1.Effect of total body irradiation on cellular senescence related indexes of bone marrow mesenchymal stem cells.

The aim of this study was to explore the changes in cellular senescence related indexes of bone marrow mesenchymal stem cells (BMMSCs) after total body irradiation (TBI). At different time points after 4 Gy irradiation, BMMSCs were isolated from male C57BL/6 mice and cultured. Morphology, senescence-associated beta-galactosidase (SA-beta-gal) staining and cell cycle analysis were used to evaluate the changes in BMMSCs at cellular level while real-time RT-PCR was used to detect the alterations in senescence related gene expression including p16INK4a, p21Cip1/Waf1, p53 and TGF-beta1. The results showed that within 4 weeks after exposure to 4 Gy TBI, the morphology of BMMSCs and the expression level of SA-beta-gal were not significantly changed, the cellular senescence-related cell cycle arrest was not occurred and the senescence related gene expression level was not increased. It is concluded that at the early stage after 4 Gy TBI, the related molecular level of cellular senescence in BMMSCs is not changed.

2.Protective effects of penehyclidine hydrochloride against acute renal injury induced by hemorrhagic shock and lipopolysaccharides in rats.

OBJECTIVETo investigate the effect of penehyclidine hydrochloride (PHC) in a rat model of renal injury induced by hemorrhagic shock and lipopolysaccharides (LPS).
METHODSForty-five healthy Wistar rats were randomized into sham operated group, model group, and 3 penehyclidine hydrochloride (PHC) dose (1, 2 and 3 mg/kg) groups (PHC1, PHC2, and PHC3 groups, respectively). The arterial blood samples were collected to determine the concentrations of serum tumor necrosis factor-α (TNF-α), interleukin-8 (IL-8), interleukin-1 (IL-1), urine creatinine (Cr) and blood urine nitrogen (BUN), and the renal tissues were collected to measure the expressions of ICAM-1 and nuclear factor-κB (NF-κB) and observe the pathological changes.
RESULTSTNF-α, IL-8, IL-1, Cr, BUN, ICAM-1 and NF-κB in the 3 PHC groups were significantly lower than those in the model group (P<0.05). TNF-α, IL-8, IL-1, Cr and BUN were significantly lower in PHC1 (P<0.05) than in the PHC2 and PHC3 groups, and ICAM-1 and NF-κB were similar between 3 PHC groups (P>0.05). Compared with the model group, the 3 PHC groups showed lessened pathological changes in the renal tubules.
CONCLUSIONPHC has protective effects against renal injury induced by hemorrhagic-endotoxin shock in rats, and treatment with 1 mg/kg PHC produces the most significant protective effect.

3.The malocclusion of primary dentition in the suburb of Chengdu: a cross-section survey.

OBJECTIVETo reveal malocclusion of primary dentition in the suburb of Chengdu.
METHODSCross-section survey and malocclusion rate calculation were performed.
RESULTSThere are 491 children with malocclusion from total 1 279 children; the rate of malocclusion is 38.38%. The most important malocclusion in primary dentition are over bite (III degrees), anterior cross bite, edge to edge bite, early loss of deciduous teeth, fused teeth.
CONCLUSIONThe rate of malocclusion of primary dentition in the suburb of Chengdu is a little bit light. Much work should be done to prevent or treat the malocclusion of primary dentition.

