2.Anti-tumor and Anti-metastasis Effects of Tumor Cells Transduced with the Gene, Cording for Human Interleukin-2

Reccent investigations demonstrated that genetically modified tumor cells could be used as vaccines against cancer. In this light, we introduced human interleukin -2 (IL-2) gene into a poorly immunogenic rat fibrosarcoma cell line WBT, a human lung abenocarcinoma cell line AGZY and a highly metastatie sub - cell line Anip973 derived from AGZY. The transduced cells showed no obvious changes in morphology, growth and colony - forming ability in soft agar.Transduction with IL-2 gene rendered the cells more sensitive than the parental cells and cells transduced with NeoR gene alone to the lysis by cytotoxic lymphocytes. High levels of IFN and TNF were detected in the supernatants of cocultured lymphocytes and tumor cells containing IL-2 gene. Tumorigenicity of WBT/IL-2 cells was greatly reduced in syngeneic rats. Metastatic capacity of Anip973 in nude mice was blocked by the transduction of IL-2 gene, but not the transduction of NeoR gene alone. Inoculation of WBT/IL-2 cells conferred rats resistance to the challenge of parental WBT cells.Splenocytes from WBT/IL-2-inoculated rats restimulated with WBT cells in vitro showed high CTL activity against WBT cells. Our results demonstrated the potential application of tumor cells transduced with IL— gene as active vaccine against cancer.

Objective:Combined laparoscopy and hysteroscopy in the diagnosis and therapy of the female infertity is and advanced means in the gynecologcial field.We analysed the causes and the treatments of the 71 infertile patients.Pelvic adhesions,endometrosis and salpingtis were the main causes of infertity in our 71 cases.Of 71 cases,29 cases were primary infertity,in whichc endometriosis,PCOS and anatomic uterine defect were the main causes,and 42 cases were secondary infertity,in which 35 cases (83%) had previous intrauterine operations.All the patients were treated for the causes.We concluded that combined laparoscopy and hysteroscopy can diagnose the causes of infertity and can observe the tubal patency by hysteroscopic tubal cathertetization and hydrotubation,which will direct the treatments and estimate the results of the therapy.Methods:Infertity;Combined Laparoscopy and Hysteroscopy.

8.Risk factors for healthcare-associated infections caused by Candida

Objective To explore the risk factors for healthcare-associated infection (HAI)caused by Candidain a hospital.Methods 64 patients with healthcare-associated(HA)Candidainfection in a hospital between January 201 1 and October 2013 were selected as case group,and 64 patients without HACandidainfection during the same period were as control group,clinical data of two groups of patients were analyzed.Results The mainCandidacau-sing HAI were Candidaalbicans(68. 75% ),followed by Candidatropicalis(15. 62% ),Candidaglabrata (9.38% )andCandidakrusei(6.25% ).The major infection sites were respiratory tract(67.19% ),urinary tract (18.75% )and digestive tract(10.94% ). Univariate analysis showed the risk factors for HA Candidainfection were age,length of hospital stay,diabetes,and malignant tumor and so on;multivariate logistic regression analysis showed that important risk factors for HACandidainfection were age(OR95% CI:2.57-33.67),length of hospi-tal stay(OR95% CI:2.17-25.37),malignant tumor(OR95% CI:1.04-15.23),chemotherapy(OR95% CI:1.76-20.63),and use of respirator(OR95% CI:4.67-96.37).Conclusion Risk factors for HACandidainfection are complicated,in order to reduce the incidence of HA Candidainfection,risk factors should be considered,and preventive measures according to risk factors should be made.
9.Efficacy of modified PLT additive solution for platelets storage at low temperature(10℃) on rabbit thrombocytopenia model

Objective To evaluate the in vivo efficacy of modified platelet(PLT) additive solution(PAS-ⅢM) with trehalose as a substitute for plasma for the storage of platelet concentrates at low temperature(10℃).Methods Rabbit platelet concentrates were collected and stored in different media and temperature conditions,plasma(22℃),70%PAS-ⅢM/30% plasma(10℃) and plasma/trehalose(-85℃).Platelets in plasma(22℃) on storage day 3,in 70%PAS-ⅢM/30% plasma(10℃) on storage day 3,7,9,and in plasma/trehalose(-85℃) thawed on day 20 were transfused to rabbit thrombocytopenia model.The in vivo recoveries and survivals of the three preserved platelets and fresh platelets were measured and evaluated after transfusion into rabit model of thrombocytopenia.Results The survival of platelets stored in 70% PAS-ⅢM/30% plasma(10℃) on day 9 was lower than that of fresh platelets(P0.05).Recovery and survival of frozen platelets were significant lower than those of the other groups(P
10.Expression of matrix metalloproteinase-1 and tissue inhibitor of metalloproteinase-1 in pathologic synovial plicae of the knee joint

Objective To explore whether or not the cartilage damage in the pathogenesis of pathologic synovial plicae is due to the matrix metalloproteinase. Methods The immunohistochemical method was used to detect the expression and distribution of matrix metalloproteinase-1(MMP-1) and tissue inhibitor of metalloproteinase-1(TIMP-1) in arthroscopically clarified pathologic synovial plicae and normal synovial plicae of the knee joint.Results The positive expression rate of MMP-1 and TIMP-1 had significant differences between the pathologic and normal synovial plicae(?~2=16.014,P=0.000;?~2=4.059,P=0.044).The expression of MMP-1 was positive in the synovial lining cells,monocytes,lymphocytes,endothelial cells,and chondrocytes,but negative in the normal synovial plicae.The TIMP-1 expression was only detected in the synovial lining cells and a small quantity of fibroblast cells.The immunohistochemical analysis revealed a greater number of positive cells and intensity of staining of MMP-1 than TIMP-1.Conclusions The development of pathologic synovial plicae may yield MMP-1 and TIMP-1 with unbalanced distributions,which may be the biological basis of the pathogenesis of cartilage destruction.
