Objective To study the effects of ? carotene on rat liver fibrosis induced by carbon tetrachloride (CCl 4). Objective To study the effect of carvedilol on heart rate variability and plasma N-terminal brain natriuretic peptide(NT-proBNP) in patients with chronic heart failure.Methods 106 patients with chronic heart fail-ure were randomly divided into the two groups ..53 patients in the control group were treated by the conventional ther-apy plus metoprolol ,while 53 patients in the observation group were treated by the conventional therapy plus carve -dilol.They were treated for six months .Heart rate variability and plasma NT-proBNP were measured before and after treatment in the two groups .Results The total effective rate of the control group was 83.0%,which was significantly lower than 94.3% of the observation group (χ2 =6.26,P<0.05).Before treatment,SDNN,SDANN,RMSSD and PNN50 between the two groups had no significant differences (t=0.284,0.360,0.410,0.302,all P>0.05).After treatment,SDNN,SDANN,RMSSD and PNN50 were significantly increased (t=3.095,9.184,3.622,4.302,2.261, 4.522,2.921,2.992,P <0.05 or P <0.01).Compared with the control group,those in the observation group increased more significantly (t=8.065,3.116,3.209,2.171,P<0.05 or P<0.01).After treatment,plasma NT-proBNP were significantly decreased in the two groups (t=7.093,9.773,all P<0.01).Compared with the control group,plasma NT-proBNP of the observation group decreased more significantly (t=4.773,P<0.01).Conclusion Carvedilol can more significantly improve heart rate variability and plasma NT-proBNP in patients with chronic heart failure.

Objective To study the impact of alprostadil on serum high-sensitivity C-reactive protein ( hs-CRP) and troponin I( cTnI) in patients with diastolic heart failure.Methods 92 patients with diastolic heart failure were randomly divided into the two groups,the control group and the observation group,46 cases in each group.The control group used the conventional treatment and the observation group on the basis of conventional therapy plus al-prostadil.Two groups were treated for 4 weeks.They were measured filling velocity ratio (E/A) in patients with early and late peak atrial contraction,stroke volume (SV),left ventricular end-diastolic diameter (LVEDD),6min walking distance and serum hs-CRP, cTnI before and after treatment.Results There were no significant difference about LVEDD,E/A and SV(t=0.,0.261,0.445,all P>0.05).After treatment for four weeks,LVEDD were (53.1 ± 5.1)mm and (47.3 ±4.7)mm,LVEDD of the two groups had significantly decreased (t=4.601,9.331,P<0.05, P<0.01),and post-treatment the observation group LVEDD decrease more significantly (t=2.914,P<0.05).After 4 weeks of treatment,the control group after treatment E/A and SV were (0.79 ±0.15) and (72.6 ±5.4),the ob-servation group E/A and SV were (0.85 ±0.14) and (79.5 ±5.9)mL,E/A and SV in both groups were significant-ly elevated (t=3.172,4.710,3.924,5.776,P<0.05,P<0.01),and E/A and SV of post-treatment the observation group increased more significantly (t=3.771,2.840,P<0.05).After treatment,patients 6min walking distance were (383.4 ±47.5)m and (426.1 ±49.1) m,6min walking distance of patients were significantly improved after treat-ment (t=8.776,12.648,P<0.01),and the observation group improved significantly more than that of the control group (t=3.019 2,P<0.05).In the control group,serum hs-CRP patients,cTnI were (21.3 ±4.6) mg/L and (2.13 ±0.52) ng/mL,(13.8 ±4.1) mg/L and (1.05 ±0.38) ng/mL in the observation group.Two groups of patients before treatment serum hs-CRP and cTnI was no significant difference (t=0.527,0.493,all P>0.05).After 4 weeks of treatment,the serum hs-CRP,cTnI were significantly lower in both groups (t=3.612,8.772,2.924, 5.164,P<0.05,P<0.01),and compared with the control,the observation group reduced more significantly (t=3.061,7.114,P<0.05).Conclusion Alprostadil can decrease serum hs-CRP and cTnI of patients with diastolic heart failure.

