1.Improvement of Amplification Method for Cryptosporidium parvum Oocysts from Mice

Objective To establish a simplified amplification method for obtaining a large number of purified Cryptosporidium parvum oocysts from infected C57BL/6N mice. Methods All mice in the experimental groups were immunosuppressed by given different concentrations of dexamethasone phosphate added in drinking water throughout the experiment. The recovery and purity of the oocysts obtained using different purification methods was compared. The infectivity of the oocysts obtained from the same origin but different animals and different purification methods in a bovine fallopian tube epithelial cell culture system was studied. Results 4.16?10 9 oocysts were obtained in 30 mice in the 3rd group with dexamethasone of 20 ?g/ml in drinking water. No significant difference in the oocyst recovery, purity and infectivity was found between methods using saturated saline floatation and sucrose density gradient centrifugation. The infectivity of the oocysts obtained from the same origin but different animals was similar. Conclusion A simplified amplification method for obtaining a large number of purified Cryptosporidium parvum oocysts from the infected mice was established.
2.The protective effects of pinacidil pretreatment on ischemic myocardium during cardiopulmonary bypass

Objective To evaluate the myocardial protective effect of pretreatment with pinacidil, a potassium channel opener, on myocardium perfused intermittently with normothermic or hypothermic high potassium crystalloid cardioplegie solution during cardiopulmonary bypass ( CPB) . Methods Eighteen dogs of both sexes weighing 10-15 kg were intubated and mechanically ventilated after induction of anesthesia. Femoral artery and vein were cannulated for direct MAP and CVP monitoring. Swan-ganz catheter was placed via right internal jugular vein. The animals were randomly divided into 3 groups: group A (hypothermic CPB) in which heart was perfused with 4℃ St Thomas solution 10 ml?kg-1 in 2 min and 30 min later perfused again with half of the initial volume(5 ml? kg-1); in group B (normotherinic CPB) and group C (hypothermic CPB) heart was pretreated with oxygenated pinacidil solution (0.083 mg?kg-1 37℃) before cardioplegia. The heart was subjected to 60 min global ischemia followed by 30 min reperfusion. The hemodynamic changes were measured before aortic cross-clamping and 15 and 30 min after release of aortic cross-clamping. Myocardial tissue was obtained from apex of left ventricle for determination of myocardial adenine nucleotide content 5 min after CPB was initiated, 30 and 60 min after aortic cross-clamping and 20 min after reperfusion. Objective:To establish early warning mechanism for enhancing the using effect of multimedia and guaranteeing quality of teaching so as to satisfy the requirement of morphology course for definition of multimedia in medical college.Methods: The archive of each multimedia equipment was established and the service times of main components and parts which existed fault were analyzed, and the anticipative occurrence time of fault was pre-deduced. And the early prevention and early detection were implemented in the early warning mechanism.Results: The each multimedia equipment could achieved optimal situation for usage, and got rid of the poor effect when equipment was normal.Conclusion: The warning management of life cycle can fast decrease fault rate and effectively reduce the maintenance cost. Therefore, the mechanism can guarantee the teaching quality of medical college and enhance the satisfaction of teachers and students.
4.The influence of two transfusion triggers on middle-and old-aged patients during tumorectomy

0.05].Conclusion To the middle-and old-aged GI cancer patients without organ dysfunction,lowering blood transfusion trigger from Hb 100 g/L to Hb 80 g/L during operation can meet the requirement of oxygen,reduce blood transfusion,and do not increase the rates of wound infection or delay wound healing.

Objective To study the curative effect of noninvasive ventilation on the patients of acute serious bronchial asthma complicated with pneumothorax. Currently, many studies illustrate that LZTS2 gene, the important candidate tumor suppressor gene, is already involved in the inhibition of tumorigenesis and aberrant proliferation of tumor cells, and other functions of tumor cells. 7.Analysis on application effect of scale score of treatment compliance of pulmonary tuberculosis patients in Guizhou province

