2.Anti-dementia effect of Tongluo Xingnao effervescent tablet based on urinary metabonomics.

Tongluo Xingnao effervescent tablet (TLXNET) is a patented prescription, which comes from modified Xionggui decoction and can improve cognitive function. However, its effect on the urine metabolites and anti-dementia mechanism in the dementia model rats induced by hippocampal injection with Aβ25-35 remains unclear. The experiment focused on the changes in trajectory and inter-relationship among the urinary metabolite of rats in the blank group, Aβ25-35 hippocampal injection dementia model group and the TLXNET intervention group, in order to determine theirs characteristic metabolic markers and explain the anti-dementia effect of TLX-NET base on the change of metabolic trajectory of these bio-markers. According to the experimental results, 5, 6-indolequinone, 4-hydroxyphenyl pyruvic acid (4-HPPA), cortisol and 3-thiosulfate lactic were preliminarily identified as the characteristic metabolic markers. They mainly participate in dopamine system, glucocorticoids and energy metabolic pathways. TLXNET can apparently downregulate the disturbances of metabolic trajectory of the four bio-markers. The experiment indicates that the dementia model induced by injecting Aβ25-3 into hippocampus has its characteristic endogenous metabolic markers in urine, and ELXNET can ameliorate dementia by down-regulating the disturbances of metabolic trajectory.

By taking clinical and basic researches for integrative medicine preventing and treating acute myocardial in-farction and sepsis as a breakthrough, authors explored key problems of Chinese medicine in improving the prognosis related diseases and patients' quality of life. In recent 3 years our department has successively become the principal unit of the national key specialties cooperative group of critical care medicine (awarded by State Administration of Traditional Chinese Medicine), the key clinical specialties (awarded by National Health and Family Planning Commission), and Guangzhou key laboratory construction unit, and achieved overall lap in clinical medical treatment, personnel training, scientific research, and social service.

5.A pharmacodynamic study of intervals of Mitomycin C treatment on human bladder cancer cell line in vitro

Objective To study the relationship between the intervals of Mitomycin C treatment and cytotoxicity, apoptosis and drug resistance for bladder cancer cells.Methods The bladder transitional cell cancer line BIU-87 was treated for two hours every time for five times with intervals of 24, 48, 72 and 96 hours respectively.Cytotoxicity was measured by MTT.p53,bcl-2,Bax and p170 expression were analyzed by Western blot.Results The IC_(50)(?g/ml)were 4.41,0.71,2.83,4.51and 6.16 with treatment intervals of 24, 48, 72 and 96 hours respectively, p53 and bcl-2 were significantly down-regulated and bcl-2/Bax was re- duced at 24 hour treatment interval but not changed at 48,72 and 96 hour intervals,p170 was not detected at 24 hour treatment interval but increasingly expressed at 48,72 and 96 hours intervals.Conclusion The in- terval of Mitomycin C treatment is closely related with cytotoxieity and apoptosis and drug resistance of blad- der cancer cells.The intervals of intravesical instillations may play an important role in the effect of chemotherapy.
6.Study of heart rate variability among 1581 normal children from age 3 to 12 in Beijing

Objective To investigate the correlation between premature beats,atrioventricular block (AVB),heart rate variability (HRV) and age as well as gender among normal children.Methods One thousand five hundred and eighty-one cases of normal children from Beijing were divided into three age groups:3 to 6-year old,~ 10-year old and ~ 12-year old.All children were evaluated with both ECG and Holter to determine arrhythmia and HRV.The arrhythmia incidence and five time-domain values of HRV were analyzed according to age,gender and the existence of ventricular premature contractions.Results (1) Both ECG and 24-hour ECG monitoring (Holter) data showed that the average heart rate (HR),the slowest HR and maximum HR of normal children decreased with age,while PR interval and QTc interval increased with age.The correlation between age and the four indices were statistically significant (P <0.01),except the PR interval.(2) The positive rate of arrhythmia with Holter and ECG in normal children had statistically significant difference(x2 =4.810,P < 0.05).(3) The incidence of premature contraction and Ⅰ ° AVB rised or fell with increasing age(P < 0.05).Compared with 3 ~6-year age group,10-year and ~ 12-year old group had highly risk tendency in premature contraction(OR =1.31,2.04).(4) The five time-domain indices of HRV were increases with age.Each age group had statistically significant difference (P < 0.01).(5) The five time-domain indexes of HRV in males were higher than those in females (P <0.01).(6) No significant differences were found with five time-domain indexes in children with or without ventricular premature contraction (P > 0.05).Conclusions (1) HR of normal children decreases with age,but QTc interval increases with age.(2) The supraventricular premature contraction is the most common arrhythmia in normal children with Holter.(3) The occurrence of premature contractions increases with age,more obvious during the preadolescence.Ⅰ ° AVB occurs with age growth on a downward trend.Sex has no effect on the same arrhythmia.(4) HRV in normal children has age difference,suggesting that the pediatric cardiac autonomic nervous system (ANS) was immature.(5) The differences of HRV in sex were most significant during the preadolescence,which showed the developmental inconsistencies in cardiac ANS between male and female of preadolescence.(6) In the majority of normal children,the ventricular premature contractions were benign.
7.A comparative study of cytology,histopathology and immunohistochemistry of malignant lymphoma of lung mucosa-associated lymphoid tissues