Objective To study the effect of ulinastatin on serum C-reactive protein (CRP) and exhaled nitric oxide(FeNO) in patients with bronchial asthma.Methods 98 patients with bronchial asthma were randomly divided into the control group (n =49 cases) and the observation group (n =49 cases).The patients in the control group were treated through the conventional treatment,while the patients in the observation group were treated through the conventional treatment plus ulinastatin.Pulmonary function,asthma symptom scores,serum CRP and FeNO were measured before and after treatment.Results FEV1％ and predicted PEF values after treatment were significantly increased (t =4.720,8.112,3.724,6.723,all P ＜ 0.05).Compared with the control group,the observation group after treatment increased more significantly (t =3.102,4.002,all P ＜ 0.05).After the treatment,the asthma symptom scores were significantly decreased (t =2.190,6.021,all P ＜ 0.05).Compared with the control group,the asthma symptom scores of the observation group significantly decreased (t =2.620,P ＜ 0.05).After treatment,FeNO and CRP were significantly decreased (t =9.124,13.076,5.772,7.064,all P ＜ 0.01).Compared with the control group,FeNO and CRP of the observation group significantly decreased (t =3.060,5.401,all P ＜ 0.05).Conclusion Ulinastatin can significantly decrease serum CRP and FeNO in patients with bronchial asthma.

Objective:To probe distribution law and pathogenesis of TCM complex symptoms of functional dyspepsia and irritable bowel syndrome.Methods:Generally recognized criteria was to establish database of comprehensive analysis of the data gained by the four methods of diagnosis,and analyze with SPSS8.0.Results:Syndrome distribution was most closely related with spleen deficiency,accounting for 66.7%;and the next,with stagnation of liver-Qi,accounting for 40.3%;the last,with the kidney,only accounting for 5.6%.Conclusion:Cause of disease and inducing factors of this disease are closely correlated with pathogenic cold, diet of lipids,mental depression,prolonged disease,wound due to abdominal operation.The disease is located at the stomach, intestine with most coexistence of deficiency and excess,and spleen deficiency is the key of pathogenesis.

AIM To explore the effects of Sini Decoction (Aconiti lateralis Radix Praeparata,Zingiberis Rhizoma,Glycyrrhizae Radix et Rhizoma) on rat myocardial fibrosis induced by isoproterenol.METHODS Forty SD rats were randomly divided into blank,model,captopril [100 mg/(kg · d)] and Sini Decoction [3.8 g/(kg · d)] groups,with ten rats in each group.Except for the blank group,the rest rats were given subcutaneous injection of isoproterenol [5 rg/(kg · d)] to prepare myocardial fibrosis model,and successive administration lasted for four weeks.At 20 h after the last administration,hemodynamics changes of heart were detected;heart weight indexes were calculated;the changes of myocardial pathological morphology were observed by HE and Masson staining;NO level in serum was measured by nitrale reduetase;SOD activity in serum was measured by xanthinoxidase method;MDA level in serum was measured by thiobarbituric acid method;the protein expressions of RhoA,ROCK1 and eNOS in myocardial tissue were detected by Western blot method.RESULTS Compared with the model group,the cardiac function of rats was significantly improved;the myocardial collagen content was significantly decreased;the MDA level in serum was obviously decreased;the NO level and SOD activity were significantly increased;the protein expressions of RhoA and ROCK1 in myocardium tissue were significantly reduced,and the protein expression of eNOS was markedly increased in the Sini Decoction group.CONCLUSION Sini Decoction can improve the isoproterenol-induced myocardial fibrosis in rats,and its mechanism may be related to inhibiting the RhoA/ ROCK signaling pathway,in turn,raising the expression of eNOS,which leads to reduced oxidative stress.