Objective To verify the accuracy of ARS scale.Methods A cohort study was conducted in 10 counties (city)the in-troduction of the ARS scale from 2013 November to 2014 May (6 months),the registrations were undertake score,and score < 4 were compliance group,more than 4 cent were nonadherence group.Follow-up observation include review at the end of 2nd month after treatment and medicine compliance.Results There was no significant difference in age,gender,nationality and whether the floating population between the compliance group and non compliance group (P >0.05),initial treatment and retreatment was sta-tistically significant (P <0.05).1 824 patients had a mean score of 3.40±1.40,including compliance group mean score was 2.34± 0.73,is not in compliance with group mean score was 4.69±0.81.Mainly concentrated in the 2-5 scores accounted for 86%.The ARS score of 2 at the end of the packet and judge review the results of test,the Kappa coefficient was 0.403 (P <0.05).The sensi-tivity was 71.1%,specificity was 70.2%,the correct index 0.41.Multivariate Logistic and review the compliance of patients re treatment ARS score,at the beginning of the results of regression analysis showed that ARS score,under the same conditions,re-treatment review compliance was initially treated patients with difference (OR=0.41,P <0.05);in the initial treatment and retreat-ment premise under the same circumstances,review of compliance with the ARS score less than 4 points.It′s 5.65 times greater than or equal to 4 points (P <0.05).Conclusion The ARS scale can accurately judge the patient compliance,with high sensitivity and specificity,suitable for application in Guizhou Province,but we should consider other interference factors of promotion.
8.The interaction between Cyclophilin A and CD147 and its clinical significance in periodontal diseases

Cyclophilin A (CypA) is the first foldable enzyme in human cells that has peptidyl proliferase-trans isomerase activity and has a strong proinflammatory effect. CD147 can act as the signal receptor of CypA. The interaction of the two through cell-surface heparin binding activates extracellular regulated protein kinases (ERK1/2) and nuclear factor kappa-B (NF-κB) signaling pathways in macrophages and increases the expression of MMPs and other inflammatory factors. The CypA/CD147 interaction regulates inflammation, promotes the inflammatory response and bone resorption and is involved in the pathological processes of a variety of systemic diseases. CypA and CD147 may take part in the chemotaxis of inflammatory cells, increase white blood cell infiltration in tissues, and increase CypA and CD147 expression in periodontitis gum tissue and gingival groove liquid with inflammation, prompting their interaction to promote the progression of periodontitis. However, the specific function of the signaling pathways in the periodontitis mechanism still requires further elucidation.
9.CT findings of invasive mucinous adenocarcinoma of the lung

Objective To analyze the CT findings of invasive mucinous adenocarcinoma of the lung.Methods The CT findings of 7 patients with invasive mucinous adenocarcinoma of the lung were retrospectively analyzed. Results Six cases manifested isolated pulmonary nodules or masses, among which circular/round shape was in 6 cases,clear tumor-lung parenchyma interface was in 6 cases,lobular sign was in 5 cases,specular sign was in 6 cases,calcifications was in 3 cases,angiogram sign of contrast enhancement was in 4 cases, and the CT value was close to the liquid density in 6 cases. One case showed consolidation of lung with multiple nodules or mass, and the lesions had angiographic signs calcification, unenhanced area with water density as well. The CT findings of angiography signs, calcification and consolidation of regional focal liquid opacity had significance for the diagnosis.One case was misdiagnosed as tuberculosis and 1 case as hamartomas. Factors which caused misdiagnosis were irregularly shaped calcifications and angiographic signs in lesion, and the enhanced CT value was less than 20 HU. Conclusions The invasive mucinous adenocarcinoma of the lung has characteristic CT findings. In addition to the malignant tumor features. When plain CT scan finds the watery density, calcifications and angiographic signs in the lesion, the invasive mucinous adenocarcinoma of the lung should be considered.The enhanced CT scan shows no enhancement or enhanced value less than 20 HU is also the characteristic of invasive mucinous adenocarcinoma of the lung. MRI check will help us to improve the diagnostic accuracy.

Objective Recent evidence has implicated Larp1,an RNA-binding protein,in cell motility and EMT therefore prompting the study of Larp1 in EOC.This project aims to examine the potential role of Larp1 in cell invasion.Methods Ovarian cancer cells SKOV-3 and OVCAR-3 were transfected with DNA constructs to overexpress Larp1 and transfect cells were used to assess the overexpression of Larp1 in cell invasion and metastasis using several functional assay.Results Larp1 overexpression in both OVCAR-3 and SKOV-3 cells appear to enhance the invasive ability of cells and the accumulation of Larp 1 in these chemoinvasive protrusions strongly suggests the potential role of Larp1 in invadopodia formation.Cell spot chemoinvasion assay demonstrated increased chemoinvasion of Larp1 overexpressing SKOV-3 and OVCAR-3 cells towards a gradient of TGF-β,EGF and bFGF.Conclusions With its potential role in EMT and cell invasion,a deeper understanding of the physiological role of Larp1 in cancer cell motility will be potentially beneficial in the diagnosis and treatments of ovarian cancer.
