Objective: To investigate the influence of allogeneic spleen tissue transplantation with low-dose FK506 on allograft tolerance in rats receiving orthotopic liver transplant and the related mechanism. Methods: Female Brown Norway (BN) rats were divided into 5 groups (eight rats in each group). Rats in Group 1 received orthotopic allogeneic liver transplantation using inbred Lewis rats as donors. Rats in Group 2 received Lewis liver transplantation+low-dose FK-506 (0.25 mg/kg FK-506 was orally given after transplantation). Rats in Group 3 received Lewis liver transplantation+Lewis spleen transplantation. Rats in Group 4 received Lewis liver transplantation+Lewis spleen transplantation+low-dose FK-506. Rats in Group 5 received Lewis liver+SD spleen transplantation+low-dose FK-506. The survival periods were recorded for each group. The forming of chimerism, the responsiveness of T cells to alloantigen and the pathological changes of transplanted liver were analyzed after treatment. Results: Compared with rats in other groups, rats in Group 4 had an obviously longer survival time, an obviously reduced T cell responsiveness to alloantigen, and obviously increased donor-positive cells (remained for 60 days after transplantation) (P<0.05). There was no significant pathological evidence of rejection. Conclusion: Allogeneic spleen transplantation combined with low-dose FK506 treatment can induce graft immune tolerance in orthotopic allogeneic liver transplant recipients through promoting the formation of chimerism and T cells hyporesonsiveness to alloantigen.

Objective To evaluate the effect of subfascial endoscopic perforating veins ablation in treating chronic venous insufficiency of lower extremities. Methods A retrospective study was carried out on 18 patients( 20 limbs) with chronic venous insufficiency treated by subfascial endoscopic perforating veins ablation. Objective To study the application of hepatamethine cyanine near-infrared fluorescence (NIRF) dye IR-783 in the mouse models of human liver cancer exenografts, and to analyze the molecular mechanisms of the NIRF dye targeting tumor cells.Methods Luciferase-tagged HepG2 cells were inoculated subcutaneously into the nude mice.We detected the correlation of NIRF intensity and bioluminescence intensity (BIL) in the tumor region.Patient-derived xenograft (PDX) model was established in mouse by subrenal capsular implantation of clinic liver cancer specimen.After injecting the IR-783 dye, the interface between mouse kidney and the xenograft tumors was confirmed by NIRF analysis, and the tumor tissue in kidney was observed by pathology using H&E staining.The expression of CEA, AFP, HIF1α and OATP3A1 in the liver cancer tissue was detected by immunohistochemical staining.The intracellular retention of NIRF dyes was observed under fluorescence microscope after adding Mito Tracker or Lyso Tracker into cultured HepG2 cells.We added IR-783 in a co-culture system of HCCs and normal liver cells to test the specifical identification ability of IR-783 of the liver cancer cells.Results There was a good correlation between NIRF intensity and BIL intensity of the subcutaneous liver cancer xenograft region in nude mice.The margin between the mouse kidney tissue and xenograft tumors was clearly identified by IR-783.Compared with normal kidney tissue, CEA, HIF1α, OATP3A1 and AFP were highly expressed in the tumor region detected by IHC staining.The NIRF dye IR-783 was mainly accumulated in the mitochondria and lysosomes of cancer cells.GFP-tagged HepG2 cells could be recognized directly, whereas red fluorescence was not detected in normal liver cells.Conclusions IR-783 is a novel near-infrared fluorescent dye with tumor targeting and imaging properties.Its targeting ability may be related to the high expression of HIF1α and OATP3A1 in the liver cancer tissue.

Objective To analyze the expression of serum anti M phospholipase A2 receptor (PLA2R)antibody in idiopathic membranous nephropathy (IMN),and to investigate its value in the diagnosis and evaluation of idiopathic membranous ne-phropathy.Methods One hundred and eighteen patients with biopsy-proved glomerular diseases were involved in this study, including 97 cases with IMN,21 cases with IgA nephropathy (IgAN)and 19 healthy people.ELISA was used to detect ser-um anti-PLA2R antibodies.Correlations of anti-PLA2R antibody level with laboratory parameters,including serum albumin, 24-hour urine protein of IMN patients were evaluated.Results The median of anti PLA2R antibody in IMN group,IgAN group and healthy group was 45.2(3.6~705.9)RU/ml,5.9(2.3~10.6)RU/ml and 1.2(0.6~9.3)RU/ml.The levels of serum anti PLA2R antibody in IMN group were higher than those in IgA nephropathy group and healthy control group.The difference was statistically significant (t=-5.027,-3.077;P=0.05).Among 97 cases with IMN,76 cases showed posi-tive anti-PLA2R antibodies (positive rate 78.35%).There was none patient showed positive anti-PLA2R antibody respec-tively in IgAN and healthy people.Furthermore,anti-PLA2R antibody level was negatively correlated with serum albumin (r=-0.453,P=0.000)and positively correlated with CREA,TC,ESR,24 hour urine protein (r=0.233,0.234,0.363, 0.586;P=0.004,0.217,0.021,0.000)in IMN patients.Conclusion Serum anti PLA2R antibody may be used as a IMN specific marker for the diagnosis of IMN,and it has important reference value for evaluating the severity of IMN.

