2.Effects of Xiongshao capsule on blood vessel collagenase gene expression in experimental rabbits with arterial restenosis.

OBJECTIVETo observe the effects of Xiongshao Capsule (XSC) on blood vessel collagenase gene expression in experimental rabbits with arterial restenosis, and to probe its mechanisms for preventing restenosis.
METHODSRestenosis rabbit model was established by injuring endothelium of abdominal aorta by balloon dilation and feeding with high fatty diet for 6 weeks. Eighty rabbits were randomly allocated into 8 groups, Group A, normal rabbit for control; Group B, rabbit with simple injured arterial endothelium; Group C, model rabbits at different times after modeling (3 days for Group C1, 2 weeks for Group C2, and 6 weeks for Group C3); Group D, model rabbit treated with Probucol for 6 weeks; Group E and F, model rabbit treated with small and large dose of XSC respectively. The effect of XSC on collagenase gene expression during the course of restenosis was observed adopting RT-PCR method and computer image analyzer, and its mechanisms in preventing RS were probed by combined analyzing the change of collagen and patho-morphological examination.
RESULTSCompensatory dilation of lumens appeared at the end of the 2nd week; while 6 weeks after modeling, the diameters of lumens obviously diminished with an apparently increased proliferation index. The cell proliferation inhibiting effect in Group D and F was significant. The total amount of collagen increased and reached the peak at the 2nd week but without conspicuous accumulation on intima, which increased gradually and reached its peak at the 6th week. In Group D-F, especially in Group F, the amount of collagen in vascular wall (intima, media and externa) was lesser than that in Groups C. MMP-1 mRNA showed weak expression in Group A and Group C1-C3; significant difference only existed in comparing Group F with C3 (P < 0.05).
CONCLUSIONXSC could markedly increase the MMP-1 mRNA expression in injured portion of vessels, suggesting that its action in preventing RS might be related with the up-regulating of MMP-1 mRNA expression, increasing collagen degradation and reducing collagen deposition in vascular wall.

4.Study on separation of sulfonamides by capillary high-performance liquid chromatography and electrochromatography.

AIMTo establish separation methods of five sulfonamides by using capillary high performance liquid chromatography(mu-HPLC) and electrochromatography. The effect of mobile phase varies such as methanol content, pH, buffer solution concentration and voltage on their chromatographic behavior and electroosmesis flow was investigated. Capillary electrochromatography (CEC) was compared with mu-HPLC at the same condition.
METHODSStationary phase was ODS, mobile phase was methanol and 2 mmol.L-1 H3PO4 buffer solution (pH 3.0-7.0), voltage was 0- -15 kV, flow rate was 10 microL.min-1, pressure was approximately 70 MPa and UV detection wavelength was 254 nm.
RESULTSSeparations on base line have been respectively accomplished for five sulfonamides by mu-HPLC with mobile phase of methanol-2 mmol.L-1 H3PO4 buffer solution (30:70) at pH 5.0 in 67 min, and CEC with the same mobile phase at -5 kV voltage in 25 min.
CONCLUSIONElectroosmesis flow of CEC decreased with the increase in methanol content, buffer solution concentration, increased with the increase in voltage and increase slightly with the increase in pH of mobile phase. Retention values (k) of solutes to be examined decreased with increasing methanol content of mobile phase in mu-HPLC and CEC. Retention values (k) of solutes increased slightly with increasing buffer solution concentration, decreased with increasing voltage in CEC. Trimethoprim(TMP) decreased obviously with increasing voltage in CEC. The effect of pH of mobile phase on retention values (k) was more complex. Five sulfonamides were separated at the same mobile phase condition by mu-HPLC and CEC. And separation speed of CEC was much faster than that of mu-HPLC. CEC was very fit for rapid separation of sulfonamides.

5.Analysis of clinicopathologic features and morphogenesis of carcinoid tumorlets in the lung with bronchiectasis.

OBJECTIVETo describe the clinicopathologic features and immunophenotypes of carcinoid tumorlets in the lung with bronchiectasis, and to study the morphogenesis of these tiny tumors.
METHODSThe histopathologic characteristics of 3 bronchiectasis cases with carcinoid tumorlets, 11 bronchiectasis and 2 normal lungs were studied. Specific markers of the tiny tumors and the number of neuroendocrine cells (NECs) in the airway mucosa were immunohistochemically detected by EnVision method.
RESULTSThe tumorlets in the lungs presented as multi-focal nodules and most were displayed only under microscopy. These cells were arranged in clusters and foci of fascicles which were situated in the surrounding bronchial wall and bronchioles adjacent to bronchiectatic lesion, or in the scar tissues. The tiny tumors were consisted of short fusiform cells and small ovoid cells. Their nuclei were circular, oval or long fusiform and the cells were strongly argyrophilic on Grimelius staining. Intensive positive immunostaining for calcitonin, chromogranin A, NSE and gastrin were detected. Weak positive for CK, EMA, S-100 and focal positive for HC, ACTH, 5-HT were also observed. Proliferative NECs in airway mucosa adjacent to the tiny tumors increased significantly in number, compared with those in the airway mucosa of bronchiectasis without tumorlets and normal lungs (P < 0.001, respectively).
CONCLUSIONSThe clinicopathologic features and immunophenotypes of carcinoid tumorlets resemble carcinoid tumors. They are the early stage of carcinoid development. Their development may be related to the chronic pulmonary damage resulting in hypoxia and stimulating the proliferation of NECs. These pulmonary carcinoid tumorlets can be used as a model to study the tumorigenesis of carcinoid carcinoma of the lung.