4.Finite element analysis on transforaminal lumbar interbody fusion treatment

Objective To analyze the clinical feasibility of unilateral transforaminal lumbar interbody fusion (TLIF) for treating lumbar degenerative diseases by finite element method. Methods Based on CT scan data, three-dimensional (3D) finite element models of the normal L3-5 segments under physiological status (intact lumbar model), L4/5 with unilateral pedicle screw fixation plus interbody fusion (unilateral TLIF model) and L4/5 with bilateral pedicle screw fixation plus interbody fusion (bilateral TLIF model) were established by using Mimics, Pro/E, ANSYS software, respectively. Preload of 500 N and load of 10 N•m torque were applied on the superior surface of the L3 segment to simulate 5 physiological activities: body upright, flexion, extension, left lateral bending and right axial rotation. The deformation and stress distributions in vertebral body, vertebral discs, pedicle screw and cage under different loads were then recorded and analyzed to compare mechanical properties of the two fixation methods. Results The deformation of L3-5 segments fixed with unilateral TLIF or bilateral TLIF decreased as compared to the intact lumbar model; the stresses in cage reached the maximum in both unilateral TLIF model and bilateral TLIF model during back extension, meanwhile peak stresses on pedicle screws in unilateral TLIF were significantly higher than those on bilateral TILF model, with the peak stress of 463.39 MPa during back extension. ConclusionsUnilateral TLIF can be selected as a method for treating lumbar degenerative diseases; however, its stability was inferior to bilateral TLIF due to the higher peak stress. Therefore, less stretch exercises may be safe for patients during rehabilitation to avoid surgery failure or pedicle screw fracture.
5.Reduction of bone marrow mesenchymal stem/progenitor cells pool and alteration of their osteogenesis potential caused by total body irradiation.

To investigate the effect of irradiation on the quantity and osteogenesis potential of BMMSCs and to explore the response of them in the irradiation stress and its contribution to long-term effects of radiation-induced bone and hematologic injury, a total body irradiation (TBI) murine model was adopted. The number of CFU-F and cell cycle profile of BMMSCs were analyzed at different time points before and after TBI. Osteogenic differentiation was evaluated by Von Kossa staining, expressions of osteogenesis-related genes and transcriptional coactivator with PDZ-binding motif (TAZ) were detected by real-time RT-PCR. The results showed that the number of CFU-F decreased greatly at day 28 after TBI. At day 3 after TBI, more cells entered cell cycle and the osteogenesis potential was greatly enhanced followed by recovery of cell cycle distribution and significant defect in osteoblast differentiation respectively, meanwhile the expression of TAZ was changed. It is concluded that TBI results in the reduction of bone marrow mesenchymal stem/progenitor cell pool and alters the osteogenesis potential of BMMSCs, which is related to the change of TAZ expression.

6.Overview of Famous Classical Formulas and Discussion on Its Exploitation

The famous classical formulas (FCF), which occupy the most important position in the prescription theory, had been brought out following the progress of traditional Chinese medicine (TCM). Its formation and exploitation, essentially the ingredient constitution and preparation technology, is an objective trend of TCM. The development of related product under the guidance of relevant national policies has its historical inevitability, as the development of formula granules in recent decades had laid a solid foundation for it. The key to exploit these FCF is to refine and standardize the clinical applications through the ages with the guidance of TCM theory, for the process design of massive industrial production. As a result of the analysis of its definition and connotation, it is clear that the core connotation of FCF is the medicinal substances with fixed formula compatibility and stable preparation process. Furthermore, the screening strategy, the exploitation value, and the applicability between ancient prescriptions and nowadays diseases, had also been discussed based on the current policy situation and research status.
7. Analysis of Quality Control Standard of “Carbonizing Retains Characteristics” Based on Quality Investigation of Commercial Platycladi Cacumen Carbonisata Pieces

Objective: To investigate the quality of Platycladi Cacumen carbonisata pieces in the market,then to explore control standard of "carbonizing retains characteristics" of Platycladi Cacumen.Method: The properties,water content and alcohol extract content of commercial Platycladi Cacumen carbonisata pieces were determined by the method in the 2015 edition of Chinese Pharmacopoeia.Thin layer chromatography(TLC) identification of quercetin and contents of quercetin and quercitin were determined.Result: The carbonizing degree of 9 batches of Platycladi Cacumen carbonisata pieces were heavier than which is specified in the 2015 edition of Chinese Pharmacopoeia.The conditions of TLC identification in the 2015 edition of Chinese Pharmacopoeia should be revised to avoid stratification;the carbonizing degree could affect the contents of quercetin and quercitin in Platycladi Cacumen carbonisata pieces;the simultaneous detection of quercetin and quercitin can be used as one of the quality control indexes of this carbonisata pieces.Conclusion: Most of commercial Platycladi Cacumen carbonisata pieces have the quality problem of excessive carbonizing degree,the established quality control standard of "carbonizing retains characteristics" can be used to effectively ensure the quality of Platycladi Cacumen carbonisata pieces.
8.Comparative study on standard decoctions of Ophiopogonis Radix and Liriopes Radix.

