1.Comparison of non-specific esterase activity in different strains and gender of Culex pipiens pallens

To explore the diversity of non-specific esterase activity in different strains of adult male and female Culex pipiens pallens,the NSE activity of single mosquito was determined by using the ?-naphthalene acetic acid ester (?-NA) as the substrate and solid blue B salt (fast blue B salt) as the developer. The results showed the NSE activities of anti-DDVP (Rd) and anti-Propoxur (Rp) strains of Culex pipiens pallens were significantly higher than that of sensitive (S) strains,and the NSE activity of anti-Cypermethrin (Rc) strains of Culex pipiens pallens was similar to that of S strain. As for the S,Rd,Rp and Rc strains of 3-day-old adult male and female Culex pipiens pallens,the NSE activity of female mosquito was higher than that of male mosquito.
2.Drug Resistance Evolution of Dichlorvos-Resistant and Cypermethrin-Resistant Strains of Culex pipiens pallens

Susceptible strain of Culex pipiens pallens was selected with dichlorvos and cypermethrin for 42 generations in the laboratory. At generation 42, the resistance level to dichlorvos and cypermethrin was 12.2-fold and 534.3- fold, respectively, in comparison to their parent generations. The dichlorvos-resistant and cypermethrin-resistant strains were then conventionally bred for another 20 generations, the resistance level to dichlorvos and cypermethrin decreased to 6.1- fold and 83.3-fold, respectively. The results indicated that the two resistant strains showed different development of resistance.
3.Clinical observation of memory impairment in patients with cereberal cysticercosis

Objective To investigate the characteristics of memory impairment in patients with cereberal cysticercosis and analyze its influence factors. Methods 62 patients with cereberal cysticercosis and S3 normal controls by age, gender, education and occupation using accepted the Clinical Memory Test. Memory score was compared to observe the contrast to patients with cerebral cysticercosis lesions in the left and right groups in different from memory effects. Results There were significant lowering in pointing memory( 12. 29 ±7.41) ,association learning(15.37 ±7.29) ,free picture recall( 11.37 ±7. 35),insignificant figure recognition( 17.72 ±6.27),portrait character association recall(14.28 ± 6.39 ) and memory quotients(71.04 ± 19.48) when that of the normal controls(P<0. 01 ) , pointing memory and free picture recall were the lowest among the five items. There were some significant differences between patients with cerebral cysticercosis in the left and the right of lesion group a-bout different memory contents(P<0.05 ~0.01), and association learning was the lowest in the left lesion group, insignificant figure recognition was the lowest in the right lesion group. Conclusion Cereberal cysticercosis could cause memory impairment. There were some significant differences between the left and the right lesion group about different memory impairment.
4.Enhancing cytotoxicity of Mitoxantrone on human breast cancer cell with microbubble contrast during low-frequency ultrasound exposure

Objective To investigate the antitumor effect and its mechanism of microbubble contrast combined with Mitoxantrone exposed to low-frequency ultrasound on human breast cancer cell MCF-7. Methods MTT method was applied to examine the growth inhibition of MCF-7 treated with Mitoxantrone. MCF-7 cells were randomly divided into 4 groups:Mitoxantrone group (group D), ultrasound+Mitoxantrone group (group U+D), ultrasound+microbuble +Mitoxantrone group (group U+M+D) and control group (group C). The cytoactive of each group was examined with MTT. The intracellular drug content in each group was measured with high performance liquid chromatography. The morphology of MCF-7 cells apoptosis was observed with transmission electron microscopy (TEM). Results The IC_(50) of Mitoxantrone was 2.87 μg/ml. The differences of cytoactive among all groups were significant (P<0.05). The intracellular drug content of group U+M+D was higher than that of group U+D, and the latter was higher than that of group D. The morphological changes of apoptosis were observed with TEM. Conclusion Low-frequency ultrasound can promote intracellular drug content as to enhance the sensitivity of chemotherapy drugs on tumor cells, and this effect can be enhanced by microbubble contrast exposure to low-frequency ultrasound.
5.Antitumor activity and immunological mechanisms of recombinant BCG

