1.Influence on quality of sputum sample of tuberculosis patients on detection of acid-fast bacilli and its nursing measure
Chinese Journal of Practical Nursing 2008;24(22):44-45
Objective To discuss the influence factors on detection of acid-fast bacilli for tubercu-lnsis patients and to seek for method to improve the quality of sputum sample. Methods A retrospective analysis was carried out in 3102 sputum sample of 1034 patients. Results The positve detection of acid-fast bacilli was found in 570 samples (18.4%). Pus sputum and bloody sputum had no evident influence on the positive detection rate(P>0.05).The other two kinds of sputum had statistical influence on the positive detection rate (P<0.01). Conclusions Attention to the intervention and instruction before collecting sputum sample proved to be important measure to ensure the quality of sputum sample and increase the positve detection rate of acid-fast bacilli.

Objective To establish and evaluate a gas chromatography-tandem mass spectrometry(GC-MS/MS)method for the detection of six common drugs(methamphetamine, meperidine, caffeine, codeine, cocaine and ketamine)in blood, and to improve the determination basis of results. Methods The above six drugs were added into the blank blood, and GC-MS/MS was used for detection after ether extraction. The collection, quantification and confirmation were carried out under the mode of multi-reaction monitoring(MRM).The qualitative results of the above six drugs were evaluated based on the maximum allowable deviation of the retention time and relative ion abundance ratio in the qualitative results of GC-MS/MS.Results There was a good linear relationship between the six common drugs, among which ketamine and caffeine had the lowest detection limit(0.01 μg/mL),methamphetamine had the highest detection limit(0.5 μg/mL).The retention time(RT)and relative retention time(RRT)of the target substance were stable under the six supplemental levels, and the absolute deviation(ΔRTabsolute)of RT was within±0.025 min.The absolute deviation of RRT(ΔRRTabsolute)was within±0.004.The relative ion abundance ratio absolute deviation(ΔIabsolute)is±20%,and the relative ion abundance ratio relative deviation(ΔIrelative)is±50%.Conclusion This study clarified the reference range for qualitative determination of six common drugs in blood matrix detected by GC-MS/MS,and effectively supplemented the qualitative determination indicators of existing instrumental analysis methods.

Objective To investigate the expression characteristics of S100 calcium binding protein P(Sl00P)and thymic stromal lymphopoietin(TSLP)in patients with breast cancer and their relationship with prognosis. Methods GEPIA database and bc-GenExMiner v4.3 were used to analyze the expression levels of S100P and TSLP in breast cancer tissues, and Kaplan-Meier Plotter was used to analyze effects of S100P and TSLP on patient prognosis. The protein interaction network analysis of S100P and TSLP were carried out by using STRING database. The co-expressed genes of S100P and TSLP were analyzed using Linked Omics database, and the role of S100P and TSLP co-expressed genes in related biological processes and signaling pathways were evaluated by gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis. The correlation between S100P and TSLP and breast cancer immunoinfiltration was analyzed by TIMER.Results The expression of S100P in breast cancer patients was significantly higher than that in normal population, and the expression of TSLP was significantly lower than that in normal population(P<0.05).The overall survival,disease-free survival and distant metastasis-free survival of patients with high S100P expression were significantly shorter than those of patients with low S100P expression(P<0.05).The distant metastasis-free survival of patients with high expression of TSLP was significantly longer than that of patients with low expression of TSLP(P<0.05).The results of protein interaction showed that S100P had a strong interaction with interleukin-11,and TSLP had a strong interaction with interleukin family proteins. The co-expressed genes of S100P and TSLP were mainly responsible for biological functions such as DNA replication and immune function.TSLP was correlated with breast cancer immune cell infiltration. Conclusion S100P and TSLP can be used as biomarkers to predict the prognosis of breast cancer, and they can also help to evaluate the tumor immunoinfiltration in breast cancer patients, and TSLP could provide theoretical basis for predicting the prognosis and tumor immunotherapy in breast cancer patients.
