Journal of Jilin University(Medicine Edition) 2014;(5):967-971
Objective To observe the influence of sirtinol,a silent information regulator 1(SIRT1)inhibitor,in the cell proliferation, cell cycle progression and the expression levels of positive regulator proteins of the cell cycle including Cyclin D1,CDK4 and pRb in prostate cancer DU145 cells, and to explore the possible mechanism of SIRT1 in occurrence of prostagte cancer. Methods The DU145 cells at logarithmic growth phase were cultured in vitro and divided into control group(DMSO)and different doses (10,25,50μmol·L-1 )of sirtinol groups. The inhibitory rate of growth of DU145 cells was detected with MTT method, the SIRT1 mRNA and protein expression levels were determined by RT-PCR and Western blotting method, and the cell cycle was measured by flow cytometry. The Cyclin D1,CDK4 and pRb protein expression levels were examined by Western blotting method. Results Compared with control group, the inhibitory rates of growth of the DU145 cells in different doses of sirtinol groups were increased markedly in a dose-dependent manner(P<0.01),and the flow cytometry analysis result showed the DU145 cells at G1 phase were increased (P<0.01 ). Compared with control group, the expression levels of SIRT1 mRNA and protein in DU145 cells in different doses of sirtinol groups were decreased significantly(P<0.01);the expression levels of Cyclin D1 and pRb proteins were decreased(P<0.01),whereas the expression levels of CDK4 had no change(P>0.05).Conclusion SIRT1 inhibition by sirtinol can inhibit the cell growth of prostate cancer DU145 cells in a dose-dependent manner and arrest the cell cycle progression, and its mechanism may be related to decreasing the CyclinD1 and pRb protein expressions.
7.Open stone surgery: is it justified in the era of minimally invasive therapies?.
Chinese Journal of Surgery 2009;47(4):244-247
OBJECTIVETo summarize the efficacy of open stone surgery for renal and ureteral calculi, and to investigate the current indications for upper urinary tract stones treated by open stone surgery.
METHODSTo analyze retrospectively the cases with upper urinary tract calculi treated by open stone surgery from January 1995 to December 2004.
RESULTSIn all, there were 667 patients treated for renal and ureteral calculi by open stone surgery, at the same time 9000 cases with upper urinary tract calculi also treated in our centre with open surgery rate 7.4%. Indications for open surgery were complex stone burden of 297 cases, nephrectomy for non-functioning kidneys of 137 cases, with anatomical abnormalities of 134 cases, ESWL or endoscopy failure of 57 cases, with co-morbid medical diseases of 30 cases, with renal carcinoma of 5 cases, with xanthogranulomatous pyelonephritis of 3 cases, with kidney injury of 2 cases, and with other ipsilateral operation of 2 cases. Of 145 cases for nephrectomy, 16 patients and 6 patients suffered from various intraoperative and postoperative complications respectively;of 522 patients treated by open stone surgery, 7 patients and 34 patients were suffered from various intraoperative and postoperative complications respectively; 57 patients with multiple renal calculi had residual calculi with overall stone-free rate 89.1%. And no case died in perioperative period.
CONCLUSIONSOpen surgery maintains its important role for treatment of renal and ureteral calculi because of its safety and efficacy. Nephrectomy for nonfunctioning kidney with renal and/or ureteral calculi, huge and complex renal calculi, anatomical abnormalities, minimal invasive therapies failure and some selective patients with co-morbid medical diseases are suitable for open stone surgery with high success and low morbidity rate.

Objective:To screen the differentially expressed miRNAs in umbilical cord tissue of children with nonsyndromic cleft lip and/or cleft palate( NSCL/P) using miRNA microarray and comprehensive bioinformatics analysis for the prediction of related the bio-logical process and signaling pathways. Methods:Umbilical cord tissues of 4 cases of healthy newborns' and 4 lip or palate tissues of 4 cases with NSCL/P without other disease aged younger than 2 years were collected. The differentially expressed miRNAs were screened by miRNA microarray. Targets of dysrugulated miRNAs were predicted by TARGETSCAN-VERT, MIRDB and RNA22-HSA. All the gene sets were analyzed by gene ontology and pathway enrichment. Results: MiRNA microarray demonstrated that 254 miRNAs were dysregulated(181 miRNAs were up-regulated and 73 downregulated,P <0. 05). The dysregulated miRNAs targets contained 5029 genes. The dysregulated miRNAs targets were enriched in anatomical structure development, cell adhesion, cell proliferation, cell motili-ty and other biological processes. The dysregulated miRNAs targets were enriched in Wnt, mTOR, cGMP-PKG, TGFβ, PI3K-Akt and other signaling pathways. Conclusion:The target genes set of miRNAs are enriched in multiple biological processes and signaling path-ways related to NSCL/P, which indicate that genetic and environmental factors may influence the development process of NSCL/P.

Objective To study the effects on berberine hydrochloride on the mitochondria membrane potential and free intracellular calcium of HaCaT cells, and elucidate the mechanism of action of berberine on keratinocytes. Methods Rhodamine-123 fluorescence (very sensitive to mitochondria membrane potential) and Fluo-3/AM fluorescence (suitabe to detect free intracellular calcium in single HaCaT cell) were measured by laser scanning confocal technique. Results Fluo-3/AM fluorescence intensity of HaCaT cells was persistently increased after treating with berberine at concentrations of 5 ? 10-5M, 2.5 ? 10-5M and 1.25 ? 10-5M, and significant differences were observed as compared with the PBS control. The intensity of rhodamine-123 fluorescence in HaCaT cells was decreased immediately when exposed to berberine, with significant difference from that of the PBS control. Conclusions It is suggested that berberine could increase free intracellular calcium and decrease mitochondria membrane potential of HaCaT cells, induce overload of intracellular calcium, influence energy metabolism, and then inhibit the proliferation of keratinocytes.
