DNA methylation is one of the regulators on the expression of genes,and the expression of genes can be ”silenced” by hypermethylation of the tumor suppressor genes,which is in close relationship with carcinogenesis.Gastric cancer is caused by multiple factors and multiple genes alteration and prolonged interaction.among them ,There is considerable relation between the hypermethylation of the tumor suppressor genes and the gastric carcinogenesis.
2.Combination of docetaxel and capecitabine in the neoadjuvant chemotherapy of breast cancer

Background and purpose:Neoadjuvant chemotherapy has become a major therapeutic strategy for the treatment of locally advanced breast cancer.Randomized trials comparing this approach to adjuvant chemotherapy have demonstrated that neoadjuvant chemotherapy was as efficacious as adjuvant chemotherapy and allowed for a higher rate of breast conservation.This study was to evaluate the efficacy and toxicity of docetaxel plus capecitabine in the neoadjuvant chemotherapy of locally advanced breast cancer(LABC) .Methods:Fifty-two patients with LABC were treated with docetaxel 75 mg/m2,d1;Capecitabine 2 000 mg/m2,d1-14.The chemotherapy was repeated every 3 weeks.Effi cacy and toxicities were reviewed after 3 to 4 cycles of chemotherapy.Results:The CR+PR rate was 80.7%.A pathological complete response in the breast was achieved in 5.8% of patients after 4 cycles of docetaxel/capecitabine.The adverse reactions were neutropenia,alopecia and hand-foot syndrome.Conclusion:The combination of docetaxel and capecitabine was well tolerated and effective in LABC for neoadjuvant chemotherapy.
3.Docetaxel in Combination with Xeloda in the Treatment of Advanced Gastric Cancer

OBJECTIVE:To evaluate the efficacy and toxicity of docetaxel plus xeloda in the treatment of advanced gastric cancer. METHODS: Forty patients with advanced gastric cancer were treated with docetaxel 75 mg?m-2 on day 1 and xeloda 2 000 mg?m-2 (in two divided oral doses) for the first 14 days. The chemotherapy repeated every 3 weeks. Efficacy and toxicities were reviewed after 3 to 4 cycles of chemotherapy (1 cycle = 21 days). RESULTS: Among the 40 evaluable patients, 1 (2.5%) showed complete remission, 23 (57.5%) partial remission, 13 (32.5%) stable and 3 (7.5%) disease progression, and the overall response rate was 60.0%. The main adverse reactions were myelosuppression, alopecia, diarrhea, and hand-foot syndrome. CONCLUSION: The combination of docetaxel and xeloda is well tolerated and effective in the treatment of advanced gastric cancer.
4.The expression of Twist in invasive ductal carcinoma of breast and its relationship with cell proliferation and angiogenesis

ObjectiveTo study the expression of Twist in invasive ductal carcinoma of breast and its relationship with cell proliferation and angiogenesis.MethodsThe expression of Twist and Ki-67 was detected in 60 cases with invasive ductal carcinoma of breast by immunohistochemistry.Ki-67 index and microvascular density(MVD) were calculated.ResultsThe positive expression rate of Twist was 56.7% (34/60) in invasive ductal carcinoma of breast.Ki-67 index and MVD in the patients with positive expression of Twist was higher than those in the patients with negative expression of Twist[(57.05 ± 16.37)% vs.(25.32 ± 16.16)%,(34.30 ± 12.25)% vs.(23.04 ± 10.45)%,P< 0.05 ].ConclusionsThe overexpression of Twist is found in invasive ductal carcinoma of breast.To stimulate cell proliferation and angiogenesis may be one of the pathways of Twist to contribute to the invasion and metastasis of breast cancer.
5.Expression of COX-2 in invasive ductal carcinoma of breast and its relationship with cell proliferation and angiogenesis

Objective To study the expression of eyelooxygenase-2(COX-2)in invasive duetal carcinoma of breast and its relationship with cell prolireration and angiogenesis. Methotis Expression of COX-2 and Ki-67 was detected in 52 cages of breast cancer tissue by immunohistochemistry. CD34 marking vascular endothelial cell was used to measure the miemvascular density(MVD).Results The positive COX-2 expression was 57.7%in invasive duetal carcinoma of breast.The Ki-67 index was (56.07±17.37)%in COX-2 positive group while(27.32±16.28)%in the negative group,and the MVD W88 32.46±12.59 in COX-2 positive group while 25.04±10.48 in the negative group.The positive group of COX-2 protein had higher Ki-67 and MVD than that in negative group(P<0.05).Conclusion The overexpression of COX-2 protein Were found in invasive ductal carcinoma of breast.To stimulate cell prolireration and angiogenesis may be one of the pathways of COX-2 protein to contribute to the invasion and metastasis in human breast cancer.
6.Intra-operative radiotherapy in breast-conserving surgery of early breast cancer patients