Objective To investigate the relationship among the proliferation of CD4+T cells, the intracellular levels of interleukin-10 (IL-10) and transforming growth factor-?1(TGF-?1), and allergic bronchial asthma. Methods Dermatophagoides farinae antigen were prepared as allergen. Twenty-five patients with asthma and 15 healthy individuals were enrolled and divided into blank control group, allergen group and self control group, respectively, after venous blood sample collection. The proliferation of CD4+T cells and the distributions of CD4+/IL-10+, CD4+/TGF-?+1 and CD4+/IL-10+/TGF-?+1 were measuredby flow cytometry (FCM). Results The distributions of CD4+/IL-10+, CD4+/TGF-?+1 and CD4+/IL-10+/TGF-?+1 could hardly be detected in the peripheral blood samples of the blank controls of the patients with asthma and healthy ones. In the allergen group of the healthy individuals, the peripheral blood CD4+T cells were significantly proliferated, and the proportions of CD4+T cells andCD4+/IL-10+ cells were much higher than the self control group, while there was no significantly increase in the proportions of CD4+/TGF-?+1 and CD4+/IL-10+/TGF-?+1 subgroups. In the allergen group of those with asthma, the proportions of peripheral blood CD4+, CD4+/IL-10+, CD4+/TGF-?+1 and CD4+/IL-10+/TGF-?+1 cells were not found significantly increased compared with those self controls. Objective To explore the relationship of thyrotropin receptor antibody (TRAb) and soluble intercellular adhesion molecule-1 ( sICAM-1 ) in patients with thyroid-associated ophthalmopathy ( TAO), and the role played by TRAb and sICAM-1 in the pathogenesis of TAO. Method Twenty-three TAO patients were assigned to groups according to the clinical activity score and EUGOGO. All patients were treated with intravenous methylprednisolone pulse. The levels of serum TRAb and sICAM-1 were evaluated by a competitive radioimmunoassay and enzyme-linked immunosorbent assay respectively before treatment and by the end of each methylprednisolone pulse. Results The differernce in serum TRAb levels was associated with activity scores of TAO (P=0. 020). The change in serum sICAM-1 was associated with durations of TAO ( P = 0.015). During methylprednisolone treatment in active TAO patients, the levels of TRAb kept on decreasing gradually and markedly declined after the third methylprednisolone pulse in active TAO patients (P＜0.05). The trends of changes in serum TRAb and sICAM-1 levels were both different in active and inactive TAO patients by tendency analysis. Conclusion TRAb level was related to the activity of TAO and might serve as a significant predictor of response to methylprednisolone therapy. The negative correlation between sICAM-1 levels and duration of TAO corroborates the role played by ICAM-1 during the early stage of TAO. Higher sICAM-1 levels are not expected to be specific to TAO and may not predict a response to methylprednisolone therapy.

Objective To investigate the experimental condition of separate purification and directed differentiation potency of pancreatic stem cells of adult rats in vitro. Methods The pancreas of adult rats were digested by collagenase V in situ perfusion. Filtration was performed by 100 mesh filter. Ficoll 400 density gradient centrifugation was used to separate pancreatic stem cells and stem cells were for cultivation. Epidermal growth factor (EGF), fibroblast growth factor (bFGF) was added to medium for in vitro cultivation.Immunofluorescence staining method was used to detect the expression of CK19, Pdx-1, nestin, insulin and glucagon, and the positive cell rate was calculated. The differentiated cell was evaluated by dithizone (DTZ)staining; insulin excretion function was determined by optical enzyme labeled ELISA staining. Results The pancreatic stem cell obtained from the study could be cultivated in vitro for 8 generations. The expression of CK19, Pdx-1 and nestin of the tem cells were all positive, and the rate of positive cells were (88.6 ± 6.2)% ,(84.6 ±8.6)% and (81.3 ±7.5)%. The differentiated cell was in brownish red color by DTZ dye. After high concentration sugar stimulation, the expression of insulin secretion was increased in the supernatant.Conclusions This method can harvest highly purified, and large amount of pancreatic duct stem cell.Artificial induction may result directed differentiate for islet-like clusters and have insulin secrete function.
