1.BC047440 gene silencing inhibits the proliferation of HepG2 cells

Objective To investigate whether the proliferation of HepG2 ceils is influenced by interfering BC047440 gene with small hairpin RNA (shRNA) expression plasmid. Methods According to the sequence of BC047440 gene, 2 pairs of BC047440 gene-specific shRNA (shRNA1 and shRNA2) were designed and synthesized. After primer annealing, they were inserted into plasmid pGenesil-1 to construct the shRNA expression plasmids. The recombinant plasmids were transfected into HepG2 cells. The expression of BC047440 gene was detected by quantitative fluorescent PCR, the proliferation of HepG2 cells by MTT assay and the changes of cell cycle by flow cytometry. Results Two shRNA expression plasmids were constructed successfully and were confirmed by restriction enzyme digestion and sequencing. Quantitative fluorescent PCR analysis showed that shRNA1 and shRNA2 could specifically inhibit the expression of BC047440 gene in HepG2 cells, with the inhibition rate of 80.22% and 58.63%, respectively. The shRNA effectively inhibited the proliferation of HepG2 cells, and arrested the cell cycle in S phase. Conclusions The shRNA significantly inhibits the expression of BC047440 gene and the proliferation of HepG2 cells. The expression of BC047440 may be correlated with the proliferation of HepG2 cells.
2.A study of mechanism of modulation of NF-κB activity of HepG2 cells by BC047440 genes

Objective To investigate the mechanism that BC047440 gene regulates nuclear fac-tor κB sigal passway and analyze the differential expression gene between HepG2 cells and HepG2 cells BC047440 gene silenced by RNAi using 35K Human Genome Array. Methods The differential expres-sion gene between HepG2 cells and HepG2 cells with BC047440 gene silenced was analyzed by 35K Human Genome Array, and the data were submitted to the database and MAS system of Capitalbio Corporation.Then TRAF6 was confirmed by RT-PCR test. Results Among the total 35000 probe sets, the expression of 59 genes was down-regulated for more than 50% and 130 genes were up-regulated more than 2 fold in the silencing group when compared with normal controls. TRAF6 mRNA was decreased for 29.5% in silicening HepG2 compared with that of wild HepG2 by RT-PCR, which is similar to human genome array(23.06%).Conclusion The high throughput and effective oligomicroarray can analyze the differential expression gene and BC047440 gene might regulate NF-κB signal pathway inderectly by TRAF6.
3.Simultaneous repair of cleft lip and closure of cleft hard palate with vomer flaps in patients with unilateral complete cleft lip and palate.

OBJECTIVEThe purpose of this study was to retrospect the prognosis of simultaneous repair of cleft lip and closure of cleft hard palate with vomer flaps in patients with unilateral complete cleft lip and palate.
METHODSA retrospective study was carried out in 47 patients with unilateral complete cleft lip and palate and, simultaneously received repair of cleft lip and closure of cleft hard palate with vomer flaps. The duration of operation, as well as the blood loss during the operation was recorded, and compared with those patients who only received cleft lip repair.
RESULTSAll the operations were successful, and the wound healed well. The procedure of simultaneous repair of cleft lip and closure of cleft hard palate with vomer flaps did not prolong the operating time, compared with simple cleft lip repair. No blood transfusion was needed due to closure of cleft hard palates with vomer flaps.
CONCLUSIONSimultaneous repairs of cleft lip and closure of cleft hard palate with vomer flaps are safe for patients with unilateral complete cleft lip and palate.

4.Experimental research on degradation and biocompatibility of super-high-molecular-weight poly-DL-lactic acid.

OBJECTIVEThe super-high-molecular-weight poly-DL-lactic acid (PDLLA), with the molecular weight of 900 kD, is a newly emerging biomaterial and potentially used in the therapy of bone fracture because of its excellent mechanical property. However the biocompatibility of this material has not been reported so far, therefore this experiment was designed to examine whether the super-high-molecular-weight PDLLA was harmful to creatures, when it was implanted in the body of animals for a long period.
METHODSThe material was prepared in small cuboids, with the size of 1.0 mm x 1.5 mm x 2.0 mm, and these blocks were implanted into the masseteric space of SD rats and, the activity of the SD-rats was monitored continuously. The animals were sacrificed in the 3rd, 6th, 9th, 12th months after the operation and, the specimens were taken out from the animals. The examination included anatomical, pathological and haematological methods. The data were analyzed with SPSS 8.0.
RESULTSThe wound healed well after the operation. Super-high-molecular-weight PDLLA degraded 6 months after the implantation. In the 3rd month after the operation, a thin fiber membrane around the materials was formed. In the 6th month, the membrane was much thinner than that in the 3rd month and completely disappeared in the 9th month. The pathological examination showed that slightly inflammatory reaction appeared in the tissue around these blocks in the 3rd month, but the inflammatory reactions were gradually remitted in the following 6th, 9th and 12th months. Further, the haematological examination did not show any abnormity during the 12-month observation period.
CONCLUSIONThe super-high-molecular-weight PDLLA can be degrade when it is implanted into the body of creatures, which proves its good biocompatibility.

