[Objective]To develop an HPLC method for the determination of berberine hydrochloride in GuZheNing liniment.[Methods]The chromatographic system consisted of Dimonsil C18 column and mobile phase of acetonitrile-50mmol?L-1 sodium dihydrogen phosphate(adjusted PH to 2.5 with H3PO4)(25:75).The flow rate was 1.0 mL?min-1,and the detective wavelength was at 348 nm.[Results]The calibration curve was linear in the range of 0.106~0.530 ?g(n=5).The method was proved to be repeatable with RSD 0.7%(n=6).4 batches of sample were analyzed and the Berberine Hydrochloride contents were 19.11,18.81,18.83,18.92mg/100mg,respectively.[Conclusion]The method is selective,simple and accurate,and could be used for the quality control of this preparation.

3.Detections of interleukin-2,-4,-10 and interferon-? secreting cells in myasthenic thymus and peripheral blood

Objective To study the immune function of the subpopulation of T helper lymphocyte(Th), including Th1 and Th2 cells in myasthenic thymus and peripheral blood (PB).Methods Enzyme-linked immunospot (ELISPOT) method is adopted to detect the acetylcholine receptor (AChR)-reactive interleukin-2(IL-2)-, interleukin-4 (IL-4)-, interleukin-10 (IL-10)-, interferon-gamma(IFN-?)-secreting cell numbers in thymus and peripheral blood (PB). Results The AChR-reactive IL-2-,IL-4-,IL-10-, IFN-?-secreting cell numbers were increased in thymus as well as the AChR-reactive IL-10-, IFN-?-secreting cell numbers in PB. There is a positive correlation between thymus and PB in MG for the AChR-reactive IL-4-, IL-10-secreting cell numbers. After treatment, the AChR-reactive IL-2-,IL-4-, IFN-?-secreting cell numbers were decreased in PB, while there were 1 case and 2 cases showing a decrease and an increase in AChR-reactive IL-10-secreting cell number, respectively.Conclusions The immune dysfunction of the Th1 and Th2 subpopulation existed in the thymus and PB of MG, and the AChR-reactive IL-4-, IL-10-secreting cell numbers might be served as parameters to evaluate the degree of the immune dysfunction in the MG thymus. The role of the IL-10 in pathogenesis of MG needs further study.

Objective To study the changes of the semen quality and the spermatogoa ultrastructure in uremic patients before and after renal transplantation. Methods The semen was analyzed and spermatogoa were investigated under scanning electron microscopy (SEM) in 8 patients before and after renal transplantation and 5 normal volunteers. Results In uremic patients, there was significant decreases in sperm motility, survival rate and sperm density. Under SEM, the deletion of the acrosome, distinct vacuoligation in the nucleus, deficiency of the postacromal and postnuclear ring and ~absence of the middle segment of mitochondrial sheath were found. 5.Relationship of brachial-ankle pulse wave velocity with carotid arterial intima-media thickness and carotid artery plaque in patients with type 2 diabetes mellitus

Objective To investigate the relationship between the brachial‐ankle pulse wave velocity (baPWV) with the ca‐rotid arterial intima‐media thickness(IMT) and carotid artery plaque in the patients with type 2 diabetes mellitus(T2DM) and to analyze the related risk factors causing atherosclerosis .Methods A total of 426 patients with T2DM were collected and conduct the records of baPWV ,carotid artery ultrasound ,biochemical index detection andrelated risk factors record .The correlation between baPWV with carotid arterial IMT was analyzed .The patients were divided into the abnormal group and normal group according to the baPWV results .The carotid artery IMT ,carotid artery plaque and related risk factors were compared between the two groups . Results The DM duration ,systolic blood pressure ,diastolic blood pressure ,pulse pressure ,low density lipoprotein cholesterol (LDL‐C) ,number of atherosclerosis risk factors had statistically significant differences between the two groups (P< 0 .05 ,P<0 .01);while the age ,waist circumference ,BMI ,uric acid ,HDL‐C ,TG ,TC ,glycosylated hemoglobin had no statistically significant differences between the two groups(P>0 .05) .The detection rate of plaque by carotid artery ultrasound had no statistically signifi‐cant difference between the two groups(P>0 .05);but the statistically significant difference was found in the number of plaque be‐tween the two groups(P<0 .01) ,the abnormal group was obviously more than the normal group;the normal IMT ratio had statisti‐cally significant difference between the two groups(P<0 .05) ,the normal group was obviously higher than the abnormal group .The IMT value had statistical difference between the two groups(P<0 .01) ,which in the abnormal group was significantly thickened compared with the normal group .The analysis of correlation between baPWV and IMT showed that IMT was positively correlated with baPWV(r=0 .228 ,P<0 .05) .Conclusion T2DM patients with reduced arterial elasticity could complicated with the obvious thickening of carotid arterial IMT and increase of plaque number .Combining baPWV with carotid arterial ultrasound examination can better conduct the arteriosclerosis evaluation .