Objective To evaluate intra-operative radiotherapy after breast conservative surgery in early breast cancer patients in terms of postoperative complications,cosmetic outcome and recurrence events.Methods From June 2007 to Dec 2010,115 early breast cancer patients received breast conserving surgery.Fifty-nine patients (study group) received intra-operative radiotherapy,compared with 56 patients (control group) receiving routine postoperative radiotherapy.Postoperative complications were evaluated 1 month after surgery; cosmetic outcome was evaluated 1 year postoperatively; recurrence and death events were followed up.Results The average wound healing time was 13 -22 days in study group and 9 - 14days in control group.In the study group,2 patients developed fat deliquescence,16 patients showed wound edema while no such side effects were found in control group.No infection or hematoma were found in either group.Overall cosmetic outcome was rated 1 year post operation.In the study group (41 cases),36 patients were graded as excellent or good,5 patients were as fair or poor.Meanwhile in the control group (37 cases),wounds in 25 patients were graded as excellent or good,that in 12 patients were as fair or poor (P =0.031).After a follow-up from 3 to 42 months(median:24 months),two patients (3.39%) in study group developed local cancer relapses,one of them( 1.7% ) died.In control group,one patient ( 1.8% )developed local relapse,and no one died.Conclusions Intra-operative radiotherapy is safe and reliable with good cosmetic outcome.

Objective To study the correlation between expression of COX-2,Ki-67 and neoadjuvant chemotherapy in breast cancer.Method COX-2 and Ki-67 were examined by immunohistochemical staining in 48 breast cancer samples.Results The overall response rate and clinical benefit rate to neoadjuvant chemotherapy were 70.8% and 95.8%,respectively.The expression of COX-2 and Ki-67 after the chemothempy [41.7% and (33.23±18.11)%] was significantly lower than those in prechemotherapy [62.5% and (46.81±23.17)%],P<0.05.Ki-67 index Was higher in COX-2 positive tumom than that in the COX-2 negative ones before and after neoadjuvant chemotherapy,P<0.01.The effect of neoadjuvant chemotherapy had a significantly negative correlation with COX-2 expression.Patients with high expression of Ki-67 were more likely to respond to treatment.Conclusion The expression of COX-2 and Ki-67 as molecular markers could be a guide for chemotherapy and prediction for neoadjuvant's response to chemotherapy in breast cancer.
8.New designed HMBA agents as inducers of erythroleukemia cell differentiation.

OBJECTIVESearching for more potent and less toxic HMBA-related agents.
METHODSHuman erythroleukemia cell K562, murine erythroleukemia cell (MEL) and its sub-line MEL DS19 were used as target cells to select a cell line which is the most sensitive to HMBA, then analyzed the activity of inducing differentiation of two new designed HMBA derivatives: HMBPA [hexamethylenebi (3-pyridin) amide] and Co-HDTA (ethylenediaminetetra acetic acid cobalt) using cell biology, cytochemical and molecular biology techniques.
RESULTSWe found that the MEL DS19 cells were most sensitive to HMBA (benzidine positive, B+ approximately 76%). Co-HDTA can inhibit the growth of MEL DS19, but induces differentiation just in a small population (B+ 2% approximately 4.5%). Between 0.02 approximately 5 micromol/L, HMBPA induces 3% approximately 8% cells committed to differentiation with little inhibition of cell proliferation. 1 micromol/L HMBPA and 2 mmol/L HMBA together, can obviously increase the percentage of differentiated cell (B+ approximately 72%), inhibit DNA synthesis and accelerate beta-globin transcription.
CONCLUSIONThe new HMBA derivatives may provide potential cancer differentiation inducers.

9.Reliability and validity of Chinese version of the neuropsychiatric inventory

Objective To assess the reliability and validity of the Chinese version of the neuropsychiatric inventory (CNPI). Methods The CNPI was administered to 219 caregivers of patients with Alzheimer's disease (AD). Each caregiver was retested 4 weeks after initial testing. Results The Cronbach a coefficient of the total symptom scale was 0.69. The Cronbach α coefficient of the total caregiver distress subscale was 0.72. The Cronbach α coefficient of the entire inventory was 0. 82. The test-retest coefficients ranged from 0.66 to 0.98 (P < 0.01). Principal axis factoring analysis of the symptom subscale yielded a five-factor solution which contributed to 67.0% of the cumulative variance. Factor 1, which included aberrant motor behavior, hallucinations, delusion and irritability had the most significant contribution to the cumulative variance. Principal axis factoring analysis of the caregiver distress subscale also yielded a five-factor solution which contributed to 70.2% of the cumulative variance. Factor 1, which included depression, delusion, sleep/night behavior, aberrant motor behavior, and irritability had the most significant contribution to the cumulative variance. Conclusion This Chinese version of NPI is a reliable and valid tool for measuring neuropsychiatric disturbances in patients with AD.