5.Early pathological morphology and clinical significance of perforated duodenal ulcer closed after non-surgical treatment

Objective To research early pathological morphology and clinical significance of perforated duodenal ulcer (PDU) closed after non-surgical treatment. Methods Observed morphological changes of duodenal ulcer (DU) lesion with gastroscopy for 302 patients of PDU with non-surgical treatment in early period,when the perforation closed and measured up the clinical indicators during this hospitalization.Results There were 255 patients to be diagnosed with DU caused the perforation. These lesions were characteristic and shown the PDU closed at the bottom and the deep concave ulcers, except for 1 case which complicated by duodenal fistula. These ulcer types were diverse according to the time difference after treatment. No case of re-perforated ulcers or recurrence of peritonitis caused by gastroscopy. Conclusions Deep concave ulcer with A1 phase mainly is an early pathological manifestations of the DU after treated the PDU with non-surgical method characteristically. The wall of the closure of the serosal side is an original form closed perforated ulcer by non-surgical treatment. The risk of perforation associated with the following factors:( 1 )A single DU is located in the anterior wall region. (2)The shape of two kissing DU. (3)The diameter of DU ≥ 1.1 cm. In this case,early diagnosis by using endoscopy is a safe way.
6.Study on injectable bioactive bone repairing material of nano-hydroxyapatite and polyamide-66 composite.

The aim of this study was to evaluate the injectability, histocompatibility, function and other properties of the injectable bioactive bone repairing material of nano-hydroxyapatite and polyamide-66 (n-HA/PA66) composite. The XRD pattern, the relationship between the injectability and liquid-powder ratio, setting time and liquid-powder ratio, compressive strength and liquid-powder ratio were assessed. The size of the composite was determined to be 70 nm in length and 30 to 50 nm in width, and the molecular weight of polyamides-66 was 18000. The diameter of pores of the composite was about 200 to 400 micrometer. To evaluate the histocompatibility and function, 8 male dogs were studied with the injectable n-HA/PA66 composite implanted in the artificial defected alveolus of mandible on only one side to be compared with the intact alveolus on the other side. The specimens were taken at 4, 8, 12, 16 months after the implantation and the results were evaluated. The XRD pattern of the solidificated n-HA/PA66 composite was the same as the powdered n-HA/PA66 composite. The injectable n-HA/PA66 composite had a good injectability, 25 to 30 minutes setting time and about 37 MPa compressive strength when the liquid-powder ratio was 0.50. The healing of the gingiva was well at the implanted areas in all animals. The height of the repaired alveolar bone was obvious higher than that of the blank control. The earlier sign of ossification was histologically observed at 16 weeks after implantation. The injectable n-HA/PA66 composite has good biocompatibility and osteoconductive property. As an injectable material, with good maneuverability, it is useful for repairing irregular bone defects, especially in oral and maxillofacial surgery.

7.The study of biocompatibility of super high molecular weight poly D,L-lactic acid implant.

OBJECTIVETo evaluate the biocompatibility of the super high molecular weight poly D,L-lactic acid (SHMW-PDLLA) implant.
METHODSThe SHMW-PDLLA plates were implanted into the SD-rats between the masseter and ramus of the mandible. The blood specimens were gained at 3, 6, 9, 12 months after the operation. The proteins, electrolyte, enzyme and other indices were tested by use of Beckman automatic biochemical analysis device. The soft tissue specimens around the SHMW-PDLLA plates were gained at 3, 6, 9, 12 months after the operation and the tissue reaction was observed with the pathological and haematological methods.
RESULTSThere were not any abnormal findings in the blood after the SHMW-PDLLA plates implanted in the body of SD-rats. The implanted SHMW-PDLLA plates were degraded gradually in 6 to 12 months after the operation. There was not any abnormal tissue reaction found to the soft tissue around the SHMW-PDLLA plates by histological and pathological observations.
CONCLUSIONSThe SHMW-PDLLA implant has a good biocompatibility to SD-rats.

8. An overview of multiple linear regression model and its application

Multiple Linear Regression (MLR) is a generalization of simple linear regression and is one of the commonly used models in multivariate statistical analysis. This article introduces the MLR model from the perspective of practical application. Four parts, including basic principle, application examples, the application condition and diagnosis, and the extension of the model, are sequentially illustrated in this article. Particularly, in the last part, alternative methods of the model are introduced when the application condition of the model is not met. We sincerely hope that this article could make our audiences have a better understanding of the MLR model in order to improve the efficiency of data utilization and statistical analysis by correctly performing this model in their research.
9. Overview of logistic regression model analysis and application

Logistic regression is a kind of multiple regression method to analyze the relationship between a binary outcome or categorical outcome and multiple influencing factors, including multiple logistic regression, conditional logistic regression, polytomous logistic regression, ordinal logistic regression and adjacent categorical logistic regression. This paper illustrates the basic principle, independent variable selection and assignment, applied condition, model evaluation and diagnosis for multiple logistic regression model. Moreover, the principle and application for polytomous logistic regression and ordinal logistic regression models were also introduced. By providing SAS codes and detailed explanations of the result for an example of obesity, readers could be able to better understand logistic regression model, and apply this method correctly to their research and daily work, so as to improve their capacity of the data analysis.
10. Overview of multivariate regression model analysis and application

Analyses of the multivariate regression model are ued very widely in the medical research. Analytical methods of the mutivariate regression model including multiple linear regression, logistic regression, Poisson regression and Cox proportional hazard model were introduced in this article. The contents of the article covered the application conditions of regression models, analytical procedures, strategies of selecting independent variables, extended discussions of regression models and application notes. It is expected that authors could understand the principle of the mutivariate regression model, accurately use these analytical methods in their research, improve the efficiency of data utilization, and enhance the level of statistical analyses.