Object To optimize the conditions for the extraction of Panax notoginseng (Burk ) F H Chen Methods Conditions for the extraction were studied by orthogonal experimental design as guided by the content of total notoginseng saponin present in the extract Results Significant effect was observed in 3 different experimental conditions Conclusion The optimum condition for the extraction of P notoginseng was to soak the drug in 75% alcohol for 24 h and then percolate at a rate of 1~3 ml/min to collect an amount corresponding to about 10 times of the quantity of P notoginseng
7.Determination of polysaccharide in TIANCI CAPSULE

Object To develop a method for the determination of polysaccharide in TIANCI CAPSULE, which is a new drug dosage form containing the bouillon of fresh water mussels with immunopotentiating activity Methods Total polysaccharide was determined by phenol sulfuric acid method with glucose as the standard, and spectrophotometer assay absorbency at 490 nm Results The linear range of polysaccharide was 10~80 ?g/mL, r=0 999 7 The average recovery rate was 98 67% (n=5), RSD=2 58% Through sample assay, the content of polysaccharide was 267 7~300 6 mg in each TIANCI CAPSULE Conclusion The method is simple, rapid and reliable, and may be used for the quality control of this preparation

The results of experiment showed that B. subtilis BR151 and MI112 containing plasmid pUB110 in LB liquid medium is more stabl than in LB slant. Stable recipient strain BSG for plasmid pUB110 was obtained by mutagenesis with NTG. The strain BSG is more efficient recipient of transformation for shuttle vectors pPTE4 and pBE2 from E.coli than its parent. The chimeric plasmids which contain small inserts are stable in strain BSG after 30 generations.
9.Exam stress and salivary immunological function

BACKGROUND: It has been verified in relevant clinical and epidemiological researches in recent years that a part of stomatopathy is associated to psychological stress factors.OBJECTIVE: To analyze the relationship between the changes in salivary secretory IgA and cortisol and psychological stress states.DESIGN: cross-sectional investigation was designed.SETTING: Department of Stomatology, Zhejiang University.PARTICIPANTS: The investigated participants were 60 students in the 2nd grade of Fuzhou №.2 Senior School selected from May to November 2003. Fifty-seven of them accomplished the investigation, of male; aged varied from 17 to 19 years, of healthy, no stomatopathy and family history of disorders in immune system and without indulgence in smoke and alcohol. All of them had participated in the 3-day terminal exam.METHODS: Before the exam, evaluation with symptom checklist (there are 90 questions, including about 10 factors, such as somatization, compulsive state, sensitive interpersonal relationships, melancholy, anxiety, hostility, fear, paranoid state, psychotic state and others. The total average score results by divided total score with 90. ) was applied. Fifty-seven students that had accomplished the investigation were divided in low-symptom group (the total score of symptom checklist < 2.0) and high-symptom group (the total score ≥ 2.0). 5 weeks before the exam, the students were called together to fill up investigation form of general situation. Four weeks before the exam and at 7: 30 am on the day of exam, the first and second saliva specimens were collected respectively to determine salivary secretory IgA and cortisol before and after exam stress. The students were called together again 3 days after exam to fill up questionnaire on psychological health to evaluate their psychological health state.stress.RESULTS: Intention management analysis was adopted, in which 57 stulist with Chinese norm: after exam, the scores of 4 factors, named compulsive state, anxiety, hostility and paranoid state, were higher than Chinese norm [(1.98±0.72, 1.69±0.61) scores; (1.79±0.31, 1.42±0.43) scores; (1.77±0.68,1.50±0.57)scores; (2.00±0.71, 1.52±0.60)scores, P < 0.01] and there was parison of salivary secretory IgA content after stress: that before exam in low symptom group was remarkably higher than that in high symptom group [(236.6±82.29),(194.55±66.29) mg/L, P < 0.05], that after exam in low symptom group was higher than that in high symptom group [(182.21±55.92,tration after stress: that after exam in low symptom group was remarkably higher than that in high symptom group [(5.96±2.46, 3.68±2.26) μg/L, P secretory IgA and cortisol in two groups: the decrease rate of secretory IgA (0.012±0.197) and increase rate of cortisol (0.199±0.654) were relevant to compulsive state and paranoid state.CONCLUSION: In evaluation with symptom checklist after exam, the scores of anxiety, compulsive state, hostility and paranoid state were higher than Chinese norm, indicating that exam stress induces increased anxiety level and variation of psychological behavior of students. The difference presented in decrease rate of salivary secretory IgA before and after exam stress between high and low symptom groups, explaining that the students with more psychological symptoms are weak to endure exam stress.
10.The diagnosis of insufficient blood supply in vertebral basilar artery by combined transcranial Doppler ultrasound and CT in elderly patients

Objective To investigate the diagnostic value of the combined transeranial Doppler ultrasound and CT for vertebrobasilar insufficiency in elderly patients.Methods Fifty elderly patients with symptoms of vertebrobasilar insufficiency were enrolled and randomly assigned to 4 groups,including examinations of TCD,CTA,DSA,combination of TCD and CTA.The diagnostic accuracy was calculated and compared.Results Among 50 cases,the vertebrobasilar insufficiency was confirmed in 42 patients (84%) by TCD,45 (90%) by CTA,48 (96%) by DSA,49(98%) by combination of TCD and CTA.There was no significant difference in diagnostic rate between TCD combined with CTA and DSA (x2 =0.34,P>0.05).Conclusions The diagnosis accuracy of the combined CTA and TCD is high,making it worth popularizing.