6.Effect of Compound Qidan Liquid on early ventricular remodeling after acute myocardial infarction in Chinese mini-pigs.

OBJECTIVETo investigate the mechanism of Compound Qidan Liquid (CQD) for intervening ventricular remodeling (VR) after acute myocardial infarction (AMI) in Chinese mini-pigs from hemodynamic and collagen metabolic views.
METHODSAMI model of Chinese mini-pigs was established by left anterior descending (LAD) coronary artery ligation. The model pigs were then randomly divided into the sham-operative group, the model group, the captopril group, the high and low dose of CQD (hCQD and lCQD) groups, the former two were treated with normal saline and the latter three treated with corresponding drugs by gastrogavage for 4 weeks after modeling. Blood pressure (BP), left ventricular pressure (LVP), maximum ascending velocity of left ventricular pressure (dp/dtmax), myocardial renin (MCR), angiotensin (Ang II), total collagen (TC), procollagen type III (PC III), collagen type IV (CIV), laminin (LM), serum hyaluronic acid (HyA) as well as pathologic changes in myocardium were observed.
RESULTSAs compared with in the model group, levels of BP, LVP and dp/dtmax were significantly higher, LM and Ang II were lower in the hCQD group (P < 0.05 or P < 0.01); LVP was higher in the lCQD group (P <0.05); LVP and dp/dtmax were higher, Ang II was lower in the captopril group (P <0.05). Besides, levels of HyA and TC were lower in all the three medicated groups (P <0.01), while the differences of PC III and CIV among groups were insignificant.
CONCLUSIONCQD has a beneficial effect in Chinese mini-pigs after AMI for increasing LVP and dp/dtmax, improving myocardial contractility and hemodynamic condition, decreasing myocardial Ang II contents, decreasing deposition of collagen so as to alleviate the pathological process of VR after AMI.

7.Application of uniform design in research of traditional Chinese medicine.

In the uniform design, all level number of experimental relevant factors are equally distributed in the range of experiment, each experimental point shows representation better. The times of experiment are reduced and the data obtained can be treated with computer. Uniform design is a potent tool in seeking for the optimal experimental condition or the optimal proportion of experiments. So, it is often used to study extraction and preparation form technology of TCM. In recent years, the uniform design has been used in the domain of traditional Chinese recipe and the compatibility of its component drugs. The study showed that the analytical results of TCM theory and that of uniform design are extremely consistent. As an optimal screening means in modernizing study on Chinese herbs, using uniform design in studying classical recipe, the "syndrome" corresponding recipe whereas it is helpful to deeply recognize the scientific connotation of "recipe syndrome".

8.The changes of micrangium density of the reverse-flow random flap with a superficial venous trunk: an experimental study using stereological methods.

OBJECTIVETo study the effects of the superficial venous trunk (SVT) and its nourishing vessels on early revascularization of the reverse-flow random flap in order to provide theoretical evidences for clinical applications.
METHODSThe morphologic changes of micrangium of the reverse-flow random flap with SVT and without SVT were observed at different phases and compared by optical microscopy and stereological methods.
RESULTSThe micrangium density of the flaps with SVT and without SVT showed a tendency of increasing at 3 days after the operation. At 5 to 10 days, the micrangium density of the flap with SVT was much higher than the flap without SVT. There was significant difference between the two groups, which was proved by the optical microscopy observation.
CONCLUSIONSThis study suggests that SVT and its nourishing vessels could promote early revascularization of the flap after transplantation. The SVT can be of benefit to the survival of the reverse-flow random flap.

10.Clinical study on effect of xiongshao capsule on restenosis after percutaneous coronary intervention.

OBJECTIVETo further evaluate the effect of Xiongshao Capsule (XSC) on the restenosis (RS) after percutaneous coronary intervention (PCI), based on previous clinical trials by which the preventive effect of XSC on RS after PCI has been proved.
METHODSAdopting the randomized, double-blind and placebo controlled method, the 124 patients enrolled, who were patients with coronary artery disease and received intracoronary stenting, were randomly assigned into two groups, the XSC group and the placebo control group, with 62 in each group. Six months of follow-up study was carried out on them to observe the intervention effect of XSC, which was predominantly evaluated by RS rate estimated by coronary angiography (CAG), combined with the recurrent rate of angina, clinical end-point event, and blood-stasis syndrome scores.
RESULTSSixty two cases of 118 effective patients were followed-up with CAG for 6 months, the follow-up rate was 52.5%. The incidence of RS in the XSC group and the control group was 24.1% and 48.5% respectively, and the incidence of RS in stent in them was 14.0% and 42.0% respectively, indicating that the incidence of RS rate in the XSC group was significantly lower than that in the control group (P < 0.05). After patients had been treated for 6 months the extent of angiostenosis and diameter of the culprit arteries, determined by CAG was 26.58 +/- 20.72% and 2.21 +/- 0.85 mm in the XSC group, and 41.19 +/- 30.92% and 1.72 +/- 0.99 mm in the control group, respectively, suggesting that the extent of angiostenosis and the diameter loss were significantly reduced in the former than those in the latter, respectively (P < 0.05). The incidence of clinical end-point event was 11.7% and 27.6% in the XSC group and the control group in the 6 months' follow-up period respectively, it was significantly lower in the XSC group than that in the control group, with the P value close to statistical significance (P = 0.051). The blood-stasis syndrome scores in the XSC group was significantly lower in comparing with that in the control group after 6 months' treatment (P < 0.01).
CONCLUSIONXSC can effectively prevent RS after PCI in combination with conventional Western medical treatment.
