Advanced technologies are used to clarify the meridian tropism theory of traditional Chinese medicine is an important part of theoretical studies of traditional Chinese medicine. In this article, modern pharmacokinetic method was used to investigate tissue distribution characteristics of psoralen and isopsoralen of Psoraleae Fructus decoction in rats, in order to provide research ideas and experimental basis for the meridian tropism theory. In this study, various tissue samples such as heart, liver, spleen, lung, kidney, brain and spermary were collected at different times after oral administration with FP decoction, in order to determine concentration of psoralen and isopsoralen by HPLC. Pharmacokinetic parameters were calculated by DAS 2.0 software. The study results showed that HPLC indexes of psoralen and isopsoralen in various tissues of rats met the determination requirements of biological samples. Both components were distributed in all of the tissues, with AUC(0-t) order of liver > lung approximately kidney > heart > brain approximately spleen > spermary. There was significant difference between liver, kidney, lung and other tissues (P < 0.05). MRT(0-t) of both psoralen and isopsoralen were about 10 h. Therefore, psoralen and isopsoralen showed stronger targeting selection in liver, kidney and lung.

3.Establishment of a mechanical injury model of rat hippocampal neurons in vitro.

OBJECTIVETo establish a simple, reproducible, and practical mechanical injury model of hippocampal neurons of Sprague-Dawley rats in vitro.
METHODSHippocampal neurons isolated from 1-2-day old rats were cultured in vitro. Mild, moderate and severe mechanical injuries were delivered to the neurons by syringe needle tearing, respectively. The control neurons were treated identically with the exception of trauma. Cell damage was assessed by measuring the Propidium Iodide (PI) uptaking at different time points (0.5, 1, 6, 12 and 24 hours) after injury. The concentration of neuron specific enolase was also measured at some time points.
RESULTSPathological examination showed that degeneration, degradation and necrosis occurred in the injured cultured neurons. Compared with the control group, the ratio of PI-positive cells in the injured groups increased significantly after 30 minutes of injury (P<0.05). More severe the damage was, more PI-positive neurons were detected. Compared with the control group, the concentration of neuron specific enolase in the injured culture increased significantly after 1 hour of injury (P<0.05).
CONCLUSIONSThe established model of hippocampal neuron injury in vitro can be repeated easily and can simulate the damage mechanism of traumatic brain injury, which can be used in the future research of traumatic brain injury.

4.Study on relationship between polymorphism sites of TIM-3 and allergic asthma in a population of adult Hans from Hubei province of China.

OBJECTIVETo investigate two single nucleotide polymorphism sites of the promoter region in T cells immunoglobulin domain and mucin domain protein-3 (TIM-3) and detect their relationship with allergic asthma in a population of adult Hans from Hubei province of China.
METHODSThe polymorphisms were detected with polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and allelic specific polymerase chain reaction (ASPCR). The genotype and allele frequencies were calculated and analyzed.
RESULTSThe genotype frequencies of CC, CT and TT in -1541 C/T polymorphism were 0.961, 0.039 and 0 respectively in the healthy population and were 0.935, 0.065 and 0 respectively in the allergic asthma population. No significant difference in genotype and alleles frequencies was found between the allergic asthma patients and control subjects (P=0.314, P=0.321). The genotype frequencies of GG, GT and TT in -574 T/G polymorphism were 0.992, 0.008 and 0 respectively in the healthy population and were 0.941, 0.059 and 0 respectively in the allergic asthma population. There was significant difference in genotype and allele frequencies between the allergic asthma patients and control subjects (P=0.046, P=0.048).
CONCLUSIONThere are polymorphism sites of the promoter region in TIM-3 , and one of these sites, the -574 G/T polymorphism site, may be associated with allergic asthma in the population of adult Hans from Hubei province of China.

5.Incidence and antimicrobial resistance of pathogens of healthcare-associated infection in a general hospital for three consecutive years

Objective To understand the occurrence of healthcare-associated infection (HAI),distribution of pathogens,and drug resistance in a general hospital in 2014-2016,provide basis for prevention and control of HAI.Methods Clinical data of hospitalized patients from January 2014 to December 2016 were collected by prospective and retrospective investigation,distribution and drug resistance of pathogens causing HAI were statistically analyzed.Results From 2014 to 2016,4 750 patients had 5 352 cases of HAI,incidence and case incidence of HAI were 2.19% and 2.46% respectively.Incidences of HAI in three years were 2.47%,2.07%,and 2.05% respectively,showing a decreased tendency,difference was statistically significant (x2 =36.217,P<0.01).Incidences of HAI were high in intensive care unit,department of neurosurgery,as well as department of burn and plastic surgery,the common HAI sites were respiratory tract,urinary tract,and surgical sites.The main pathogens causing HAI were gram-negative bacteria (76.10%).Resistance rates of Escherichia coli to cephalosporins and fluoroquinolones were relatively higher (>60%);resistance rates of Klebsiella pneumoniae to carbapenems were relatively higher;resistance rates of Pseudomonas aeruginosa to carbapenems showed a increased tendency year by year (x2 =15.175,P =0.001);antimicrobial resistance rates of Acinetobacter baumannii were all>50 %.Methicillin-resistant Staphy lococcus aureus (SA) accounted for about 60% of SA,methicillin-resistant coagulase negative Staphylococcus(CNS) accounted for more than 80% of CNS,vancomycin-and linezolid-resistant Staphylococcus spp.were not found.Conclusion The common pathogens causing HAI in this hospital are higher.Scientific monitoring on HAI and regular analysis of clinical data are of great significance for guiding rational use of antimicrobial agents,controlling multidrug-resistant organisms,and reducing the occurrence of HAI.
6.Effect of high amounts of medium chain triglyceride and protein enteral nutrition on nutritional status in patients after major abdominal operation.