Objective:To study the antitumor activity and immunological mechanisms of rBCG including GM-CSF and EB virus LMP2A gene fusion.Methods: Animal models of EB virus-positive tumors was built.The formation time of tumors in mice,survival time,tumor weight was analyzed to detect rBCG anti-tumor activity;ELISA method was used to detect the specific antibodies which was produced in the mice stimulated by rBCG,specific CTL killing effect was detected by lactic dehydrogenase assay,ELISPOT was used to assay the secretion of IFN-γand flow cytometry, HE staining of tumor tissue was used to detected lymphocyte infiltration in mice immunized with recombinant BCG.Statistical methods were used for rBCG immunization effect preliminary analysis and evaluation.Results:Comparing to other control,tumor formation time was significantly delayed and tumor growth was slow, survival time of mice prolonged .ELISA test results showed that the rBCG immunization groups of mice could produce specific IgG antibodies of GM-CSF and LMP2A.Specific CTL activity was detected in mice immunized with rBCG.IFN-γsecretion was detected by ELISPOT method, flow cytometry and morphological observation detected tumor tissue infiltration of lymphocytes growth in mice immunized with rBCG.Conclusion:The rBCG induced a humoral and cellular immune responses and induced C57BL/6 mice to produce a strong anti-tumor effect and the EB virus-positive tumor cells was significantly inhibited.
6.Bioeffects of canine myocardium under microbubble destruction via diversity of ultrasonic intension

Objective To explore the bioeffects of canine myocardium under microbubble destruction via diversity of ultrasonic intension, in order to optimize ultrasonic intension for experiments. Methods Nine mongrel dogs were randomly divided into 3 groups. Ultrasound (1 MHz) in diversity of different intension (0.5 W/cm~2, 1.0 W/cm~2, 2.0 W/cm~2) was applied to expose canine myocardium after intravenous injection microbubbles of 2.0 ml. All the dogs were killed after being exposed for 5 min. The myocardium was harvested for HE staining and observed with transmission electron microscope for the tissue microstructures. Results The myocardium of hyperemia, disfiguration and necrosis wer observed in all groups. Myocardial edema but not hemorrhage appeared with 0.5 W/cm~2 , mild myocardial hemorrhage and slight inflammatory cell infiltration happened with 1.0 W/cm~2, whereas obvious hemorrhage and certain degree of inflammatory cell infiltration occurred with intension of 2.0 W/cm~2. With the augmentation of ultrasonic intension, myocardium trend to aggravate. Conclusion Ultrasound of diversity intension can induce different bioeffects of canine myocardium. Ultrasound mediated microbubble destruction with the intensity of 1.0-2.0 W/cm~2 can provoke a certain degree of inflammatory reaction with mild myocardial damage.
7.Experimental study on cross-resistance of Culex pipiens pallens to 3 kinds of chemical pesticides

Objective To understand the cross-resistance of Culex pipiens pallens to common pesticides,so as to provide the evidence for improving the application of chemical pesticides. Methods The IV instar larvae of DDVP-resistant,propoxur-resistant and cypermethrin-resistant strains as well as the sensitive strain of Culex pipiens pallens were collected to detect the re-sistance to DDVP,propoxur and cypermethrin based on the WHO bioassay method. Results The resistance coefficients of DDVP-resistant strain to DDVP,propoxur and cypermethrin were 14.47,8.96 and 207.27 respectively. The resistance coeffi-cients of propoxur-resistant strain to DDVP,propoxur and cypermethrin were 3.27,6.93 and 8.65 respectively. The resistance coefficients of cypermethrin-resistant strain to DDVP,propoxur and cypermethrin were 2.93,1.61 and 501.11 respectively. Con-clusion The resistance and cross-resistance could be generated during the long-term application of a single kind of chemical pesticide,and we should pay more attention to the varieties and dosages of them.
8. The research of rBCG inhibiting function on EB virus positive tumors

Objective:
The inhibition function on EB virus positive tumors with rBCG (recombinant BCG) was researched.
Methods:
After cancer models of EB virus positive tumor cells(GT39)were established in C57BL/6 mice, the mice survival conditions, tumor weight and tumor formation time were analyzed, flow cytometry was used to research the proliferation of CD4+ T. Inhibition of rBCG to cancer was researched, HE staining of the mice tumor tissue was used to detect and analyze lymphocyte infiltration. Single factor analysis method of variance (One-way ANOVA) was processed for rBCG′s inhibition evaluation by SPSS 11.0 statistical software.
Results:
Compared with the control group, rBCG group significantly slowed tumor growth, tumor formation time was delayed, rBCG significantly prolonged survival time of the mice.
Conclusions
rBCG had inhibition to EB virus positive tumors in mice.