Objective: To explore the cytopathologic features and differential diagnostic essentials of the lung mucosa-associated lymphoid tumor(MALT).Methods: The fine needle aspiration(FNA) tissues and bronchi smears of 4 cases of MALT were examined by cytology,histology and immunohistochemistry respectively.Results: Cytologically,3 of the 4 cases were definitely diagnosed as lymphoid tumor and 1 suspected of small cell undifferentiated carcinoma,while histopathologically,all were diagnosed as MALT.The neoplastic cells were positive for CD20 and CD79a,but negative for CD3,CD5,CD10,CD45RO,CKpan and EMA.Conclusion: MALT is a rare pulmonary lymphoma difficult to be diagnosed.The definite diagnosis of pulmonary MALT depends on the cytology of FNA and bronchi smears,histopathology,and immunohistochemistry.
8.Varieties textual research and usage investigation of white flos gentianae, a Tibetan medicine.

OBJECTIVETo clarity the original plants and the main application varieties of White Flos Gentianae.
METHODHerbal textual research, wild specimen collection, investigation and collection of the samples from Tibetan hospital, Tibetan pharmaceutical factory and medical material market were carried out simultaneously to identify the original plants of White Flos Gentianae.
RESULTThe results of varieties textual research and specimen identification showed that Gentiana szechenyii, G. purdomii and G. algida were in accord with the record of Tibetan herbal textual The three species above were the original plants of White Flos Gentianae. The identification of 20 batches samples showed that G. szechenyii was the main application variety. The other varieties were only used in Tibetan hospitals. All the samples above were flowering branches.
CONCLUSIONIt was necessary to strengthen the research on variety systematization of White Flos Gentianae make a further discussion on the taxonomy position of G. purdomii, G. algida and the white flos population. Its was also nessary to establish and improve the quality standard of different variety based on the principle of "one species, one name". The quality specification of White Flos Gentianae should be established and improved to standard clinical utilization and produce feeding. More study of resources investigation and cultivation of G. szechenyii should be carried on to meet the demand of produce and clinic.

9.Identification of pyrrosiae folium and its adulterants based on psbA-trnH sequence.

In this study, the psbA-trnH sequence as DNA barcode was used to evaluate the accuracy and stability for identification pteridophyte medicinal material Pyrrosiae Foliumas from adulterants. Genomic DNA from 106 samples were extracted successfully. The Kimura 2-Parameter (K2P) distances and ML tree were calculated using software MEGA 6.0. The intra-specific genetic distances of 3 original plants were lower than inter-specific genetic distances of adulterants. The ML tree indicated that Pyrrosiae Folium can be distinguished from its adulterants obviously. Therefore, the psbA-trnH sequence as a barcode of the pteridophyte, can accurately and stably distinguish Pyrrosiae Folium from its adulterants.

10.Changes in B-type natriuretic peptide in plasma and its clinical significance in patients with severe burn during shock stage.

OBJECTIVETo study the plasma content of B-type natriuretic peptide (BNP) in patients with severe burn during shock stage and probe its clinical significance.
METHODSForty-two patients aged 18-60 years, with total burn surface area 30% TBSA or full-thickness burn area > or = 10% TBSA, hospitalized within 4 hours after burn, were divided into A group (with total burn surface area 30%-50% TBSA or full-thickness burn area 10%-20% TBSA, n = 21), and B group (with total burn surface area > 50% TBSA or full-thickness burn area > 20% TBSA, n = 21). Twenty patients admitted during the same time for plastic surgery were enrolled as control group. The plasma levels of BNP, creatine kinase (CK), CK-MB, troponin I (TnI) of all patients were determined on admission. The levels of BNP, TnI and fluid resuscitation volume were examined at 8, 16, 24, 48 post burn hour (PBH) in A and B groups. Analysis of correlation between BNP and fluid resuscitation volume was performed.
RESULTSOn admission: BNP level in A group (68 +/- 19 ng/L) and B group (99 +/- 38 ng/L), respectively, was increased as compared with that in control group (17 +/- 7 ng/L, P < 0.01). TnI level in A group (2.13 +/- 0.67 microg/L) and B group (2.98 +/- 0.58 microg/L), respectively, was increased as compared with that in control group (0.12 +/- 0.03 microg/L, P < 0.01). There was no obvious difference in CK, CK-MB levels among A, B, and control groups (P > 0.05). BNP levels in A, B groups continuously rose during 8-48 PBH, and they were positively correlated with fluid resuscitation volume. TnI level peaked at 24 PBH, and decreased at 48 PBH.
CONCLUSIONSThe plasma level of BNP is sensitive to reflect changes in myocardial ischemia and hypoxia as a rise in level of TnI in shock stage of severe burn, and it was positively correlated with fluid resuscitation volume. BNP can be used to guide fluid resuscitation during shock stage.