OBJECTIVETo investigate the effect of Nutrition MCT and Nutrition MF enteral nutrition on nutritional status of patients after major abdominal operation.
METHODSIn a double- blinded and randomized cross- cover study, Nutrition MCT and Nutrition MF enteral nutrition were fed in patients when the gut function restored after operation. The total calorie was 104.6 kJ(25 kcal) x kg(-1) x d(-1) and the period of full dose of enteral nutrition was 5 days. The blood samples were collected before operation,before enteral nutrition and the sixth day after full dose of enteral nutrition for the measurement of pre- albumin, total protein,albumin, transferrin and triglyceride. The urine, stool and drainage fluid were collected to analyze nitrogen balance.
RESULTSThe plasma protein and fat were obviously dropped in patients after abdominal operation and improved after the enteral nutrition support in two groups. However, the pre- albumin level increased more in patients of Nutrition MCT than Nutrition MF.
CONCLUSIONNutrition MCT can obviously improve the nutritional status of patients after major abdominal operation.

7.Diagnostic value of LP(a),D-D combined LAD for LATH in atrial fibrillation

Objective:To study diagnostic value of left atrial diameter(LAD),lipoprotein(a)[LP(a)]combined D-dimer(D-D)for left atrial thrombus(LATH)in patients with atrial fibrillation(AF).Methods:According to results of transesophageal echocardiography,a total of 367 AF patients treated in our hospital were divided into LATH group(n=67)and no LATH group(n=300).General clinical data,LAD,serum Lp(a)level and positive D-D rate were compared between two groups,and diagnostic value of above indexes for LATH in AF was ana-lyzed.Results:Compared with no LATH group,there were significant rise in LAD[(40.93±4.69)mm vs.(44.65±4.31)mm],serum Lp(a)level[(17.05±2.31)mg/dl vs.(27.42±3.06)mg/dl]and positive D-D rate(5.67%vs.22.34%)in LATH group(P=0.001 all).ROC curve indicated that AUC of Lp(a),positive D-D,LAD single detection and triple combined detection diagnosing LATH in AF was 0.711,0.584,0.728 and 0.801 respectively;sensitivity was 43.28%,22.39%,64.18%and 65.67%respectively;and specificity was 92.00%,94.33%,76.67%and 80.67%respectively.AUC of triple combined detection was significantly higher than that of any single detection,and sensitivity and specificity were significantly higher than those of Lp(a)and positive D-D single detection(P<0.05 or<0.01).Conclusion:Combined detection of Lp(a),positive D-D and LAD possess high diagnostic value for left atrial thrombus in atrial fibrillation.
9.Application of real-case-based training and examination method in ICU nurse training

Objective To investigate the effect of real-case-based training and examination method in ICU nurses training. Methods A test group of multi-level real-case-based training was established and took the examination of nurses in ICU. The examination included the ability of disease observation and assessment, clinical thinking, application of nursing procedure and so on. The frequency of assessment was divided into three parts:once a month, every three months and every six months. Results After the real-case-based training and examination program, the comprehensive score of ICU nurses at excellent and favorable level were improved. The excellent and favorable rates of the first and the fourth season were 31. 6% and 59. 6%, which had a significant difference (χ2 =9. 161,P<0. 05). The nursing quality of the second half of year was significantly improved. There was no case of pressure sore, unplanned extubation, out for inspection. The compliance of hand washing and bed raise was improved. The incidence of ventilator associated pneumonia, catheter related bloodstream infection and urinary tract infection was decreased. The nurse training examination, education level and learning atmosphere were significantly improved (P <0. 05). Conclusions The real-case-based training and examination model can effectively improve the ICU nurses′ clinical nursing competency, and it also can promote the enthusiasm and the quality of nursing.
10.Effects of Xuesaitong injection on inflammation induced by OGD/reoxygenation in BV2 microglia.

The present study aims to investigate the effects of Xuesaitong (XST)injection on inflammation induced by OGD/reoxygenation in BV2 microglia cell and explore the underlying mechanisms.The effects of XST injection were evaluated in terms of cell viability, secretion of TNF-α, IL-1β, IL-6 and IL-10 into culture media, protein expression of p-ERK1/2, p-JNK and p-p38, and nuclear translocation of NF-κB p65. The results showed that XST injection significantly increased cell viability, suppressed release of TNF-α, IL-1β, IL-6 and IL-10 and down-regulated p-JNK1/2 and p-p38 MAPK expression in BV2 microglia cells induced by OGD/R injury, whereas it had no effect on p-ERK1/2 expression. Furthermore, XST injection suppressed nuclear translocation of NF-κB p65 in BV2 microglia after OGD/R injury. These data indicate that the neuroprotective effects of XST injection on OGD/R injury are associated with its inhibition of pro-inflammatory mediator production, down-regulation of JNK1/2 and p38 MAPK activation, and suppression of NF-κB p65 nuclear translocation in BV2 microglia cells.
