27.74/27.36 J/K·mol) at certain temperatures were obtained,indicating that the binding force between them is mainly the static in nature.The binding locality was an area of 4.09 nm away from tryptophan residue in gelatin based on F(o)rster's non-radiation energy transfer mechanism.The results provide information for exploring the chemical mechanism of interaction between nanoparticle and this kind of biological macromolecule.
7.Electrocardiographic Characteristics in Patients With Cardiac Amyloidosis

Objective: To study the diagnostic characteristics of cardiac amyloidosis (CA) by non-invasive electrocardiography (ECG) in relevant patients. Methods: We retrospectively analyzed 60 CA patients diagnosed in our hospital from 2008-08 to 2013-12 for their clinical and ECG characteristics. Results: There were 48 male and 12 female patients with the ratio of 4: 1. The ifrst time diagnosis rate was low and the average age for conifrmed diagnosis was at (54. 5±14. 2) years.①There were 32 (53. 3%) cases combining heart failure, 12 (20%) with pleural effusion, 20 (33. 3%) with atrial arrhythmia, 8 (13. 3%)with ventricular arrhythmia, 4 (6. 7%)with sino-atrial block, 15 (25%)with atrio-ventricular block, 4 (6. 7%) with left bundle branch block (LBBB), 5 (8. 3%)with RBBB and 8 (13. 3%)with intra-ventricular block.②There were 32 (53. 3%) cases with low voltage on limb leads, 52 (86. 7%) with pseudo-infarct pattern, 48 (60%) with ST-T abnormality and 30 (50%) combining low voltage on limb leads with pseudo-infarct pattern.③The patients combining pleural effusion and with pseudo-infarct pattern had the increased ratio of low voltage on limb leads, while there were still 22 (45. 8%) cases without pleural effusion had low voltage on limb leads.④ ECG characteristics for 60 CA patients were as follows: QRS duration (104±26) ms, QT interval (404±34) ms, QTc (462±35) ms; the R wave of avR 0. 17 mV, QRS wave 0.30 mV; the R wave of limb leads and V1-3 were all<0.5mV, the S wave of V1-3 were 0. 62mV, 1. 61mV, 1. 56mV; the R/S ratio of V1-3 were 0. 19, 0. 12, 0. 20 respectively. Conclusion: CA patients had the highest incidence of pseudo-infarct pattern; meanwhile, combining with low voltage on limb leads, pseudo-infarct with long Q or S wave and ST-T abnormality but normal QRS duration was helpful for differential diagnosis of CA in clinical practice.
8.The distribution of human papillomavirus in different age groups and its correlation with the degree of cervical lesions

Objective To investigate the distribution of human papillomavirus(HPV)in different age groups and its correlation with the degree of cervical lesions.Methods 2 031 patients with HPV screening specimens in our hospital from May 2012 to May 2015 were chosen as study subjects.To acquire patient cervical squamous columnar epithelial cells at the junction to detect HPV DNA types.HPV -positive patients used ultra -thin liquid -based cervical cytology technology (TCT)to detect.Patients with abnormal TCT detection were performed with electronic biopsy,and the diagnosis was made by pathology biopsy,and analyzed the distribution of HPV in different age groups and different degree of cervical lesions.Results In different age groups,≤20 years of age groups of patients with high -risk HPV positive rate was 19.7%,which was significantly higher than that of >20 -30 years,>30 -40 years and >40 -50 years three groups (14.4%,13.9%,15.0%)(χ2 =4.259,5.724,3.988,all P <0.05).There was no significant difference with the 50 years old group (17.1% )(χ2 =1.724,P >0.05).There was no significant difference in HPV positive rate among the ≤20 years,>20 -30 years,>30 -40 years,>40 -50 years,>50 years ages patients with high risk HPV infection group and low risk HPV infection group(χ2 =0.679,1.021,0.968,0.736, 0.668,all P >0.05).In the high -risk HPV infected group,the risk of single type infection in >20 -30 years,>30 -40 years,>40 -50 years,>50 years ages patients were significant higher than the mixed types infection(χ2 =4.213,3.894,4.256,5.330,5.666,all P <0.05).Infected patients of all ages with high -risk HPV were HPV16, HPV52,HPV58 type -based,low -risk type HPV43 type places mainly.More than 31 to 40 years age group of patients infected with high -risk HPV types in cervical lesions mainly cervicitis and 40 years and older group of patients with high -risk HPV infection incidence of invasive cervical cancer were significantly higher.Conclusion Different age groups of women for HPV genotyping is important for diagnosis of cervical lesions.
9.Internet Game Addiction and Influential Factors of Middle School Students in Guangzhou

Objective:To investigate the internet game addiction disorder and influential factors in the middle school students of Guangzhou.Methods:A total of 900 middle school students of Guangzhou selected by cluster sampling were investigated with unified questionnaires.Results:Of the middle school students tested,4.9%were diagnosed internet game addiction.There were remarkable differences of the addiction to network game in demography variable,such as age, educational background of father and family economic situation.Between online game addicts and non-addicts,there were significant discrepancies in social support,subjective support,use of support,self-identity,self investment today,and parent-child conflict.Gender,subjective support,parent-child conflict,self investment now,educational background of father and family economic situation entered the regression model.Conclusion:Internet game addiction is closely associated with social support,parent-child relationship,and self-identity.
10.Blocking NR2B Receptor Reversed the Process of Cytotoxicity to Spiral Ganglion Neurons Induced by Sodium Salicylate in Guinea Pig

Objective To investigate whether blocking NR2B receptor can reverse the process of cytotoxicity to spiral ganglion neurons induced by sodium salicylate in guinea pig by applying ifenprodil (a NR2B antagonist) at the round window niche .Methods Sixty healthy guinea pigs provided by the experimental animal center of Guangxi medical university were randomly and evenly divided into a control group (Group I ,no treatment) ,an APL group (Group II ,60μl APL directly applied to the round window ) ,a sodium salicylate group (Group III ,60 μl APL di-rectly applied to the round window and then be given intraperitoneal sodium salicylate injection ) ,and an ifenprodil group (Group IV ,60μl of 10μmol/l ifenprodil in APL directly applied to the round window and then be given intra-peritoneal sodium salicylate injection ) .Sodium salicylate was given at 400 mg · kg -1 · d-1 for 7 days .Auditory brainstem responses (ABRs) were recorded before animal sacrifice by decapitation .The left cochlea was removed and prepared for detection of caspase -3 expression in spiral ganglion neurons via immunohistochemistry .From each group ,6 cochleae were used to test apoptosis index in spiral ganglion neurons using the TUNEL technique .Results Before salicylate administration ,the ABR threshold was less than 40 dB SPL in all animals .After salicylate ad-ministration ,the ABR threshold was 33 .33 ± 5 .17 dB SPL in Group II ,64 .17 ± 7 .36 dB SPL in Group III and 49 .17 ± 5 .85 dB SPL in Group IV ,in contrast to 31 .67 ± 5 .16 dB SPL in Group I (controls) .The caspase -3 ex-pression was not changed obviously in Group I and Group II ,but was significantly changed in Group III and Group IV (P<0 .01) .The caspase-3 expression appeared to be decreased in Group IV compared to those in Groups III (P<0 .05) ,but still increased compared to those of in Group I and II (P<0 .05) .The apoptosis index among spiral ganglion neurons in Groups III and IV increased significantly compared to those of in Group I and II (P<0 .001) .It was however ,lower in Group IV than in Group III (P<0 .01) .Conclusion Blocking NR2B receptor with specificity can reverse the process of cytotoxicity to spiral ganglion neurons induced by sodium salicylate in guinea pig .
