1.FABRICATION OF BONE WITH POLY-BETA-HYDROXYBUTYRATE AND BONE MARROW STROMAL CELL EMPLOYING THE METHOD OF TISSUE ENGINEERING

To investigate the feasibility of using poly-beta-hydroxybutyrate as scaffolds for bone tissue engineering. Marrow stromal cells were harvested from New Zealand rabbit′s iliac bone. After being cultured and multiplied in vitro,with the use of dexamethasone to promote the osteoblastic phenotype of the cells, the cells were seeded into poly-beta-hydroxybutyrate. The cells/ poly-beta-hydroxybutyrate construction was implanted subcutaneously in nude mice. In control animals, poly-beta-hydroxybutyrate alone was implanted. Osteogenesis was assessed by histological and roentgenographic analysis. Four weeks after implantation, osteoid tissue had been observed in the specimens of the composite; 8 weeks after implantation, large amount of new bone had been found, and lamellar bone had been observed also. In the control, only fibrous tissue had been found. It suggested that poly-beta-hydroxybutyrate can be used as a scaffold material for bone tissue engineering.

4.Differences on the parameters of the optic disc between the amblyopic eye and non-amblyopic eye in adolescent with anisometropic amblyopia

AIM: To compare the peripapillary retinal nerve fiber layer ( RNFL ) thickness and peripapillary topographic map's parameters between amblyopic eyes and non -amblyopic eyes in adolescent with anisometropic amblyopia by optical coherence tomography(OCT).
METHODS: Thirty - four juveniles with anisometropic amblyopia were selected. Peripapillary RNFL thickness and peripapillary topographic map were measured by frequency domain OCT with both eyes in all participants, and the differences between amblyopic eyes and non -amblyopic eyes were compared.
RESULTS: There was no significant difference in average thickness of peripapillary RNFL and in any other region of peripapillary RNFL. The disc area in amblyopic eyes was bigger than that in non - amblyopic eyes ( t =2. 8054,P= 0. 0263). The disc area in amblyopic eyes were significantly related to the thickness of nasal RNFL and the rim area(r= 0. 7592,0. 7501;P= 0. 0289,0. 0321).
CONCLUSION: There existed some difference in peripapillary structure between amblyopic eyes and non-amblyopic eyes in adolescent with anisometropic amblyopia.
5.Study on non-enzymatic separation and culture method of adipose-derived stem cells

Objective To explore a separation and culture method of human adipose-derived stem cells(ADSCs) suitable for the clinical application .Methods The non-enzymatic method and the collagenase digestion method were adopted to isolate and culture the cells from the human adipose tissue in the individuals with liposuction .The characteristics of isolated mesenchymal stem cells were comparatively analyzed .Results The required time in the non-enzymatic method was one third of that in the collagenase di-gestion method and the cellular morphology ,reproductive capacity ,immunophenotype and differentiation potential of the isolated cells were consistent to those isolated by the collagenase digestion method .Conclusion The no-enzymatic method may isolate and culture ADSCs from the adipose tissue in the individual with liposuction ,which is a safety and reliable isolation and culture method of human adipose tissue-derived stem cells suitable for clinical application .
6.The Role of Androgen in High-Salt Induced Hypertension in Rats

Objective To study the effects of high salt intake on blood pressure and renin-angiotensin in male rats with different plasm androgen levels.Methods Thirty male Wistar rats were randomized to sham(n=10)or operated(n=10),or castration(n=10),or testosterone replacement after castarion(26.7 mg/kg,n=10)and fed with 8% NaCl for 8 weeks.Tall arterial pressure were recorded before,4 and 8 weeks after experiment.Serum PRA,plasma angiotensin Ⅱ(Ang Ⅱ)and testosterone(T)were determined by radioimmunoassey respectively. 7.Clinical curative effect observation of ultra pulse CO2 laser combined with collagen dressing in treatment of primary cutaneous amyloidosis

Objective To investigate the clinical efficacy and safety of ultra pulse CO2 laser combined with collagen dressing in treatment of cutaneous amyloidosis.Methods A total of 68 cases of primary cutaneous amyloidosis patients according to the different treatment methods was divided into two groups:the control group (n =33 cases) that was given acitretin capsules oral and topical compound flumetasone ointment packet treatment,and observation group (n =35 cases) with ultra pulse CO2 laser combined with collagen dressing external treatment.Two groups were observed in patients with curative effect and adverse reaction.Results Disease of integral observation group after treatment was significantly lower than the control group(t =3.12,P <0.05).The observation group had the efficiency of [88.6% (31/35)] that was significantly higher than that of control group[17/33(51.5%),x2 =11.23,P <.0.01].The observation group had significantly shorter healing time [(5.38 ± 1.81) d] compared to the control group [(10.75 ±2.06)d,t =11.39,P < 0.01].The observation group did not have recurrence,which was significantly different from the control group (x2 =4.90,P < 0.05).Conclusions Ultra pulse CO2 laser combined with collagen dressing in treatment of primary cutaneous amyloidosis is a kind of effective treatment methods with remarkable curative effect and less adverse reactions.It is worthy of clinical application.
8.Kinematics Modeling and Analysis of Central-driven Robot for Upper Limb Rehabilitation after Stroke.

