1.Research on the expression difference of hot shock protein 70 before and after laparoscopic cholecystectomy for acute cholecystitis patients

Objective To compare the expression difference of heat shock protein 70 (HSP70)!in blood of acute cholecystitis (AC) patients before and after laparoscopic cholecystectomy, and to explore the relationship between HSP70 and AC at molecular and proteinic level.Methods Subjects in the study included 85 in-hospital AC patients who received laparoscopic cholectectomy. Blood before and after laproscopic operation were obtained. Ezyme linked immunosorbent assay (ELISA) was used to determine the quantity of HSP70. Four subjects were randomly selected and the total RNA and protein in blood were extracted. RT-PCR and Western-blotting were applied to determine the expression of HSP70. 3.The clinical study of different dose of dezocine in preventing postoperative pain after painless artificial abortion

Objective To explore the lowest and most effective dose of dezocine in preventing postoperative pain after painless artificial abortion.Methods A total of 180 ASA Ⅰ ~ Ⅱ patients who accepted painless artificial abortion were randomly divided into 6 groups (n =30 patients per group).Group 1 (control) was given the placebo (0.9% NS; 2 ml) by vein while other groups were given dezocine by vein with 0.01 mg/kg (group 2),0.02 mg/kg (group 3),0.03 mg/kg (group 4),0.04 mg/kg (group 5),and 0.05 mg/kg (group 6),respectively.The dosage of propofol,BP,HR,SpO2 during the operation,operation time,consciousness recovery time,analgesic effect (VAS score),leaving time,defecation feeling,and untoward reaction of each group were recorded and compared.Results The dosage of propofol in the 5 dezocine groups were significantly lower than that in the control group,respectively (t =2.38,4.72,6.03,4.57,3.96,P < 0.05),while the difference in the operation time,consciousness recovery time and leaving time between the 5 dezocine groups and the control group had not statistical significance (P >0.05).The effect of postoperative analgesia and defecation feeling were apparently improved in the group 3,4,5 and 6,compared to the control group(group 3:x2 =3.36,6.94,P < 0.05;group 4:x2 =7.17,15.01,P < 0.05 ; group 5:x2 =15.5,24.3,P < 0.01 ; group 6:x2 =13.01,20.24,P < 0.01).Conclusions The lowest and most effective dose of dezocine was 0.05 mg/kg in preventing postoperative pain after painless artificial abortion.
4.Factors and mechanism influencing elastin-like polypeptides self-assembled into micron-sized particles.

Many factors influence the elastin-like polypeptides (ELPs) self-assembled into micron-sized particles. However, few efforts were made to investigate these factors. Using the ELPs [KV8F]n as the target, we studied systematically the factors with the dynamic light scattering. Our results show that the particle size increased and the uniform of particles decreased with the increase of the molecular weight. The analysis of size variation in self-assembled ELPs in response to changes in salt concentration indicated that the size increased with increasing the salt concentration, and the opposite response was observed when the concentration was above 0.4 mol/L. Under these conditions, the particles are micron-sized and larger than 1.1 μm. However, when the fusions containing the same ELPs and xylanase or 1,3-propanediol oxidoreductase, the size of the self-assembled ELPs particles decreased dramatically, which was only about 1/10 of that of the free ELPs. We proposed that the solvent accessible charged area of the enzymes could interact with the ELPs, the sterical hindrance of the enzymes prevent the aggregation of the ELPs. This might be the most important parameter in altering the particle size sharply.

5.Ultrasound and blood indicators in diagnosis of early cirrhosis for patients with chronic hepatitis B

Objective To investigate the application of ultrasound and blood indicators in diagnosis of early liver cirrhosis (S4) for patients with chronic hepatitis B (CHB).Methods Data of blood and ultrasound examinations of 631 patients with CHB who received liver biopsy in the Sixth People' s Hospital of Shenyang during April 2002 and March 2011 were collected.Logistic regression analysis was performed to determine the factors independently correlated with early liver cirrhosis,and the diagnostic model was established using these indicators.The diagnostic value of the established model was assessed by using area under receiver operating characteristic curve (AUROC) and compared with FIB-4 index,aspartate aminotransferase (AST)/platelet (PLT) ratio index (APRI) and S index.Results Logistic regression analysis indicated that age,PLT,albumin globulin ratio (A/G) and splenic square area (SPS) in ultrasound findings were independently correlated to early liver cirrhosis (Wald =10056,46.236,3.751 and 10.669,P <0.01).AUROC of the model based on the above factors in diagnosing early liver cirrhosis was 0.908,which was higher than those of FIB-1 index,APRI index and S index (Z =8.322,4.334 and 4.087,P < 0.05).Taking 0.063 as cut-off value,the sensitivity,specificity,positive predict value and negative predict value of the established model in diagnosis of early liver cirrhosis were 90.1%,77.8%,50.0% and 97.1%,respectively.Taking <0.060 and ≥0.110 as the cut-off values to exclude,and diagnose early liver cirrhosis,69.7% (440/631) patients could avoid liver biopsy.Conclusion The model based on age,PLT,A/G and SPS can effectively predict early liver cirrhosis,and can reduce liver biopsy.
6.Comparative analysis of ultrasoundgraphy and CT guided percutaneous paracentesis for liver abscess

