Object To study the effect of rare-earth elements La3+ on growth of hairy roots and normal roots of Rheum palmatum L and their yield of anthraquinones. Methods The biomass and an- thraquinone yield of two hairy clones DH5c, DH7a, and normal root NOR cultures of R. palmatum were evaluated statistically after various concentration of La3+ were administrated into culture medium. Results Significant differences of biomass yield and anthraquinone yield were shown among six La3+ concentrations, in which 10 mg/L showed obvious inhibitory effect for root growth and 1. 0 mg/L was the best for anthraquinone yield. In general, aloe-emodin and rhein were predominant in five anthraquinones after treated with the rare-earth element. Conclusion The growth of hairy root cultures of R. palmatum and their anthraquinone yield are greatly promoted by addition of La3+ to the medium.

Objective To investigate the chemical constituents of red algae Hypnea charoides. MethodsThe constituents were isolated by column chromatography and their structures were elucidated by chemical properties and spectroscopic analyses. Results Three compounds were isolated and their structures were identified as 3, 6-dimethyl-8-(4-methyleneheptan-3-yloxy) octane-1-amine (Ⅰ), palmitate-K (Ⅱ), and (2R, 1′S, 2′S, 3′R)-N-(1′-hydroxymethyl-2′, 3′-dihydroxy-heptadecenyl)-2-hydroxy-12, 13-methylene-tetracosanamide (Ⅲ). Conclusion Compounds Ⅰ and Ⅲ are new compounds named as charoidesine and charoidesamide.

Objective To study the effects of phytohormones and elicitors on the growth and anthraquinone production of Cassia obtusifolia hairy roots. Methods Blades and petioles of C. obtusifolia〖WTBZ〗 were infected with Agrobacterium rhizogenes LBA9402 and the in vitro culture system of the hairy root was established. Results Hairy roots were induced by A. rhizogenes LBA9402. The results showed that the addition of 0.5% NAA to the hairy roots increased the content of five kinds of anthraquinones by four times, and Aspergillus niger elicitor had no much effect and JA had slightly inhibitory effect on root growth, but both of them significantly promoted the anthraquinone accumulation, the anthraquinone yield increased more than two times with the treatment of 2.0 mL elicitor/50 mL medium and by 73% with the treatment of 10 ?mol/L jasmin acid (JA), respectively. JA had little inhibitory effect on the growth of fairy roots. Conclusion The fundation has been established for further oplimizing the proper cultural system for C. obtusifolia hairy roots and regulating the secondary melabolite.

Objective To study the effect of optimized media and clones on growth and anthraquinone yield in hairy root cultures of Rheum palmatum. Methods The four monoclones and four media were optimized statistically for studying the effect on the biomass accumulation and five anthraquinones yield in hairy root cultures of R.palmatum.Results Significant differences in biomass were shown among various clones and media,in which monoclone DH7a was the highest and WP medium was the best.Chrysophanol and physcion were predominant among five free anthraquinones in hairy root cultures of R.palmatum.Notable differences of total five free anthraquinones yield were observed among different clones, in which DH7a was the highest.Reciprocity analysis showed that the highest yield of five free anthraquinones is in the combinations of DH7a with B5 medium.DH5a less and DH5a the least with WP medium,DH7a and B5 medium were predominent for the yield of above five free anthraquinones with exception of DH5c which has no significant difference in four media. Conclusion The study lays the foundation for mass production of anthraquinone by hairy root culture of R.palmatum.

Objective To investigate the biotransformation of podophyllotoxin by cell suspension culture and root culture systems of Rheum palmatum. Methods Plant tissue culture technology was employed. The products were isolated by silica gel column chromatography and their structures were elucidated by spectral methods. Results Adding of podophyllotoxin had no obvious effects on the pH value of cell suspension culture and root culture systems of R.palmatum.In the case of present study,73.8% of podophyllotoxin was converted by cell suspension culture and 56.3% by root culture. The products were different between the two culture system.Podophyllotoxin was converted into picropodophyllin with cell suspension culture system and many other products besides epipodophyllotoxin with root culture system. Conclusion Both of cell suspension culture and root culture systems of R.palmatum are able to convert podophyllotoxin.

Objective To measure the mRNA expression of interleukin receptors (IL-2R?IL-4R and IL-10R) in peripheral blood mononuclear cells (PBMCs) from patients with chronic idiopathic urticaria (CIU). Methods Thirty CIU patients and 30 controls were enrolled in this study. PBMCs were separated from the peripheral blood specimens. Reverse transcription-polymerase chain reaction (RT-PCR) technique was applied to semi-quantitatively analyze the mRNA expression. Results The expression level of IL-10R mRNA was significantly increased in patients with CIU than that in the healthy controls, while that of IL-2R and IL-4R mRNA in PBMCs showed no significant difference. Conclusion Our results suggest that IL-10R might be involved in the pathogenesis of CIU.

Objective: To explore the changes of pulmonary function in type 2 diabetes and its related factors. Methods: The pulmonary functions of 72 patients with type 2 diabetes and 22 healthy objects were measured, and the related factors, such as duration of DM,HbA1c,BMI,were analysed with linear regression analysis.Ninteen of them underwent a 12-week-long intensive insulin therapy and pulmonary function tests before and after the treatment. Results: Vital capacity(VC),forced vital capacity(FVC),forced expiratory volume in the first second(FEV1),total lung capacity(TLC) and carbon monoxide diffusion in the lung(DLco) were significantly decreased in the diabetes patients.Correlation analysis revealed that DLco was negatively correlated with the duration of DM.which was shown by linear regression analysis to be the only significant predictor. After a 12-week-long intensive insulin therapy,DLco and DLco/VA decreased significantly. Conclusion:Patients with type 2 diabetes have abnormal pulmonary ventilatory function and impaired pulmonary diffusive function, and the latter is related to the time of hyperglycemia, and can not ameliorated by short-term glycemic control.

Objective To explore the clinical application value for calculating the volume of splenic embolization by splenic artery branch diameter ratio method. Methods Data of 20 patients with cirrhosis and hypersplenism who underwent partial operation of splenic artery embolization were retrospectively analyzed.Based on the angiography of splenic artery during operation, the diameter of branch vessels of splenic artery was measured and the percentage of embolism volume in spleen was calculated.CT scans were performed on all patients after one month of the operations.Based on VR reconstruction image, the percentage of embolism volume in the spleen was calculated.Postoperative complications and adverse responses were observed.Peripheral blood red cells, white blood cells and platelets were tested repetitively three days before the operation, and a week,a month, three months after the operation. Moreover, the statistical analysis was performed. Results The difference between the percentage of embolism volume in the spleen calculated by splenic artery branch diameter ratio method ([52.15±3.29]%) and calculated by CT ([49.99±6.02]%) was not statistical (t=-1.630,P=0.120).All the patients had moderate or below moderate pain on left upper quadrant, and had symptoms of nausea, vomiting and fever after the operation. They all got better after symptomatic treatment.Peripheral blood red cells, white blood cells and platelets all had significant differences among three days before the operation,a week,a month and three months after the operation (all P<0.001).Conclusion During the partial splenic embolization, the application of ratio method on branch vessels of splenic artery to measure the volume of embolism in the spleen is convenient, prompt and relatively accurate. It is worthy to be expanded in clinic.