The present paper proposed a central-driven structure of upper limb rehabilitation robot in order to reduce the volume of the robotic arm in the structure, and also to reduce the influence of motor noise, radiation and other adverse factors on upper limb dysfunction patient. The forward and inverse kinematics equations have been obtained with using the Denavit-Hartenberg (D-H) parameter method. The motion simulation has been done to obtain the angle-time curve of each joint and the position-time curve of handle under setting rehabilitation path by using Solid Works software. Experimental results showed that the rationality with the central-driven structure design had been verified by the fact that the handle could move under setting rehabilitation path. The effectiveness of kinematics equations had been proved, and the error was less than 3° by comparing the angle-time curves obtained from calculation with those from motion simulation.

9.Relationship between plasma exosomal miR-346 and treatment outcomes in patients with multidrug resistant pulmonary tuberculosis

Abstract: Objective To investigate the relationship between plasma exosomal microRNA (miRNA)-346 and treatment outcomes in patients with multidrug-resistant tuberculosis (MDR-TB), to provide more reference basis for the treatment of MDR-TB patients. Methods A total of 406 patients with MDR-TB admitted to Tuberculosis Control and Prevention Institute of Shaanxi Provincial between January 2018 and May 2021 were selected as the study subjects. General clinical data of the patients were collected and analyzed. The expression level of plasma exosomal miR-346 was detected by real-time fluorescence quantitative polymerase chain reaction. The predictive value of plasma exosomal miR-346 for treatment outcome was analyzed using receiver operating characteristic (ROC) curve analysis. Furthermore, the relationship between the expression of plasma exosomal miR-346 and treatment outcome was analyzed using a multivariable logistic regression model. After standard treatment, patients were divided into good treatment outcome group (n=226) and poor treatment outcome group (n=180) according to the treatment outcome. Results Typical exosomes were identified by transmission electron microscopy, particle size analysis and Western blot, that is, plasma exosomes were successfully extracted. In the poor treatment outcome group, more patients were complicated with diabetes or HIV infection, and the proportion of patients with pulmonary cavity, acid-fast bacilli smear positive rate >1+, previous treatment history and fluoroquinolone resistance was also significantly increased, and the levels of white blood cells, neutrophils, and monocytes were significantly increased, while the level of albumin was significantly decreased, with statistical significance (P>0.05). Compared with the good treatment outcome group [0.61 (0.46, 0.74)], the expression level of plasma exosomal miR-346 in the poor treatment outcome group [1.23 (0.60, 2.02)] was significantly higher (Z=-13.185, P<0.001). ROC curve analysis showed that the area under the curve of plasma exosomal miR-346 to predict adverse outcomes of MDR-TB treatment was 0.881 (95%CI: 0.846-0.915), with a sensitivity of 78.3% and specificity of 86.7%. The corresponding cut-off value was 0.81. Multivariate logistic regression analysis showed that diabetes, AIDS virus infection, pulmonary cavity, AFB smear positive degree>1+, previous treatment history, fluoroquinolones resistance and high expression of plasma exosomal miR-346 were independent influencing factors for poor treatment results of MDR-TB (P<0.05). Conclusions High expression of plasma exosomal miR-346 is associated with the high risk of adverse outcome of MDR-TB treatment, and it is promising to be a useful marker for predicting the outcome of MDR-TB treatment.
10.Effect on Expression of APPmRNA in Cellular Model of AD by Therapy of Replenishing Kidney-essence and Removing Phlegm to Benefit Wisdom

This study successfully established in v itro model of Alzheimerˊs disease ( AD ) in order to investigate effects and mechanisms on AD by Bushen-Huatan-Yizhi Recipe (BSHTYZR, replenishing kidney-essence and removing phlegm to benefit wisdom ) . The PC-12 cells affected by Aβ1-42 incubated were produced for neuron cell model of AD . Then , they were intervened by the serum containing BSHTYZR , which were compared to the serum containing indomethacin . The cell morphology was observed . The neuronal survival was assessed by counting MTT assay . The expression of APP mRNA was detected with RT-PCR . And the protective effect of BSHTYZR on neuronal cells and neurotoxic effects of anti-Aβ was observed . The results showed that 2 . 5 mmol/L Aβ1-42 can significantly decrease the cell viability . The RT-PCR was used on semi-quantitative analysis for expression of APPmRNA. It showed that expression of APP mRNA was increased in each group, and the BSHTYZR group can reduce the expression significantly ( P < 0 . 01 ) . It was concluded that the BSHTYZR method can protect nerve cells , suppress the expression of APP mRNA , and reduce the toxic effects .