Objective To explore differences of ultrasoundgraphy and CT guided percutaneous paracentesis for liver abscess and provide references for the paracentesis treating of liver abscess .Methods 68 patients with liver abscess arranged ultrasoundgraphy guided percutaneous paracentesis were regarded as ultrasound group ,and 38 patients accepted CT guided percutaneous paracentesis were considered as CT group .The succeed rate of paracentesis ,operation period ,treatment effectiveness ,complications of the 2 groups were compared .Results The succeed rate of paracentesis in ultrasound group (81 .7% ) was not different obviously to CT group(82 .6% ) (χ2 =0 .016 2 ,P=0 .898 6) .The operation period of ultrasound group(22 .3 ± 5 .6)min was shorten than CT group (25 .6 ± 4 .8)min ,t= 3 .057 4 ,P< 0 .05 .The heal rate of the 2 groups(91 .2% ,92 .1% ) was not different (χ2 = 0 .027 1 ,P=0 .869 3) .There was no different of the complications in the 2 groups(5 .88% ,8 .57% )(χ2 =0 .010 1 ,P=0 .920 1) .Conclusion Either ultrasoundgraphy or CT guided percutaneous paracentes is feasible and safe for liver abscess .Compared to CT ,the ultra-soundgraphy is more convenient and economic .
7.Effect of tooth wear on morphological features of the mandibular first molars

Objective:To investigate the effect of tooth wear on the morphological features of mandibular first molars. Methods:15 extracted mandibular first molars with tooth wear and 15 without wear were investigated. Data about the morphological features of tooth were collected through the general measurement and the image measurement by electronic meter. Results:①In non-tooth-wear group, mesiodistal width of tooth was greater than buccolingual width(P0.05); ②The vertical distance between the lowest point of pulp chamber roof and occlusal surface in non-tooth-wear group was greater than that in tooth-wear group(P05); ④The distance between distolingual pulp horn and distal proximal plane was greater than that between mesiolingual pulp horn and mesial proximal plane in both two groups(P0.05). Conclusion:For different parts of tooth, the effect of tooth wear differs. As for mandibular first molars, the distal part of occlusal surface and the buccal part of distal proximal plane are susceptible to tooth wear, which is mainly due to occlusal factors and the features of biomechanical system.
8.The correlation between mild cognitive impairment in the elderly and impaired fasting glucose tolerance and low fasting blood glucose

Objective:To investigate the correlation between mild cognitive impairment(MCI)and abnormal glucose metabolism and thus to provide a basis for MCI prevention.Methods:A total of 1 074 elderly outpatients with normal cognitive function and without confirmed diabetes mellitus, hyperlipoidemia or gout were enrolled.During a five-year follow-up period, 121 subjects were diagnosed with MCI based on the mini mental state examination(MMSE)and the Montreal cognitive assessment(MoCA). Furthermore, annual blood glucose and glycated hemoglobin monitoring was carried out to examine the long-term effects of abnormal glucose metabolism on MCI risk.Results:According to cognitive function, 1 074 subjects were divided into the MCI group and the non-MCI group.Compared with the non-MCI group, the mean values of fasting blood glucose(FBG), glycosylated hemoglobin(HbA1c), triglycerides(TG)and total cholesterol(TC)in the MCI group were elevated( P<0.05). The receiver operating characteristic(ROC)curve showed that the cut-off value of FBG was 6.2 mmol/L for the hyperglycemia group(sensitivity: 84.1%, specificity: 90.9%, area under curve: 0.875, P<0.001)and 4.5mmol/L for the hypoglycemic group(sensitivity: 77.4%, specificity: 87.3%, area under curve: 0.823, P<0.001); the cut-off value of HbA1c was 5.5%(sensitivity: 76.0%, specificity: 87.0%, area under curve: 0.815, P<0.001). Multiple Logistic regression analysis showed that increased risk of MCI was associated with the mean values of fasting blood glucose <4.5 mmol/L( RR: 1.69, 95% CI: 1.11-2.59)or ≥6.2 mmol/L( RR: 1.81, 95% CI: 1.15-2.86)and of glycosylated hemoglobin ≥ 5.5%( RR: 2.13, 95% CI: 1.51-2.99). Conclusions:Impaired fasting glucose tolerance and low fasting blood glucose are independent risk factors for MCI in the elderly.
9.Study and Compatible Design of Two Types of Liquid Auto Sampler for Gas Chromatographer

We carried out parallel design and development of two differently structured auto sampler based on a multi-axis and multi-mode high-precision closed-loop servo control system. An integrated embedded control drive module was developed based on the idea of compatibility and inter-changeability, so that DC motor and encoder were standardized into uniform models. Meanwhile, electric and mechanical interfaces were uniformed to a same standard. This allows the direct exchange of above-mentioned components between the two models. A 1-μL manual sample injection syringe was installed on both standard 110-sample and platform 40-sample liquid auto sample injectors connected with gas chromatographer. Approximately 0. 5μL of cetane-isooctyl was sampled for 6 consecutive times at six different positions in the sample bottle. The repeatability RSDs of the injection peak areas of the two systems were 1. 1% and 1. 5%, respectively. A linear correlation coefficient (0. 9947) of peak area with injection volume was achieved based on the gradient sampling volume of 0. 1, 0. 3, 0. 5, 0. 7 and 0. 9 μL.