According to relevant national laws and regulations, practitioner training was included into laboratory animal science teaching reform.By adjusting the training content and teaching method and use of animal models of typical human diseases, the transformation of training mode was realized and improved.By the assessment of basic theory in combination with practical operation, the thinking ability and hands-on skill of the practitioners are much improved. Through classroom instruction, experimental teaching, quality assessment and tracking survey, the evaluating process of the training quality of training teaching is performed.Therefore, the teaching reform of the laboratory animal science based on the training of practitioners is established.

Objective To investigate the possibility to create versatile system recovery disc for PACS workstation using the software Ghost 11. Methods Firstly, PACS and Windows software was installed into a PACS workstation. Secondly, S&R&S V9.7 1112F intelligent system -packaging tool and DllCacheManager_V1.6 Dllcache backup -restore tool were used to package the system. Finally, Ghost 11 was adopted to produce mirror image of the system. Results The image could recover system easily. Conclusion The system-recovery disc created by Ghost11 can significantly minimized the workload when the PACS workstation runs into error and improves the maintenance efficiency.

Objective To investigate the effect of continuous positive airway pressure (CPAP) on seizure frequency in epilepsy patients complicated with obstructive sleep apnea (OSA). Methods Subjects were divided into CPAP group (20 subjects) and medication group (22 subjects) according to whether they can tolerate CPAP. CPAP group were treated with CPAP combined with antiepileptic drugs. Medication group were treated with antiepileptic drugs alone. Seizure frequency and apnea-hypopnea index (AHI) were compared between groups before and after 4 weeks and 24 weeks of treatment. Results Baseline seizure frequency and apnea-hypopnea index (AHI) were compared between groups(P>0.05). A sig?nificant reduction of seizure frequency was observed in CPAP group after 4 weeks and 24 weeks of treatment compared to that before treatment (P<0.01). In medication group, no significant difference in seizure frequency was noted betweem 4 weeks and 24 weeks after treatment compared to that before treatment(P>0.05). A significant reduction of AHI was ob?served in the CPAP group after 24 weeks of treatment compared to that before treatment (P<0.01), but no change of AHI by treatment was observed in medication group (P>0.05). Conclusion Treatment of OSA in patients complicated with epilep?sy may improve seizure control in short and long term, but longer observation time and more samples are needed for further research.

Objective To establish a patient-derived gastric cancer xenograft( PDX) model in nude mice and to in-vestigate the application of near infrared fluorescent ( NIRF) dye IR-783 in in vivo imaging of gastric cancer xenograft mod-els.Methods Fresh human gastric cancer tissue was taken and transplanted into the subrenal capsule of nude mice to es-tablish the xenograft model.When the transplanted tumors grew,took part of the tumor tissue to do HE staining and compare the structural characteristics with the primary tumor.Another portion of the tumor was xenografted into nude mice subcutane-ously.Twenty days later,the tumor-bearing mice were injected intraperitoneally with IR-783 dye (10μM) in a dose of 100 mg/20 g.The intensity of the tumor image was monitored by optical NIRF imaging.The correction between tumor volume and fluorescence intensity was analyzed.Finally,the expression of OATP1B3 and HIF1αin the xenografted tumor tissue was detected by immunohistochemistry.Results We successfully established three patient-derived xenograft ( PDH) models of human gastric cancer.The transplanted tumor tissues maintained the histological characteristics of the primary tumor well.NIRF signal can be detec ted in subrenal capsule of the xenografted nude mice.The correlation between tumor size and fluorescence intensity in the PDX models reached higher than 98%.Strong positive expressions of HIF1αand OATP1B3 in the tumor tissues were detected.Conclusions NIRF dye IR-783 can be specifically accumulated at the tumor site,therefore, can be used to detect PDX in vivo early.The tumor targeting property may be related to the expression of OATP1B3 and HIF1α.