Standard decoction is the core of the pharmacodynamics for water-soluble substance of Chinese materia medica. Its research is of great significance to the research and development of some single ingredients and the classical prescriptions,and it is the only way to transform traditional medication experience into industrial products. In this article,standard decoction research strategies were used for the comparison analysis of Ophiopogonis Radix from Zhejiang province(ZMD),Ophiopogonis Radix from Sichuan province(CMD),and Liriopes Radix(SMD). Regularities were present among different grades of CMD; potential quality markers and pH differences associated with SO2 residues were also found. Finally,the extract powder of Ophiopogonis Radix prepared by mass production process was analyzed and validated,and the results showed that the standard system could be used for the quality control of intermediates and final products. In conclusion,this study can provide reference for the clinical application of Ophiopogonis Radix medicines and provide testing method for higher quality with higher price.

9.Sorafenib in liver function impaired advanced hepatocellular carcinoma.

OBJECTIVETo explore the efficacy and safty of sorafenib in Child-Pugh class B to class C hepatocellular carcinoma (HCC).
METHODSIn this three-center open-label study from November 2011 to May 2013, we randomly assigned 189 patients with advanced Child-Pugh class B or C HCC patients into two groups, one group with 95 patient to receive sorafenib (400 mg a time, twice a day) and the other group with 94 patients to receive best supportive care. The primary end points were progression-free survival and overall survival.
RESULTSThe median progression-free survival was 2.2 months and 1.9 months in the sorafenib group and best supportive care group respectively (Hazard ratio in the sorafenib group, 0.55; 95% confidence interval, 0.40-0.75; P=0.002). The median overall survival was 4.0 months and 3.5 months in the sorafenib group and best supportive care group respectively (Hazard ratio in the sorafenib group, 0.48; 95% confidence interval, 0.35-0.68; P<0.001). The main adverse effect of sorafenib was rash and acne of the skin (in 51.7% patients). The incidences of severe rash, diarrhea, and dry skin were 5.6%, 5.6%, and 2.2% in the sorafenib group. One patient reached partial response in the sorafenib group.
CONCLUSIONSSorafenib is safe in patients with liver function impaired advanced HCC. It is effective in terms of progression-free survival and overall survival compared with best supportive care. Liver functions are the important predictive factors.

OBJECTIVE:
This study was aimed at investigating the carrier rate of, and molecular variation in, α- and β-globin gene mutations in Hunan Province.
METHODS:
We recruited 25,946 individuals attending premarital screening from 42 districts and counties in all 14 cities of Hunan Province. Hematological screening was performed, and molecular parameters were assessed.
RESULTS:
The overall carrier rate of thalassemia was 7.1%, including 4.83% for α-thalassemia, 2.15% for β-thalassemia, and 0.12% for both α- and β-thalassemia. The highest carrier rate of thalassemia was in Yongzhou (14.57%). The most abundant genotype of α-thalassemia and β-thalassemia was -α 3.7/αα (50.23%) and β IVS-II-654/β N (28.23%), respectively. Four α-globin mutations [CD108 (ACC>AAC), CAP +29 (G>C), Hb Agrinio and Hb Cervantes] and six β-globin mutations [CAP +8 (C>T), IVS-II-848 (C>T), -56 (G>C), beta nt-77 (G>C), codon 20/21 (-TGGA) and Hb Knossos] had not previously been identified in China. Furthermore, this study provides the first report of the carrier rates of abnormal hemoglobin variants and α-globin triplication in Hunan Province, which were 0.49% and 1.99%, respectively.
CONCLUSION
Our study demonstrates the high complexity and diversity of thalassemia gene mutations in the Hunan population. The results should facilitate genetic counselling and the prevention of severe thalassemia in this region.
